RESUMO

The Auditory Steady-State Response (ASSR) is a type of auditory evoked potential (AEP) generated in the auditory system that can be automatically detected by means of objective response detectors (ORDs). ASSRs are usually registered on the scalp using electroencephalography (EEG). ORD are univariate techniques, i.e. only uses one data channel. However, techniques involving more than one channel - multi-channel objective response detectors (MORDs) - have been showing higher detection rate (DR) when compared to ORD techniques. When ASSR is evoked by amplitude stimuli, the responses could be detected by analyzing the modulation frequencies and their harmonics. Despite this, ORD techniques are traditionally applied only in its first harmonic. This approach is known as one-sample test. The q-sample tests, however, considers harmonics beyond the first. Thus, this work proposes and evaluates the use of q-sample tests using a combination of multiple EEG channels and multiple harmonics of the stimulation frequencies and compare them with traditional one-sample tests. The database used consists of EEG channels from 24 volunteers with normal auditory threshold collected following a binaural stimulation protocol by amplitude modulated (AM) tone with modulating frequencies near 80 Hz. The best q-sample MORD result showed an increase in DR of 45.25% when compared with the best one-sample ORD test. Thus, it is recommended to use multiple channels and multiple harmonics, whenever available.

An Auditory Steady-State Response (ASSR) is a valuable tool for determining auditory thresholds in individuals who are either unable or unwilling to cooperate with conventional behavioral testing methods. This study proposes a sequential test technique for automatic detection of ASSRs, incorporating a non-detection stopping criterion. The electrophysiological thresholds of a normal hearing volunteer were established using data collected from multichannel EEG signals. The detection probabilities and critical values were obtained via Monte Carlo simulations. Remarkably, application of the non-detection stopping criterion resulted in a 60% reduction in exam time in the absence of a response. These findings clearly demonstrate the significant potential of the sequential test in enhancing the performance of automatic audiometry.

The multichannel objective response detection (MORD) techniques are statistical methods, which use information from more than one electroencephalography (EEG) channel, to infer the presence of evoked potential. However, the correlation level between the channels can lead to a decrease in MORD performance, such as an increase in the false positive (FP) rate and/or a decrease in the detection rate (DR). The present study aims to propose a method to deal with the correlations in the multichannel EEG. The method consists of making an adjustment in the Monte Carlo simulation, considering the information between channels. The MORD techniques with and without the new method were applied to an auditory steady-state response (ASSR) database, composed of the EEG multichannel of eleven volunteers during multifrequency stimulation. The proposed method kept the FP rate at values equal to or less than the significance level of the test and led to an increase of 8.51% in the DR in relation to non-application of the method. Results of this study indicate that the proposed method is an alternative to deal with the effect of the correlation between channels in situations where MORD techniques are applied.

PURPOSE: In the present study, a new procedure to perform automatic audiometry using multifrequency Auditory Steady-State Response (ASSR) is proposed. METHODS: The automatic audiometry procedure consists of detecting the presence of multifrequency ASSR in real-time using the sequential test strategy and by adjusting the stimulus intensity independently. The ASSR audiometric thresholds of 18 adult volunteers with normal hearing were determined by automatically (four simultaneous frequencies per ear) at modulation frequencies in the 80 Hz range. The exam time and the difference between ASSR thresholds and pure-tone behavioural hearing thresholds were estimated as performance measures. RESULTS: The results showed that automatic audiometry can reduce the number of intensity levels used to obtain the ASSR threshold by up to 58% when compared to audiometry without using the techniques applied in automatic audiometry. In addition, the average of the difference between ASSR thresholds and Pure-Tone Behavioural Hearing thresholds was around 19 dB, which is similar to the results reported in similar studies. CONCLUSIONS: The audiometric procedure proposed in this study is fully automatic, i.e., does not require any human supervision throughout the exam, and is able to significantly reduce the conventional exam time.

Auditory steady-state response (ASSR) is useful for hearing threshold estimation. The ASSR is usually detected with objective response detectors (ORD). The performance of these detectors depends on the signal-to-noise ratio (SNR) as well as the signal length. Since it is undesirable to increase the signal length, then, this work provides a multivariate technique for improving the SNR and consequently the detection power. We propose the insertion of a short calibration step before the detection protocol, in order to perform a search among the available electroencephalogram (EEG) derivations and select the derivation with the highest SNR. The ORD used in this work was the magnitude-squared coherence (MSC). The standard detection protocol is to use the same EEG derivation in all exams. Using 22-scalp positions, the new technique achieved a detection rate higher than that obtained in 99.13% of the standard detection protocol. When restrictions were applied to the search, a superior performance was achieved. Thus, the technique proposed was able to track the best EEG derivations before exams and seems to be able to deal with the variability between individuals and between sessions.

Objective: Objective Response Detection (ORD) can be used for auditory steady-state response (ASSR) detection. In conventional ORD methods, the statistical tests are applied at the end of data collection ('single-shot tests'). In sequential ORD methods, statistical tests are applied repeatedly, while data is being collected. However, repeated testing can increase False Positive (FP) rates. One solution is to infer that response is present only after the test remains significant for a predefined number of consecutive detections (NCD). Thus, this paper describes a new method for finding the required NCD that control the FP rate for ASSR detection.Design: NCD values are estimated using Monte Carlo simulations.Study sample: ASSR signals were recorded from 8 normal-hearing subjects.Results: The exam time was reduced by up to 38.9% compared to the single-shot test with loss of approximately 5% in detection rate. Alternatively, lower gains in time were achieved for a smaller (non-significant) loss in detection rate. The FP rates at the end of the test were kept at the nominal level expected (1%).Conclusion: The sequential test strategy with NCD as the stopping criterion can improve the speed of ASSR detection and prevent higher than expected FP rates.

Objective response detection (ORD) techniques such as the magnitude-squared coherence (MSC) are mathematical methods tailored to detect potentials evoked by an external periodic stimulation. The performance of the MSC is directly proportional to the signal-to-noise ratio (SNR) of the recorded signal and the time spent for collecting data. An alternative to increasing the performance of detection techniques without increasing data recording time is to use the information from more than one signal simultaneously. In this context, this work proposes two new detection techniques based on the average and on the product of MSCs of two different signals. The critical values and detection probabilities were obtained theoretically and using a Monte Carlo simulation. The performances of the new detectors were evaluated using synthetic data and electroencephalogram (EEG) signals during photo and auditory stimulation. For the synthetic signals, the two proposed detectors exhibited a higher detection rate when compared to the rate of the traditional MSC technique. When applied to EEG signals, these detectors resulted in an increase of the mean detection rate in relation to MSC for visual and auditory stimulation of at least 25% and 13.21%, respectively. The proposed detectors may be considered as promising tools for clinical applications. Graphical Abstract.

ABSTRACT Introduction The structural and mechanical adaptations of the right ventricular (RV) myocytes in response to hypertension associated with low-intensity endurance training (LIET) have not been studied in experimental models. Objective To determine the effects of LIET on the structural and mechanical properties of RV myocytes in spontaneously hypertensive rats (SHRs). Methods Male SHRs and normotensive Wistar rats (age: 16 weeks) were allocated to groups (n=7): WIS (Wistar Controls); SHR-C (SHR Controls) and SHR-T (SHR Trained; 60 min/day, 50-60% of maximal exercise capacity, 5 days/week for 8 weeks). Systolic arterial pressure (SAP), isolated RV myocyte dimensions, contractility, intracellular Ca2+ transient ([Ca2+]i), and ventricular Ca2+ regulatory proteins were measured. The statistical analysis was performed by one-way ANOVA followed by the Tukey post hoc test (α=5%). Results LIET reduced the SAP in SHR animals (SHR-C, 164 ± 2 mmHg vs. SHR-T, 152 ± 4 mmHg; P<0.05). Hypertension increased cell length (WIS, 156.8 ± 2.7 µm; SHR-C, 166.6 ± 3.1 µm; P<0.05) but did not affect cell width or volume (P>0.05). LIET did not change the cell dimensions in the SHR-T. Neither hypertension nor LIET affected myocyte contractility or the expression of Ca2+ regulatory proteins in the RV of the SHR-C and SHR-T groups. Hypertension did not affect the amplitude of the [Ca2+]i transient or the time to half resting level (P>0.05), but increased the time to peak (WIS, 58 ± 1 ms vs. SHR-C, 79 ± 2 ms; P<0.05). LIET increased the amplitude of the [Ca2+]i transient (WIS, 2.28 ± 0.07 F/F0 and SHR-C, 2.48 ± 0.08 F/F0 vs. SHR-T, 2.87 ± 0.08 F/F0 P<0.05), but did not alter the times to peak or to half resting level. Conclusion LIET had no effect on the structural and mechanical properties of RV myocytes in the SHRs, although it increased the amplitude of the [Ca2+]i transient and reduced the SAP. RESUMO Introdução As adaptações estruturais e mecânicas de miócitos do ventrículo direito (VD) em resposta à hipertensão associada ao treinamento aeróbio de baixa intensidade (TABI) não foram estudadas em modelos experimentais de ratos. Objetivo Determinar os efeitos do TABI sobre as propriedades estruturais e mecânicas de miócitos do VD em ratos espontaneamente hipertensos (SHR). Métodos Ratos SHR e ratos Wistar machos e normotensos (idade: 16 semanas) foram distribuídos em grupos (n = 7): WIS (Wistar controle); SHR-C (SHR controle) e SHR-T (SHR treinados; 60 min/dia, 50% a 60% da capacidade máxima de exercício, 5 dias/semana por 8 semanas). Procedeu-se à medição de pressão arterial sistólica (PAS), dimensões dos miócitos isolados do VD, contratilidade, transiente intracelular de Ca2+ ([Ca2+]i) e de proteínas reguladoras de Ca2+. A análise estatística foi realizada por ANOVA one-way, seguida por teste de Tukey post hoc (α = 5%). Resultados O TABI reduziu a PAS nos animais SHR (SHR-C, 164 ± 2 mmHg vs. SHR-T, 152 ± 4 mmHg; P < 0,05). A hipertensão aumentou o comprimento celular (WIS, 156,8 ± 2,7 µm; SHR-C, 166,6 ± 3,1 µm; P < 0,05), mas não afetou a largura ou o volume (P > 0,05). O TABI não alterou as dimensões celulares nos SHR-T. Nem a hipertensão nem o TABI afetaram a contratilidade dos miócitos ou a expressão das proteínas reguladoras do Ca2+ no VD dos grupos SHR-C e SHR-T. Métodos Ratas SHR y ratas Wistar machos y normotensas (edad: 16 semanas) fueron distribuidas en grupos (n = 7): WIS (Wistar controle); SHR-C (SHR controle) y SHR-T (SHR entrenado; 60 min/día, el 50% a 60% de la capacidad máxima de ejercicio, 5 días/semana por 8 semanas). Se midieron presión arterial sistólica (PAS), dimensiones de los miocitos aislados del VD, contractilidad, transitorio intracelular de Ca2+ ([Ca2+]i) y proteínas reguladoras de Ca2+. El análisis estadístico se realizó mediante ANOVA one-way seguida por la prueba de Tukey post hoc (α = 5%). BACKGROUND AND OBJECTIVE: The local spectral F-test (SFT) corresponds to a statistical way of assessing whether the spectrum of a signal is flat in the vicinity of a specific frequency. The power of this univariate test (comparing one frequency component  against its neighbours using only one signal) depends on the signal-to-noise ratio, which is fixed in the case of electroencephalogram (EEG) analysis. However, this limitation could be overcome by considering more signals in the analysis. Thus, this work presents an alternative multivariate approach for estimating the local SFT. METHODS: Probabilities of detection and false alarm studies were performed for this new detector using Monte Carlo simulations and theoretically whenever possible. The application was illustrated in recorded EEG data collected during photic stimulation. RESULTS: The results showed that it is worth using more channels if available, since the probability of detecting a response tends to increase with increasing number of signals. In the application to the EEG during photic stimulation, the best results were obtained by using N > 2 signals (around 30% more accurate when compared with the univariate case. The false positive levels were maintained below 5%). CONCLUSION: Consequently, it is conjectured that it is always better to apply the proposed method if more than one EEG signal with the same signal-to-noise ratio (SNR) is available. For the case where the SNRs are different, a guideline has been given to improve the detection.

BACKGROUND AND OBJECTIVE: The use of objective detection techniques applied to the auditory steady-state responses (ASSRs) for the assessment of auditory thresholds has been investigated over the years. The idea consists in setting up the audiometric profile without subjective inference from patients and evaluators. The challenge encountered is to reduce the detection time of auditory thresholds reaching high correlation coefficients between the objective and the conventional thresholds, as well as reducing difference between thresholds. METHODS: This paper evaluated the use of the Multiple Magnitude-Squared Coherence (MMSC) in Auditory Steady-State Responses (ASSRs) evoked by amplitude modulated tones around 40 Hz, attaining objective audiograms, which were, later, compared to conventional audiograms. It was proposed an analysis of the electroencephalogram signals of ten subjects, monaurally stimulated, in the intensities 15, 20, 25, 30, 40 and 50 dB SPL, for carrier frequencies of 0.5, 1, 2 and 4 kHz. After the detection protocol parameters variation, two detectors were selected according to behavioral thresholds. RESULTS: The method of this study resulted in a Maximum detector with correlation coefficient r = 0.9262, mean difference between the objective and behavioral thresholds of 6.44 dB SPL, average detection time per ear of 49.96 min and per stimulus of 2.08 min. Meanwhile, the Fast detector presented coefficient r = 0.8401, mean difference of 6.81 dB SPL, average detection time of 28.20 min per ear and 1.18 per stimulus. CONCLUSIONS: The results of this study indicate that the MMSC use in the auditory responses detection might provide a reliable and efficient estimation of auditory thresholds.

Abstract Background: The lack of cardiac β1-adrenergic receptors (β1-AR) negatively affects the regulation of both cardiac inotropy and lusitropy, leading, in the long term, to heart failure (HF). Moderate-intensity aerobic exercise (MCAE) is recommended as an adjunctive therapy for patients with HF. Objective: We tested the effects of MCAE on the contractile properties of left ventricular (LV) myocytes from β1 adrenergic receptor knockout (β1ARKO) mice. Methods: Four- to five-month-old male wild type (WT) and β1ARKO mice were divided into groups: WT control (WTc) and trained (WTt); and β1ARKO control (β1ARKOc) and trained (β1ARKOt). Animals from trained groups were submitted to a MCAE regimen (60 min/day; 60% of maximal speed, 5 days/week) on a treadmill, for 8 weeks. P ≤ 0.05 was considered significant in all comparisons. Results: The β1ARKO and exercised mice exhibited a higher (p < 0.05) running capacity than WT and sedentary ones, respectively. The β1ARKO mice showed higher body (BW), heart (HW) and left ventricle (LVW) weights, as well as the HW/BW and LVW/BW than WT mice. However, the MCAE did not affect these parameters. Left ventricular myocytes from β1ARKO mice showed increased (p < 0.05) amplitude and velocities of contraction and relaxation than those from WT. In addition, MCAE increased (p < 0.05) amplitude and velocities of contraction and relaxation in β1ARKO mice. Conclusion: MCAE improves myocyte contractility in the left ventricle of β1ARKO mice. This is evidence to support the therapeutic value of this type of exercise training in the treatment of heart diseases involving β1-AR desensitization or reduction.


Resumo Fundamento: A falta de receptores β1-adrenérgicos (β1-AR) cardíacos afeta negativamente a regulação de inotropismo e lusitropismo cardíacos, levando, no longo prazo, a insuficiência cardíaca (IC). Recomenda-se exercício aeróbico contínuo de intensidade moderada (EACM) como adjuvante no tratamento de pacientes com IC. Objetivo: Testar os efeitos do EACM nas propriedades contráteis de miócitos do ventrículo esquerdo (VE) de camundongos com nocaute para o receptor β1-adrenérgico (β1ARKO). Método: Camundongos machos com 4 a 5 meses de idade, wild-type (WT) e β1ARKO foram divididos em grupos: WT controle (WTc) e treinado (WTt); e β1ARKO controle (β1ARKOc) e treinado (β1ARKOt). Os grupos treinados foram submetidos a regime de EACM (60 min/dia; 60% da velocidade máxima, 5 dias/semana) em esteira rolante, por 8 semanas. Adotou-se P ≤ 0,05 como nível de significância em todas as comparações. Resultados: Os animais β1ARKO (β1ARKOc + β1ARKOt) correram uma distância maior do que os animais WT (WTc + WTt) (p < 0,05). Os camundongos β1ARKO apresentaram maiores pesos corporal (PC), do coração (PCo) e do ventrículo esquerdo (PVE), assim como PCo/PC e PVE/PC do que os camundongos WT. Entretanto, o EACM não afetou tais parâmetros. Os miócitos do VE de camundongos β1ARKO apresentaram maiores (p < 0,05) amplitude e velocidades de contração e relaxamento do que os dos camundongos WT. Além disso, o EACM aumentou (p < 0,05) a amplitude e as velocidades de contração e relaxamento nos camundongos β1ARKO. Conclusão: O EACM melhora a contratilidade do miócito do VE de camundongos β1ARKO. Tal achado confirma o valor terapêutico desse tipo de treinamento físico para o tratamento de doenças cardíacas envolvendo dessensibilização ou redução de β1-AR.

BACKGROUND: The lack of cardiac ß1-adrenergic receptors (ß1-AR) negatively affects the regulation of both cardiac inotropy and lusitropy, leading, in the long term, to heart failure (HF). Moderate-intensity aerobic exercise (MCAE) is recommended as an adjunctive therapy for patients with HF. OBJECTIVE: We tested the effects of MCAE on the contractile properties of left ventricular (LV) myocytes from ß1 adrenergic receptor knockout (ß1ARKO) mice. METHODS: Four- to five-month-old male wild type (WT) and ß1ARKO mice were divided into groups: WT control (WTc) and trained (WTt); and ß1ARKO control (ß1ARKOc) and trained (ß1ARKOt). Animals from trained groups were submitted to a MCAE regimen (60 min/day; 60% of maximal speed, 5 days/week) on a treadmill, for 8 weeks. P ≤ 0.05 was considered significant in all comparisons. RESULTS: The ß1ARKO and exercised mice exhibited a higher (p < 0.05) running capacity than WT and sedentary ones, respectively. The ß1ARKO mice showed higher body (BW), heart (HW) and left ventricle (LVW) weights, as well as the HW/BW and LVW/BW than WT mice. However, the MCAE did not affect these parameters. Left ventricular myocytes from ß1ARKO mice showed increased (p < 0.05) amplitude and velocities of contraction and relaxation than those from WT. In addition, MCAE increased (p < 0.05) amplitude and velocities of contraction and relaxation in ß1ARKO mice. CONCLUSION: MCAE improves myocyte contractility in the left ventricle of ß1ARKO mice. This is evidence to support the therapeutic value of this type of exercise training in the treatment of heart diseases involving ß1-AR desensitization or reduction.

Abstract The aim of this study was to propose a mechanical device that could increase the mechanical load during the entire eccentric (ECC) action on a knee flexor machine and to compare the muscular activity when the device was used during a testing protocol. Fifteen physically active women were recruited, and they performed two protocols: control and eccentric overload. Control protocol was performed with concentric (CON) and ECC actions with similar load (60% of one repetition maximum) whilst eccentric overload protocol consisted of ECC actions with 40% more load than CON actions. Muscular activation was measured using surface electromyography of the biceps femoris (BF) and the gastrocnemius medialis (GM) muscles. ECC actions presented a higher muscular activation during eccentric overload protocol than control protocol for BF (p = 0.032), but not for the GM (p = 0.439). The mechanical device increased the mechanical load during the ECC muscle action and consequently increased the amplitude of the neural drive to the BF muscle; however, it did not increase the amplitude of the neural drive to the GM muscle.


Resumo O objetivo deste estudo foi propor um dispositivo mecânico que acoplado a uma cadeira flexora aumente a carga mecânica durante toda a ação excêntrica (ECC) e comparar a atividade muscular durante a utilização do dispositivo proposto. Quinze mulheres fisicamente ativas foram recrutadas e realizaram dois diferentes protocolos (controle e sobrecarga ECC). O protocolo controle foi realizado com ações concêntricas (CON) e ECC com carga iguais (60% de uma repetição máxima), enquanto o protocolo de sobrecarga ECC consistiu em ações ECC com 40% mais de carga do que as ações CON. A ativação muscular dos músculos biceps femoris (BF) e gastrocnemius medialis (GM) foi medida por meio de eletromiografia de superfície. Durante as ações ECC foi observada uma maior ativação do BF durante o protocolo de sobrecarga quando comparado ao protocolo controle (p = 0,032). No entanto, GM não apresentou diferenças na ativação entre os protocolos (p = 0,439). O dispositivo mecânico proposto foi capaz de aumentar a carga durante a ação ECC e, consequentemente, a atividade muscular do BF.

The spectral local F-test has been applied for detecting evoked responses to rhythmic stimulation that are embedded in the ongoing electroencephalogram (EEG). Based on the sampling distribution of a flat spectrum at the neighbourhood of the stimulation frequency, spectral peaks in an EEG signal that are due to the stimulation may be readily assessed. Nevertheless, the performance of the technique is strongly affected by both the signal-to-noise ratio (SNR) of the responses and the number of data segments used in the estimation. The present work aims at both deriving and evaluating a multivariate extension of local F-test by including the EEG collected at a second distinct derivation. The detection rate with this multivariate detector was found to be greater than that using a single channel in case of equal SNR in both signals. Monte Carlo simulation results showed that the probability of detection with this new detector saturates for signal-to-noise ratios above 12 dB and indicated a greater detection rate in practical situations, even when smaller SNR-values are found in the added signal (e.g. 5 dB for 16 neighbouring frequencies used in the estimation). The technique was next applied to the EEG from 12 subjects during intermittent, photic stimulation leading to superior performance in comparison with the univariate local F-test. Since a higher detection rate with the proposed technique is achieved without the need of increasing the number of data segments, it allows evoked responses to be detected faster, once the same detection rate may be accomplished with less segments. This might be useful in clinical practice.

INTRODUCTION: The development of Brain Computer Interfaces based on Auditory Selective Attention allows patients unable to voluntarily control eye movement to interact with the interface, because such systems are independent of vision. An alternative technique suitable for Brain Computer Interface applications is Spatial Coherence, an objective method to detect the side where a subject is focusing attention. This method takes into consideration the Coherence Function and the topographic distribution of responses between EEG electrodes. In this work, we further study the Information Transfer Rate and the effects of overlapping windows in the calculations. The idea behind the overlapping is to decrease the duration of the test in order to augment the Information Transfer Rate. METHOD: EEG signals were collected from fourteen adult men aged between 19 and 28 years. Amplitude-modulated tones were used for stimulation, with 32 and 38 Hz modulation and 500 and 2000 Hz carrier frequencies, on the left and right ears, respectively. Spatial coherence was used in an online Brain Computer Interface system using auditory steady-state responses modulated by Auditory Selective Attention. RESULTS: The obtained hit rates and the Information Transfer Rate may be considered appropriate, with a maximum value of 82% and 1.89 bits/min. The better detector regarding sensitivity versus specificity can be obtained by using a 50% overlap between consecutive data windows. CONCLUSION: We conclude that the Spatial Coherence successfully detected the focus of attention, and it seemed useful as a classifier of the attention condition for vision-free Brain Computer Interface.


O desenvolvimento de interfaces cérebro computador (BCI), com base em atenção seletiva auditiva (ASA), auxilia pacientes que não conseguem controlar voluntariamente o movimento do olho para interagir com interfaces visuais, uma vez que tais sistemas são independentes de visão. A Coerência Espacial é técnica alternativa adequada para aplicações BCI, tal técnica é usada em eletroencefalograma (EEG) para detectar o lado, ou hemisfério, em que o sujeito está focalizando atenção. Este método leva em consideração a função de coerência e a distribuição topográfica das respostas entre os eletrodos de EEG. Neste estudo foram coletados sinais EEG de 14 homens adultos com idade entre 19 e 28 anos com aplicação de estímulos sonoros, tons em amplitude modulada (AM) com frequência de modulação e portadora 32/500 Hz na orelha esquerda e 38/2000 Hz na orelha direita. A coerência espacial foi usada em um sistema BCI on-line com ASSR modulada pela atenção seletiva auditiva. Neste trabalho, foi abordado a taxa de transferência de informação (ITR) e os efeitos de sobreposição de janelas nas estimativas da coerência com objetivo de reduzir a duração do ensaio e aumentar a ITR. Conclui-se que a coerência espacial pode útil para detecção do foco de atenção, aplicado a BCI. As taxas de sensibilidade e ITR máximas são de 82% e 1,89 bits/min. Neste caso a melhor sensibilidade e especificidade foram obtidas com 50% de sobreposição entre as janelas nas estimativas da coerência.

OBJECTIVES: The present study was performed to investigate 1) whether aerobic exercise training prior to myocardial infarction would prevent cardiac dysfunction and structural deterioration and 2) whether the potential cardiac benefits of aerobic exercise training would be associated with preserved morphological and contractile properties of cardiomyocytes in post-infarct remodeled myocardium. METHODS: Male Wistar rats underwent an aerobic exercise training protocol for eight weeks. The rats were then assigned to sham surgery (SHAM), sedentary lifestyle and myocardial infarction or exercise training and myocardial infarction groups and were evaluated 15 days after the surgery. Left ventricular tissue was analyzed histologically, and the contractile function of isolated myocytes was measured. Student's t-test was used to analyze infarct size and ventricular wall thickness, and the other parameters were analyzed by the Kruskal-Wallis test followed by Dunn's test or a one-way analysis of variance followed by Tukey's test (p<0.05). RESULTS: Myocardial infarctions in exercise-trained animals resulted in a smaller myocardial infarction extension, a thicker infarcted wall and less collagen accumulation as compared to myocardial infarctions in sedentary animals. Myocardial infarction-induced left ventricular dilation and cardiac dysfunction, as evaluated by +dP/dt and -dP/dt, were both prevented by previous aerobic exercise training. Moreover, aerobic exercise training preserved cardiac myocyte shortening, improved the maximum shortening and relengthening velocities in infarcted hearts and enhanced responsiveness to calcium. CONCLUSION: Previous aerobic exercise training attenuated the cardiac dysfunction and structural deterioration promoted by myocardial infarction, and such benefits were associated with preserved cardiomyocyte morphological and contractile properties.

We tested the effects of low-intensity endurance training (LIET) on the structural and mechanical properties of right (RV) and left ventricular (LV) myocytes. Male Wistar rats (4 mo old) were randomly divided into control (C, n = 7) and trained (T, n = 7, treadmill running at 50-60% of maximal running speed for 8 wk) groups. Isolated ventricular myocyte dimensions, contractility, Ca(2+) transients {intracellular Ca(2+) concentration ([Ca(2+)]i)}, and ventricular [Ca(2+)]i regulatory proteins were measured. LIET augmented cell length (C, 152.5 ± 2.0 µm vs. T, 162.2 ± 2.1 µm; P < 0.05) and volume (C, 5,162 ± 131 µm(3) vs. T, 5,506 ± 132 µm(3); P < 0.05) in the LV but not in the RV. LIET increased cell shortening (C, 7.5 ± 0.3% vs. T, 8.6 ± 0.3%; P < 0.05), the [Ca(2+)]i transient amplitude (C, 2.49 ± 0.06 F/F0 vs. T, 2.82 ± 0.06 F/F0; P < 0.05), the expression of sarcoplasmic reticulum Ca(2+)-ATPase 2a (C, 1.07 ± 0.13 vs. T, 1.59 ± 0.12; P < 0.05), and the levels of phosphorylated phospholamban at serine 16 (C, 0.99 ± 0.11 vs. T, 1.34 ± 0.10; P < 0.05), and reduced the total phospholamban-to-sarcoplasmic reticulum Ca(2+)-ATPase 2a ratio (C, 1.19 ± 0.15 vs. T, 0.40 ± 0.16; P < 0.05) in the LV without changing such parameters in the RV. In conclusion, LIET affected the structure and improved the mechanical properties of LV but not of RV myocytes in rats, helping to characterize the functional and morphological changes that accompany the endurance training-induced cardiac remodeling.

The aim of the present study was to verify the effects of low-intensity endurance training and detraining on the mechanical and molecular properties of cardiomyocytes from spontaneously hypertensive rats (SHRs). Male SHRs and normotensive control Wistar rats at 16-weeks of age were randomly divided into eight groups of eight animals: NC8 and HC8 (normotensive and hypertensive control for 8weeks); NT8 and HT8 (normotensive and hypertensive trained at 50-60% of maximal exercise capacity for 8weeks); NC12 and HC12 (normotensive and hypertensive control for 12weeks); NDT and HDT (normotensive and hypertensive trained for 8weeks and detrained for 4weeks). The total exercise time until fatigue (TTF) was determined by a maximal exercise capacity test. Resting heart rate (RHR) and systolic arterial pressure (SAP) were measured. After the treatments, animals were killed by cervical dislocation and left ventricular myocytes were isolated by enzymatic dispersion. Isolated cells were used to determine intracellular global Ca(2+) ([Ca(2+)]i) transient and cardiomyocyte contractility (1Hz; ~25°C). [Ca(2+)]i regulatory proteins were measured by Western blot, and the markers of pathologic cardiac hypertrophy by quantitative real-time polymerase chain reaction (q-RT-PCR). Exercise training augmented the TTF (NC8, 11.4±1.5min vs. NT8, 22.5±1.4min; HC8, 11.7±1.4min vs. HT8, 24.5±1.3min; P<0.05), reduced RHR (NT8initial, 340±8bpm vs. NT8final, 322±10bpm; HT8initial, 369±8bpm vs. HT8final, 344±10bpm; P<0.05), and SBP in SHR animals (HC8, 178±3mmHg vs. HT8, 161±4mmHg; P<0.05). HC8 rats showed a slower [Ca(2+)]i transient (Tpeak, 83.7±1.8ms vs. 71.7±2.4ms; T50%decay, 284.0±4.3ms vs. 264.0±4.1ms; P<0.05) and cell contractility (Vshortening, 86.1±6.7µm/s vs. 118.6±6.7µm/s; Vrelengthening, 57.5±7.4µm/s vs. 101.3±7.4µm/s; P<0.05), and higher expression of ANF (300%; P<0.05), skeletal α-actin (250%; P<0.05) and a decreased α/ß-MHC ratio (70%; P<0.05) compared to NC8. Exercise training increased [Ca(2+)]i transient (NC8, 2.39±0.06F/F0 vs. NT8, 2.72±0.06F/F0; HC8, 2.28±0.05F/F0 vs. HT8, 2.82±0.05F/F0; P<0.05), and cell contractility (NC8, 7.4±0.3% vs. NT8, 8.4±0.3%; HC8, 6.8±0.3% vs. HT8, 7.8±0.3%; P<0.05). Furthermore, exercise normalized the expression of ANF, skeletal α-actin, and the α/ß-MHC ratio in HT8 rats, augmented the expression of SERCA2a (NC8, 0.93±0.15 vs. NT8, 1.49±0.14; HC8, 0.83±0.13 vs. HT8, 1.32±0.14; P<0.05) and PLBser16 (NC8, 0.89±0.18 vs. NT8, 1.23±0.17; HC8, 0.77±0.17 vs. HT8, 1.32±0.16; P<0.05), and reduced PLBt/SERCA2a (NC8, 1.21±0.19 vs. NT8, 0.50±0.21; HC8, 1.38±0.17 vs. HT8, 0.66±0.21; P<0.05). However, all these adaptations returned to control values within 4weeks of detraining in both SHR and normotensive control animals. In conclusion, low-intensity endurance training induces positive benefits to left ventricular myocyte mechanical and molecular properties, which are reversed within 4weeks of detraining.

FUNDAMENTO: A capacidade aeróbica é fundamental para o desempenho físico, e a baixa capacidade aeróbica está relacionada ao desencadeamento de diversas doenças cardiovasculares. OBJETIVO: Comparar a contratilidade e a morfologia de cardiomiócitos isolados de ratos com baixo desempenho e desempenho padrão para o exercício físico. MÉTODOS: Ratos Wistar, com 10 semanas de idade, foram submetidos a um protocolo de corrida em esteira até a fadiga, e foram divididos em dois grupos: Baixo Desempenho (BD) e Desempenho Padrão (DP). Em seguida, após eutanásia, o coração foi removido rapidamente e, por meio de dissociação enzimática, os cardiomiócitos do ventrículo esquerdo foram isolados. O comprimento celular e dos sarcômeros e a largura dos cardiomiócitos foram medidos usando-se um sistema de detecção de bordas. Os cardiomiócitos isolados foram estimulados eletricamente a 1 e 3 Hz e a contração celular foi medida registrando-se a alteração do seu comprimento. RESULTADOS: O comprimento celular foi menor no grupo BD (157,2 ± 1,3µm; p < 0,05) em relação ao DP (161,4 ± 1,3 µm), sendo o mesmo resultado observado para o volume dos cardiomiócitos (BD, 25,5 ± 0,4 vs. DP, 26,8 ± 0,4 pL; p < 0,05). Os tempos para o pico de contração (BD, 116 ± 1 vs. DP, 111 ± 2ms) e para o relaxamento total (BD, 143 ± 3 vs. DP, 232 ± 3 ms) foram maiores no grupo BD. CONCLUSÃO: Conclui-se que os miócitos do ventrículo esquerdo dos animais de baixo desempenho para o exercício físico apresentam menores dimensões que os dos animais de desempenho padrão, além de apresentarem perdas na capacidade contrátil.


BACKGROUND: Aerobic capacity is essential to physical performance, and low aerobic capacity is related to the triggering of various cardiovascular diseases. OBJECTIVE: To compare the morphology and contractility of isolated rat cardiomyocytes with low performance and standard performance for exercise. METHODS: Wistar rats with 10 weeks of age underwent a protocol of treadmill running to fatigue, and were divided into two groups: Low Performance (LP) and Standard Performance (SP). Then, the animals were sacrificed, the heart was quickly removed and, by means of enzymatic dissociation, left ventricular cardiomyocytes were isolated. The cell and sarcomeres length and width of cardiomyocytes were measured using an edge detection system. The isolated cardiomyocytes were electrically stimulated at 1 and 3 Hz and cell contraction was measured by registering the change of their length. RESULTS: The cell length was shorter in the LP group (157.2 1.3µm; p < 0.05) compared to SP (161.4 1.3µm), and the same result was observed for the volume of cardiomyocytes (LP, 25.5 0.4. vs. SP, 26.8 ± 0.4 pL; p < 0.05). The time to peak contraction (LP, 116 1 vs. SP 111 2ms) and total relaxation (LP, 143 3 vs. SP 232 3ms) were higher in the LP group. CONCLUSION: We conclude that left ventricular myocytes of animals with low performance for exercise are smaller than animals with standard performance. In addition to that, they present losses in contractile capacity.
