RESUMO

Straightforward access to enantiomerically pure 3,4-diamino-3,4-dideoxyphytosphingosines, as novel analogues of natural d-ribo-phytosphingosine was accomplished, starting from two available chirons: dimethyl l-tartrate and d-isoascorbic acid. A sequential Overman rearrangement followed by late-stage introduction of the alkyl side chain moiety via olefin cross-metathesis is the cornerstone of this approach. The preliminary evaluation study of the synthesised sphingomimetics, based on their ability to inhibit a proliferation of human cancer cells, showed promising cytotoxicity against Jurkat and HeLa cells for (2R,3R,4S)-2,3,4-triaminooctadecan-1-ol trihydrochloride.

RESUMO

A straightforward approach to a novel phytosphingosine-like ceramide has been accomplished. The cornerstone features of this divergent synthesis are a cascade Overman rearrangement of tris(imidate) to introduce three desired stereogenic centres via sequential chirality transfer and an effective olefin cross-metathesis to install a long side chain. The final unusual phytoceramides were evaluated for their capacity to inhibit the proliferation of cancer cell lines. The preliminary results revealed that compound 21 exhibits promising anticancer activity against HeLa and HCT-116 cells as well as the excellent selectivity in cytotoxicity (malignant vs non-malignant cell lines).

A novel lithiated high-entropy oxychloride Li0.5(Zn0.25Mg0.25Co0.25Cu0.25)0.5Fe2O3.5Cl0.5 (LiHEOFeCl) with spinel structure belonging to the cubic Fd3̄m space group is synthesized by a mechanochemical-thermal route. Cyclic voltammetry measurement of the pristine LiHEOFeCl sample confirms its excellent electrochemical stability and the initial charge capacity of 648 mA h g-1. The reduction of LiHEOFeCl starts at ca. 1.5 V vs. Li+/Li, which is outside the electrochemical window of the Li-S batteries (1.7/2.9 V). The addition of the LiHEOFeCl material to the composite of carbon with sulfur results in improved long-term electrochemical cycling stability and increased charge capacity of this cathode material in Li-S batteries. The carbon/LiHEOFeCl/sulfur cathode provides a charge capacity of 530 mA h g-1 after 100 galvanostatic cycles, which represents ca. 33% increase as compared to the charge capacity of the blank carbon/sulfur composite cathode after 100 cycles. This considerable effect of the LiHEOFeCl material is assigned to its excellent structural and electrochemical stability within the potential window of 1.7 V/2.9 V vs. Li+/Li. In this potential region, our LiHEOFeCl has no inherent electrochemical activity. Hence, it acts solely as an electrocatalyst accelerating the redox reactions of polysulfides. This can be beneficial for the performance of Li-S batteries, as evidenced by reference experiments with TiO2 (P90).

Surface modification of magnetic nanoparticles with poly-l-lysine, proline, and tryptophan was used to design potential theranostic agents for the application in cancer diagnosis and radionuclide-hyperthermia therapy. Characterization of bare and functionalized magnetic nanoparticles was performed in detail. The transparency of the examined magnetic nanoparticles was measured in the non-alternating magnetic field for a complete and better understanding of hyperthermia. For the first time amino acid-functionalized magnetic nanoparticles were labeled with theranostic radionuclides 131I and 177Lu. The specific absorption rate (SAR) procured for poly-l-lysine functionalized magnetic nanoparticles (SAR values of 99.7 W/g at H0 = 15.9 kA/m and resonant frequency of 252 kHz) demonstrated their possible application in magnetic hyperthermia. Poly-l-lysine functionalized magnetic nanoparticles labeled with 177Lu showed the highest radiochemical purity (>99.00 %) and in vitro stability in saline and serum (>98.00 % up to 96 h). The in vivo analysis performed after their intravenous administration in healthy Wistar rats presented good in vivo stability for several days. Encouraging results as well as magnetic and radiochemical properties of 177Lu-PLL-MNPs (80 °C) justify their further testing toward the potential use as theranostic agents for diagnostic and combined radionuclide-hyperthermia therapeutic applications.

SPD (several plastic deformations) methods make it possible to obtain an ultrafine-grained structure (UFG) in larger volumes of material and thus improve its mechanical properties. The presented work focuses on the structural and mechanical changes of aluminium alloy AlMgSi0.5 (EN AW 6060) during processing by repeated extrusion through the ECAP rectangular channel. After a four-pass extrusion, the samples' microstructures were observed using an optical microscope, where refinement of the material grains was confirmed. Tensile tests determined the extrusion forces and allowed interpretation of the changes in the mechanical properties of the stressed alloy. The grain size was refined from 28.90 µm to 4.63 µm. A significant improvement in the strength of the material (by 45%) and a significant deterioration in ductility (to 60%) immediately after the first extrusion was confirmed. The third pass through the die appeared to be optimal for the chosen deformation path, while after the fourth pass, micro-cracks appeared, significantly reducing the strength of the material. Based on the measurement results, new analytical equations were formulated to predict the magnitude or intensity of the volumetric and shape deformations of the structural grain size and, in particular, the adequate increase in the strength and yield point of the material.

Copper, antimony and sulfur in elemental form were applied for one-pot solid-state mechanochemical synthesis of skinnerite (Cu3SbS3) in a laboratory mill and an industrial mill. This synthesis was completed after 30 min of milling in the laboratory mill and 120 min in the industrial mill, as corroborated by X-ray diffraction. XRD analysis confirmed the presence of pure monoclinic skinnerite prepared in the laboratory mill and around 76% monoclinic skinnerite, with the secondary phases famatinite (Cu3SbS4; 15%), and tetrahedrite (Cu11.4Sb4S13; 8%), synthesized in the industrial mill. The nanocrystals were agglomerated into micrometer-sized grains in both cases. Both samples were nanocrystalline, as was confirmed with HRTEM. The optical band gap of the Cu3SbS3 prepared in the laboratory mill was determined to be 1.7 eV with UV-Vis spectroscopy. Photocurrent responses verified with I-V measurements under dark and light illumination and Cu3SbS3 nanocrystals showed ~45% enhancement of the photoresponsive current at a forward voltage of 0.6 V. The optical and optoelectrical properties of the skinnerite (Cu3SbS3) prepared via laboratory milling are interesting for photovoltaic applications.

The datasets presented here are related to the research paper entitled "Disordered Gd6UO12-δ with the cation antisite defects prepared by a combined mechanochemical-thermal method"[1]. The datasets complement the findings [1] on the effect of the combined mechanochemical-thermal processing of the stoichiometric mixture of solid precursors (3Gd2O3 + UO2) on the formation of Gd6UO12-δ phase. In this article, we provide (i) X-ray diffraction (XRD) data of the 3Gd2O3 + UO2 mixture milled for 12 h, (ii) the refined XRD data of the non-milled 3Gd2O3 + UO2 mixture after annealing at 1282 °C for 3 h in air, and (iii) the thermogravimetric and differential thermal analysis (TG-DTA) data for non-milled and mechanically preactivated 3Gd2O3 + UO2 mixture measured in air at a heat rate of 10 K/min.

BACKGROUND: Aceruloplasminaemia is a very rare autosomal recessive disorder caused by a mutation in the ceruloplasmin gene, which is clinically manifested by damage to the nervous system and retinal degeneration. This classical clinical picture can be preceded by diabetes mellitus and microcytic anaemia, which are considered to be early manifestations of aceruloplasminaemia. CASE PRESENTATION: In our report, we describe the case of a patient with aceruloplasminaemia detected in an early stage (without clinical symptoms of damage to the nervous system) during the search for the cause of hepatopathy with very low values of serum ceruloplasmin. Molecular genetic examination of the CP gene for ceruloplasmin identified a new variant c.1664G > A (p.Gly555Glu) in the homozygous state, which has not been published in the literature or population frequency databases to date. Throughout the 21-month duration of chelatase treatment, the patient, who is 43 years old, continues to be without neurological and psychiatric symptomatology. We observed a decrease in the serum concentration of ferritin without a reduction in iron deposits in the brain on magnetic resonance imaging. CONCLUSION: Currently, there is no unequivocal recommendation of an effective treatment for aceruloplasminaemia. Early diagnosis is important in the neurologically asymptomatic stage.

The synthesis of novel sphingoid base-like compounds with a quaternary stereocentre was achieved in a sequence featuring [3,3]-sigmatropic rearrangements and olefin cross-metathesis transformation as the key reaction steps, which were accompanied by the rational selection of suitable functional group transformations. The stereochemistry of the desired tetra-substituted carbon bearing nitrogen functionality was determined via NOESY experiments of the advanced oxazolidine-2-thiones. Cell viability experiments revealed significant antiproliferative/cytotoxic activity of the target compounds 7, ent-7 and 29 against the Jurkat cell line, with the IC50 values of 6.6 µM, 5.6 µM and 6.1 µM, respectively.

An electrochemical sensor is described for the determination of L-dopa (levodopa; 3,4-dihydroxyphenylalanine). An inkjet-printed carbon nanotube (IJPCNT) electrode was modified with manganese dioxide microspheres by drop-casting. They coating was characterized by field emission scanning electron microscopy, Fourier-transform infrared spectroscopy and X-ray powder diffraction. The sensor, best operated at a working voltage of 0.3 V, has a linear response in the 0.1 to 10 µM L-dopa concentration range, a 54 nM detection limit, excellent reproducibility, repeatability and selectivity. The amperometric approach was applied to the determination of L-dopa in spiked biological fluids and displayed satisfactory accuracy and precision. RESUMO

Nanostructured nickel ferrite (NiFe2O4) is prepared via high-energy ball milling of the bulk counterpart at ambient temperature. The structure of the as-prepared nanoferrite is characterized by Raman spectroscopy and 57Fe Mössbauer spectroscopy. Due to the ability of these spectroscopic techniques to probe the local environment of ions, valuable complementary insight into the nature of the local structural disorder of nanosized NiFe2O4 is provided. For the first time, evidence is given of the tetrahedrally coordinated nickel cations in the nanomaterial.

Bacillus anthracis protective antigen (PA) is a well known and relevant immunogenic protein that is the basis for both anthrax vaccines and diagnostic methods. Properly folded antigenic PA is necessary for these applications. In this study a high level of PA was obtained in recombinant Escherichia coli. The protein was initially accumulated in inclusion bodies, which facilitated its efficient purification by simple washing steps; however, it could not be recognized by specific antibodies. Refolding conditions were subsequently analyzed in a high-throughput manner that enabled nearly a hundred different conditions to be tested simultaneously. The recovery of the ability of PA to be recognized by antibodies was screened by dot blot using a coefficient that provided a measure of properly refolded protein levels with a high degree of discrimination. The best refolding conditions resulted in a tenfold increase in the intensity of the dot blot compared to the control. The only refolding additive that consistently yielded good results was L-arginine. The statistical analysis identified both cooperative and negative interactions between the different refolding additives. The high-throughput approach described in this study that enabled overproduction, purification and refolding of PA in a simple and straightforward manner, can be potentially useful for the rapid screening of adequate refolding conditions for other overexpressed antigenic proteins.


El antígeno protector de Bacillus anthracis (protective antigen, PA) es una importante proteína inmunogénica, en la que se basan tanto las vacunas contra el ántrax/carbunclo como varios métodos diagnósticos. Para estas aplicaciones es esencial que el PA mantenga sus propiedades antigénicas, para lo cual debe estar correctamente plegado. En este estudio se obtuvieron altos niveles del PA en Escherichia coli recombinante. Inicialmente, la proteína se acumuló desnaturalizada en cuerpos de inclusión, lo que facilitó su eficiente purificación en simples pasos de lavado, pero no fue reconocida por anticuerpos específicos. Se analizaron las condiciones de replegado con un sistema de alto rendimiento, evaluando simultáneamente casi un centenar de condiciones diferentes. La recuperación de la capacidad del PA de ser reconocido por los anticuerpos se evaluó por dot blot utilizando un coeficiente que proporcionó una medida de los niveles de proteína correctamente plegada, con un alto grado de discriminación. Las mejores condiciones de renaturalización permitieron un aumento de diez veces en la intensidad de los dot blots con respecto del control. El único aditivo que produjo buenos resultados de forma constante fue la L-arginina. El análisis estadístico de las interacciones entre los diferentes aditivos de replegado permitió identificar tanto interacciones cooperativas como negativas. El enfoque de alto rendimiento descripto en este trabajo, que permitió la sobreproducción, purificación y plegado del PA de una manera sencilla y directa, puede ser potencialmente útil para el rápido screening de las condiciones adecuadas de replegado cuando se sobreexpresan otras proteínas antigénicas.

RESUMO

Direct electrical stimulation (DES) of the optic radiation (OR) during an awake tumor resection has been repeatedly reported. In all cases, visual function monitoring was performed in patients with open eyes which were looking at a picture. We report a new modification of the standard method, OR stimulation in patient with closed and covered eyes. To the best of our knowledge, this method was not presented before. According to our first experience, this methodology may be in some cases a potentially more sensitive form of neuromonitoring than the OR stimulation in patients with open eyes, as the phosphenes elicited by DES may be more distinct in patients with covered eyes. The technique is discussed, and a literature review on intraoperative identification of the OR is presented as well. However, a future prospective study is needed to confirm the relevance of our finding.

BACKGROUND: The postoperative biological behavior of nonfunctioning pituitary adenomas (NFPAs) is variable. Some residual NFPAs are stable long-term, others grow, and some recur despite complete removal. The usual histological markers of tumor aggressiveness are often similar between recurring, regrowing, and stable tumors, and therefore are not reliable as prognostic parameters. In this study, the clinical utility of proliferation indices (labeling index, Li) based on immunohistochemistry targeted at antigens Ki-67 and High-mobility group A1 (HMGA-1) for prediction of NFPA prognosis was investigated. METHODS: Fifty patients with NFPAs were investigated. In each patient, Ki-67 and HMGA-1 Li were evaluated. Based on postoperative magnetic resonance images, patients were classified as tumor-free (18 patients), or harboring a residual tumor (32 patients). The latter group was further subdivided into groups with stable tumor remnants (11 patients) or progressive tumor remnants (21 patients). RESULTS: The median follow-up period was 8 years. No significant relationship between HMGA-1 Li and residual tumor growth was found. Growing residual tumors showed a trend towards higher Ki-67 Li compared with stable ones (p = 0.104). All tumor remnants with Ki-67 Li above 2.2% were growing. The relationship between residual tumor growth and Ki-67 Li exceeding the cutoff value of 2.2% was significant (p = 0.01 in univariate, p = 0.044 in multivariate analysis). CONCLUSIONS: The prognostic significance of the HMGA-1 antigen was not confirmed. In contrast, the Ki-67 Li provides useful and valuable information for the postoperative management of NFPAs. In residual adenomas with a Ki-67 Li above 2.2%, regrowth should be expected, and these tumors may require shorter intervals of follow-up magnetic resonance imaging (MRI) and/or early adjuvant therapy. Future larger studies are needed to confirm the results of this study.

Nanostructured fayalite (α-Fe(2)SiO(4)) with a large volume fraction of interfaces is synthesized for the first time via single-step mechanosynthesis, starting from a 2α-Fe(2)O(3) + 2Fe + 3SiO(2) mixture. The nonequilibrium state of the as-prepared silicate is characterized by the presence of deformed polyhedra in the interface/surface regions of nanoparticles.

The 5-10 nm Ce(1-x)Eu(x)O(2-delta) (0 < or = x < or = 0.30) nanoparticles with fluorite structure were synthesized by thermal decomposition of Eu- and Ce-2,4-pentanedione complexes mixtures. X-ray line broadening analysis of mixed samples Ce(1-x)Eu(x)O(2-delta) (0.05 < or = x < or = 0.30) showed that the crystallite size was lower and root mean square strain higher than in pure ceria. However, within mixed samples Ce(1-x)Eu(x)O(2-delta) (0.05 < or = x < or = 0.30) crystallite size and root mean square strain were independent of Eu3+ concentration. Raman spectroscopy results indicated that europium ions yield disorder by breaking the phonon propagation and therefore making the non-centre Brillouin zone modes Raman active. The absorption bands in the spectra of mixed oxides were blue-shifted in comparison to pure CeO(2-delta) nanopowder. The samples show red emission typical for Eu ions. The biggest photoluminescent intensity was observed for the highest Eu3+ concentration (x = 0.30) and further enhanced with the increase in crystallinity.
