RESUMO

Background: Platycodon grandiflorus belongs to the genus Platycodon and has many pharmacological effects, such as expectorant, antitussive, and anti-tumor properties. Among transcription factor families peculiar to eukaryotes, the basic leucine zipper (bZIP) family is one of the most important, which exists widely in plants and participates in many biological processes, such as plant growth, development, and stress responses. However, genomic analysis of the bZIP gene family and related stress response genes has not yet been reported in P. grandiflorus. Methods: P. grandiflorus bZIP (PgbZIP) genes were first identified here, and the phylogenetic relationships and conserved motifs in the PgbZIPs were also performed. Meanwhile, gene structures, conserved domains, and the possible protein subcellular localizations of these PgbZIPs were characterized. Most importantly, the cis-regulatory elements and expression patterns of selected genes exposed to two different stresses were analyzed to provide further information on PgbZIPs potential biological roles in P. grandiflorus upon exposure to environmental stresses. Conclusions: Forty-six PgbZIPs were identified in P. grandiflorus and divided into nine groups, as displayed in the phylogenetic tree. The results of the chromosomal location and the collinearity analysis showed that forty-six PgbZIP genes were distributed on eight chromosomes, with one tandem duplication event and eleven segmental duplication events identified. Most PgbZIPs in the same phylogenetic group have similar conserved motifs, domains, and gene structures. There are cis-regulatory elements related to the methyl jasmonate (MeJA) response, low-temperature response, abscisic acid response, auxin response, and gibberellin response. Ten PgbZIP genes were selected to study their expression patterns upon exposure to low-temperature and MeJA treatments, and all ten genes responded to these stresses. The real-time quantitative polymerase chain reaction (RT-qPCR) results suggest that the expression levels of most PgbZIPs decreased significantly within 6 h and then gradually increased to normal or above normal levels over the 90 h following MeJA treatment. The expression levels of all PgbZIPs were significantly reduced after 3 h of the low-temperature treatment. These results reveal the characteristics of the PgbZIP family genes and provide valuable information for improving P. grandiflorus's ability to cope with environmental stresses during growth and development.

BACKGROUND: R2R3-MYB genes comprise one of the largest and most important gene families in plants, and are involved in the regulation of plant growth and development as well as responses to abiotic stresses. However, the functions of R2R3-MYB genes in Dendrobium nobile remains largely unknown. METHODS: Here, a comprehensive genome-wide analysis of D. nobile R2R3-MYB genes was performed, in which phylogenic relationships, gene structures, motif composition, chromosomal locations, collinearity analysis, and cis-acting elements were investigated. Moreover, the expression patterns of selected DnMYB genes were analyzed in various tissues and under different abiotic stresses. RESULTS: In total, 125 DnMYB genes were identified in the D. nobile genome, and were subdivided into 26 groups based on phylogenetic analysis. Most genes in the same subgroup showed similar exon/intron structure and motif composition. All the DnMYB genes were mapped to 19 chromosomes with the co-linearity relationship. Reverse transcription-quantitative real-time PCR (RT-qPCR) results showed that 8 DnMYBs exhibited different expression patterns in different plant tissues, and were differentially expressed in response to abscisic acid, methyl jasmonate, low-temperature stress. CONCLUSIONS: This work contributes to a comprehensive understanding of the R2R3-MYB gene family in D. nobile, and provides candidate genes for future research on abiotic stress in this plant.

While thermosets with high mechanical properties and shape memory capabilities have been developed in recent years, two main bottlenecks persist in their intrinsic nonreusability and flammability, especially for those shape memory thermosets used in recycling-required field with high glass transition temperatures ( Tg) and thus risky high temperatures to trigger shape recovery. Here, we report a new shape memory epoxy thermoset integrated with excellent fire retardancy, recyclability, high mechanical performance, and 100% shape recovery ratio. The shape memory effect of this new thermoset was directly triggered by high-temperature flame for the first time. Furthermore, the survival thermoset can be recycled by a simple solid-state recycling method and reused as reinforcing fillers for polyester. The highest recycling efficiency reached 85.4%, and the reinforced composite presented about four times higher storage modulus compared to that of neat sample. This work may open a door for application of thermoset shape memory polymers in many lightweight engineering structures and devices where fire hazard is a concern. The newly proposed concept of flame-triggered shape memory effect may also find applications in fire-protecting system.

Low output in stress and energy in rubbery state has been a bottleneck for wide-spread applications of thermoset shape memory polymers (SMPs). Traditionally, stress or energy storage in thermoset network is through entropy reduction by mechanical deformation or programming. We here report another mechanism for energy storage, which stores energy primarily through enthalpy increase by stretched bonds during programming. As compared to entropy-driven counterparts, which usually have a stable recovery stress from tenths to several MPa and energy output of several tenths MJ/m3, our rubbery network achieved a recovery stress of 17.0 MPa and energy output of 2.12 MJ/m3 in bulk form. The giant stress and energy release in the rubbery state will enhance applications of thermoset SMPs in engineering structures and devices.

Amylopectin is a naturally hyperbranched biopolymer with an extremely high molecular weight. Furthermore, this material is non-toxic in nature, and exhibits good biocompatibility and biodegradability properties. Herein, we describe the development of a one-step reaction strategy for the synthesis of amphiphilic high-molecular-weight hyperbranched amylopectin derivatives with hydrophobic shells and large hydrophilic cores. The chemical structures of the resulting materials were characterized using FTIR spectroscopy, solid-state (13)C cross-polarization/magic angle spinning NMR spectroscopy and gas chromatography. The results from transmission electron microscopy, fluorescence spectroscopy, and UV-vis analysis confirmed that the hyperbranched amylopectin derivatives were composed of hydrophobic shells with cholesteryl residues and hydrophilic amylopectin cores. These amylopectin derivatives exhibited high encapsulation capabilities toward water-soluble molecules, and could be used as functional nanovehicles for the controlled release of water-soluble molecules, and the in situ synthesis of metallic nanoparticles.

Unique "micro-bean sprouts" with "nano-tails" were electrohydrodynamically prepared from poly[(propylmethacryl-heptaisobutyl-polyhedral oligomeric silsesquioxane)-co-(methylmethacrylate)] (POSS-MMA). The nano/microstructured POSS-MMA substrates reveal superhydrophobic nature, with contact angles >160°. Contact angle versus time plots show that water droplet evaporation from the substrates with "micro-bean sprouts" falls into two stages: the initial stage, marked by steadily decreasing contact angles and the later stage, marked by rapidly decreasing contact angles. The turning point differentiating these two stages occurs at ~120°-130°. The substrates consisting of "micro-bean sprouts" with larger "heads" experience pinned triple-line phase during the early stage of droplet evaporation. In contrast, the triple line for a droplet placed on "micro-bean sprouts" with smaller "heads" can move easily, leading to a rapid decrease in contact diameter. Meanwhile, the contact angle decreases only slightly during the rapidly moving triple-line phase. The contact angles of the fibrous substrates measured during the initial stage of water droplet evaporation are much lower than those of "micro-bean sprouts", even though the time period of the initial stage is extended. Both the architectures and the sizes of these POSS-MMA nano/microstructures were shown to affect the energy barrier for the triple-line motion during water droplet evaporation and therefore the contact angle hysteresis.

AIM: to investigate the ability of drug-loaded N,O-carboxymethyl chitosan (CMCS) hydrogels to modulate wound healing after glaucoma filtration surgery. METHODS: the drug-loaded CMCS hydrogels were in situ synthesized using genipin as the crosslinker in the presence of 5-fluorouracil (5FU) or bevacizumab. Their structures were characterized by FTIR, ultraviolet-visible (UV-vis) spectroscopy and scanning electron microscopy (SEM). In-vitro drug release experiments and in vivo evaluation in rabbits were performed. RESULTS: the results of FTIR, UV-vis spectroscopy and SEM analyses indicated that 5FU was encapsulated into the CMCS hydrogels that were crosslinked by genipin. The in vitro drug release experiments showed that nearly 100% of 5FU was released from the drug-loaded hydrogels within 8 h, but less than 20% bevacizumab was released after 53 h. The in vivo evaluation in rabbits indicated that the drug-loaded CMCS hydrogels were nontoxic to the cornea and were gradually biodegraded in the eyes. Furthermore, the drug-loaded CMCS hydrogels effectively inhibited conjunctival scarring after glaucoma filtration surgery and controlled postoperative intraocular pressure (IOP). CONCLUSION: the drug-loaded CMCS hydrogels provide a great opportunity to increase the therapeutic efficacy of glaucoma filtration surgery.
