RESUMO

The placenta becomes one of the most diversified organs during placental mammal radiation. The main in vitro model for studying mouse trophoblast development is the 2D differentiation model of trophoblast stem cells, which is highly skewed to certain lineages and thus hampers systematic screens. Here, we established culture conditions for the establishment, maintenance, and differentiation of murine trophoblast organoids. Murine trophoblast organoids under the maintenance condition contain stem cell-like populations, whereas differentiated organoids possess various trophoblasts resembling placental ones in vivo. Ablation of Nubpl or Gcm1 in trophoblast organoids recapitulated their deficiency phenotypes in vivo, suggesting that those organoids are valid in vitro models for trophoblast development. Importantly, we performed an efficient CRISPR-Cas9 screening in mouse trophoblast organoids using a focused sgRNA (single guide RNA) library targeting G protein-coupled receptors. Together, our results establish an organoid model to investigate mouse trophoblast development and a practicable approach to performing forward screening in trophoblast lineages.

The stability and toxicity problems have haunted the development and applications of metal halide perovskite materials, for which the lead-free inorganic double perovskite Cs2AgBiBr6 has emerged as a promising substitute in recent years. However, poor film quality has severely limited its photovoltaic performance that could have been induced by some key factors such as high annealing temperature. Herein, we present a facile strategy to fabricate high-quality pinhole-free Cs2AgBiBr6 films with large grain sizes by introducing carbohydrazide (CBH) into the precursor. Detailed characterizations have shown that the carbonyl group (C═O) in CBH plays the critical role in coordinating with Ag+ and Bi3+ cations during the film formation process. As another consequence, the as-fabricated devices have exhibited significantly higher reproducibility for fabrication. By optimizing the amount of CBH, the power conversion efficiency (PCE) relatively increased 37 to 1.57%, which remained 95.0% in an ambient environment for a 1000-h test. Hopefully, this work could facilitate the current technologies in the exploration of high-performance lead-free perovskites such as Cs2AgBiBr6 and better understanding of the mechanism in the additive engineering as well.

SiO2 nanomaterials are widely used for antireflection and self-cleaning, but the preparation process is usually complex and time-consuming. Hence, we present a facile one-step synthesis of a hydrophobic two-dimensional SiO2 nanomesh by tuning the reaction temperature using dodecylamine as a catalyst. SiO2 nanomesh has the advantages of an adjustable refractive index, simple preparation process, and low cost, which affords both antireflection and self-cleaning functions for solar cells. Two types of perovskite solar cells were used to verify the stability and universality of the SiO2 nanomesh coatings. The antireflection effect of the SiO2 nanomesh is found to increase the current density of both perovskite solar cells fabricated at 500 °C and 150 °C, with the efficiency increased by 4.48% and 4.79%, respectively.

RESUMO

Organic room temperature phosphorescence (RTP) materials with ultralong lifetime possess the remarkable advantage in bioimaging for elimination of background noise by characteristic time scale. However, most of RTP luminogens need to be excited by the harmful ultraviolet (UV) lamp, and exhibit green or yellow emission with shallow tissue penetration, constraining the in vivo bioimaging for further application in clinical diagnosis and pathological study. In this text, the much safer excitation process by sunlight and mobile phone flashlight is realized by organic luminogens with various electronic pull-push systems. Moreover, the bright red RTP emission with lifetime up to 344 ms is achieved by optimizing molecular geometry and electronic property. Especially, the mobile phone flashlight-excited red afterglow imaging of lymph nodes in living mice has been realized for the first time, affording a safe and conventional approach to achieve the afterglow imaging of living mice with deep issue penetration and high signal-to-noise ratios.

A series of triphenylethylene derivatives exhibited multi-photoresponsive properties, including photochromism, photodeformation and room temperature phosphorescence (RTP), which are strongly related to molecular conformations and packing in the aggregated states. Accordingly, these properties can be subtly adjusted by substituents to the center double bond and/or peripheral phenyl moieties. The introduction of bromine atom was beneficial to photochromism and photodeformation properties as a result of the additional C-H⋯Br interactions and electron-withdrawing property. Also, it can promote the RTP effect via heavy atom effect, resulting in persistent afterglow lasting up to 150 min as detected by chemiluminescent imaging system.

Tembusu Virus (TMUV), a pathogenic member of Flavivirus family, acts as the causative agent of egg-laying and has severely threatened the duck industry over the past few years. Thus far, the pathogenicity of such virus has been extensively studied, whereas TMUV on immune system has been less comprehensively assessed, especially on ducklings that exhibit more susceptible to TMUV attack. Accordingly, in the present study, 5-day-old ducklings were infected with TMUV-TC2B (104 TCID50) via intravenous injection, and mock ones were inoculated with phosphate-buffered saline (PBS) in identical manner as control. At 1 day-post inoculation (dpi), the innate immunity was strongly activated, and reacted rapidly to TMUV invasion, which was reflected as the significantly up-regulated IFN-stimulated genes (ISGs), especially in immune organs (e.g., thymus, bursa of Fabricius (BF) and spleen). Subsequently, under the continuous monitoring, the levels of IgA, IgM and IgG acting as the representative immunoglobulins (Igs) were constantly higher than those of mock ducklings, demonstrating that humoral immunity also played a major role in anti-virus infection. Despite the immune system activated positively, TMUV still caused systemic infection, and in particular, the immune organs were subject to severe damage in the early infection. With our constant observation, the injury of spleen and BF turned out to be getting more serious, and at 6 dpi, TMUV antigen was widely detected in both of two immune organs by immunohistochemistry (IHC) and main histopathological lesion presented as lymphocytopenia. Moreover, the elevated apoptosis rate of splenic lymphocytes and the alteration of immune organ index also revealed the damage of lymphoid organs and similarly, it is worth noting that severe damages were detected in thymus of TMUV-infected ducklings as well. In brief, the present study systematically described the dynamic damage of immune system after being attacked by TMUV and presented insights into the research of pathogenicity.

Fowl adenovirus (FAdV) has posed a grave threat to the health of poultry, and the sudden outbreak highlights the importance of the new rapid diagnostic method for the control and prevention of transmission. Hence, in the present study, a novel recombinase polymerase amplification (RPA) assay, which was suitable for all 12 serotypes (FAdV-1 to 8a and 8b to 11) had been successfully launched to detect FAdV. Also, the entire amplification process could be completed in the isothermal condition when temperature ranged from 26 to 42°C within no more than 14 min, which was remarkably superior to endpoint polymerase chain reaction (98 min) with the same detecting sensitivity (as low as 0.1 fg viral DNA), avoiding sophisticated thermal cyclers with simple operation. Additionally, the same primers did not produce positive reactions with other viruses tested, demonstrating that the specificity of the RPA assay was acceptable. Moreover, this developed method could be efficiently used in the diagnosis of FAdV references and epidemic strains from different avian origins, thus making it a rapid, reliable, and point-of-care FAdV diagnostics tool, as well as an alternative to endpoint PCR.

Following the development of organic mechanoluminescence (ML) materials, molecular packing was proved as the key point to the emission process under an external force. In this text, with the introduction of spiro-(fluorene-9-9'-xanthene) (SFX) unit as the building block, the molecular packing of the resultant compound (BSFXA) was optimized with the interlaced mode, directly leading to the efficient ML effect. The key role of SFX with a spiro-structure can be further confirmed by the ML inactivity of reference compound BFA with the replacement of SFX unit by dimethyl fluorene (MeF), which provided a novel strategy to construct organic ML luminogens.
