RESUMO

Propylparaben (PrP) and dichloropropylparaben (diClPrP) are found in soil worldwide, mainly due to the incorporation of urban sludge in crop soils and the use of non-raw wastewater for irrigation. Studies on the adverse effects of PrP on plants are incipient and not found for diClPrP. PrP and diClPrP were evaluated at concentrations 4, 40, and 400 µg/L for their phytotoxic potential to seeds of Allium cepa (onion), Cucumis sativus (cucumber), Lycopersicum sculentum (tomato), and Lactuca sativa (lettuce), and cytotoxic, genotoxic potential, and for generating oxygen-reactive substances in root meristems of A. cepa bulbs. PrP and diClPrP caused a significant reduction in seed root elongation in all four species. In A. cepa bulb roots, PrP and diClPrP resulted in a high prophase index; in addition, PrP at 400 µg/L and diClPrP at the three concentrations significantly decreased cell proliferation and caused alterations in a significant number of cells. Furthermore, diClPrP concentrations induced the development of hooked roots in onion bulbs. The two chemical compounds caused significant changes in the modulation of catalase, ascorbate peroxidase, and guaiacol peroxidase, disarming the root meristems against hydroxyl radicals and superoxides. Therefore, PrP and diClPrP were phytotoxic and cytogenotoxic to the species tested, proving dangerous to plants.

The benzophenone-3 (BP-3) sunscreen is recurrently released into the environment from different sources, however, evaluations of its adverse effects on plants do not exist in the literature. In this study, BP-3 was evaluated, at concentrations 2; 20, and 200 µg/L, regarding phytotoxicity, based on germination and root elongation in seeds, in Lactuca sativa L., Cucumis sativus L. and Allium cepa L., and phytotoxicity, cytogenotoxicity and oxidative stress in A. cepa bulb roots. The BP-3 concentrations, except for the 200 µg/L concentration in L. sativa, caused no significant reduction in seed germination. All concentrations tested significantly reduced the elongation of roots from seeds and roots from bulbs. The 20 and 200 µg/L concentrations caused oxidation in cells, disturbances in the cell cycle, and alterations in prophase and metaphase, as well as the induction of micronuclei, in A. cepa root meristems. Furthermore, the three concentrations induced a high number of prophases in root tips. Such disorders were caused by excess H2O2 and superoxide produced in cells due to exposure to BP-3, which triggered significant phytotoxicity, cytotoxicity, and genotoxicity in root meristems. Thus, the recurrent contamination of agricultural and non-agricultural soils with BP-3, even at a concentration of 2 µg/L, represents an environmental risk for plants. These results point to the impending need to set limits for the disposal of this sunscreen into the environment since BP-3 has been used in industry for several decades.

ABSTRACT: This research aimed to morphologically characterize and estimate the genetic diversity of 21 Capsicum accessions belonging to the Capsicum Germplasm Active Bank at the Universidade Federal do Piauí (BAGC-UFPI) using uni- and multivariate analysis. The experiment was carried out in a greenhouse, by completely randomized experimental design with four repetitions, with one plant per plot. Analysis of variance (ANOVA) and the comparison of means for seven quantitative variables were performed, followed by clustering the averages by the Scott-Knott test (P < 0.05). The analysis of the seven quantitative and thirteen qualitative descriptors was estimated based on the Gower distance. Later, it was performed the principal component analysis and the UPGMA hierarchical cluster method. Results characterized and identified a wide intra- and interspecific genetic variability related to the fruit size, colors, and shapes among the Brazilian Capsicum genotypes belonging to the BAGC-UFPI. The descriptors used in this research were effective in the discrimination of the pepper accessions, especially the closely related C. frutescens and C. chinense species.

O experimento foi conduzido em telado, utilizando-se o delineamento inteiramente ao acaso, com quatro repetições, sendo uma planta por parcela. Realizou-se análise de variância (ANOVA) e comparação da média para as setes variáveis quantitativas, seguidas do agrupamentos de médias pelo teste Scott-Knott (P < 0,05). A análise dos sete descritores quantitativos e treze qualitativos foi estimada com base na distância de Gower. Posteriormente, foi realizada a análise de componentes principais e o método de agrupamento hierárquico UPGMA. Os resultados caracterizaram e identificaram uma ampla variabilidade genética intra e interespecífica relacionada ao tamanho, cor e formato dos frutos entre os genótipos brasileiros de Capsicum do banco de germoplasma de pimentas BAGC-UFPI. Os descritores utilizados foram eficientes na discriminação dos acessos de pimentas, especialmente para espécies proximamente relacionadas C. frutescens e C. chinense.

Molecular markers are important tools in the characterization of plant genetic diversity and can provide support for conservation strategies for endangered populations. The different molecular techniques involve the evaluation of many individuals; therefore, it is crucial to have fast, efficient, and inexpensive methods for DNA extraction. Given the importance of the Aroeira (Myracrodruon urundeuva Fr. All.) it is pertinent to optimize a protocol that allows the obtainment of intact and pure DNA, aiming to assist conservation strategies for this species that is threatened with extinction. Thus, this study aimed to compare five DNA extraction methods: Dellaporta et al. (1983), Doyle and Doyle (1987) modified, Ferreira and Grattapaglia (1995), Romano and Brasileiro (2015), and Khanuja et al. (1999) and optimize the most efficient protocol for M. urundeuva. The modified DNA extraction protocol proposed by Doyle and Doyle (1987), using 100 mg of leaf tissue and 6 µl of ß-mercaptoethanol was the protocol that presented the sharpest bands after DNA electrophoresis and after the reactions of amplification employing Polymerase Chain Reaction (PCR). Therefore, it is suggested to use the protocol described by Doyle and Doyle (1987) modified for the extraction of DNA from young M. urundeuva leaves to carry out techniques involving molecular markers.

Cytogenomic resources have accelerated synteny and chromosome evolution studies in plant species, including legumes. Here, we established the first cytogenetic map of V. angularis (Va, subgenus Ceratotropis) and compared this new map with those of V. unguiculata (Vu, subgenus Vigna) and P. vulgaris (Pv) by BAC-FISH and oligopainting approaches. We mapped 19 Vu BACs and 35S rDNA probes to the 11 chromosome pairs of Va, Vu, and Pv. Vigna angularis shared a high degree of macrosynteny with Vu and Pv, with five conserved syntenic chromosomes. Additionally, we developed two oligo probes (Pv2 and Pv3) used to paint Vigna orthologous chromosomes. We confirmed two reciprocal translocations (chromosomes 2 and 3 and 1 and 8) that have occurred after the Vigna and Phaseolus divergence (~9.7 Mya). Besides, two inversions (2 and 4) and one translocation (1 and 5) have occurred after Vigna and Ceratotropis subgenera separation (~3.6 Mya). We also observed distinct oligopainting patterns for chromosomes 2 and 3 of Vigna species. Both Vigna species shared similar major rearrangements compared to Pv: one translocation (2 and 3) and one inversion (chromosome 3). The sequence synteny identified additional inversions and/or intrachromosomal translocations involving pericentromeric regions of both orthologous chromosomes. We propose chromosomes 2 and 3 as hotspots for chromosomal rearrangements and de novo centromere formation within and between Vigna and Phaseolus. Our BAC- and oligo-FISH mapping contributed to physically trace the chromosome evolution of Vigna and Phaseolus and its application in further studies of both genera.

"Sucupira branca" is a plant found in the Brazilian Cerrado and is adapted to low fertility soils, and its fruit extract has anti-inflammatory, healing, antiulcerogenic, antimicrobial, cercaricidal, leishmanicidal and antioxidant activities. Furthermore, it provides protection against oxidative stress, is a natural biocontrol agent of Aedes aegypti, has very resistant wood, is a melliferous plant and has been used in reforestation programs. The development of conservation strategies is important for maintaining diversity in natural populations of "sucupira branca" since these populations are in the process of genetic erosion. Molecular biology techniques, which are important for characterizing the genetic diversity of plants to develop conservation strategies, require sufficient high-quality genomic deoxyribonucleic acid (DNA). This study aimed to compare five methods to extract DNA from "sucupira branca". The quality and concentration of DNA were revealed by agarose gel electrophoresis, and only the protocols of Dellaporta, Wood and Hicks et al. (1983) and Khanuja et al. (1999) did not result in satisfactory quantities of DNA. When PCR (Polymerase Chain Reaction) was performed with three inter-simple sequence repeat (ISSR) primers, DNA was successfully amplified from extractions performed with the protocols proposed by Doyle and Doyle (1987), Romano and Brasileiro (1998) and Ferreira and Grattapaglia (1995), which are less expensive than commercial purification kits. These protocols resulted in DNA of sufficient quality and quantity after the amplification reactions were performed.

Mitotic prophase chromosome condensation plays an essential role in nuclear division being therefore regulated by highly conserved mechanisms. However, degrees of chromatin condensation in prophase-prometaphase cells may vary along the chromosomes resulting in specific condensation patterns. We examined different condensation patterns (CPs) of prophase and prometaphase chromosomes and investigated their relationship with genome size and distribution of histone H4 acetylated at lysine 5 (H4K5ac) in 17 plant species. Our results showed that most species with small genomes (2C < 5 pg) (Arachis pusilla, Bixa orellana, Costus spiralis, Eleutherine bulbosa, Indigofera campestris, Phaseolus lunatus, P. vulgaris, Poncirus trifoliata, and Solanum lycopersicum) displayed prophase chromosomes with late condensing terminal regions that were highly enriched in H4K5ac, and early condensing regions with apparently non-acetylated proximal chromatin. The species with large genomes (Allium cepa, Callisia repens, Araucaria angustifolia and Nothoscordum pulchellum) displayed uniformly condensed and acetylated prophase/prometaphase chromosomes. Three species with small genomes (Eleocharis geniculata, Rhynchospora pubera, and R. tenuis) displayed CP and H4K5ac labeling patterns similar to species with large genomes, whereas a forth species (Emilia sonchifolia) exhibited a gradual chromosome labeling, being more acetylated in the terminal regions and less acetylated in the proximal ones. The nucleolus organizer chromatin was the only chromosomal region that in prometaphase or metaphase could be hyperacetylated, hypoacetylated or non-acetylated, depending on the species. Our data indicate that the CP of a plant chromosome complement is influenced but not exclusively determined by nuclear and chromosomal DNA contents, whereas the CP of individual chromosomes is clearly correlated with H4K5ac distribution.

The goal of the present study was to evaluate the cytotoxicity and genotoxicity of artificial synthetic flavoring agents cookie and tutti-frutti. To this end, root meristem cells of Allium cepa L. were exposed to these substances in exposure times of 24 and 48 hour using individual doses of 0.3; 0.6 and 0.9 mL and doses combined as follows: 0.3 mL + 0.3 mL; 0.6 mL and 0.9 mL + 0.6 mL + 0.9 mL. After applying the treatments, root meristems were fixed, hydrolyzed, stained and analyzed a total of 5,000 cells using an optical microscope to evaluate each dose and combined treatment. All three doses of cookie flavoring and combined treatments significantly inhibited cell division of the tissue studied. Doses of tutti-frutti caused no change in cell division rate. In addition, doses of both flavorings and treatments combining these solutions induced cell aberrations in a significant number of cells to the A. cepa system. Therefore, under these analytical conditions, cookie flavoring and combined doses were cytotoxic and genotoxic, and tutti-frutti flavoring, although non-cytotoxic, demonstrated genotoxic action.


Objetivou-se neste trabalho avaliar a citotoxicidade e a genotoxicidade de aditivos aromatizantes sintéticos artificiais de biscoito e tutti-frutti. Essa avaliação se deu por meio das células meristemáticas de raízes de Allium cepa L., nos tempos de exposição de 24 e 48 horas, em que estas substâncias foram analisadas individualmente, nas doses de 0,3; 0,6 e 0,9 mL, e associadas entre si, da seguinte forma: 0,3 mL + 0,3 mL; 0,6 mL + 0,6 mL e 0,9 mL + 0,9 mL. Os meristemas de raízes foram fixados, hidrolisados e corados, e em seguida analisados em microscópio óptico, no qual se avaliou para cada dose e tratamento associado um total de 5.000 células. A partir dos resultados obtidos verificou-se que as três doses do aromatizante de biscoito e os tratamentos em associação inibiram significativamente a divisão celular dos tecidos em estudo. Já as doses de tutti-frutti não alteraram o índice de divisão celular das raízes em questão. Verificou-se também que as doses dos dois aromatizantes e os tratamentos em associação, induziram aberrações celulares em número significativo. Portanto, nestas condições de análise, o aditivo de biscoito e as doses associadas foram citotóxicos e genotóxicos, e o de tutti-frutti, apresentou apenas ação genotóxica.

The chromosomal phosphorylation of histone H3 during mitosis and meiosis seems to play a fundamental role in the control of cell division in all eukaryotes. In plants, the temporal and spatial distribution of H3S10 phosphorylated (H3S10ph) is currently known only for chromosomes of a few angiosperms. In the present study, we analyzed the pattern of H3S10ph in mitotic chromosomes of 14 plant species, including 2 bryophytes and 12 tracheophytes. In all species, the phosphorylation of H3S10 was cytologically detected first in prophase and disappeared in late anaphase. Two patterns of chromosomal phosphorylation were observed: (1) only the pericentromeric regions were hyperphosphorylated, whereas the remaining chromosome arms appeared weakly and diffusely immunolabeled, and (2) the whole chromosomes appear uniformly phosphorylated, increasing the labeling intensity at metaphase. The first pattern was observed in all tracheophytes with monocentric chromosomes, whereas the second one was restricted to the bryophytes and angiosperms with holokinetic chromosomes. In both cases, no particular kind of chromatin such as NORs or heterochromatic bands were differentially labeled. Based on this data and previous analyses in other eukaryotes, we suggest that hyperphosphorylation of the whole mitotic metaphase chromosome represents the ancestral condition for eukaryotic chromosomes, and the change to the pericentromeric pattern occurred in the transition from bryophytes to tracheophytes. The meaning and possible implications of these results are discussed in the light of recent and classical experiments.

Eukaryotic chromosomes are organized into two large and distinct domains, euchromatin and heterochromatin, which are cytologically characterized by different degrees of chromatin compaction during interphase/prophase and by post-synthesis modifications of histones and DNA methylation. Typically, heterochromatin remains condensed during the entire cell cycle whereas euchromatin is decondensed at interphase. However, a fraction of the euchromatin can also remain condensed during interphase and appears as early condensing prophase chromatin. 5S and 45S rDNA sites and telomere DNA were used to characterize these regions in metaphase and interphase nuclei. We investigated the chromosomal distribution of modified histones and methylated DNA in the early and late condensing prophase chromatin of two species with clear differentiation between these domains. Both species, Costus spiralis and Eleutherine bulbosa, additionally have a small amount of classical heterochromatin detected by CMA/DAPI staining. The distribution of H4 acetylated at lysine 5 (H4K5ac), H3 phosphorylated at serine 10 (H3S10ph), H3 dimethylated at lysine 4 or 9 (H3K4me2, H3K9me2), and 5-methylcytosine was compared in metaphase, prophase, and interphase cells by immunostaining with specific antibodies. In both species, the late condensing prophase chromatin was highly enriched in H4K5ac and H3K4me2 whereas the early condensing chromatin was very poor in these marks. H3K9me2 was apparently uniformly distributed along the chromosomes whereas the early condensing chromatin was slightly enriched in 5-methylcytosine. Signals of H3S10ph were restricted to the pericentromeric region of all chromosomes. Notably, none of these marks distinguished classical heterochromatin from the early condensing euchromatin. It is suggested that the early condensing chromatin is an intermediate type between classical heterochromatin and euchromatin.

O conhecimento da biologia floral é de suma importância para o desenvolvimento da cultura do cajueiro (Anacardium occidentale L.). Com relação aos aspectos botânicos, as características morfológicas das flores contribuíram efetivamente para a determinação das espécies do gênero Anacarduim conhecidas. No presente trabalho, objetivou-se estudar a biologia floral dos cajueiros anão precoce e comum. A pesquisa foi desenvolvida na área experimental do Departamento de Fitotecnia, Centro de Ciências Agrárias, Universidade Federal do Piauí, em Teresina, PI, avaliando-se nove clones de cajueiro anão ("CAP 14", "Embrapa 09", "Embrapa 50", "Embrapa 51", "Embrapa 76", "Embrapa 183", "Embrapa 189", "FAGA 01", "FAGA 11") e um clone de cajueiro comum ("CCA"), utilizando-se quatro panículas por planta, cada uma com orientação norte, sul, leste e oeste. Os tipos varietais, cajueiro comum e anão precoce, apresentam pouca variação para a maioria dos caracteres avaliados. A proporção entre flores hermafroditas e o total de flores, em cajueiro comum, pode levá-lo a uma maior produção de frutos por panícula do que nos clones de cajueiro anão precoce analisados. O número de frutos desenvolvidos é bastante reduzido nos dois tipos varietais. As panículas situadas em diferentes orientações cardeais são semelhantes em todos os clones estudados quanto aos aspectos relacionados à biologia floral do cajueiro.

The common cashew presents a larger proportion between the number of hermaphrodite flowers and the total number of flowers than the early dwarf cashew clones; this may facilitate a larger production of fruits in the common cashew. The number of developed fruits is very low in both varietal types. The paniculae placed in different cardinal orientations are similar in all the clones in relation to the aspects related to the floral biology of the cashew.
