RESUMO

BALB/C mice were treated with epsilon aminocaproic acid (EACA), 0.1 to 3.2 mg/g body weight, subcutaneously every 24 hours for six days, starting on the second day after intraperitoneal transplantation of L5178Y lymphoma. Complete involution was observed in 20 to 90 per cent of the ascitic tumor and in 50 per cent of the solid tumor 72 hours after EACA was discontinued, tumoral involution and dosage were directly proportional. One hundred per cent of the non treated controls died 10 +/- 0.6 days after intraperitoneal lymphoma transplantation. During tumor involution malignant cells underwent autolysis and were attacked or phagocytized by activated macrophages. Since EACA is a proteolytic enzyme inhibitor, inhibition during treatment and activation of these enzymes after EACA was discontinued could have been the cause of autolysis of the L5178Y lymphoma cells.

Lymphoma L5178Y cells were treated with neuraminidase of Vibrio cholerae, potassium iodine, dithiotreitol (DTT), mercaptoethanol, glutaraldehyde, iodoacetamide, merthiolate, sodium periodate, urea, papaine, trypsine and EDTA, to increase immunoreaction in tumor cells. Mice were immunized with modified tumor cells every week for one month. Thereafter non modified tumor cells were transplanted to previously immunized mice. Only the immunization with neuraminidase-treated cells rejected the tumor. Although the immunization with cells treated with potassium iodine, DTT and mercaptoethanol did not reject tumor, prolonged significantly span of life. The other reactives had neither effect on tumor rejection nor on span of life.

In special designed vials using coverslides glued to segments of polyethilene test tubes of 0.15 ml of fibrin free blood were incubated during 45 min to 37 degrees C in a humid chamber. Blood samples were obtained from patients with disseminated Lupus erythematosus (DLE) and normal subjects (N). Adhered polymorpho-nuclear cells (PNC) to glass were washed with Hank's solution; immediately 0.15 ml of DLE or N serum containing 5X10(6) lymphocytes (L) were added to culture cells and incubated at 37 degrees C during 30 min. Lymphocytes were previously incubated at 37 degrees C during 30 min with either N serum or DLE serum. Thereafter the segment of polyethilene test tube was detached from coverslides and cells attached to glass was washed with Hank's solution and stained with Wright solution. PMN of DLE and N in presence of L of DLE and N incubated with fresh serum of DLE formed 5 to 15% of LE cells. Determining factor for LE cells formation is the serum of DLE. Slides contained only PMN and LE cells which make easy the observation of results. All possible combinations with PMN and DLE serum and N serum allowed inclusion of several negative or positive control groups.

Since 1975, the authors have been working in immunochemotherapy in acute myeloblastic leukemic adult patients. Autologous leukemic leucocytoblasts have been cultured with Vibrio cholerae neuraminidase (NVC). Thereafter, neoplastic cells were radiated with cobalt bomb and frozen to--70degreesC and afterwards 5 X 10(6) of these leucoblasts were intradermally injected to each patients every eight days. All patients received between six to ten injections. So far, ten such patients have been followed during eight months and since two months after immunization no leukemic leucocytoblasts in bone marrow and blood have been detected. Hemoglobin and platelets have increased more rapidly than usual and no patient has developed side effects. Phagocytosis tests carried out in vitro during remission, have demonstrated that only in immunized patients neutrophils and monocytes were capable of phagocytosis of autologous cultured malignant leucocytoblasts. The nucleus of phagocytized malignant cells was small and basophylic. These so far undescribed phagocytic cells were named post-immunization cells.

RESUMO

The presence of virus particles and their possible mechanism of excretion was studied in cells of murine lymphoma L 5178Y grown intraperitoneally. Malignant cells obtained from 10 mice bearing this lymphoma were studied with the electron microscope. In 96% of the cells 1 to 30 viroid particles (VP) type C (VP-C) clumped or isolated, averaging six per cell were observed. Isolated VP-C were commonly seen within endoplasmic reticulum or budding off from cell surface. Such a finding was regarded as the mechanism of excretion. However, with the present findings it is not possible to draw any conclusions upon the oncogenic nature of the virus particles described herein.

Cervical smears and serum samples were obtained from 28 ABO compatible patients with carcinoma of the cervix (CaCx) in O to IV clinical stages. Serum IgG antibodies against autologous CaCx cells were detected by immunofluorescence reactions in 24/24 patients with carcinoma of the cervix in I to IV clinical stages. These antibodies also reacted against alogeneic CaCx cells 53/54. Negative reactions were obtained n 4/4 cases of carcinoma of the cervix in stage O, and in control group, 10 normal persons and in 33 neoplastic sera from patients with other malignancies. Hence, detection of serum antibodies against CaCx cells could be an additional tool for CaCx detection.

Phagocytosis was evaluated in nine patients with splenctomy undergoing renal transplant, eight renal transplanted patients without splenectomy, three immunodepressed patients with terminal renal failure, four patients with splenectomy and six healthy volunteers. For phagocytosis evaluation, neutrophils adhered to glass were used. They were cover with 4% latex and albumin containing culture medium with or without phagocytosis stimulating tetrapeptide extracted from IgG in each case. Phagocytosis was decreased in all groups studied. Immunodepressed patients showed the highest diminishment of phagocytosis and patients with splenectomy and immunodepression the lesser. It was speculated that such a finding is due to a decrease of tetrapeptide in serum.
