RESUMO

The ventral tegmental area (VTA) is a nodal link in reward circuitry. Based on its striatal output, it has been subdivided in a caudomedial part which targets the ventromedial striatum, and a lateral part which targets the ventrolateral striatum [Ikemoto S (2007) Dopamine reward circuitry: two projection systems from the ventral midbrain to the nucleus accumbens-olfactory tubercle complex. Brain Res Rev 56:27-78]. Whether these two VTA parts are interconnected and to what extent the VTA innervates the substantia nigra compacta (SNc) and retrorubral nucleus (RR) are critical issues for understanding information processing in the basal ganglia. Here, VTA projections to the VTA-nigral complex were examined in rats, using Phaseolus vulgaris leucoagglutinin (PHA-L) as anterograde tracer. The results show that the dorsolateral VTA projects to itself, as well as to the dorsal tier of the SNc and RR, largely avoiding the caudomedial VTA. The ventrolateral VTA innervates mainly the interfascicular nucleus. The components of the caudomedial VTA (the interfascicular, paranigral and caudal linear nuclei) are connected with each other. In addition, the caudomedial VTA (especially the paranigral and caudal linear nuclei) innervates the lateral VTA, and, to a lesser degree, the SNc and RR. The caudal pole of the VTA sends robust, bilateral projections to virtually all the VTA-nigral complex, which terminate in the dorsal and ventral tiers. Modest inputs from the medial supramammillary nucleus to ventromedial parts of the VTA-nigral complex were also identified. In double-immunostained sections, PHA-L-labeled varicosities were sometimes found apposed to tyrosine hydroxylase-positive neurons in the ventral mesencephalon. Overall, the results underscore that VTA projections to the VTA-nigral complex are substantial and topically organized. In general, these projections, like the spiralated striato-nigro-striatal loops, display a medial-to-lateral organization. This anatomical arrangement conceivably permits the ventromedial striatum to influence the activity of the lateral striatum. The caudal pole of the VTA appears to be a critical site for a global recruitment of the mesotelencephalic system.

A set of guidelines are presented for the definition of monitoring plans in coastal and transitional (estuarine and lagoonal) systems subject to the European Union Water Framework Directive - WFD (2000/60/EC). General principles of best practice in monitoring are outlined, including (a) the definition of three types of broad management objectives: water quality, conservation, and human use, to which the general public may easily relate. These will define the core and research indicators (WFD quality elements) to be used for monitoring; (b) priorities and optimisation in a (financially and logistically) resource-constrained environment; (c) quality assurance; and (d) assessment of monitoring success: this should focus on the outputs, i.e. the internal audit of the monitoring activity, and on the outcomes. The latter component assesses programme effectiveness, i.e. environmental success based on a set of clearly-defined targets, and informs management action. The second part of this work discusses the approach and actions to be carried out for implementing WFD surveillance, operational and investigative monitoring. Appropriate spatial and temporal scales for surveillance monitoring of different indicators are suggested, and operational monitoring is classified into either screening or verification procedures, with an emphasis on the relationship between drivers, pressure, state and response. WFD investigative monitoring is interpreted as applied research, and thus guidelines cannot be prescriptive, except insofar as to provide examples of currently acceptable approaches. Specific case studies are presented for both operational (coastal eutrophication control) and investigative monitoring (harmful algal blooms), in order to illustrate the practical application of these monitoring guidelines. A spatially dynamic model for the productivity of spores and adults of green macroalgae (Enteromorpha sp.) was developed for a mesotidal estuary (Mondego estuary, Portugal). Many of the algal processes and parameters included in the model were experimentally obtained. Model predictions were compared to a real time series (1993-1997) of macroalgal biomass variation and the two sets show a good agreement (ANOVA, P<0.001). Results suggest that algal growth is highly sensitive to small changes in depth and exhibits different patterns of variation in different seasons. On a yearly basis, global calculations for the south channel of the estuary (137 ha) suggest that during bloom years, macroalgal biomass may reach about 21,205 ton DW compared to 240 ton DW in regular years. On a seasonal basis, the difference may be even more significant. The consequences of such variations on the nitrogen and phosphorus loading of the system and the adjacent coastal area are discussed.

The ventral tegmental area (VTA) is crucially involved in brain reward, motivated behaviors, and drug addiction. This district is functionally heterogeneous, and studying the connections of its different parts may contribute to clarify the structural basis of intra-VTA functional specializations. Here, the efferents of the rostral linear nucleus (RLi), a midline VTA component, were traced in rats with the Phaseolus vulgaris leucoagglutinin (PHA-L) technique. The results show that the RLi heavily innervates the olfactory tubercle (mainly the polymorph layer) and the ventrolateral part of the ventral pallidum, but largely avoids the accumbens. The RLi also sends substantial projections to the magnocellular preoptic nucleus, lateral hypothalamus, central division of the mediodorsal thalamic nucleus, lateral part of the lateral habenula and supraoculomotor region, and light projections to the prefrontal cortex, basolateral amygdala, and dorsal raphe nucleus. A similar set of projections was observed after injections in rostromedial VTA districts adjacent to RLi, but these districts also send major outputs to the lateral ventral striatum. Overall, the data suggest that the RLi is a distinct VTA component in that it projects primarily to pallidal regions of the olfactory tubercle and to their diencephalic targets, the central division of the mediodorsal thalamic nucleus and the lateral part of the lateral habenula. Because the rat RLi reportedly contains a lower density of dopaminergic neurons as compared with most of the VTA, its unusual projections may reflect a non-dopaminergic, putative GABAergic, phenotype, and this distinctive cell population seemingly extends beyond RLi boundaries into the laterally adjacent VTA. By being connected to the central division of the mediodorsal thalamic nucleus (directly and via ventral striatopallidal system) and to the magnocellular preoptic nucleus, the RLi and its surroundings may play a role in olfactory-guided behaviors, which are part of the approach responses associated with appetitive motivational states.

ACTH is known to act through the activation of cAMP/PKA in adrenocortical cells, but it has been suggested that it could also act via other pathways such as the ERK 1/2 cascade. To determine the effects of ACTH administration at sequential time points on the activation of ERKs 1/2, groups of rats (n = 6/group) were subjected to i.p. injections of either ACTH (Synacthen Depot-0.2 mg/Kg), or saline (Ct). The animals were sacrificed and the adrenal glands collected at different timings after ACTH injection (2 h, 18 h and 24 h). Two additional groups were injected daily until sacrifice (3 days and 15 days). Blood was collected for analysis and the adrenals were used for immunohistochemistry or Western Blot (WB) analysis. Immunoreactivity was scored by counting the mean number of zonae fasciculata (ZF) and reticularis (ZR) positive cells/section (mean +/- SEM). Adrenal weight was increased by ACTH in comparison with Ct. Corticosterone levels, as expected, were higher in ACTH treated animals than in Ct. The number of pERK positive cells increased in a time-dependent manner until 3d, and declined although not significantly in the 15 days animals (Control--48.13 +/- 9.0; ACTH 2 h--125.93 +/- 14.5; ACTH 18 h-139.46 +/- 10.0; ACTH 24 h--185.28 +/- 13.3; ACTH 3 days--198.47 +/- 18.6; ACTH 15 days--158.58 +/- 15.1). Comparable results were obtained with WB analysis. Our data shows that ACTH induces the activation of the MAPK/ERKs 1/2 cascade, especially in the ZF, consistent with this zone being more responsive to ACTH.

A dynamic model for prediction of bioaccumulation in aquatic macrofauna is described. The model, entitled SHRIMP (Simulation of Heavy-metal Rate of Intake for Macrofaunal sPecies), consists of five coupled submodels, which simulate individual growth, population dynamics, metal transport in the abiotic part of the system, metal bioaccumulation at the organism level, and upscaling of individual contamination to the population scale. The sublethal effects of metal toxicity on individual and population levels are shown to act through the reduction of individual growth rates. The model was tested for cadmium and mercury using epibenthic crustacea from an estuarine system. Individual and population patterns of bioaccumulation were analyzed by comparing the simulated results of five different scenarios of dissolved metal concentrations. Model results suggest that the subtle effect of growth reduction due to metal toxicity is sufficient to cause a decrease on population numbers simply by affecting the growth of the mature females and males.

RESUMO

Acute adrenal haemorrhage (AAH) is a rare disorder with different aetiologies. Aiming to discuss this condition, this report deals with four different cases that will be analysed and examined below, each one of them confirmed by biopsy or surgery and followed clinically and radiologically. In these cases it was found that the patients suffered from localized abdominal pain (4/4) and fever (2/4); one patient had adrenal insufficiency due to bilateral massive AAH. Therefore we concluded that AAH is an uncommon condition with variable clinical manifestations.
