RESUMO

Adult stem cells (SCs) represent the regenerative capacity of organisms throughout their lifespan. The maintenance of robust SC populations capable of renewing organs and physiological systems is one hallmark of healthy aging. The local environment of SCs, referred to as the niche, includes the nutritional milieu, which is essential to maintain the quantity and quality of SCs available for renewal and regeneration. There is increased recognition that SCs have unique metabolism and conditional nutrient needs compared to fully differentiated cells. However, the contribution of SC nutrition to overall human nutritional requirements is an understudied and underappreciated area of investigation. Nutrient needs vary across the lifespan and are modified by many factors including individual health, disease, physiological states including pregnancy, age, sex, and during recovery from injury. Although current nutrition guidance is generally derived for apparently healthy populations and to prevent nutritional deficiency diseases, there are increased efforts to establish nutrient-based and food-based recommendations based on reducing chronic disease. Understanding the dynamics of SC nutritional needs throughout the life span, including the role of nutrition in extending biological age by blunting biological systems decay, is fundamental to establishing food and nutrient guidance for chronic disease reduction and health maintenance. This review summarizes a 3-day symposium of the Marabou Foundation (www.marabousymposium.org) held to examine the metabolic properties and unique nutritional needs of adult SCs and their role in healthy aging and age-related chronic disease.

Knee meniscus is a hydrated tissue; it is a fibrocartilage of the knee joint composed primarily of water. We present results of interferometric surface monitoring by which we measure physical properties of human knee meniscal cartilage. The physical response of biological tissue to a short laser pulse is primarily thermomechanical. When the pulse is shorter than characteristic times (thermal diffusion time and acoustic relaxation time) stresses build and propagate as acoustic waves in the tissue. The tissue responds to the laser-induced stress by thermoelastic expansion. Solving the thermoelastic wave equation numerically predicts the correct laser-induced expansion. By comparing theory with experimental data, we can obtain the longitudinal speed of sound, the effective optical penetration depth and the Grüneisen coefficient. This study yields information about the laser tissue interaction and determines properties of the meniscus samples that could be used as diagnostic parameters.

The potential ecotoxicity of fluorescent dyes used in tracing, and their possible effects on human health, were evaluated by reviewing available toxicological information for 12 dyes - fluorescein, Lissamine Flavine FF, Rhodamine WT, Rhodamine B, Sulpho Rhodamine G, Sulpho Rhodamine B, eosin, pyranine, Phorwite BBH Pure, Tinopal 5BM GX, Tinopal CBS-X, and Diphenyl Brilliant Flavine 7GFF - and a dye-intermediate, amino G acid. This evaluation used available toxicological information, test data on analogous substances, and mathematical expressions for biological activity. Based on set criteria for human health and acute ecotoxicity, the evaluation indicated that these tracers have low to moderate levels of concern. The use of these tracers for the study of groundwater flow is appropriate if consideration is given to the overall human health and environmental effects. Their use in the environment requires tracer concentrations not exceeding 1-2 mg 1(-1) persisting for a period in excess of 24 h in the groundwater at the point of groundwater withdrawal or discharge. A simple calculated potential dose was used in a comparison of the estimated acute toxicity of Rhodamine WT in rats to the known acute oral toxic dose in humans for several known acutely toxic chemicals. This comparison showed that none of the fluorescent dyes evaluated would present an acutely toxic threat at or substantially above the recommended 2 mg 1(-1) concentration.

To calculate the diffuse reflectance from a semi-infinite slab of tissue, we introduce a probability distribution function, f(n)(g), that a photon will escape from the tissue after n scattering events. This approach permits the separation of the phase dependence of scattering, described by the anisotropy coefficient, g, from the absorption, micro(alpha), and scattering, micro(s), coefficients in the calculation of diffuse reflectance. We demonstrate that f(n)(g) and g are related to each other through a universal probability function. The analytical form of this probability function is explored and used to obtain the diffuse reflectance from tissue. The diffuse reflectance calculated with this method is in excellent agreement with Monte Carlo simulations over the parameter range typically found in human tissue, even for the values in which diffusion theory is a poor approximation.
