BACKGROUND: Recent studies have hypothesized that circulation of classical swine fever virus (CSFV) variants when the immunity induced by the vaccine is not sterilizing might favour viral persistence. Likewise, in addition to congenital viral persistence, CSFV has also been proven to generate postnatal viral persistence. Under experimental conditions, postnatal persistently infected pigs were unable to elicit a specific immune response to a CSFV live attenuated vaccine via the mechanism known as superinfection exclusion (SIE). Here, we study whether subclinical forms of classical swine fever (CSF) may be present in a conventional farm in an endemic country and evaluate vaccine efficacy under these types of infections in field conditions. RESULTS: Six litters born from CSF-vaccinated gilts were randomly chosen from a commercial Cuban farm at 33 days of age (weaning). At this time, the piglets were vaccinated with a lapinized live attenuated CSFV C-strain vaccine. Virological and immunological analyses were performed before and after vaccination. The piglets were clinically healthy at weaning; however, 82% were viraemic, and the rectal swabs in most of the remaining 18% were positive. Only five piglets from one litter showed a specific antibody response. The tonsils and rectal swabs of five sows were CSFV positive, and only one of the sows showed an antibody response. After vaccination, 98% of the piglets were unable to clear the virus and to seroconvert, and some of the piglets showed polyarthritis and wasting after 36 days post vaccination. The CSFV E2 glycoprotein sequences recovered from one pig per litter were the same. The amino acid positions 72(R), 20(L) and 195(N) of E2 were identified in silico as positions associated with adaptive advantage. CONCLUSIONS: Circulation of chronic and persistent CSF infections was demonstrated in field conditions under a vaccination programme. Persistent infection was predominant. Here, we provide evidence that, in field conditions, subclinical infections are not detected by clinical diagnosis and, despite being infected with CSFV, the animals are vaccinated, rather than diagnosed and eliminated. These animals are refractory to vaccination, likely due to the SIE phenomenon. Improvement of vaccination strategies and diagnosis of subclinical forms of CSF is imperative for CSF eradication.

Cuba is a country in transition with a considerable potential for economic growth. Soils are recipients and integrators of chemical pollution, a frequent negative side effect of increasing industrial activities. Therefore, we established a soil monitoring network to monitor polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) in soils of Mayabeque, a Cuban province southeast of Havana. Concentrations of the sum of the 16 US EPA PAHs and of the seven IRMM PCBs in soils from 39 locations ranged from 20 to 106 µg kg-1 and from 1.1 to 7.6 µg kg-1, respectively. While such concentrations can be considered as low overall, they were in several cases correlated with the distance of sampling sites to presumed major emission sources, with some of the concomitantly investigated source diagnostic PAH ratios, and with black carbon content. The presented data adds to the limited information on soil pollution in the Caribbean region and serves as a reference time point before the onset of a possible further industrial development in Cuba. It also forms the basis to set up and adapt national environmental standards.

Although avian influenza (AI) virus of H5 and H7 subtypes has the potential to mutate to a highly pathogenic form and cause very high mortalities in some poultry species, most AI infections in poultry are due to low pathogenic AI (LPAI). Hence serological surveys, coupled with passive surveillance activities, are essential to detect sub-clinical infections by LPAI viruses, H5 and H7 subtypes. However the proper planning of an active surveillance system should be based on a careful estimation of its performance. Therefore, the sensitivity of the active surveillance system for AI in the western region of Cuba was assessed by a stochastic model quantifying the probability of revealing at least one animal infected by H5 or H7 subtype. The diagnostic sensitivity of the haemagglutination inhibition assay and different levels of within-flock prevalence (5%, 12% and 30%) were considered. The sensitivity of the surveillance system was then assessed under five different samples size scenarios: testing 20, 30, 40, 50 or 60 animals in each flock. Poultry flock sites in the western region of Cuba with a size ranging from 10,000 to 335,000 birds were included in the study.

Classical swine fever virus produces a huge mortality in infected herds during recurrent outbreaks, predominantly in tropical and subtropical areas. In this scenario, it is common that cold-chain related issues affect the efficacy of virus attenuated-derived vaccines, which are frequently used in eradication programs. In the present work, the stability and protective capacity of a recombinant vaccine preparation, based on goat milk derived E2 glycoprotein extracellular domain, were both analyzed after incubation at 4 degrees C or 37 degrees C for 1 week. Differences in the viscosity and in the homodimeric form of the antigen were observed after comparing physicochemical properties of stressed and not stressed vaccine formulations. However, these differences did not affect the immunogenicity and protective capacity of such preparations. Noticeably, pigs immunized with the E2-based vaccine subjected to thermal stress became totally protected from the viral infection, after a challenge with 10(5) PLD(50) of a high virulent classical swine fever strain. This result supports the practical value of this vaccine preparation mostly for those regions in which cold-chain related failures tend to affect the protective capability of conventional virus attenuated vaccines.
