The present study aims to investigate if Cimicifuga racemosa (L.) Nutt extract (CIMI) reduces deleterious effects of dexamethasone (DEXA) in ovaries cultured in vitro. Mouse ovaries were collected and cultured in DMEM+ only or supplemented with 5 ng/mL of CIMI, or 4 ng/mL DEXA, or both CIMI and DEXA. The ovaries were cultured at 37.5°C in 5% CO2 for 6 days. Ovarian morphology, follicular ultrastructure, and the levels of mRNA for Bax, Bcl-2, and Caspase-3 were evaluated. The results showed that DEXA reduced the percentage of morphologically normal follicles, while CIMI prevented the deleterious effects caused by DEXA. In addition, DEXA negatively affected the stromal cellular density, while CIMI prevented these adverse effects. Ovaries cultured with DEXA and CIMI showed similar levels of mRNA for Bax, Bcl-2, and Caspase-3 compared to those cultured in control medium, while ovaries cultured with DEXA had increased expression of the above genes. Additionally, the ultrastructure of the ovaries cultured with CIMI was well preserved. Thus, the extract of CIMI was able to prevent the deleterious effects caused by DEXA on cultured mouse ovaries.

In the drying experiment, the Melanoxylon brauna seeds were submitted to two drying conditions: a forced air circulation chamber (40.18 °C ± 0.13 and 28.48% ± 3.95 RH) and a silica gel desiccator (27.19 °C ± 1.28 and 26.19% ± 0.94 RH) for different times (0, 12, 24, 36, 72, and 144 hours). A completely randomized design in a 2 (drying methods) × 5 (drying times) factorial scheme plus control and 4 replications of 25 seeds was used. The following variables were evaluated before and after drying: seed moisture content, percentage of germinated seeds, germination speed index, percentage of mortality, normal and abnormal seedlings. In the storage experiment the seeds were divided into two batches: pre-dried (at 5.0% humidity) and without drying (control at 8.9% humidity). The seeds were then stored in plastic bags in three environments: refrigerator at 5 °C, freezer at 20 °C and room temperature (29 °C). The seeds were removed every four months and submitted to the humidity and germination test for 24 months. Data from this storage experiment were analyzed considering a randomized block design in a 2 (drying levels: presence and absence) × 3 (storage environments: refrigerator, freezer or room temperature) factorial scheme + 2 controls (with and without drying at baseline) and 4 repetitions of 25 seeds. Drying reduced initial seed water content from 8.9% to 5.0%, without loss of viability. Drying in the chamber at 40 °C was faster and more efficient than in silica gel. The results enable classifying the seeds of this species as orthodox, i.e. tolerant to desiccation. O objetivo deste trabalho foi avaliar a sensibilidade das sementes da leguminosa arbórea Melanoxylon brauna Schott à dessecação e ao armazenamento. No experimento de secagem as sementes de braúna foram submetidas a duas condições de secagem: câmara com circulação forçada de ar (40,18 °C ± 0,13 e 28,48% ± 3,95 UR) e dessecador com sílica gel (27,19 °C ± 1,28 e 26,19% ± 0,94 UR), por diferentes tempos (0, 12, 24, 36, 72, 144 horas). Foi utilizado delineamento experimental inteiramente casualizado, em esquema fatorial 2 (método de secagem) × 5 (tempo de secagem), mais a testemunha, com 4 repetições de 25 sementes. A cada quatro meses as sementes foram retiradas e submetidas ao teste de umidade e de germinação durante 24 meses. Os resultados foram avaliados por meio do delineamento em blocos casualizados, com 4 repetições de 25 sementes, em esquema fatorial 2 (secagem) × 3 (ambiente de armazenamento) + 2 testemunhas. A secagem proporcionou a redução do teor de água inicial das sementes de 8,9% até 5,0%, sem perda da sua viabilidade. A secagem na câmara a 40 °C foi mais rápida e eficiente do que na sílica gel. Os resultados permitem classificar as sementes desta espécie como ortodoxas, ou seja, tolerantes à dessecação. A geladeira e o freezer foram eficientes para o armazenamento das sementes de braúna, até 24 meses, independente da secagem prévia das sementes, enquanto o armazenamento das sementes a temperatura ambiente é mais duradouro quando as sementes são submetidas previamente à secagem, podendo durar até 16 meses com secagem ou 12 meses sem secagem.(AU)

BACKGROUND: Even though mental disorders represent a major public health problem for women and respective children, there remains a lack of epidemiological longitudinal studies to assess the psychological status of women throughout pregnancy and later in life. This epidemiological cohort study assessed the relationship between mental disorders of 409 Brazilian women in pregnancy and 5-8 years after delivery. METHODS: The women were followed from 1997 to 2000 at 17 health services, and subsequently from 2004 to 2006 at their homes. Mental disorders were investigated by the Perceived Stress Scale-PSS, General Health Questionnaire-GHQ and State-Trait Anxiety Inventories-STAI. The relationship between scores of the PSS, GHQ and STAI 5-8 years after delivery and in pregnancy was assessed by multivariate linear regression analysis, controlling for the following confounders: maternal age, education, per capita income, family size, work, marital status and body mass index. RESULTS: Scores of the PSS, GHQ and STAI 5-8 years after delivery were positively associated with scores of the PSS, GHQ and STAI in the three trimesters of pregnancy, and inversely associated with maternal age and per capita income (adj. R2 varied from 0.15 to 0.37). PSS, GHQ and STAI scores in the 3rd trimester of pregnancy were positively associated with scores of the PSS, GHQ and STAI in the 1st and 2nd trimesters of pregnancy (adj. R2 varied from 0.31 to 0.65). CONCLUSIONS: The results of this study reinforce the urgency to integrate mental health screening into routine primary care for pregnant and postpartum women.

In this study, we described, for the first time, specific aspects of an anti-Leishmania immune response in a Brazilian Xakriabá indigenous community. Induction of an intracellular NO pathway, triggered by the binding of IgE to CD23 receptor in IFN-γ/IL-4 cytokines environment, was evaluated in localized cutaneous leishmaniasis (LCL) carriers and positive Montenegro skin test (MST) individuals without skin lesion (MT(+) SL(-)). Our data demonstrated that the higher frequency of CD23(+) CD14(+) monocytes and the increased serum levels of IgE observed in the LCL group were even higher in LCL carriers with late lesions (LCL≥60). Furthermore, patients with LCL presented increased NO production after Leishmania (Viannia) braziliensis stimulation and this NO profile was independent of the time of the lesion (recent LCL<60 or late LCL≥60). We also showed that the increased frequency of IFN-γ(+) and IL-4(+) CD4(+) T cells is related to the MT(+) SL(-) group. The results of biomarker signature curves demonstrated that in the MT(+) SL(-) group, the index signature was characterized by DAF-2T(+) CD14(+)/IL-4(+) CD8(+)/IFN-γ(+) CD4(+)/IL-4(+) CD4(+). On the other hand, the LCL group presented a higher index of DAF-2T(+) CD14(+)/CD23(+) CD14(+)/IL-4(+) CD8(+), associated with a lower index of IFN-γ(+) CD8(+). Considering the time of lesion, data analysis demonstrated that the main differences observed were highlighted in LCL<60 patients, with a higher index of CD23(+) CD14(+), which was also present in LCL≥60 patients. In conclusion, our data suggest that the protective immune response involving CD23-IgE-mediated NO release is a hallmark of patients with LCL. However, in MT(+) SL(-) individuals, another different leishmanicidal mechanism seems to be involved.

The manufacturing of boards and roof tiles is one of the routes to reuse waste from the recycled-carton-packaging process. Such a process requires knowledge of the hygroscopic behaviour of these carton-packaging waste mixtures in order to guarantee the quality of the final product (e.g. boards and roof tiles). Thus, with four carton-packaging waste mixtures of selected compositions (A, B, C and D), the sorption isotherms were obtained at air temperature of 20, 40 and 60 degrees C by using the static method. This permits one to investigate which model can relate the equilibrium moisture content of the mixture with that of a pure component through the mass fraction of each component in the mixtures. The results show that the experimental data can be well described by the weighted harmonic mean model. This suggests that the mean equilibrium moisture content of the carton-packaging mixture presents a non-linear relationship with each single, pure compound.

OBJECTIVE: The aim of this study was to assess bone changes and mobility in temporomandibular joints (TMJs) using cone beam CT (CBCT) in a population sample in Recife, PE, Brazil. METHODS: The TMJ images of patients treated by a radiologist at a private dental radiology service over a period of 1 year were retrieved from the computer database and assessed using a computer with a 21-inch monitor and the iCAT Cone Beam 3D Dental Imaging System Workstation program (Imaging Sciences International, Hatfield, PA). The Pearson χ(2) test was used to analyse the differences in percentage of bone changes among the categories of mobility (p ≤ 0.05). The McNemar test was used to compare the presence of bone changes in TMJs on the right and left sides (p ≤ 0.05). RESULTS: An adjusted logistic regression model was used to assess the effect of age and gender on the occurrence of bone changes (p ≤ 0.05). Bone changes were present in 227 (71%) patients. Age group and gender showed a statistically significant association with presence of bone changes (p ≤ 0.05). There was no significant difference between the right and left sides (p = 0.556) and in condylar mobility (p = 0.925) with regard to the presence of degenerative bone changes. CONCLUSIONS: There is a high prevalence of degenerative bone alteration in TMJs, which is more frequent in women and mostly located in the condyle. The prevalence of degenerative bone changes increases with age. There is no correlation between condylar mobility and the presence of degenerative bony changes in TMJs.

Essential oils of Origanum majorana L. (marjoram), Illicium verum Hook. f. (star-anise) and Cinnamomum zeylanicum Blume (cinnamon) were obtained by steam distillation using a modified Clevenger device. The antimicrobial activity of each oil was evaluated against the bacteria Staphylococcus aureus, Escherichia coli and the fungi Aspergillus flavus and Aspergillus parasiticus by observing their growth and/or mycelial inhibition through comparison with the standard dish (without oil). The essential oils were analyzed using a gas chromatograph coupled to a mass spectrometer for identification and coupled to a flame ionization detector for quantification. The major constituents of marjoram, star-anise and cinnamon essential oils were 4-terpineol, trans-anetole and cinnamic aldehyde, respectively. In in vitro tests, essential oils of marjoram and cinnamon promoted an inhibitory effect on the bacteria S. aureus and E. coli, while the essential oil of star-anise presented activity only against E. coli. Marjoram, star-anise and cinnamon oils were effective against the studied fungi, presenting an inhibitory effect. The minimal inhibitory concentration for the mycelial growth of A. parasiticus was 1 and 0.01 µL mL-1 for star-anise and cinnamon oils, respectively. The minimal inhibitory concentration for A. parasiticus was 0.25, 2 and 2 µL mL-1 for cinnamon, star-anise and marjoram oils, respectively.


Óleos essenciais de Origanum majorana L. (manjerona), Illicium verum Hook. f. (anis estrelado) e Cinnamomum zeylanicum Blume (canela) foram obtidos pela técnica de arraste a vapor d'água com aparelho de Clevenger modificado. Foram avaliadas as atividades antimicrobianas de cada um sobre as bactérias Staphylococcus aureus, Escherichia coli e para os fungos Aspergillus flavus e Aspergillus parasiticus, observando o crescimento e/ou inibição micelial, comparando-se estes com a placa-padrão (sem óleo). Os óleos essenciais foram analisados em cromatógrafo gasoso acoplado a espectrômetro de massa para a identificação e cromatógrafo gasoso com detector de ionização de chamas para a quantificação dos compostos. Os principais constituintes dos óleos essenciais de manjerona, anis-estrelado e canela foram o 4-terpineol, trans-anetol e aldeído cinâmico, respectivamente. Nos testes in vitro, os óleos essenciais de manjerona e canela promoveram efeito inibitório sobre as bactérias S. aureus e E. coli, enquanto o óleo essencial de anis estrelado apresentou atividade apenas frente E. coli. Os óleos de manjerona, anis estrelado e canela foram efetivos sobre os fungos estudados, apresentando efeito inibitório. A concentração mínima inibitória pra o crescimento micelial de A. parasiticus foi de 1 e 0,01 µL mL-1 para os óleos de anis-estrelado e canela, respectivamente. Enquanto a concentração mínima inibitória para A. parasiticus foi de 0,25; 2 e 2 µL mL-1 para os óleos de canela, anis-estrelado e manjerona, respectivamente.

This study analyses the anti-proliferative effect of lemongrass essential oil and its main constituent (citral) on all 3 evolutive forms of Trypanosoma cruzi. Steam distillation was used to obtain lemongrass essential oil, with chemical composition determined by gas chromatography (GC) and GC coupled to mass spectrometry (GC-MS). The IC50/24 h (concentration that reduced the parasite population by 50%) of the oil and of citral upon T. cruzi was determined by cell counting in a Neubauer chamber, while morphological alterations were visualized by scanning and transmission electron microscopy. Treatment with the essential oil resulted in epimastigote growth inhibition with IC50=126.5 microg/ml, while the IC50 for trypomastigote lysis was 15.5 microg/ml. The IC50/48 h for the Association Index (% macrophage infection x number of amastigotes per cell) was 5.1 microg/ml, with a strong inhibition of intracellular amastigote proliferation. Ultrastructural analysis demonstrated cytoplasmic and nuclear extraction, while the plasma membrane remained morphologically preserved. Our data show that lemongrass essential oil is effective against T. cruzi trypomastigotes and amastigotes, and that its main component, citral, is responsible for the trypanocidal activity. These results indicate that essential oils can be promising anti-parasitic agents, opening perspectives to the discovery of more effective drugs of vegetal origin for treatment of parasitic diseases. However, additional cytotoxicity experiments on different cell lines and tests in a T. cruzi-mouse model are needed to support these data.

In this work, the moisture adsorption and desorption isotherms were determined with the aim of defining the range of moisture content for storage of sludge powder. Equilibrium moisture content provides the basis for information not only on how much water has been taken out of a system but also on how fast drying is taking place (drying rate). Once the drying process is accomplished, the main concern rests on the storage of the dried final product. Still, the equilibrium moisture content is valuable information in that it has a major effect on the product physical and chemical properties. The present work also addresses the problem of selecting the best fit for equilibrium moisture content of sludge powder out of six well-known correlations for moisture sorption isotherms of solids: Henderson, Henderson-Thompson, Chung-Pfost, Chen-Clayton, Modified Halsey and Oswin. The equilibrium moisture content was determined by the static method (saturated salt solutions), in which the atmosphere surrounding the product is in equilibrium with the product without mechanical movement of the air or the product. Experiments were carried out under isothermal conditions at 20 and 40 degrees C. By calculating the regression coefficient, the residuals and the bias measure of Box for the equilibrium moisture content, the study showed that the Oswin Model was the most suitable. The range enclosed within the adsorption isotherm at 40 degrees C and the desorption isotherm at 20 degrees C defines the moisture extremes for storage in most tropical areas of the world.

RESUMO

This case report presents a 51 year old female patient who had a train accident in 1999. She suffered bilateral trans-tibial and bilateral trans-radial amputations. In this paper, the evolution of the right transradial amputation where eventually a Krukenberg procedure was performed, is described as is its good functional outcome after rehabilitation treatment. After this first procedure the patient also asked for the Krukenberg procedure for her left arm.

RESUMO

There is a severe shortage of trained microbiologists in Brazil which is hampering the development and expansion of microbiology/biotechnology research, the development of the biotechnology industries and environmental control agencies. Even some medical areas are affected. The reason for this situation lies in the deficient teaching of microbiology, where undergraduate courses in the biological sciences contain little or no microbiology, and in the low number of professionals that are produced. There are a few post-graduates courses in microbiology but these have high costs and little capacity for expansion. There is a need for incentives for the establishment of more undergraduate university courses, more practical training in specific areas, courses for retraining or updating of professionals already at work and specific programmes by the funding agencies.

To investigate the effect of substrates during oral rehydration therapy, we studied intestinal cation cotransport (ICC) with glutamine (Gln) alanine (Ala) and glucose (Glu). The specific aims were to determine the biological effects of these three different cotransport systems on intestinal function. Isolated rabbit ileal mucosa preparations mounted in Ussing chambers were studied. ICC was determined by measuring short-circuit current (Isc) and potential difference (PD) while monitoring toissue resistance (TR). The data are reported as the mean ñ SEM of 4-6 experiments for each amino acid concentration. Increasing concentrations of Gln (10-5 to 10-2 M), Ala (10-5 to 10-1 M) and Glu (10-5 to 10-2 M) caused a significant (P<0.05) increase in ICC. Glin (30 mM) and Ala (0.1 M) had a maximal effects (Em (Glin)=100% anmd Em (Ala)=66%, P<0.05) which was higher than that obtained with 30 mM Glu (Em (Glu)=35%). When sodium was replaced with choline on the mucosal side. Ringer solution completeley abolished the response with Gln, Ala and Glu. The presence of all three substrates (10-2 M gln, 10-1 M Ala and 10-2 M Glu) in Ringer solution on the mucosal side caused a significant increase in ICC ( increase of short circuit current = III ñ 43 uA, P<0.05). These results demonstrate that Glin, Ala and Glu each increased sodium-dependent cation cotransport, and that sodium-dependent intestinal cation cotransport was higher with Gln than with Ala or Glu

Hyperbolic and allosteric mathematical equations are used extensively to describe cumulative dose-response curves in pharmacology. In 1954, using an axiom described by Clark, Ariëns suggested the following equation for cumulative dose-response curves: Em/E-1 = Kd/[D], where Em = maximum effect, E = effect, Kd = dissociation constant of the drug-receptor complex and [D] = drug concentration. To evaluate the validity of the modified Ariëns equation on the dose-response curves with a Hill coefficient statistically different from one, we used sodium intestinal cotransport (SIC) with glutamine (Gln) and glucose (Glu). Increasing doses (10 microM to 0.1 M) of Gln and Glu were added to the mucosal side of the isolated rabbit ileum mucosal membrane mounted on Ussing chambers. Sodium cotransport was recorded by measuring short-circuit current (Isc), potential difference (PD) and tissue resistance (TR) at 20-min intervals for 2 h. The Em and ED50 for SIC with glucose were 25% and 150 microM, respectively, using the method proposed by Lineweaver-Burk. The same method could not be used to estimate the Em and ED50 in SIC with glutamine, suggesting a specific limitation of the Gln cotransport kinetic curve in the equation. Therefore, we modified the Ariëns equation to Em/E-1 = (Kd/[D])p. With p = Log (S), Log (Kd) = -pD2 and Log[D] = -pDx, we obtain E = Em/(1 + S(pD x -pD2)), where S is a hyperbolic constant.(ABSTRACT TRUNCATED AT 250 WORDS)

Malnutrition and dehydration are the immediated consequences of diarrheal diseases. To investigate the biological significance ofglutamine, alanine and glucose in the intestinal mucosa, we have used Ussing chambers to determine electrolyte transport by measuring short-circuit current (Isc), potential difference (PD) and tissue resistance (TR) in rabbit intestinal mucosa. Increasing doses (10-5 M to 10-1 M)of glutamine,alanine and glucose cause a significant increase in intestinal cation cotransport. Although glucose had a slightly earlier effect, 30mM glutamine and 0.1 M alanine had a maximal effect which was more than two times that caused by 30 nM glucose. The pD2 values for glucose, glutamine and alanine were 3.0, 2.5, and 2.0, respectively. The dose-response curves of these substrates suggest that the intestinal cotransport kinetics for glutamine is differentfrom that of glucose and alanine. Our results demonstrated that all three substrates cause a significant increase in Isc or PD, suggesting an increase in the intestinal mucosa cation cotransport. Glutamine has a larger effect on cation cotransport than alanine and glucose. These combinations should be studied further for the development of an oral rehydrating solution for diarrhea treatment which could prevent the resulting malnutrition, especially in those cases of prolonged diarrheal diseases

A 68 year-old white male patient with previous diagnosis of pulmonary emphysema was submitted to acupuncture. The needles were inserted into the precordial area and the patient immediately complained of worsening dyspnea. Four days later pneumothorax was detected by chest X-rays. A thoracic tube was inserted with total lung expansion.
