A new Continuous Flow Analysis (CFA) system coupled with Fast Liquid Chromatography - tandem Mass Spectrometry (FLC-MS/MS) has been recently developed for determining organic markers in ice cores. In this work we present an upgrade of this innovative technique, optimized for the detection of levoglucosan in ice cores, a crucial tracer for reconstructing past fires. The upgrade involved a specific optimization of the chromatographic and mass spectrometric parameters, allowing for a higher sampling resolution (down to 1 cm) and the simultaneous collection of discrete samples, for off-line analysis of water stable isotopes and additional chemical markers. The robustness and repeatability of the method has been tested by the analysis of multiple sticks of ice cut from the same shallow alpine ice core, and running the system for several hours on different days. The results show similar and comparable trends between the ice sticks. With this upgraded system, a higher sensitivity and a lower limit of detection (LOD) was achieved compared to discrete analysis of alpine samples for levoglucosan measurements. The new LOD was as low as 66 ng L-1, a net improvement over the previous LOD of 600 ng L-1.

North-Eastern Italy and in particular Veneto Region, stands out as a major centre of agriculture and viticulture which has rapidly expanded in the last decade with high productivity indexes. In this context, assessing atmospheric pollution caused by crop spraying with pesticides in rural areas and their transport to high-altitude remote sites is crucial to provide a basis for understanding possible impacts on the environment and population health. We aim to improve existing methods with a highly sensitive technique by using high pressure anion exchange chromatography coupled to a triple quadrupole mass spectrometer. Thus, a total of fourteen polar pesticides were determined in aerosol samples collected from August to December 2021 at Roncade (Venetian plain) and Col Margherita Observatory (Dolomites). The observatory was chosen as the background site as it is representative of the surrounding alpine region. Some samples revealed a substantial amount of cyanuric acid mainly at Roncade (mean concentration of 10 ± 10 ng m-3), glyphosate and fosetyl-aluminium (0.1 ± 0.2 and 0.1 ± 0.1 ng m-3, respectively). Surprisingly, some pesticides have been also found at Col Margherita, a high mountain background site, with concentrations an order of magnitude lower than at Roncade. This is the first time that fourteen polar pesticides have been assessed in the aerosol phase of the Po' Valley and detected at a high-altitude remote site, and consequently this study provides the first data on their occurrences in Italian aerosols. It represents a basis for the assessment of risks for humans.

The development of new analytical systems and the improvement of the existing ones to obtain high-resolution measurements of chemical markers in samples from ice cores, is one of the main challenges the paleoclimatic scientific community is facing. Different chemical species can be used as markers for tracking emission sources or specific environmental processes. Although some markers, such as methane sulfonic acid (a proxy of marine productivity), are commonly used, there is a lack of data on other organic tracers in ice cores, making their continuous analysis analytically challenging. Here, we present an innovative combination of fast liquid chromatography coupled with tandem mass spectrometry (FLC-MS/MS) to continuously determine organic markers in ice cores. After specific optimization, this approach was applied to the quantification of vanillic and syringic acids, two specific markers for biomass burning. Using the validated method, detection limits of 3.6 and 4.6 pg mL-1 for vanillic and syringic acids, respectively, were achieved. Thanks to the coupling of FLC-MS/MS with the continuous flow analytical system, we obtained one measurement every 30 s, which corresponds to a sampling resolution of a sample every 1.5 cm with a melting rate of 3.0 cm min-1. To check the robustness of the method, we analyzed two parallel sticks of an alpine ice core over more than 5 h. Vanillic acid was found with concentrations in the range of picograms per milliliter, suggesting the combustion of coniferous trees, which are found throughout the Italian Alps.

A recent outbreak of anthrax disease, severely affecting reindeer herds in Siberia, has been reportedly associated to the presence of infected carcasses or spores released from the active layer over permafrost, which is thawing and thickening at increasing rates, thus underlying the re-emerging nature of this pathogen in the Arctic region because of warming temperatures. Anthrax is a global zoonotic and epizootic disease, with a high case-fatality ratio in infected animals. Its transmission is mediated by environmental contamination through highly resistant spores which can persist in the soil for several decades. Here we develop and analyze a new epidemiological model for anthrax transmission that is specifically tailored to the Arctic environmental conditions. The model describes transmission dynamics including also herding practices (e.g. seasonal grazing) and the role of the active layer over permafrost acting as a long-term storage of spores that could be viable for disease transmission during thawing periods. Model dynamics are investigated through linear stability analysis, Floquet theory for periodically forced systems, and a series of simulations with realistic forcings. Results show how the temporal variability of grazing and active layer thawing may influence the dynamics of anthrax disease and, specifically, favor sustained pathogen transmission. Particularly warm years, favoring deep active layers, are shown to be associated with an increase risk of anthrax outbreaks, and may also foster infections in the following years. Our results enable preliminary insights into measures (e.g. changes in herding practice) that may be adopted to decrease the risk of infection and lay the basis to possibly establish optimal procedures for preventing transmission; furthermore, they elicit the need of further investigations and observation campaigns focused on anthrax dynamics in the Arctic environment.

This study explores the factors affecting the biodiversity of diatoms, vegetation with focus on bryophytes, and invertebrates with focus on water mites, in a series of 16 spring-habitats. The springs are located primarily from the mountainous part of the Emilia-Romagna Region (Northern Apennines, Italy), and two pool-springs from agricultural and industrial lowland locations. Overall, data indicate that biological diversity (Shannon-Wiener, α-diversity) within individual springs was relatively low, e.g.: Sdiatoms = 0-46, Swater-mites = 0-11. However, when examined at the regional scale, they hosted a very high total number of taxa (γ-diversity; Sdiatoms = 285, Swater-mites = 40), including several new or putatively-new species, and many Red-List taxa. This pattern suggested there is high species turnover among springs, as well as high distinctiveness of individual spring systems. A key goal was to assess the hydrogeological and hydrochemical conditions associated with this high regional-pool species richness, and to provide a guide to future conservation strategies. There was a striking variety of geological conditions (geodiversity, captured mainly with lithotype and aquifer structure) across the study region, which led to wide variation in the hydrosphere, especially in conductivity and pH. Agriculture and industrial activities (anthroposphere) in the lowlands resulted in nutrient enrichment and other forms of pollution. Across all 16 spring-systems, several hydrogeological conditions most strongly influenced the presence or absence of particular biota and were determinants of species importance: spring-head morphology, hydroperiod, discharge, current velocity, and elemental concentration. These findings have important practical consequences for conservation strategies. Our data show that it is imperative to protect entire regional groups of springs, including representatives of the different ecomorphological spring types, lithologies, and degrees of human influence. These findings suggest that springs, when studied from an ecohydrogeological perspective, are excellent systems in which to further investigate and understand geo-biodiversity relationships.

The present study aims to reconstruct vegetation development, climate changes and human impact using an ombrotrophic peat core from the Coltrondo bog in the eastern Italian Alps. Evidence from pollen, micro-charcoal, major and trace elements, and lead isotopes from this 7,900 years old peat deposit has been combined, and several climatic oscillations and phases of human impact detected. In particular, human presence was recorded in this area of the Alps from about 650 cal bc, with periods of increased activity at the end of the Middle Ages and also at the end of the 19th century, as evidenced by both human-related pollen and the increase in micro-charcoal particles. The enrichment factor of lead (EFPb) increased since the Roman period and the Middle Ages, suggesting mainly mining activities, whereas the advent of industrialization in the 20th century is marked by the highest EFPb values in the whole core. The EFPb data are strongly supported by the 206Pb/207Pb values and these are in general agreement with the historical information available. Therefore, the multi-proxy approach used here has allowed detection of climatic events and human impact patterns in the Comelico area starting from the Iron Age, giving new insights into the palaeoecology as well as the course of the interaction among humans, climate and ecosystems in this part of the eastern Italian Alps.

BACKGROUND: A critical aspect regarding the global dispersion of pathogenic microorganisms is associated with atmospheric movement of soil particles. Especially, desert dust storms can transport alien microorganisms over continental scales and can deposit them in sensitive sink habitats. In winter 2014, the largest ever recorded Saharan dust event in Italy was efficiently deposited on the Dolomite Alps and was sealed between dust-free snow. This provided us the unique opportunity to overcome difficulties in separating dust associated from "domestic" microbes and thus, to determine with high precision microorganisms transported exclusively by desert dust. RESULTS: Our metagenomic analysis revealed that sandstorms can move not only fractions but rather large parts of entire microbial communities far away from their area of origin and that this microbiota contains several of the most stress-resistant organisms on Earth, including highly destructive fungal and bacterial pathogens. In particular, we provide first evidence that winter-occurring dust depositions can favor a rapid microbial contamination of sensitive sink habitats after snowmelt. CONCLUSIONS: Airborne microbial depositions accompanying extreme meteorological events represent a realistic threat for ecosystem and public health. Therefore, monitoring the spread and persistence of storm-travelling alien microbes is a priority while considering future trajectories of climatic anomalies as well as anthropogenically driven changes in land use in the source regions.

Reconstructing the past variability of Arctic sea ice provides an essential context for recent multi-year sea ice decline, although few quantitative reconstructions cover the Holocene period prior to the earliest historical records 1,200 years ago. Photochemical recycling of bromine is observed over first-year, or seasonal, sea ice in so-called "bromine explosions" and we employ a 1-D chemistry transport model to quantify processes of bromine enrichment over first-year sea ice and depositional transport over multi-year sea ice and land ice. We report bromine enrichment in the Northwest Greenland Eemian NEEM ice core since the end of the Eemian interglacial 120,000 years ago, finding the maximum extension of first-year sea ice occurred approximately 9,000 years ago during the Holocene climate optimum, when Greenland temperatures were 2 to 3 °C above present values. First-year sea ice extent was lowest during the glacial stadials suggesting complete coverage of the Arctic Ocean by multi-year sea ice. These findings demonstrate a clear relationship between temperature and first-year sea ice extent in the Arctic and suggest multi-year sea ice will continue to decline as polar amplification drives Arctic temperatures beyond the 2 °C global average warming target of the recent COP21 Paris climate agreement.

To provide a reliable tool for investigating diffusion processes of the specific components of the human odor 3-hydroxy-3-methylhexanoic acid and 3-methyl-3-sulfanylhexan-1-ol through the snowpack, we developed and optimized an analytical method based on direct immersion solid-phase microextraction followed by gas chromatography with mass spectrometry. Direct immersion solid-phase microextraction was performed using polyacrylate fibers placed in aqueous solutions containing 3-hydroxy-3-methylhexanoic acid and 3-methyl-3-sulfanylhexan-1-ol. After optimization, absorption times of 120 min provided a good balance to shorten the analysis time and to obtain suitable amounts of extractable analytes. The extraction efficiency was improved by increasing the ionic strength of the solution. Although the absolute extraction efficiency ranged between 10 and 12% for 3-hydroxy-3-methylhexanoic acid and 2-3% for 3-methyl-3-sulfanylhexan-1-ol, this method was suitable for analyzing 3-hydroxy-3-methylhexanoic acid and 3-methyl-3-sulfanylhexan-1-ol concentrations of at least 0.04 and 0.20 ng/mL, respectively. The precision of the direct immersion solid-phase microextraction method ranged between 8 and 16%. The variability within a batch of six fibers was 10-18%. The accuracy of the method provided values of 88-95 and 86-101% for 3-hydroxy-3-methylhexanoic acid and 3-methyl-3-sulfanylhexan-1-ol, respectively. The limit of detection (and quantification) was 0.01 ng/mL (0.04 ng/mL) for 3-hydroxy-3-methylhexanoic acid and 0.06 ng/mL (0.20 ng/mL) for 3-methyl-3-sulfanylhexan-1-ol. The signal versus concentration was linear for both compounds (r(2) = 0.973-0.979). The stability of these two compounds showed that 3-hydroxy-3-methylhexanoic acid was more stable in water than 3-methyl-3-sulfanylhexan-1-ol. We applied the method to environmental samples in correspondence with an olfactory target buried previously.

Ombrotrophic peatlands are remarkable repositories of high-quality climatic signals because their only source of nutrients is precipitation. Although several analytical techniques are available for analysing inorganic components in peat samples, they generally provide only low-resolution data sets. Here we present a new analytical approach for producing high-resolution data on main and trace elements from ombrotrophic peat cores. Analyses were carried out on a 7-m-long peat core collected from Danta di Cadore, North-Eastern Italy (46° 34' 16″ N, 12° 29' 58″ E). Ca, Ti, Cr, Fe, Cu, Zn, Ga, Sr, Y, Cd, Ba and Pb were detected at a resolution of 2.5 mm with a non-destructive X-ray fluorescence core scanner (XRF-CS). Calibration and quantification of the XRF-CS intensities was obtained using collision reaction cell inductively coupled plasma quadruple mass spectrometry (CRC-ICP-QMS). CRC-ICP-QMS measurements were carried out on discrete samples at a resolution of 1 cm, after dissolution of 150-mg aliquots with 9 ml HNO3 and 1 ml HF at 220 °C in a microwave system. We compare qualitative XRF-CS and quantitative CRC-ICP-MS data and, however the several sources of variability of the data, develop a robust statistical approach to determine the R (2) and the coefficient of a simple regression model together with confidence intervals. Perfect positive correlations were estimated for Cd, Cr, Pb, Sr, Ti and Zn; high positive correlations for Ba (0.8954), Y (0.7378), Fe (0.7349) and Cu (0.7028); while moderate positive correlations for Ga (0.5951) and Ca (0.5435). With our results, we demonstrate that XRF scanning techniques can be used, together with other well-established geochemical techniques (such as ICP-MS), to produce high-resolution (up to 2.5 mm) quantitative data from ombrotrophic peat bog cores.

In the presence of different environmental stressors, diatoms can produce frustules presenting different types of deformities. Metals and trace elements are among the most common causes of these teratological forms. Metal enrichment in water bodies can be attributed to the geological setting of the area or to pollution. The widespread benthic diatom Achnanthidium minutissimum (ADMI) is one of the most metal-tolerant species. In the present study, ADMI teratologies were defined from samples taken from eight very diverse, widely-distributed inland-water habitats: streams affected by active and abandoned mining areas, a metal-contaminated stream, a spring in an old chalcopyrite mine, a mineral-water fountain, and a sediment core taken from a lake affected by metal contamination in the past. Deformed frustules of ADMI were characterised mainly by one (sometimes two) more or less bent off ending, conferring to the specimens a cymbelloid outline (cymbelliclinum-like teratology, CLT). Marked teratologies were distinguished from slight deformities. Hydrochemical analyses, including metals and trace elements, were carried out and enrichment factors (EF) relative to average crustal composition were calculated. To improve our knowledge on the potential of different metals and trace elements to trigger the occurrence of ADMI CLT, we carefully selected 15 springs out of 110 (CRENODAT dataset) where both ADMI and above-average metal or metalloid concentrations occurred, and re-analysed these samples. The results from the eight widely-distributed core sites as well as from the 15 selected CRENODAT springs led to the hypothesis that two metals (copper and zinc) and a metalloid (antimony) were the most likely triggers of ADMI CLT formation. From a quantitative point of view, it is worth noting that the lowest concentrations triggering ADMI CLT can be fairly low, particularly in the case of copper contamination. The antimony-rich site was characterised by a marked-teratology variant where both ends of ADMI were bent off.

Biomass burning is one of the most important sources of organic matter in the atmosphere as it affects the absorption and scattering of solar radiation, creates cloud condensation nuclei and possibly influences ice and snow albedo. Here we created and validated an analytical method using HPLC/(-)-ESI-MS/MS to determine phenolic compounds (PCLCs): vanillic acid, isovanillic acid, homovanillic acid, syringic acid, syringaldehyde, ferulic acid, p-coumaric acid, and coniferyl aldehyde at trace levels in particulate matter. We analyzed eighteen high-volume air samples from Ny Ålesund (Svalbard) collected during the boreal spring and summer of 2010. Biomass burning molecules including PCLCs (<0.49 µm, mean atmospheric concentration 6 pg m(-3)), levoglucosan (0.004 to 0.682 ng m(-3)) and acrylamide (32 fg m(-3) to 166 fg m(-3)) were present in the sampled aerosols. Levoglucosan concentrations, an unambiguous cellulose combustion tracer, derived from 2010 Russian fires. PCLCs levels in the Ny Alesund atmosphere in different size fractions reflected both long-range transport linked to biomass burning and a terrigenous local source.

The occurrence and temporal variation of 18 perfluoroalkyl substances (PFASs) and 8 polybrominated diphenyl ethers (PBDEs) in the European Alps was investigated in a 10 m shallow firn core from Colle Gnifetti in the Monte Rosa Massif (4455 m above sea level). The firn core encompasses the years 1997-2007. Firn core sections were analyzed by liquid chromatography-tandem mass spectrometry (PFASs) and gas chromatography-mass spectrometry (PBDEs). We detected 12 PFASs and 8 PBDEs in the firn samples. Perfluorobutanoic acid (PFBA; 0.3-1.8 ng L(-1)) and perfluorooctanoic acid (PFOA; 0.2-0.6 ng L(-1)) were the major PFASs while BDE 99 (

Iodine and bromine species participate in key atmospheric reactions including the formation of cloud condensation nuclei and ozone depletion. We present a novel method coupling a high-performance liquid chromatography with ion chromatography and inductively coupled plasma mass spectrometry, which allows the determination of iodine (I) and bromine (Br) species (IO(3)(-), I(-), Br(-), BrO(3)(-) ) at the picogram-per-gram levels presents in Antarctic ice. Chromatographic separation was achieved using an IONPAC® AS16 Analytical Column with NaOH as eluent. Detection limits for I and Br species were 5 to 9 pg g(-1) with an uncertainty of less than 2.5% for all considered species. Inorganic iodine and bromine species have been determined in Antarctic ice core samples, with concentrations close to the detection limits for iodine species, and approximately 150 pg g(-1) for Br(-). Although iodate (IO(3)(-)) is the most abundant iodine species in the atmosphere, only the much rarer iodide (I(-)) species was present in Antarctic Holocene ice. Bromine was found to be present in Antarctic ice as Br(-).

Inductively coupled plasma-mass spectrometry coupled with cation exchange matrix separation has been optimised for the direct determination of platinum group element (PGE) and trace element emissions from a diesel engine car. After matrix separation method detection limits of 1.6 ng g(-1) for Pd, 0.4 ng g(-1) for Rh and 4.3 ng g(-1) for Pt were achieved, the method was validated against the certified reference material BCR 723, urban road dust. The test vehicle was fitted with new and aged catalytic converters with and without diesel particulate filters (DPF). Samples were collected after three consecutive New European Driving Cycle (NEDC) of the particulate and "soluble" phases using a home-made sampler optimised for trace element analysis. Emission factors for the PGEs ranged from 0.021 ng km(-1) for Rh to 70.5 ng km(-1) for Pt; when a DPF was fitted, the emission factors for the PGEs actually used in the catalysts dropped by up to 97% (for Pt). Trace element emission factors were found to drop by a maximum of 92% for Ni to a minimum of 18% for Y when a DPF was fitted; a new DPF was also found to cause a reduction of up to 86% in the emission of particulate matter.

PAH concentrations have been determined in 47 seasonal snowpack samples collected in the Valbelluna valley and in the Bellunesi Dolomites National Park, in the Italian North-Eastern Alps, during the winter of 2005. The SigmaPAH concentration in high-altitude alpine sites (above 1700 m) was 32+/-20 ng/kg while in valley bottom urban areas it was 165+/-54 ng/kg with maximum values of 290 ng/kg. The GIS mapping technique was employed to produce a PAH spatial distribution. The urbanized Valbelluna valley, and in particular the SW part, had the highest accumulation of all PAH, with values an order of magnitude more than those in rural and alpine areas. This behaviour is consistent with urban air quality data, and is due to geo-morphological and meteorological factors such as the deeper shape of the valley at the position of the town of Feltre and the low altitude of the boundary layer during the winter season.

RESUMO

The occurrence of organic pollutants in European Alpine snow/ice has been reconstructed over the past three centuries using a new online extraction method for polycyclic aromatic hydrocarbons (PAH) followed by liquid chromatographic determination. The meltwater flow from a continuous ice core melting system was split into two aliquots, with one aliquot directed to an inductively coupled plasma quadrupole mass spectrometer for continuous trace elements determinations and the second introduced into a solid phase C18 (SPE) cartridge for semicontinuous PAH extraction. The depth resolution for PAH extractions ranged from 40 to 70 cm, and corresponds to 0.7-5 years per sample. The concentrations of 11 PAH were determined in dated snow/ice samples to reconstruct the atmospheric concentration of these compounds in Europe for the last 300 years. The PAH pattern is dominated by phenanthrene (Phe), fluoranthene (Fla), and pyrene (Pyr), which represent 60-80% of the total PAH mass. Before 1875 the sum of PAH concentration (SigmaPAH) was very low with total mean concentrations less than 2 ng/kg and 0.08 ng/kg for the heavier compounds (SigmaPAH*, more than four aromatic rings). During the first phase of the industrial revolution (1770-1830) the PAH deposition showed a weak increase which became much greater from the start of the second phase of the industrial revolution at the end of 19th Century. In the 1920s, economic recession in Europe decreased PAH emissions until the 1930s when they increased again and reached a maximum concentration of 32 ng/kg from 1945 to 1955. From 1955 to 1975 the PAH concentrations decreased significantly, reflecting improvements in emission controls especially from major point sources, while from 1975 to 2003 they rose to levels equivalent to those in 1910. The Fla/(Fla+Pyr) ratio is often used for source assignment and here indicates an increase in the relative contribution of gasoline and diesel combustion with respect to coal and wood burning from 1860 to the 1980s. This trend was reversed during the last two decades.
