RESUMO

In the monitoring process 110 representatives of different age groups held a comparative analysis of the efficacy and safety of several geroprotective techniques, including the use of dry carbon dioxide baths, hyperbaric oxygen therapy, therapeutic massage and receiving Oligopeptide preparations containing complexes lysyl-glutamyl-asparagin and glutamyl-asparagin-arginine (Vezugen and Pinealon). The most pronounced positive impact on indicators of biological age were detected during the combined use of these two Oligopeptide complexes. The most safe from the point of view of influence on a number of biochemical, immunological parameters, clinical condition of the patients were therapeutic massage and Oligopeptide drugs. The use of dry carbon dioxide baths and hyperbaric oxygenation with an undoubtedly positive influence on the indices of biological age has a number of limitations and contraindications regarding its security.

In the course of inspection 1136 persons of different age and sex data on dynamics of the main components of lipid profile, the concentration of brain-derived neurotrophic factor (BDNF) in blood plasma and indicators of levels of free cortisol, melatonin sulfate, metanephrine and normetanephrine in urine daily were obtained. Clear age and gender differences related both to the content of atherogenic fractions of lipids in the blood, and types of dyslipidemia were revealed. Fundamental differences in the age dynamics of indicators of stress-realizing systems in men and women and the correlation of these indicators with the level of low density lipoproteins and atherogenic coefficient were identified. The data obtained may indicate different mechanisms of development of atherosclerosis and its associated pathological aging in people of different sex and age, which enables the practical use of research results for the earliest diagnosis and prognosis of a number of associated with age and pathological conditions.

Among 330 patients with severe mental retardation who were consulted at random for medico-genetic purposes (excluding phenylketonuria and Down's disease), 27,3% constituted retarded patients due to genetic diseases, 10%--to exogenous factors. Families with a high genetical risk (25%-50% for sibs) constituted 20%. In the group of patients with severe mental retardation, but without signs of organic lesion in the neurological state or multiple congenital anomalies, there was a higher percentage of consanguineous parents and affected sibs, than in the group of patients with residual-organic neurological changes and in the group of patients with the so-called malformation retardation syndromes.

RESUMO

The results of modelling and study of persistent infection of pig embryo kidney cell cultures (PEK) with MF variant of tick-borne encephalitis (TBE) virus are presented. In the life of chronically infected cultures periods of formation and stabilization were observed which were characterized by different types of relationships between the persisting virus and the cells. By the characters under study the persisting virus did not differ significantly from the initial MF variant. Inoculation of PEK and BHK-21 cell monolayers with samples of DNA from chronically infected cultures permitted to isolate transfected agents. Their identification by the neutralization tests and the indirect immunofluorescence procedure confirmed that they belonged to TBE virus. It is suggested that the mechanism of TBE virus persistence in PEK-MF system is based on the integration of TBE virus genome with DNA of chronically infected PEK cells.

RESUMO

A persistent infection of the continuous embryonal pig kidney cell cultures induced by a rat parvovirus (the Kilhem virus) did not alter morphological or karyological characteristics of the cultures, and caused no transformation of these. The data obtained suggests the resistance of the pig karyotype to the virus under investigation.

RESUMO

From HEp-2 cells chronically infected with tick-borne encephalitis virus clones were obtained which produced no virus antigens and contained no sequences homologous to virus RNA. Expression of oncornavirus in the system under study was demonstrated. The possibility of transduction of cell genes by a defective virus strain was established. Preliminary experiments showed the possibility of integration of virus genome in tick egg DNA in transovarial transmission.

RESUMO

Duck embryo cell (DEC) cultures were used for the study of arbovirus reproduction. High sensitivity of DEC cultures to Japanese encephalitis, West Nile, Tyuleniy, Sakhalin and Baku viruses was established. DEC cultures are recommended for investigation of arbovirus reproduction.

RESUMO

Reproduction of human cytomegalovirus (CMV) and Japanese encephalitis virus (JE) was studied in organ cultures of human embryo intestine under conditions of mono- and combined infection. The titer of JE virus in monoinfection reached its maximum level (10(7) TCD50/ml) whereas the titer of CMV did not exceed 10(2) TCD50/ml. In combined infection the titer of CMV increased to 10(5.5) TCD50/ml and the titer of JE virus remained at the same level (10(7) TCD50/ml). The results obtained in light and scanning electron microscopy studies indicate that in monoinfection CMV causes considerably earlier and more profound degenerative changes in mesanchymal elements of embryonal intestinal tissues than does JE virus. At the same time the combined infection of organ cultures with these viruses is not cytocidal.

RESUMO

Variants of tick-borne encephalitis virus (TBA) isolated by inoculation of pig embryo kidney cell cultures (SPEV) with infectious DNA from chronically infected HEp-2-Soph cells produced cytopathic effect in SPEV cell cultures. In suckling mice inoculated intracerebrally they induced inapparent infection. Titers of TBE virus variants in SPEV cell cultures treated with DEAE-dextran increased considerably. As a result of filtration a variant was obtained replicating in SPEV cell cultures without any CPE. The cytopathic effect developed only when SPEV cells were treated with DEAE dextran. This variant is assumed to consist of two subpopulations 1 of which produces the cytopathic effect in DEAE-dextran-untreated SPEV cells while the other needs DEAE-dextran treatment of cells for its CPE manifestation.

Reproduction of Japanese encephalitis virus isolated at various passage levels from chronically infected GMS-1-K33 cell cultures was maximum in chick embryo fibroblast cultures at a temperature of 28 degrees C. A temperature of 42 degrees C was found to be nonpermissive for the majority of variants and restrictive for the 20th passage virus. Some decline in titers was also observed at 37 degrees C. The ts mutants derived from the population of the persisting virus had lower pathogenicity for laboratory animals, were sensitive to RNA-ase and resistant to 4 M urea. The data obtained by us and other authors suggest that appearance and selection of ts mutants in persistent infection in cell cultures in a regular phenomenon playing a significant role both in establishment and maintenance of persistently infected systems.

The brain cells of the suckling mice line MSB-1-K-33 chronically infected by an attenuated variant of the Japanese encephalitis virus and of cell clones isolated from the later, had mainly a neartetraploid keryotype (the modal class 70-71 chromosomes). In metaphases of cloned cell populations, an increase of number of the chromosomes was especially obvious in clones 3 and 4 (modal classes in both cases were 76-77 chromosomes). Cell population of clone 1 differed insignificantly from that of the parental line MSB-1-K-33 in respect of its cytogenetic characteristics. In metaphases of the line MSB-1-K-33 and clones 1, 3, and 4, a high frequency of chromosomal damages was observed. The most frequent type of structural chromosome aberrations were chromatid breaks. The karyotypes of clones 3 and 4 were characterized by presence of a large marker metacentric chromosome in 50-53% of the cells. The data obtained suggest a continuous effect of the Japanese encephalitis virus on the karyotype of cells in infected culture.

RESUMO

Japanese encephalitis virus contained in the medium of chronically infected VERO-K43 cell cultures was shown to be sensitive to the effect of RNA-ase, resistant to treatment with urea, to have no hemagglutinating activity. After 2 passages in suckling mouse brain the virus lost its sensitivity to RNA-ase, was inactivated by urea, and its hemagglutinating activity was restored. The importance of Japanese encephalitis virus variability in the mechanism of chronic infection of cell cultures is discussed.

RESUMO

The authors studied the clinical features in 19 patients with the mosaic variety of Down's disease and 63 patients with trisomia in relation to the 21st chromosome. It was demonstrated that mild forms of mental retardation are mainly seen in mosaicism rather than in the trisome variety, although severe forms of retardation may be seen in the first group as well. A full correlation between the severity of mental retardation and the % of the aneuploid cell content in the peripheral blood was not always observed.
