RESUMO

A dual color, frequency and pulse duration agile laser system, capable of delivering laser pulses in arbitrary temporal profiles with ∼1 ns to ∼1 µs pulse duration, chirping rates of ∼27 MHz/ns with an achievable chirping range of several GHz across the pulse duration, and with energies ranging from a few nJ to hundreds of mJ per pulse has been developed. The flexibility and capability of this laser system provide a wide range of laser parameters that can be exploited to optimize operational conditions in various experiments ranging from laser diagnostics to spectroscopy and optical manipulation of matter. The developed system is successfully demonstrated to obtain coherent Rayleigh-Brillouin scattering (CRBS) in both single and dual color configuration, in an effort to expand the non-intrusive accessibility towards lower pressure regime for neutral gas and plasma diagnostics.

RESUMO

We present a series of twelve patients, bearing a wide range of solid malignancies, who received either PD-L1 or a combination of PD-L1 and CTLA-4 inhibitors. Following immunotherapy administration, they exhibited the clinical signs indicative of renal toxicity, including increased serum creatinine levels, proteinuria, nephrotic syndrome and/or hematuria. All patients underwent renal biopsy. Results: All cases demonstrated some degree of interstitial inflammation and tubular injury, while in five patients, glomerular alterations consistent with a specific glomerulopathy were also observed: secondary "lupus-like" membranous glomerulopathy in two cases and membranoproliferative glomerulonephritis, IgA glomerulonephritis and secondary AA amyloidosis in each of the remaining three patients. The two patients with "lupus-like" nephritis and the one with amyloidosis experienced nephrotic syndrome, while their creatinine was within normal range. In the remaining nine cases, deterioration of renal function was the main manifestation. Conclusion: Our findings harmonize with bibliographical data that identify tubulointerstitial nephritis as the most frequent histological lesion related to ICIs administration. The preferential involvement of tubulointerstitial tissue could be associated with the reported higher expression levels of PD-L1 on tubular epithelial cells, compared to glomeruli. On the other hand, glomerular involvement is probably a consequence of a systemic immune system reconstruction, induced by immune-checkpoints inhibition.

RESUMO

We present a novel concept for a Thomson scattering diagnostic, based on a high-speed fiber optic spectrometer. The high-speed fiber optic spectrometer presented here translates a spectral measurement from the frequency domain into the time domain, thus requiring the use of only a single photodetector for spectral acquisition. The high temporal precision offered by the instrument gives rise to a number of advantages over traditional spectrometers, such as nearly background-free measurements and multiple uses of the same injected beam. Multiple uses of the same beam would enable greatly increased measurement rates, in the range of 10-100 MHz. The spectral range and resolution of the fiber spectrometer can be easily tailored to be optimized for the light source and experimental conditions by selecting different lengths of fiber, thus allowing for the proposed technique to exhibit high dynamic range when measuring many points simultaneously. Finally, due to the temporal separation of the background from the signal, these improvements are possible without the need for increased average input laser power.
