RESUMO

Caffeine is widely present in food and drinks, such as teas and coffees, being also part of some currently commercialized medicines, but despite its enhancement on several functions of human body, its exceeding use can promote many health problems. In order to develop new fast approaches for the caffeine sensing, graphite-epoxy composite electrodes (GECE) were used as substrate, being modified by different diazonium salts, synthetized as their tetraflouroborate salts. An analytical method for caffeine quantification was developed, using sware wave voltammetry (SWV) in Britton-Robinson buffer pH 2.0. Detection limits for bare electrode and 4-benzenesulfonic modified electrode were observed circa 145 µmol·L-1 and 1.3 µmol·L-1, respectively. The results have shown that the modification shifts the oxidation peaks to lower potential. Kinetics of the reaction limited by diffusion was more expressive when caffeine was added to the solution, resulting in decreases of impedance, characterized by lower R ct. All results for caffeine determination were compared to a reference chromatographic procedure (HPLC), showing no statistical difference. Analytical parameters for validation were suitably determined according to local legislation, leading to a linear behaviour from 5 to 150 µmol·L-1; precision of 4.09% was evaluated based on the RDC 166/17, and accuracy was evaluated in comparison with the reference method, with recovery of 98.37 ± 2.58%.

RESUMO

Forages are usually inoculated with homofermentative and facultative heterofermentative lactic acid bacteria (LAB) to enhance lactic acid fermentation of forages, but effects of such inoculants on silage quality and the performance of dairy cows are unclear. Therefore, we conducted a meta-analysis to examine the effects of LAB inoculation on silage quality and preservation and the performance of dairy cows. A second objective was to examine the factors affecting the response to silage inoculation with LAB. The studies that met the selection criteria included 130 articles that examined the effects of LAB inoculation on silage quality and 31 articles that investigated dairy cow performance responses. The magnitude of the effect (effect size) was evaluated using raw mean differences (RMD) between inoculated and uninoculated treatments. Heterogeneity was explored by meta-regression and subgroup analysis using forage type, LAB species, LAB application rate, and silo scale (laboratory or farm-scale) as covariates for the silage quality response and forage type, LAB species, diet type [total mixed ration (TMR) or non-TMR], and the level of milk yield of the control cows as covariates for the performance responses. Inoculation with LAB (≥105 cfu/g as fed) markedly increased silage fermentation and dry matter recovery in temperate and tropical grasses, alfalfa, and other legumes. However, inoculation did not improve the fermentation of corn, sorghum, or sugarcane silages. Inoculation with LAB reduced clostridia and mold growth, butyric acid production, and ammonia-nitrogen in all silages, but it had no effect on aerobic stability. Silage inoculation (≥105 cfu/g as fed) increased milk yield and the response had low heterogeneity. However, inoculation had no effect on diet digestibility and feed efficiency. Inoculation with LAB improved the fermentation of grass and legume silages and the performance of dairy cows but did not affect the fermentation of corn, sorghum, and sugar cane silages or the aerobic stability of any silage. Further research is needed to elucidate how silage inoculated with homofermentative and facultative heterofermentative LAB improves the performance of dairy cows.

RESUMO

PURPOSE: The relatively recent advent of polyphenol supplement for exercise studies has been tested in a variety of forms and doses. However, the dose-response on adipokines and oxidative stress biomarker effect remains unknown. The aim of the present study was to assess the effect of intense, long-duration (48-h) exercise, and a single dose of proanthocyanidin, on plasma leptin, adiponectin, and electronegative low-density lipoprotein (LDL(-)) concentrations. METHODS: Fifty-four healthy male army cadets (22 ± 2 years) participated in a double-blind, randomized, placebo-controlled study and were distributed between control (CG; n = 27) and supplemented groups (SG; n = 27). Immediately before the start of the exercise, both CG and SG groups received a capsule containing starch (200 mg) or proanthocyanidin (dry Vitis vinifera extract, 200 mg), respectively. Following a 12-h fasting period, the plasma adiponectin, leptin, and LDL(-) concentrations were measured prior to the start of the exercise after 24 and 48 h of military training, and after 24 h of rest. The effects of the proanthocyanidin (supplement), exercise (time), and their interaction were investigated using factorial two-way ANOVA. RESULTS: Plasma leptin concentration was only influenced by exercise (p = 0.001). Plasma adiponectin concentration was influenced by exercise (p = 0.037), and by the exercise x supplement interaction (p = 0.033). LDL(-) was influenced by the supplement (p = 0.001), exercise (p = 0.001), and their interaction (p = 0.001). CONCLUSIONS: A single dose of proanthocyanidin (200 mg) was able to reduce LDL(-) concentration and increase plasma adiponectin concentration after 24 h of rest in SG group, indicating its potential protective action.

RESUMO

We report herein a systematic investigation on the effect of the size of silver (Ag) nanoparticles employed as starting materials over the morphological features and catalytic performances of AgPt nanoshells produced by a combination of galvanic replacement between Ag and PtCl6(2-) and PtCl6(2-) reduction by hydroquinone. More specifically, we focused on Ag nanoparticles of four different sizes as starting materials, and found that the outer diameter, shell thickness, and the number of Pt surface atoms of the produced nanoshells increased with the size of the starting Ag nanoparticles. The produced AgPt nanoshells were supported into SiO2, and the catalytic performances of the AgPt/SiO2 nanocatalysts toward the gas-phase oxidation of benzene, toluene, and o-xylene (BTX oxidation) followed the order: AgPt 163 nm/SiO2 > AgPt 133 nm/SiO2 > AgPt 105 nm/SiO2 > AgPt 95 nm/SiO2. Interestingly, bigger AgPt nanoshell sizes lead to better catalytic performances in contrast to the intuitive prediction that particles having larger outer diameters tend to present poorer catalytic activities due to their lower surface to volume ratios as compared to smaller particles. This is in agreement with the H2 chemisorption results, and can be assigned to the increase in the Pt surface area with size due to the presence of smaller NPs islands at the surface of the nanoshells having larger outer diameters. This result indicates that, in addition to the overall diameters, the optimization of the surface morphology may play an important role over the optimization of catalytic activities in metal-based nanocatalysts, which can be even more pronounced that the size effect. Our data demonstrate that the control over surface morphology play a very important role relative to the effect of size to the optimization of catalytic performances in catalysts based on noble-metal nanostructures.

RESUMO

This study aimed to compare the biochemical markers of muscle and liver injury and total antioxidant capacity in army cadets after a traditional army physical training program and to correlate these effects with aerobic conditioning. Male army cadets (n = 87; age, 20 ± 2 years) were evaluated 12 hours before the start of training (T0), 12 hours after a 30-km march (T1), and 48 hours after military training (T2). Creatine kinase (CK), aspartate aminotransferase (AST), and alanine aminotransferase (ALT) serum levels were measured using an autoanalyzer. Total antioxidant capacity was determined by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (Sc%). Seventy-four percent of army cadets were classified as having excellent aerobic conditioning (53.9 ± 3.0 ml · kg(-) · min(-1) predicted VO2max from the Cooper test). The median serum concentration of all enzymes increased 12 hours after marching (T1: CK 675%; ALT 59%; AST 336%; AST/ALT ratio 85%; p = 0.001) and 48 hours after the end of training (T2: CK 878%; ALT 256%; AST 418%; AST/ALT ratio 180%; p = 0.001). Sc% was higher in T2 (31.1 ± 9.8%; p = 0.01) than in T0 (3.4% change). Maximal oxygen consumption (ml · kg(-1) · min(-1)) correlated negatively with CK (r = -0.25; p = 0.009) and ALT (r = -0.21; p = 0.03) serum levels, and positively with the change in Sc% (r = 0.22; p = 0.04) at T2. The results indicate that intense military training can cause liver and muscle injury and that aerobic conditioning can be considered as a protective factor for these injuries.

RESUMO

This paper presents, for the first time, the recycling and use of spent Li-ion battery cathode tape as a catalyst in the degradation of an organic dye. In our proposal, two major environmental problems can be solved: the secure disposal of cell phone batteries and the treatment of effluents with potentially toxic organic dyes. The spent Li-ion battery cathode investigated in this paper corresponds to 29% of the mass of Li-ion batteries and is made up of 83% LiCoO2, 14.5% C and less than 2.5% Al, Al2O3 and Co3O4. The use of spent Li-ion battery cathode tape increased the degradation velocity constant of methylene blue in the absence of light by about 200 times in relation to pure H2O2. This increase can be explained by a reduction in the activation energy from 83 kJ mol(-1) to 26 kJ mol(-1). The mechanism of degradation promoted by LiCoO2 is probably related to the generation of superoxide radical (O2(-)). The rupture of the aromatic rings of methylene blue was analyzed by ESI-MS.

RESUMO

A Brazilian field isolate (IBV/Brazil/PR05) of avian infectious bronchitis virus (IBV), associated with development of nephritis in chickens, was previously genotyped as IBV variant after S1 gene sequencing. The aim of this study was to evaluate the levels of IL-6 in kidneys and trachea of birds vaccinated and challenged with IBV/Brazil/PR05 strain, correlating these results with scores of microscopic lesions, specific IBV antigen detection and viral load. The up-regulation of IL-6 and the increased levels of viral load on renal and tracheal samples were significantly correlated with scores of microscopic lesions. Reduced levels of viral load were detected in kidneys of birds previously vaccinated and challenged, compared to non-vaccinated challenged group, although markedly microscopic lesions were observed for both groups. The expression of IL-6, present both in the kidney and in the tracheas, was dependent on the load of the virus present in the tissue, and the development of lesions was related with IL-6 present in the tissues. These data suggest that variant IBV/Brazil/PR05 can induce the expression of proinflammatory cytokines in a manner correlated with viral load and increased IL-6 is involved in the tissue with the influx of inflammatory cells and subsequent nephritis. This may contribute with a model to the development of immunosuppressive agents of IL-6 to prevent acute inflammatory processes against infection with IBV and perhaps other coronaviruses, as well as contribute to the understanding of the immunopathogenesis of IBV nephropatogenic strains.

Objective: To evaluate the influence of different luting protocols on the push-out bond strength of fiber posts cemented to bovine root dentin. Method: Forty bovine root canals (length: 16 mm) were prepared up to 12 mm using #3 (RTD) preparation burs. Each canal had its apical 4 mm embedded in acrylic resin and the roots were assigned to four groups (n=10), according to the cementation procedures: G1: etch-and-rinse multi-bottle adhesive system + #3 fiber post; G2: etch-and-rinse multibottle adhesive system + #1 fiber post; G3: etch-and-rinse multi-bottle adhesive system + #1 fiber post + accessory fiber posts; G4: #3 fiber post without etching and bonding procedures to dentin. Except for G4, root dentin was treated with the etch-and-rinse multi-bottle adhesive system All Bond 2 (Bisco). The adhesive system was applied with a microbrush (Dentsply) and excess material was removed withabsorbent paper points. The quartz fiber posts (Macro-Lock Illusion/RTD) were cemented to the root dentin with a dual-cure resin cement (Duolink/Bisco) and were stored in distilled water (for 24 h at 37 °C) before the mechanical tests. Each sample was cut into four slices (1.8 mm thick) that were subjected to push-out tests in a universaltesting machine (1 mm/min). The data (in MPa) were analyzed statistically by one-way ANOVA. Results: The mean push-out bond strength values (±SD) were: G1: 5.4 ± 1.3 MPa; G2: 4.2 ± 2.4 MPa; G3: 4.6 ± 1.5 MPa and G4: 3.3 ± 1.7 MPa. One-way ANOVA showed no statistically significant differences among the groups. The tested hypothesis was discarded. Conclusion: The push-out bond strength of fiber posts to root dentin was not influenced by the evaluated luting protocols...
