RESUMO

Knee braces are commonly used to support the knee joint and improve function. However, brace sliding caused by the misalignment between brace and knee during motion is a common problem, which reduces the therapeutic effect and leads to brace abandonment. To investigate the effect of mechanism combinations on sliding, an experimental brace was designed to isolate the mechanism as the sole variable. Ten healthy participants were recruited, each of whom worn four combinations of lateral/medial mechanisms: lateral and medial single-axis (SA), lateral super gear (SG) and medial non-circular gear (NCG), lateral four-bar linkage (FL) and medial SG, and lateral FL and medial NCG. The knee flexion angle was collected using inertial measurement units, and brace sliding was measured by 3D motion capture system. All combinations had significant changes in peak sliding of thigh and shank compared to the SA combination (p < 0.05), but lateral FL and medial NCG combination had the lowest peak and final sliding during walking and leg-raising, with significant reductions of 40.7 and 85.3% in peak sliding of thigh, and significant reductions of 56.3 and 72.0% in peak sliding of shank, respectively (p < 0.05). Moreover, the mechanism combination did not significantly impact the knee range of motion (p > 0.05). The mechanism combination that fit the instantaneous center of rotation of lateral/medial condyle of knee joint demonstrates a significant reduction in brace sliding. Additionally, the peak sliding during motion is significantly higher than the final sliding.

Ciliates in the order Pleurostomatida are found free-living in many habitats including within biofilms, but some (e.g. Pseudoamphileptus spp.) are ectocommensal on various hosts. Due to issues involving overall undersampling, the exact diversity and molecular phylogeny of this group remain largely underexplored. To combat this deficiency, detailed investigations were undertaken in northern China. As a result of these studies, we provide the morphological descriptions of two new species. Pseudoamphileptus apomacrostoma sp. nov., a new ectocommensal species, is characterized by the broadly oval cell shape, numerous scattered contractile vacuoles, and unique densely bounded extrusomes; Amphileptus qingdaoensis sp. nov., a marine form, is characterized by possessing oblong extrusomes with a conical anterior end, a single contractile vacuole and 5-7 left and 18-23 right kineties. In addition, a new population of Amphileptus orientalis Zhang et al., 2022, a freshwater representative, was documented and an improved diagnosis is provided. The phylogenetic analyses based on the SSU rDNA sequences imply that the genus Pseudoamphileptus is monophyletic whereas the genus Amphileptus is paraphyletic. The new molecular sequences presented here further support the establishment of two new species.

Background: Valgus braces are prescribed as a common conservative treatment option for patients with medial gonarthrosis to improve their quality of life. Many studies had reviewed the effects of the valgus braces on patients with medial gonarthrosis, while they mainly focused on the knee adduction moment (KAM), with less attention paid to other parameters such as spatiotemporal and morphological parameters. Objectives: The purpose of this study was to review the effects of valgus braces on the spatiotemporal, kinematic/kinetic, morphological, and muscle parameters. Methods: Based on the selected keywords, a survey of literatures was performed in Web of Science, PubMed, Scopus, and Google Scholar using the PRISMA methods, and the search period was established from January 2000 to March 2022. Results: Thirty-four articles were included. According to the conclusion of these articles, the valgus brace can be used to relieve the symptoms of patients with medial gonarthrosis by decreasing the varus angle, decreasing the KAM, and redistributing the knee compartment loads. However, the effects of valgus braces on other biomechanical parameters (e.g., walking speed, cadence, joint angle, and joint space) had not reached a consensus. Conclusions: The valgus knee brace can effectively relieve the symptoms of medial gonarthrosis through multiple mechanisms, while there is still some confusion about the effectiveness of the valgus brace on the other biomechanical parameters.

Ciliated protists represent a morphologically and genetically distinct group of single-celled eukaryotes which can reproduce asexually and sexually. Morphogenesis occurs in both asexual and sexual modes of reproduction which is of interest for researchers investigating cell differentiation, regeneration, systematics and evolution. However, studies of morphogenesis have concentrated almost entirely on the asexual mode. Here we use protargol staining to investigate the morphogenetic processes during sexual reproduction in the model species Euplotes vannus (Müller). The major events include: (1) two rounds of morphogenesis occur during sexual reproduction, i.e., conjugational and postconjugational reorganization; (2) in both processes the oral primordium is generated de novo in a pouch beneath the cortex; (3) the frontoventral-transverse cirri anlagen are formed de novo and fragment in a 3:3:3:3:2 pattern; (4) the leftmost cirrus and the paroral membrane do not change during conjugational morphogenesis, but reorganize de novo during postconjugational morphogenesis; (5) marginal cirral anlagen are formed de novo in both morphogenetic processes; (6) two or three caudal cirri are formed at the ends of the rightmost two or three old dorsal kineties; (7) the dorsal kineties are retained entirely. These results can serve as reference to investigate the morphogenetic events in the different stages of sexual reproduction.

Ciliates form a distinct group of single-celled eukaryotes that host two types of nuclei (micro and macronucleus) in the same cytoplasm and have a special sexual process known as conjugation, which involves mitosis, meiosis, fertilization, nuclear differentiation, and development. Due to their high species diversity, ciliates have evolved different patterns of nuclear events during conjugation. In the present study, we investigate these events in detail in the marine species Euplotes raikovi. Our results indicate that: (i) conjugation lasts for about 50 hours, the longest stage being the development of the new macronucleus (ca. 36 hours); (ii) there are three prezygotic micronuclear divisions (mitosis and meiosis I and II) and two postzygotic synkaryon divisions; and (iii) a fragment of the parental macronucleus fuses with the new developing macronucleus. In addition, we describe for the first time conjugation in amicronucleate E. raikovi cells. When two amicronucleate cells mate, they separate after about 4 hours without evident nuclear changes; when one amicronucleate cell mates with a micronucleate cell, the micronucleus undergoes regular prezygotic divisions to form migratory and stationary pronuclei, but the two pronuclei fuse in the same cell. In the amicronucleate cell, the parental macronucleus breaks into fragments, which are then recovered to form a new functional macronucleus. These results add new information on the process of conjugation in both micronucleate and amicronucleate Euplotes cells.
