RESUMO

Copper pyrithione (CuPT) is used as a co-biocide in new antifouling paints but its toxicity remains little known. To compare the toxicity of copper-based compounds, rainbow trout (Oncorhynchus mykiss) larvae were exposed for 8-day to CuPT and CuSO4 at equivalent copper concentrations. CuPT exposure led to the greatest accumulation of Cu in larvae. Exposure to 10 µg.L-1 CuPT induced 99% larval mortality but only 4% for CuSO4-exposed larvae. The larval development and growth were affected by CuPT (from 0.5 µg.L-1 Cu) but not by CuSO4. Lipid peroxidation was not induced by either contaminant. The expression of genes involved in oxidative stress defence, detoxification and copper transport was induced in larvae exposed to CuSO4 and CuPT but at higher concentrations for CuPT. This study highlights the marked toxicity of CuPT for early life stages of fish and raises the question of the possible environmental risks of this antifouling compound.

In fish, the gut microbiome plays a crucial role in homeostasis and health and is affected by several organic and inorganic environmental contaminants. Amphidromous fish are sentinel species, particularly exposed to these stressors. We used whole metagenome sequencing to characterize the gut microbiome of wild European eels (Anguilla anguilla) at a juvenile stage captured from three sites with contrasted pollution levels in term of heavy metals and persistent organic pollutants. The objectives were to identify what parameters could alter the gut microbiome of this catadromous fish and to explore the potential use of microbiota as bioindicators of environment quality. We identified a total of 1079 microbial genera. Overall, gut microbiome was dominated by Proteobacteria, Firmicutes and Actinobacteria. Alpha and beta diversity were different amongst sites and could be explained by a reduced number of environmental and biological factors, specifically the relative abundance of fish preys in eels' diet, PCB101, γHCH (lindane), transnonachlor and arsenic. Furthermore, we identified a series of indicator taxa with differential abundance between the three sites. Changes in the microbial communities in the gut caused by environmental pollutants were previously undocumented in European eels. Our results indicate that microbiota might represent another route by which pollutants affect the health of these aquatic sentinel organisms.

Behavioral parameters are increasingly considered sensitive and early bioindicators of toxicity in aquatic organisms. A video-tracking tool was specifically developed to monitor the swimming behaviour of D-larvae of the Mediterranean mussel, Mytilus galloprovincialis, in controlled laboratory conditions. Both maximum and average swimming speeds and trajectories were recorded. We then investigated the impact of copper and silver with or without a moderate rise of temperature on swimming behavior and acetylcholinesterase (AChE) activity of mussel D-larvae and the possible mechanistic link between both biological responses. Our results showed that copper and/or silver exposure, as well as temperature increase, disrupts the swimming behavior of mussel larvae which could compromise their dispersal and survival. In addition, the combined effect of temperature and metals significantly (p < 0.05) increased AChE activity in mussel larvae. Pearson's correlation analysis was performed and results showed that the AChE activity is positively correlated with maximum speeds (r = 0.71, p < 0.01). This study demonstrates the value of behavioral analyzes of aquatic invertebrates as a sensitive and integrate marker of the effects of stressors.

Since 2011, the Caribbean Islands have witnessed unprecedented massive stranding of a pelagic brown algal Sargassum spp. inducing damages for coastal ecosystems and economy. By accumulating heavy metals, Sargassum can play a role in contaminant transportation from offshore to the coast. In 2019, three genotypes of Sargassum (S. fluitans III, S. natans I, and VIII) were sampled in seven stations along a 3400 km transect in the Atlantic Ocean. Concentrations of 15 heavy metal(loid)s elements were analyzed by Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES). Mean metal concentrations were ranked following descending order: As >Fe > Al > Mn > Cd > Zn > Ni > V > Cu > Cr > Hg. The metalloid As was the most abundant contaminant with a maximum value of 115 ppm, previously observed in the Caribbean area (80-150 ppm). At Atlantic Ocean Basin-scale, metallic element concentrations do not present spatial longitudinal gradients. Genotypes S. fluitans III and S. natans (I and VIII), present differents metal(loid)s contamination distinct patterns.

Over the last decade, increasing proliferations of Atlantic Sargassum populations have led to massive beaching with disastrous environmental consequences. This study is a preliminary assessment of open ocean Sargassum spp. element concentration to assess their potential contribution on coastal ecosystems. Sargassum spp. samples from seven sites, collected along a transect from the center of the Atlantic Ocean to near the coast of Martinique (French West Indies), were analyzed to determine their potential metal and metalloid enrichment. Mean element concentrations from the Sargassum spp. samples were ranked in the following descending order: As > Fe > Mn > Al > Zn > V > Ni > Cu > Cr > Cd > Hg. Element concentrations are relatively low compared to previous results of beached Sargassum spp. except for As that need to be carefully considered before reusing Sargassum spp.

The aim of this study was to analyze the impact of three concentrations of a pesticide mixture on the first development stages of rainbow trout (Oncorhynchus mykiss). The mixture was made up of three commonly used pesticides in viticulture: glyphosate (GLY), chlorpyrifos (CPF) and copper sulfate (Cu). Eyed stage embryos were exposed for 3 weeks to three concentrations of the pesticide mixture. Lethal and sub-lethal effects were assessed through a number of phenotypic and molecular endpoints including survival, hatching delay, hatching success, biometry, swimming activity, DNA damage (Comet assay), lipid peroxidation (TBARS), protein carbonyl content and gene expression. Ten target genes involved in antioxidant defenses, DNA repair, mitochondrial metabolism and apoptosis were analyzed using real-time RT-qPCR. No significant increase of mortality, half-hatch, growth defects, TBARS and protein carbonyl contents were observed whatever the pesticide mixture concentration. In contrast, DNA damage and swimming activity were significantly more elevated at the highest pesticide mixture concentration. Gene transcription was up-regulated for genes involved in detoxification (gst and mt1), DNA repair (ogg1), mitochondrial metabolism (cox1 and 12S), and cholinergic system (ache). This study highlighted the induction of adaptive molecular and behavioral responses of rainbow trout larvae when exposed to environmentally realistic concentrations of a mixture of pesticides.

The increase in the global population, coupled with growing consumption of Rare Earth Elements (REEs), has led to increasing transfer of these emerging contaminants into the environment, particularly through the effluents from wastewater treatment plants (WWTP). The objectives of this study were to determine the geochemical quality of a French river subject to strong urban pressure (the Jalle River in the Bordeaux area) and to examine the bioavailability of natural and anthropogenic REEs in a model species of freshwater bivalve, the Asian clam Corbicula fluminea. To this end, two fractions (dissolved and total) of the water from the Jalle River were sampled and the bivalves were exposed by in situ caging during a three-month monitoring period. The REE patterns obtained showed the presence of Gadolinium (Gd) anomalies in the dissolved and total fractions as well as in Corbicula fluminea. The apparent bioavailability of natural REEs was in the following order for the dissolved fraction: Medium REEs (MREEs) > Light REEs (LREEs) > Heavy REEs (HREEs) and for the particulate fraction: MREEs > LREEs = HREEs. These results highlight the importance of the particulate fraction in the study of the bioavailability of REEs in bivalves. An increase of anthropogenic Gd (Gdanth) was observed in the dissolved fraction between the upstream site (3.4 ng.L-1) and the WWTP Downstream site (48.4 ng.L-1). The Gd anomaly observed in the water was also observed in Corbicula fluminea with a significant increase in the bioaccumulation of Gdanth, from 1.5 ± 1 ng.gDW-1 upstream to 4.1 ± 0.7 ng.gDW-1 downstream of the WWTP effluents, thus confirming the enhanced bioavailability of medical-origin Gd to freshwater bivalves. This study strongly suggests that Corbicula fluminea can be used as a sentinel species in the monitoring of Gd contamination of medical origin. It would thus appear important to consider the potential entry of this contaminant into the human food chain via other, commercially exploited bivalve species.

This study describes an image analysis method that has been used to analyze the swimming behavior of native oyster D-larvae (Crassostrea gigas) from the Arcachon Bay (SW, France). In a second time, this study evaluated the impact of copper and S-metolachlor pollutants on D-larvae swimming activity and the possible relationship between developmental malformations and abnormal swimming behavior. Analyses in wild and cultivated oyster D-larvae were investigated during two breeding-seasons (2014 and 2015) at different sampling sites and dates. In controlled conditions, the average speed of larvae was 144 µm s-1 and the maximum speed was 297 µm s-1 while the trajectory is mainly rectilinear. In the presence of environmental concentration of copper or S-metolachlor, no significant difference in maximum or average larval speed was observed compared to the control condition but the percentage of circular trajectory increased significantly while the rectilinear swimming larvae significantly declined. The current study demonstrates that rectilinear trajectories are positively correlated to normal larvae while larvae with shell anomalies are positively correlated to circular trajectories. This abnormal behavior could affect the survival and spread of larvae, and consequently, the recruitment and colonization of new habitats.

The European sturgeon, Acipenser sturio, is a highly endangered species that almost disappeared in the last decades. Thanks to yearly restocking of the population, this species is still found in the Gironde estuary (France), where juveniles grow during several years before leaving to the ocean. The aims of this study were to evaluate the pressure exerted on these fish by known organic and inorganic contaminants during their stay at the Gironde estuary, and to get information on the fish's health in this context. Monthly captures over the year 2014 provided 87 fish from the cohorts 2012 and 2013 mainly, and from cohorts 2008, 2009, and 2011, all fish born in hatchery. We report the very first analyses of contaminant levels and of biological markers measured in the blood of these fish. Low inorganic contamination was found, composed of seven metals mainly Zn (< 5 µg mL-1), Fe (< 1.5 µg mL-1), Cu (< 0.8 µg mL-1), Se (< 0.8 µg mL-1), As (< 0.25 µg mL-1), Co (< 0.14 µg mL-1), and Mn (< 0.03 µg mL-1). Concerning persistent organic contaminants, the sum of seven PCBs varied from 1 to 10 ng g-1 plasma, that of eight OCPs from 0.1 to 1 ng g-1, and that of eight PBDEs from 10 to 100 pg g-1. Higher levels of contaminants were measured during spring as compared to summer. The sex steroid hormone plasma levels (estradiol, testosterone, and 11-ketotestosterone) were quite low, which was predictable for juveniles. The transcription of reproduction-involved genes (EstR, AR, LHR, sox9) in blood cells was demonstrated for the first time. Some of them were correlated with organic contaminant levels PCBs and OCPs. Other gene transcriptions (sodCu and bax) were correlated with PCBs and OCPs. However, the DNA damage level measured here as comet tail DNA and micronuclei ratio in red blood cells were in the very low range of the values commonly obtained in fish from pristine areas. The data presented here can serve as a reference base for future monitoring of this population of sturgeons.

Very few studies have characterized the concentrations of pollutants in bioturbating species. These species are considered as ecosystem engineers and characterizing stressors, such as contaminants, that impact them could lead to a better understanding of the functioning of ecosystems. In addition to contaminants, bioturbators are affected by a wide range of stressors, which can influence their physiological status and their ability to accumulate pollutants. Among these stressors, parasitism is of particular concern due to the ubiquity of parasites in natural environments and their influence on the fitness of their host. This study aims to assess the relationship between parasitism and metal accumulation in the bioturbating mud shrimp Upogebia cf. pusilla. A one-year seasonal survey was conducted in Arcachon Bay, France, with the aims of (1) characterizing the levels of metals in the mud shrimp and (2) evaluating the influence of two macroparasites (a bopyrid isopod and a trematode) on the variation of the metal content in mud shrimp. The bopyrid parasite castrates its female host and a particular attention has therefore been paid to the reproductive cycle of female mud shrimp by quantifying the expression of the vitellogenin gene that encodes the major yolk protein in female crustaceans. The levels of contaminants in mud shrimp appeared low compared to those reported in other crustaceans in areas of higher pollution. Even at these low contamination levels, we observed a significant impact by the bopyrid parasite that depends on season: bopyrid-infested organisms are generally more contaminated than their uninfested conspecifics except in summer when the opposite trend was observed. We suggest that the bopyrid indirectly interferes with the metal accumulation process by altering the reproductive capabilities of the mud shrimp. On the opposite, very low influence of the trematode parasite on the metal content of the host was found.

This study investigates the combined effects of increased temperatures and copper on the early live stages of the Mediterranean mussel Mytilus galloprovincialis and the blue mussel Mytilus edulis as well as their hybrids. For this purpose, developmental abnormalities was measured after 48 h of exposure as well as a battery of oxidative stress markers such as, antioxidant enzyme activities, lipid peroxidation measured as malondialdehyde accumulation (MDA) and metallothionein contents (MT) and related gene expression pattern. Embryotoxicity and metal accumulation in tissues of mussel larvae exposed to a sublethal concentration of copper (10 µg/L) along with a slight temperature increase from 18 °C to22 °C were significantly increased after 48 h of exposure. Co-exposure to Cu and elevated temperatures significantly increased the antioxidant enzyme activities termed as, glutathione-S-transferase (GST), catalase (CAT), superoxide dismutase (SOD) and being maintained at 22 °C only in pure M. galloprovincialis (PG) larvae and female galloprovincialis x male edulis hybrid larvae (HFG). A significant decrease in mRNA abundance of cat, sod, gst gene transcription levels was showed in the same species. Furthermore, metallothionein accumulation increased significantly in PG D-larvae exposed to copper at 22 °C. The same pattern was observed in term of gene expression of MTs cognates (mt-10 and mt-20). Significant increase of MDA levels in pure M. edulis (PE) larvae and their female M. edulis x male M. galloprovincialis hybrid larvae (HFE) combined with a low MTs content were observed. Overall, this study provides clues about the relatively higher resistance and resilience of M. galloprovincialis species compared to M. edulis species under environmental pollution and future climate change scenarios.

The aim of this work was to study the impact of copper during a sub-chronic exposure to environmental concentrations in the early life stages of rainbow trout (Oncorhynchus mykiss). Eyed-stage embryos of rainbow trout, at 265 °D, were exposed in semi-static conditions to sub-lethal concentrations of CuSO4 up to the larval stage (528 °D) under laboratory-controlled conditions. During 3 weeks, they were exposed to the environmentally-realistic concentration of 2 µg/L Cu and to a 10-fold higher concentration, 20 µg/L Cu. Several biological (survival, hatching success, malformation, growth) and behavioral (swimming activity) and molecular endpoints (genotoxicity and gene transcription) were studied. Exposure to 20 µg/L Cu had an inhibitory effect on hatching and increased half-hatched embryos (25%). At the end of the exposure, no significant differences were observed in growth of the larvae exposed to the highest Cu concentration. However, larvae exposed to 2 µg/L Cu exhibited increased growth in comparison with non-exposed larvae. The percentage of malformed larvae was significantly higher for both copper conditions, with skeletal malformations being the most observed. Expression of several genes was evaluated in whole larvae using quantitative real-time PCR. Genes involved in detoxification (gst, mt1 and mt2) and in cell cycle arrest (p53) were significantly repressed in both copper conditions when compared to control. In addition, potential genotoxic effects on larvae were investigated by the comet assay on blood cells, but this test did not demonstrate any significant DNA damage on larvae exposed to copper. This study confirms the adverse effects of copper on early life stages of rainbow trout even at the lowest environmentally relevant tested concentration.

The increase in human population and urbanization are resulting in an increase in the volume of wastewater and urban runoff effluents entering natural ecosystems. These effluents may contain multiple pollutants to which the biological response of aquatic organisms is still poorly understood mainly due to mixture toxicity and interactions with other environmental factors. In this context, RNA sequencing was used to assess the impact of a chronic exposure to wastewater treatment plant and stormwater effluents at the whole-transcriptome level and evaluate the potential physiological outcomes in the Asian clam Corbicula fluminea. We de-novo assembled a transcriptome from C. fluminea digestive gland and identified a set of 3,181 transcripts with altered abundance in response to water quality. The largest differences in transcriptomic profiles were observed between C. fluminea from the reference site and those exposed to wastewater treatment plant effluents. On both anthropogenically impacted sites, most differentially expressed transcripts were involved in signaling pathways in relation to energy metabolism such as mTOR and FoxO, suggesting an energy/nutrient deficit and hypoxic conditions. These conditions were likely responsible for damages to proteins and transcripts in response to wastewater treatment effluents whereas exposure to urban runoff might result in immune and endocrine disruptions. In absence of comprehensive chemical characterization, the RNAseq approach could provide information regarding the mode of action of pollutants and then be useful for the identification of which parameters must be studied at higher integration level in order to diagnose sites where the presence of complex and variable mixtures of chemicals is suspected.

The past two million years of eastern African climate variability is currently poorly constrained, despite interest in understanding its assumed role in early human evolution1-4. Rare palaeoclimate records from northeastern Africa suggest progressively drier conditions2,5 or a stable hydroclimate6. By contrast, records from Lake Malawi in tropical southeastern Africa reveal a trend of a progressively wetter climate over the past 1.3 million years7,8. The climatic forcings that controlled these past hydrological changes are also a matter of debate. Some studies suggest a dominant local insolation forcing on hydrological changes9-11, whereas others infer a potential influence of sea surface temperature changes in the Indian Ocean8,12,13. Here we show that the hydroclimate in southeastern Africa (20-25° S) is controlled by interplay between low-latitude insolation forcing (precession and eccentricity) and changes in ice volume at high latitudes. Our results are based on a multiple-proxy reconstruction of hydrological changes in the Limpopo River catchment, combined with a reconstruction of sea surface temperature in the southwestern Indian Ocean for the past 2.14 million years. We find a long-term aridification in the Limpopo catchment between around 1 and 0.6 million years ago, opposite to the hydroclimatic evolution suggested by records from Lake Malawi. Our results, together with evidence of wetting at Lake Malawi, imply that the rainbelt contracted toward the Equator in response to increased ice volume at high latitudes. By reducing the extent of woodland or wetlands in terrestrial ecosystems, the observed changes in the hydroclimate of southeastern Africa-both in terms of its long-term state and marked precessional variability-could have had a role in the evolution of early hominins, particularly in the extinction of Paranthropus robustus.

Marine sediments are an important source of contaminants since they are susceptible to be remobilized to the water column. By modifying the physical and biogeochemical characteristics of sediments, bioturbation can influence contaminants remobilization. Within bioturbators, mud shrimp are considered as among the most influential organisms in marine soft-bottom environments. The physiological state of mud shrimp can be impaired by bopyrid parasites. The present study aims to evaluate the influence of bopyrid-uninfested and bopyrid-infested mud shrimp on sediment contaminants resuspension. Through a 14-days ex-situ experiment we showed a moderate effect of mud shrimp on contaminant remobilization compared to molecular diffusion without the bioturbator. Conversely, parasite presence significantly impaired contaminant bioaccumulation in mud shrimp and interfered with genetic expression. The weak effect of mud shrimp on contaminant remobilization may be due to trace metal accumulation and thus we suggested to evaluate the influence of contaminants on activities of bioturbating species.

Fish are currently used for the assessment of chemical toxicity. The REACh regulation and the European directive on the protection of animals used for scientific purposes both recommend the use of methods other than animal testing. In view of this, fish cell lines are increasingly used to provide fast and reliable toxic and ecotoxic data on new chemicals. The sensitivity of the Rainbow trout liver cell line RTL-W1 and Japanese medaka embryos cell line OLCAB-e3 were used with different toxicity endpoints, namely cytotoxicity, EROD activity, ROS production and DNA damage for various classes of pollutants displaying different modes of action but also with complex environmental mixtures. Toxicity tests were coupled with chemical analysis to quantify the chemical concentrations in cell cultures. Differences in sensitivity were found between fish cell lines. MTT reduction assay revealed that OLCAB-e3 cells were more sensitive than RTL-W1 cells. On the contrary, RTL-W1 gave higher response levels for the Fpg-modified comet assay and ROS assay. The OLCAB-e3 cell line did not express EROD activity unlike RTL-W1. This study highlights the capacity of the two different fish cell lines to measure the toxicity of individual toxicants but also environmental mixtures. Then, results obtained here illustrate the interest of using different cell lines and toxicity endpoints to assess the toxicity of complex or unknown mixture of chemicals.

This work addressed the trophic transfer and effects of functionalized gold nanoparticles (AuNPs) from periphytic biofilms to the crustacean Gammarus fossarum. Biofilms were exposed for 48 h to 10 nm positively charged functionalized AuNPs at two concentrations, 4.6 and 46 mg/L, and crustaceans G. fossarum grazed on these for 7 days, with daily biofilm renewal. Gold bioaccumulation in biofilm and crustacean were measured to estimate the trophic transfer ratio of these AuNP, and, for the first time, a transcriptomic approach and transmission electron microscopy observations in the crustacean were made. These two approaches showed cellular damage caused by oxidative stress and, in particular, an impact of these AuNPs on mitochondrial respiration. Modulation of digestive enzyme activity was also observed, suggesting modifications of digestive functions. The damage due to these nanoparticles could then have vital consequences for the organisms during chronic exposure.

This study evaluates the combined effects of two contaminants (copper and a herbicide S-metolachlor) and temperature on the early life stages of the Pacific oyster Crasssotrea gigas, which is native to Arcachon Bay (southwest France). The responses of D-larvae, obtained from wild and cultivated oysters, were investigated for one year during the oyster breeding period at different sampling sites and compared with the response of D-larvae from a commercial hatchery. Embryotoxicity was measured by considering the percentage of abnormal D-shaped larvae. Normal development of embryos occurred only from 22°C to 26°C. There were synergistic effects of copper at low and high temperatures. Native oysters appear particularly sensitive to an increase in water temperature, suggesting a future increase in the percentage of larval abnormalities as a result of global climate change. Hatchery oysters represent a good alternative model for studying the effects of both pollutants and climate change stressors.

The present study aims to elucidate the stress response of early life stages of Mytilus galloprovincialis to the combine effects of selected metals and elevated temperature. For this purpose, we investigated the response of a large panel of oxidative stress markers such as catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST) activities and lipid peroxidation (thiobarbituric acid reactive substrates (TBARS) concentration) and metallothionein accumulation (MT) as well as selected gene transcription level and metal accumulation in mussels larvae exposed to a sub-lethal concentration of Cu (9.54µg/L), Ag (2.55µg/L) and mixture of the two metals (Cu (6.67µg/L)+Ag (1.47µg/L)) along with a temperature gradient (18, 20 and 22°C) for 48h. Cu and Ag applied as single or mixture were differentially accumulated in mussel larvae according to the exposure temperature. Sod, cat, gst and mt-10 gene transcription levels showed an important increase in larvae exposed to Cu, Ag or to the mix compared to the control condition at 18°C. The same pattern but with higher induction levels was recorded in larvae co-exposed to metals at 20°C. At 22°C, a significant decrease in mRNA abundance of cat, gst and sod and a significant up-regulation of mts targets (mt10 and mt20) were observed. RESULTS: suggest that co-exposure to metals and moderate elevated temperature (20 and 22°C) significantly increased the antioxidant enzyme activities of catalase (CAT), and glutathione-S-transferase (GST) and caused an increase of metal and metallothionein concentrations. In contrast, no significant change in lipid peroxidation products measured as TBARS content was observed indicating a protective response of anti-oxidative system. This study provides first evidences of the early and efficient protective response of antioxidant defense mechanisms in mussel's early life stages facing in multi stressors situations.

The present work aimed to assess the effects of two widespread metallic pollutants, copper and silver, along with environmentally-realistic temperature increases, on embryo-larval development of the Mediterranean mussel Mytilus galloprovincialis. First, mussel embryos upon fertilization were exposed for 48 h to increasing concentrations of Cu (0.5-500 µg/L) and Ag (0.1-100 µg/L) at different temperatures (18, 20, 22 or 24 °C) in order to characterize toxicity of each toxicant at the different tested temperatures. Increasing concentrations of a Cu-Ag mixture were then tested in order to assess the mixture effect at different temperatures (18, 20 or 22 °C). Embryotoxicity was measured after 48 h of exposure (D-larvae stage) considering both the percentage of abnormalities and developmental arrest in D-larvae. The results suggest that the optimum temperature for mussel larvae development is 18 °C (12.65± 1.6% malformations) and beyond 20 °C a steep increase of abnormal larvae was observed up to 100% at 24 °C. Ag was more toxic than Cu with a 50% effective concentration (EC50) at 18 °C of 6.58 µg/L and 17.6 µg/L, respectively. Temperature increased the toxicity of both metals as proved with the EC50 at 20 °C at 3.86 µg/L and 16.28 µg/L for Ag and Cu respectively. Toxic unit calculation suggests additive effects of Cu and Ag in mixture at 18 and 20 °C. These results highlight a possible impairment of M. galloprovincialis reproduction in the Mediterranean Sea in relation to increase of both pollutants and water temperature due to global warming.
