RESUMO

BACKGROUND: This two-study paper developed a climate change risk perception model that considers the role of posttraumatic growth (i.e., a reappraisal of life priorities and deeper appreciation of life), resource loss, posttraumatic stress, coping, and social support. METHOD: In Study 1, participants were 332 persons in the Philippines who experienced Super Typhoon Haiyan. In Study 2, participants were 709 persons in Fiji who experienced Cyclone Winston. Climate change can increase the size and destructive potential of cyclones and typhoons as a result of warming ocean temperatures, which provides fuel for these storms. Participants completed measures assessing resource loss, posttraumatic stress, coping, social support, posttraumatic growth, and climate change risk perception. RESULTS: Structural equation modeling was used to develop a climate change risk perception model with data collected in the Philippines and to confirm the model with data collected in Fiji. The model showed that climate change risk perception was influenced by resource loss, posttraumatic stress, coping activation, and posttraumatic growth. The model developed in the Philippines was confirmed with data collected in Fiji. CONCLUSIONS: Posttraumatic growth played a central role in climate change risk perception. Public health educational efforts should focus on vividly showing how climate change threatens life priorities and that which gives life meaning and can result in loss, stress, and hardship. Disaster response organizations may also use this approach to promote preparedness for disaster threats.

Breast cancers are divided into subtypes with different prognoses and treatment responses based on global differences in gene expression. Luminal breast cancer gene expression and proliferation are driven by estrogen receptor alpha, and targeting this transcription factor is the most effective therapy for this subtype. By contrast, it remains unclear which transcription factors drive the gene expression signature that defines basal-like triple-negative breast cancer, and there are no targeted therapies approved to treat this aggressive subtype. In this study, we utilized integrated genomic analysis of DNA methylation, chromatin accessibility, transcription factor binding, and gene expression in large collections of breast cancer cell lines and patient tumors to identify transcription factors responsible for the basal-like gene expression program. Glucocorticoid receptor (GR) and STAT3 bind to the same genomic regulatory regions, which were specifically open and unmethylated in basal-like breast cancer. These transcription factors cooperated to regulate expression of hundreds of genes in the basal-like gene expression signature, which were associated with poor prognosis. Combination treatment with small-molecule inhibitors of both transcription factors resulted in synergistic decreases in cell growth in cell lines and patient-derived organoid models. This study demonstrates that GR and STAT3 cooperate to regulate the basal-like breast cancer gene expression program and provides the basis for improved therapy for basal-like triple-negative breast cancer through rational combination of STAT3 and GR inhibitors. SIGNIFICANCE: This study demonstrates that GR and STAT3 cooperate to activate the canonical gene expression signature of basal-like triple-negative breast cancer and that combination treatment with STAT3 and GR inhibitors could provide synergistic therapeutic efficacy.

This study aimed to develop risk scores based on clinical characteristics at presentation to predict intensive care unit (ICU) admission and mortality in COVID-19 patients. 641 hospitalized patients with laboratory-confirmed COVID-19 were selected from 4997 persons under investigation. We performed a retrospective review of medical records of demographics, comorbidities and laboratory tests at the initial presentation. Primary outcomes were ICU admission and death. Logistic regression was used to identify independent clinical variables predicting the two outcomes. The model was validated by splitting the data into 70% for training and 30% for testing. Performance accuracy was evaluated using area under the curve (AUC) of the receiver operating characteristic analysis (ROC). Five significant variables predicting ICU admission were lactate dehydrogenase, procalcitonin, pulse oxygen saturation, smoking history, and lymphocyte count. Seven significant variables predicting mortality were heart failure, procalcitonin, lactate dehydrogenase, chronic obstructive pulmonary disease, pulse oxygen saturation, heart rate, and age. The mortality group uniquely contained cardiopulmonary variables. The risk score model yielded good accuracy with an AUC of 0.74 ([95% CI, 0.63-0.85], p = 0.001) for predicting ICU admission and 0.83 ([95% CI, 0.73-0.92], p<0.001) for predicting mortality for the testing dataset. This study identified key independent clinical variables that predicted ICU admission and mortality associated with COVID-19. This risk score system may prove useful for frontline physicians in clinical decision-making under time-sensitive and resource-constrained environment.

Cultivation of the filamentous chlorophyte Oedogonium in municipal wastewater effluent is known to improve water quality and yield lipid- and protein-rich biomass for industrial applications. Chlorophyte celluloses, whose molecular organization and physical traits differ from those of plants, represent yet another valuable extractive, and algal oxygen production is of economic value in wastewater treatment. Consequently, we explored cellulose and oxygen production from Oedogonium biomass batch-cultivated in treated secondary municipal wastewater effluent. We compared biomass, cellulose, and oxygen production outside and within an adjacent greenhouse, under differing dissolved CO2 and pH conditions, and during temperate-zone seasonal change from summer through fall. Overall production did not differ within or outside the greenhouse, but outside production was higher in summer and lower in fall as air temperatures declined. Batch cultivation offered advantages, but high levels of mixing and CO2 were essential to maintain neutral pH for optimal algal growth and oxygen production.

This study is a reliability generalization meta-analysis that reviews 5 of the most frequently used continuous measures of adult attachment security: the Adult Attachment Scale, Revised Adult Attachment Scale, Adult Attachment Questionnaire, Experiences in Close Relationships, and Experiences in Close Relationships-Revised. A total of 313,462 individuals from 564 studies provided 1,629 internal consistency reliability estimates for this meta-analysis. We present the average internal consistency reliability of scores for each measure and test the consistency of score reliabilities across a wide variety of sample characteristics. In light of this, we highlight several issues in the measurement of adult attachment security and make concrete recommendations for researchers seeking to measure adult attachment.

Periphyton dominated by the cellulose-rich filamentous green alga Cladophora forms conspicuous growths along rocky marine and freshwater shorelines worldwide, providing habitat for diverse epibionts. Bacterial epibionts have been inferred to display diverse functions of biogeochemical significance: N-fixation and other redox reactions, phosphorus accumulation, and organic degradation. Here, we report taxonomic diversity of eukaryotic and prokaryotic epibionts and diversity of genes associated with materials cycling in a Cladophora metagenome sampled from Lake Mendota, Dane Co., WI, USA, during the growing season of 2012. A total of 1,060 distinct 16S, 173 18S, and 351 28S rRNA operational taxonomic units, from which >220 genera or species of bacteria (~60), protists (~80), fungi (6), and microscopic metazoa (~80), were distinguished with the use of reference databases. We inferred the presence of several algal taxa generally associated with marine systems and detected Jaoa, a freshwater periphytic ulvophyte previously thought endemic to China. We identified six distinct nifH gene sequences marking nitrogen fixation, >25 bacterial and eukaryotic cellulases relevant to sedimentary C-cycling and technological applications, and genes encoding enzymes in aerobic and anaerobic pathways for vitamin B12 biosynthesis. These results emphasize the importance of Cladophora in providing habitat for microscopic metazoa, fungi, protists, and bacteria that are often inconspicuous, yet play important roles in ecosystem biogeochemistry.

This study examines variables associated with posttraumatic stress symptoms (PTS) and posttraumatic growth among 2 independent samples of survivors following the Indian Ocean tsunami in Khao Lak, Thailand. Participants were exposed to unprecedented horror and loss of life and property. At 3 months participants (N = 248) were living in temporary shelters, and at 15 months a second sample (N = 255) was living in homes built after the tsunami. Prior traumatic experiences, life threat, loss of personal characteristic resources and condition resources, somatic problems, and social support accounted for close to half of the variance in PTS in each sample. At 3 months, emotion-focused coping and concerns about government favoritism also contributed to PTS. At 15 months, lack of prior disaster experience and loss of energy resources also contributed to PTS. Distress was higher among participants surveyed at 3 months than among those surveyed at 15 months. Posttraumatic growth was positively associated with social support and problem-focused coping in both samples. The findings support conservation of resources stress theory ( Hobfoll, 2012 ) and underscore how systemic issues affect mental health. The implications of the findings are discussed, as is the educational International Tsunami Museum designed by the first author to address systemic stressors.

The beneficial effect of social support on the well-being of individuals and romantic relationships has been extensively studied in married heterosexual relationships. The direct effects model suggests that social support is directly associated with well-being, while the social buffering model describes how social support can protect individual well-being from the negative impact of stress. In the present study, we seek to test the extent to which these processes apply to gay and lesbian couples. We use a sample of 111 gay, lesbian, and married heterosexual couples to test the predictions of the social buffering models across relationship types. Irrespective of sexual orientation, the results suggest that social support from family and friends is directly related to well-being, while support provided by one's romantic partner buffered individual well-being from the negative impact of stress. The direct and buffering effects of partner support on romantic relationship quality were likewise consistent across couple types. While the amount of social support received from family members had a beneficial effect on the relationship quality of heterosexual couples, family support was unrelated to relationship quality in same-sex couples. Furthermore, the relation between friend support and relationship quality differed across couple types. We discuss the results in the context of the unique challenges faced by same-sex couples.

The green algal genus Cladophora forms conspicuous nearshore populations in marine and freshwaters worldwide, commonly dominating peri-phyton communities. As the result of human activities, including the nutrient pollution of nearshore waters, Cladophora-dominated periphyton can form nuisance blooms. On the other hand, Cladophora has ecological functions that are beneficial, but less well appreciated. For example, Cladophora has previously been characterized as an ecological engineer because its complex structure fosters functional and taxonomic diversity of benthic microfauna. Here, we review classic and recent literature concerning taxonomy, cell biology, morphology, reproductive biology, and ecology of the genus Cladophora, to examine how this alga functions to modify habitats and influence littoral biogeochemistry. We review the evidence that Cladophora supports large, diverse populations of microalgal and bacterial epiphytes that influence the cycling of carbon and other key elements, and that the high production of cellulose and hydrocarbons by Cladophora-dominated periphyton has the potential for diverse technological applications, including wastewater remediation coupled to renewable biofuel production. We postulate that well-known aspects of Cladophora morphology, hydrodynamically stable and perennial holdfasts, distinctively branched architecture, unusually large cell and sporangial size and robust cell wall construction, are major factors contributing to the multiple roles of this organism as an ecological engineer.

Until recently, biodiesel production has been derived from terrestrial plants such as soybean and canola, leading to competition between biodiesel production and agricultural production for source materials. Microalgae have the potential to synthesize 30 times more oil per hectare than terrestrial plants without competing for agricultural land. We examined four genera (Cyclotella, Aulacoseira, Fragilaria, Synedra) of common freshwater diatoms (Bacillariophyceae) for growth and lipid content in defined medium (sD11) that replicates hypereutrophic conditions in lakes and wastewater treatment plant effluents and optimized the medium for silicon content. Cyclotella and Aulacoseira produced the highest levels of total lipids, 60 and 43 µg total lipids/ml, respectively. Both diatoms are rich in fatty acids C14, C16, C16:1, C16:2,7,10, and C22:5n3. Of the diatoms examined, Cyclotella reached the highest population density (>2.5 × 10(6) cells/ml) in stationary phase when many of the cells appeared to be filled entirely with oil. Silicon enrichment studies indicated that for optimal utilization of phosphorus and nitrogen by diatoms growing in wastewater effluent, the amount of silicon present or added to the effluent should be 17.5 times the mass of phosphorus in the effluent. With high growth rates, high lipid contents, and rapid settling rates, Cyclotella and Aulacoseira are candidates for biodiesel production.

We conducted a reliability-generalization meta-analysis of 7 of the most frequently used measures of relationship satisfaction: the Locke-Wallace Marital Adjustment Test (LWMAT), the Kansas Marital Satisfaction Scale (KMS), the Quality of Marriage Index, the Relationship Assessment Scale, the Marital Opinion Questionnaire, Karney and Bradbury's (1997) semantic differential scale, and the Couples Satisfaction Index. Six hundred thirty-nine reliability coefficients from 398 articles and 636,806 individuals provided internal consistency reliability estimates for this meta-analysis. We present the average score reliabilities for each measure, characterize the variance in score reliabilities across studies, and consider sample and study characteristics that are predictive of score reliability. Overall, the KMS and the LWMAT appear to be the strongest and weakest measures, respectively, from a reliability perspective. We discuss the importance of considering reliability invariance when making cross-group comparisons and provide recommendations for researchers when electing a measure of relationship satisfaction.

In the face of the rising number of doctoral recipients in professional psychology, many have voiced concerns about the quality of nontraditional training programs. Past research suggests that, on a variety of outcomes, graduates from clinical PhD programs outperform graduates from clinical PsyD and, to a lesser extent, counseling PhD programs. We examine an aggregate archival dataset to determine whether student or university characteristics account for the differences in outcomes among programs. The data show meaningful differences in the outcomes of clinical PhD, PsyD, and counseling PhD programs. Furthermore, graduates from research-intensive universities perform better on the psychology licensure exam and are more likely to become American Board of Professional Psychology diplomates. The available data support the notion that the ability to conduct research is an essential component of graduate education. In this light, PsyD programs represent a unique opportunity to train students in the types of evaluation and outcomes assessments used by practicing psychologists. We discuss implications for graduate-level training in professional psychology.

Here, we respond to a letter and a commentary on our article in the present issue, welcoming this opportunity to continue the scientific dialogue about an issue that has long been stimulating and controversial. For the reader's benefit, we first briefly describe the recent scholarly context of our article.

PREMISE OF THE STUDY: The goal of this study was to illuminate the evolutionary history and ecological importance of plant mixotrophy-the uptake and utilization of exogenous organic compounds. • METHODS: We quantitatively assessed the effect of sugar amendments on laboratory growth of Sphagnum compactum as a representative emergent peat moss and two species of ecologically associated zygnematalean algae, Cylindrocystis brebissonii and Mougeotia sp. • KEY RESULTS: Together with observations published elsewhere, our results suggest that under carbon or light limitation, the uptake of exogenous sugars by cells of charophycean algae and peat mosses may help these organisms maintain positive carbon balance. Utilization of 1% glucose by aquatic-grown algae helped to relieve dissolved inorganic carbon limitation, enhancing photoautotrophic growth by factors of 9.0 and 1.7, respectively. After an 8-wk growth period, amendments of 1% and 2% glucose enhanced air-grown moss biomass by 28 and 39 times, respectively, that of controls lacking sugar amendments. After 9 wk, 1% fructose enhanced biomass by 21 times, and 2% sucrose enhanced biomass by 31 times. • CONCLUSION: Our results indicate that plant mixotrophy is an early-evolved trait. The results also indicate that quantitative differences in sugar utilization by bryophytes and charophycean algae correlate with relative investments in protective cell-wall polyphenolics measured in previous studies, suggesting that sugar utilization may subsidize the cost of producing phenolic wall compounds in bryophytes.

New structural, nutritional, and stable carbon isotope data may resolve a long-standing mystery-the biological affinities of the fossil Prototaxites, the largest organism on land during the Late Silurian to Late Devonian (420-370 Ma). The tree trunk-shaped specimens, of varying dimensions but consistent tubular anatomy, first formed prior to vascular plant dominance. Hence, Prototaxites has been proposed to represent giant algae, fungi, or lichens, despite incompatible biochemical and anatomical observations. Our comparative analyses instead indicate that Prototaxites formed from partially degraded, wind-, gravity-, or water-rolled mats of mixotrophic liverworts having fungal and cyanobacterial associates, much like the modern liverwort genus Marchantia. We propose that the fossil body is largely derived from abundant, highly degradation-resistant, tubular rhizoids of marchantioid liverworts, intermixed with tubular microbial elements. Our concept explains previously puzzling fossil features and is consistent with evidence for liverworts and microbial associates in Ordovician-Devonian deposits, extensive ancient and modern marchantioid mats, and modern associations of liverworts with cyanobacteria and diverse types of fungi. Our interpretation indicates that liverworts were important components of Devonian ecosystems, that some macrofossils and microfossils previously attributed to "nematophytes" actually represent remains of ancient liverworts, and that mixotrophy and microbial associations were features of early land plants.

It is believed that beta-amyloid aggregation is an important event in the development of Alzheimer's disease. In the course of our studies to identify beta-amyloid aggregation inhibitors, a series of N-phenyl anthranilic acid analogs were synthesized and studied for beta-amyloid inhibition activity. The synthesis, structure-activity relationship, and in vivo activity of these analogs are discussed.

The self-expansion model of close relationships posits that when couples engage in exciting and activating conjoint activities, they feel connected with their partners and more satisfied with their relationships. In the present study, the experience sampling method was used to examine the predictions of the self-expansion model in couples' momentary experiences. In addition, the author generated several new hypotheses by integrating the self-expansion model with existing research on flow. Over the course of 1 week, 20 couples were signaled at quasi-random intervals to provide data on 1,265 unique experiences. The results suggest that the level of activation experienced during an activity was positively related to experience-level relationship quality. This relationship was consistent across free-time and nonfree-time contexts and was mediated by positive affect. Activation was not found to predict later affect unless the level of activation exceeded what was typical for the individual. Also examined was the influence of interpersonal context and activity type on self-expansion. The results support the self-expansion model and suggest that it could be considered under the broader umbrella of flow.

RESUMO

As part of an on-going effort to investigate the chemical space requirements for D(2)/5-HT(2A) receptor antagonists as atypical antipsychotics, new 1-aminoindanes were synthesized. The replacement of the heterocycle (oxindole) in ziprasidone with a carbocycle (indane) was well tolerated and was found to retain binding affinities for dopamine D(2), serotonin 5-HT(2A), and serotonin 5-HT(1A). Such compounds hold promise as a new chemical motif with atypical antipsychotic properties for the treatment of schizophrenia and related disorders.
