RESUMO

Two billion tonnes of alkaline metallurgical waste is generated per year as a product of industry, mining, and metal processing. Filtercake is one such residue formed as a bi-product of steelmaking. Metal rich bi-products can be both an environmental concern and potential resource. High concentrations of heavy metals, if accessed, could be utilised and reprocessed reducing both pollution and the demand for raw metal ores. Phytoextraction is one such method of recovering metals from contaminated mediums. Research interest in Lemna sp. has grown due to their phytoremediation potential. Facilitated by rapid growth and accumulation of nutrients and metals, Lemna minor has been described as one of the most effective macrophytes for remediating contaminated water. The present study outlines a system using L. minor to extract Zn from filtercake when submerged in static water. To facilitate phytoremediation, CO2 carbonation can be employed to solubilise elements and utilise this greenhouse gas, another a bi-product of steel industry. The addition of CO2 to vessels of water containing filtercake lowered the pH from as high as 8.8 to 5.6 and significantly increased Zn in solution compared to vessels receiving no CO2. Results suggest the potential of L. minor to accumulating 68.7 kg Zn per year from 20.5 Mt. filtercake ha-1. This system facilitates a circular economy with re-use of multiple existing bi-products. In addition, the potential employment of biomass in biofuel production and use of remediated filtercake in carbon sequestration adds further environmental and socio-economic impact. The extent to which the approach was consistent with circular economy was discussed and its wider integration considered.

Life-cycle assessment (LCA) is the preferred methodology to assess carbon footprint per unit of milk. The objective of this case study was to apply an LCA method to compare carbon footprints of high-performance confinement and grass-based dairy farms. Physical performance data from research herds were used to quantify carbon footprints of a high-performance Irish grass-based dairy system and a top-performing United Kingdom (UK) confinement dairy system. For the US confinement dairy system, data from the top 5% of herds of a national database were used. Life-cycle assessment was applied using the same dairy farm greenhouse gas (GHG) model for all dairy systems. The model estimated all on- and off-farm GHG sources associated with dairy production until milk is sold from the farm in kilograms of carbon dioxide equivalents (CO2-eq) and allocated emissions between milk and meat. The carbon footprint of milk was calculated by expressing GHG emissions attributed to milk per tonne of energy-corrected milk (ECM). The comparison showed that when GHG emissions were only attributed to milk, the carbon footprint of milk from the Irish grass-based system (837 kg of CO2-eq/t of ECM) was 5% lower than the UK confinement system (884 kg of CO2-eq/t of ECM) and 7% lower than the US confinement system (898 kg of CO2-eq/t of ECM). However, without grassland carbon sequestration, the grass-based and confinement dairy systems had similar carbon footprints per tonne of ECM. Emission algorithms and allocation of GHG emissions between milk and meat also affected the relative difference and order of dairy system carbon footprints. For instance, depending on the method chosen to allocate emissions between milk and meat, the relative difference between the carbon footprints of grass-based and confinement dairy systems varied by 3 to 22%. This indicates that further harmonization of several aspects of the LCA methodology is required to compare carbon footprints of contrasting dairy systems. In comparison to recent reports that assess the carbon footprint of milk from average Irish, UK, and US dairy systems, this case study indicates that top-performing herds of the respective nations have carbon footprints 27 to 32% lower than average dairy systems. Although differences between studies are partly explained by methodological inconsistency, the comparison suggests that potential exists to reduce the carbon footprint of milk in each of the nations by implementing practices that improve productivity.

Life cycle assessment (LCA) and the Intergovernmental Panel on Climate Change (IPCC) guideline methodology, which are the principal greenhouse gas (GHG) quantification methods, were evaluated in this study using a dairy farm GHG model. The model was applied to estimate GHG emissions from two contrasting dairy systems: a seasonal calving pasture-based dairy farm and a total confinement dairy system. Data used to quantify emissions from these systems originated from a research study carried out over a 1-year period in Ireland. The genetic merit of cows modelled was similar for both systems. Total mixed ration was fed in the Confinement system, whereas grazed grass was mainly fed in the grass-based system. GHG emissions from these systems were quantified per unit of product and area. The results of both methods showed that the dairy system that emitted the lowest GHG emissions per unit area did not necessarily emit the lowest GHG emissions possible for a given level of product. Consequently, a recommendation from this study is that GHG emissions be evaluated per unit of product given the growing affluent human population and increasing demand for dairy products. The IPCC and LCA methods ranked dairy systems' GHG emissions differently. For instance, the IPCC method quantified that the Confinement system reduced GHG emissions per unit of product by 8% compared with the grass-based system, but the LCA approach calculated that the Confinement system increased emissions by 16% when off-farm emissions associated with primary dairy production were included. Thus, GHG emissions should be quantified using approaches that quantify the total GHG emissions associated with the production system, so as to determine whether the dairy system was causing emissions displacement. The IPCC and LCA methods were also used in this study to simulate, through a dairy farm GHG model, what effect management changes within both production systems have on GHG emissions. The findings suggest that single changes have a small mitigating effect on GHG emissions (<5%), except for strategies used to control emissions from manure storage in the Confinement system (14% to 24%). However, when several management strategies were combined, GHG emissions per unit of product could be reduced significantly (15% to 30%). The LCA method was identified as the preferred approach to assess the effect of management changes on GHG emissions, but the analysis indicated that further standardisation of the approach is needed given the sensitivity of the approach to allocation decisions regarding milk and meat.

Determining bioequivalence for solution pressurized metered dose inhalers (pMDI) is difficult because the critical characteristics of such products are poorly defined. The aim of this study was to elucidate the non-aerodynamic properties of the emitted aerosol particles from two solution pMDI products that determine their biopharmaceutical differences after deposition. Novel particle capture and analysis techniques were employed to characterize the physicochemical and biopharmaceutical properties of two beclomethasone dipropionate (BDP) products: QVAR and Sanasthmax. The BDP particles emitted from the Sanasthmax inhaler were discernibly different those emitted from QVAR in terms of size (50% larger, less porous), solid state (less crystalline) and dissolution (20-fold slower). When deposited onto the surface of respiratory epithelial cell layers, QVAR delivered ∼50% more BDP across the cell layer in 60 min than Sanasthmax. Biopharmaceutical performance was not attributable to individual particle properties as these were manifold with summative and/or competing effects. The cell culture dissolution-absorption model revealed the net effect of the particle formed on drug disposition and was predictive of human systemic absorption of BDP delivered by the test inhalers. This illustrates the potential of the technique to detect the effect of formulation on the performance of aerosolized particles and contribute to assessment of bioequivalence.

BACKGROUND: The high prevalence of obesity in children in the UK warrants continuing public health attention. 'Families for Health' is a family-based group programme for the treatment of childhood obesity. Significant improvements in body mass index (BMI) z-score (-0.21, 95% CI: -0.35 to -0.07, P = 0.007) and other health outcomes were seen in children at a 9-month follow-up. AIM: To undertake a 2-year follow-up of families who attended 'Families for Health' in Coventry, to assess long-term outcomes and costs. METHODS: 'Families for Health' is a 12-week programme with parallel groups for parents and children, addressing parenting skills, healthy lifestyles and emotional well-being. The intervention was delivered at a leisure centre in Coventry, England, with 27 overweight or obese children aged 7-13 years (18 girls, 9 boys) and their parents, from 21 families. A 'before-and-after' evaluation was completed with 19 (70%) children followed up at 2 years. The primary outcome was change in BMI z-score from baseline; secondary outcomes were children's quality of life, parent-child relationships, eating/activity habits and parents' mental health. Costs to deliver the intervention and to families were recorded. RESULTS: Mean change in BMI z-score from baseline was -0.23 (95% CI: -0.42 to -0.03, P = 0.027) at the 2-year follow-up and eight (42%) children had a clinically significant reduction in BMI z-score. Significant improvements were seen in children's quality of life and eating habits in the home, while there were sustained reductions in unhealthy foods and sedentary behaviour. Fruit and vegetable consumption and parent's mental health were not significantly different at 2 years. Costs of the programme were £517 per family (£402 per child), equivalent to £2543 per unit reduction in BMI z-score. CONCLUSIONS: Improvements in BMI z-score and certain other outcomes associated with the 'Families for Health' programme were sustained at the 2-year follow-up. 'Families for Health' is a promising new childhood obesity intervention, and a randomized controlled trial is now indicated.

This experiment measured variations in plasma concentrations of metabolic hormones and metabolites in cows undergoing extended lactations of up to 670 d at 2 planes of nutrition. Thirty-seven Holstein-Friesian cows that calved in late winter were selected for varying milk yield and then managed for a lactation of 670 d by delaying breeding until approximately 450 d in milk (DIM). Cows grazed fresh pasture supplemented with pasture silage or hay and crushed wheat or triticale grain. Dietary intake was reduced by approximately 1.8 kg (dry matter) grain/cow per day for 19 of the cows from 300 DIM until the end of lactation to assess the effect of restricted energy intake on the persistency of milk production. Samples of blood were collected monthly from each cow to measure plasma concentrations of selected hormones and metabolites. Dietary restriction beyond 300 DIM reduced yields of milk, protein, and fat, but did not alter the proportion of cows reaching the 670-d lactation target. Dietary restriction had no effect on cow BW or plasma concentrations of any hormones or metabolites. Overall, blood plasma concentrations of insulin-like growth factor-I, leptin, and glucose were elevated from 301 to 600 DIM compared with 0 to 300 DIM, whereas concentrations of growth hormone and nonesterified fatty acids were lower after 300 DIM. Plasma concentrations of insulin and prolactin were unaffected by stage of lactation, but prolactin concentrations increased during summer. These changes were consistent with a decrease in milk yield and an increase in the partitioning of nutrients to body tissue gain, primarily adipose tissue, throughout the later stages of the extended lactation. Cows that continued milking beyond 600 DIM had increased plasma concentrations of growth hormone and decreased concentrations of glucose and leptin compared with cows that milked <600 DIM. These differences, coupled with reduced body weight gain, indicated an increased priority for nutrient partitioning to milk production at the expense of body tissue gain throughout the extended lactation period in cows with greater lactation persistency.

Feed conversion efficiency of dairy cattle is an important component of the profitability of dairying, given that the cost of feed accounts for much of total farm expenses. Residual feed intake (RFI) is a useful measure of feed conversion efficiency, as it can be used to compare individuals with the same or differing levels of production during the period of measurement. If genetic variation exists in RFI among dairy cattle, selection for lower RFI could improve profitability. In this experiment, RFI was defined as the difference between an animal's actual feed intake and its expected feed intake, which was determined by regression of dry matter (DM) intake against mean body weight (BW) and growth rate. Nine hundred and three Holstein-Friesian heifer calves, aged between 5 and 7 mo, were measured for RFI in 3 cohorts of approximately 300 animals. Calves were housed under feedlot style conditions in groups of 15 to 20 for 85 to 95 d and had ad libitum access to a cubed alfalfa hay. Intakes of individual animals were recorded via an electronic feed recording system and BW gain was determined by weighing animals once or twice weekly, over a period of 60 to 70 d. Calves had DM intake (mean ± SD) of 8.3±1.37 kg of DM/d over the measurement period with BW gains of 1.1±0.17 kg/d. In terms of converting feed energy for maintenance and growth, the 10% most efficient calves (lowest RFI) ate 1.7 kg of DM less each day than the 10% least efficient calves (highest RFI) for the same rate of growth. Low-RFI heifers also had a significantly lower rate of intake (g/min) than high-RFI heifers. The heritability estimate of RFI (mean ± SE) was 0.27 (±0.12). These results indicate that substantial genetic variation in RFI exists, and that the magnitude of this variation is large enough to enable this trait to be considered as a candidate trait for future dairy breeding goals. A primary focus of future research should be to ensure that calves that are efficient at converting feed energy for maintenance and growth also become efficient at converting feed energy to milk. Future research will also be necessary to identify the consequences of selection for RFI on other traits (especially fertility and other fitness traits) and if any interactions exist between RFI and feeding level.

An experiment was conducted to measure the effect of diet on circulating concentrations of metabolic hormones and metabolites in cows undergoing extended lactations. Two groups of 6 Holstein-Friesian cows managed for lactations of 670 d were used in the experiment. One group was fully fed on a total mixed ration (TMR), whereas the other group grazed fresh pasture supplemented with grain (P+G). On 7 occasions between 332 and 612 d in milk, concentrations of metabolic hormones and glucose were measured in the blood plasma of each cow. Cows fed TMR gained more weight and body condition than P+G cows, but did not produce more milk during the study period. Only 3 of the TMR cows continued to lactate until 612 d in milk compared with all 6 of the P+G cows. Blood plasma from cows fed TMR had higher concentrations of glucose, insulin, glucagon, insulin-like growth factor 1, and leptin, but lower concentrations of growth hormone, than that from P+G cows. These changes were consistent with the preferential deposition of energy into adipose tissue at the expense of milk production and presumably were induced by a diet that provided precursors for gluconeogenesis that were in excess of the requirements for maintenance and prevailing milk production. The mechanism responsible for some TMR cows putting on excess weight and reducing or ceasing milk production is uncertain, but this observation has important implications for the nutritional management of cows in extended lactation programs.

The primary objective of our research was to determine the effect of a high dose of monensin supplementation on enteric CH(4) emissions of dairy cows offered a ryegrass pasture diet supplemented with grain. An additional objective was to evaluate effects on milk production and rumen function, because a commensurate improvement in milk production could lead to adoption of monensin as a profitable strategy for methane abatement. Two experiments were conducted (grazing and respiratory chambers) and in both experiments monensin (471 mg/d) was topdressed on 4 kg (dry matter)/d of rolled barley grain offered in a feed trough twice daily at milking times. In the grazing experiment, 50 Holstein-Friesian cows were assigned randomly to 1 of 2 groups (control or monensin). Cows grazed together as a single herd on a predominantly ryegrass sward and received monensin over a 12-wk period, during which time measurements of milk production and body weight change were made. The SF(6) tracer technique was used to estimate methane production of 30 of the 50 cows (15 control cows and 15 monensin cows) for 3 consecutive days in wk 3, 5, 8, and 12 of treatment. Samples of rumen fluid were collected per fistula from 8 of the 50 cows (4 per diet) on 2 consecutive days in wk 3, 5, 8, and 12 of treatment and analyzed for volatile fatty acids and ammonia-N. In the metabolic chamber experiment, 10 pairs of lactating dairy cows (control and monensin) were used to determine the effects of monensin on methane emissions, dry matter intake, milk production, and body weight change over a 10-wk period. Methane emissions were measured by placing cows in respiration chambers for 2 d at wk 5 and 10 of treatment. Cows received fresh ryegrass pasture harvested daily. Monensin did not affect methane production in either the grazing experiment (g/d, g/kg of milk) or the chamber experiment (g/d, g/kg of dry matter intake, g/kg of milk). In both experiments, milk production did not increase with addition of monensin to the diet. Monensin had no effect on body weight changes in either experiment. Monensin did not affect volatile fatty acids or ammonia-N in rumen fluid, but the acetate to propionate ratio tended to decrease. Monensin did not improve milk production of grazing dairy cows and no effect on enteric methane emissions was observed, indicating that monensin cannot be promoted as a viable mitigation strategy for dairy cows grazing ryegrass pasture supplemented with grain.

The purpose of this study was to model the effect of 3 divergent strains of Holstein-Friesian cows in 3 pasture-based feed systems on greenhouse gas (GHG) emissions. The 3 strains of Holstein-Friesian compared were high-production North American (HP), high-durability North American (HD), and New Zealand (NZ). The 3 feed systems were a high grass allowance system (MP, control); high stocking rate system (HS); and high concentrate supplementation system (HC). The MP system had an overall stocking rate of 2.47 cows/ha and received 325 kg of dry matter concentrate per cow in early lactation. The HS system had a similar concentrate input to the MP system, but had an overall stocking rate of 2.74 cows/ha. The HC system had a similar overall stocking rate to the MP system, but 1,445 kg of dry matter concentrate was offered per cow. A newly developed integrated economic-GHG farm model was used to evaluate the 9 milk production systems. The GHG model estimates on-farm (emissions arising within the farm's physical boundaries) and production system (incorporating all emissions associated with the production system up to the point milk leaves the farm gate) GHG emissions. Production system GHG emissions were always greater than on-farm emissions, and the ranking of the 9 systems was usually consistent under both methods. The exception was the NZ strain that achieved their lowest GHG emission per unit of product in the HC system when indirect emissions were excluded, but their lowest emission was in the HS system when indirect emissions were included. Generally, the results showed that as cow strain changed from lower (HD and NZ) to higher genetic potential (HP) for milk production, the GHG emission per kilogram of milk solids increased. This was because of a decline in cow fertility in the HP strain that resulted in a higher number of nonproductive animals, leading to a lower total farm milk solids production and an increase in emissions from nonproductive animals. The GHG emission per hectare increased for all strains moving from MP to HS to HC feed systems and this was associated with increases in herd total feed intake. The most profitable combination was the NZ strain in the HS system and this combination resulted in a 12% reduction in production system GHG emission per hectare compared with the NZ strain in the HC system, which produced the highest emissions. This demonstrates that grass-based systems can achieve high profitability and decreased GHG emissions simultaneously.

The objective of our work was to supplement a forage and cereal diet of lactating dairy cows with whole cottonseed (WCS) for 12 wk and to determine whether the expected reduction in CH(4) would persist. A secondary objective was to determine the effect of supplementing the diet with WCS on milk yield and rumen function over the 12-wk feeding period. Fifty lactating cows were randomly allocated to 1 of 2 diets (control or WCS). The 2 separate groups were each offered, on average, 4.2 kg of DM/cow per day of alfalfa hay (a.m.) and 6.6 kg of DM/cow per day of ryegrass silage (p.m.) on the ground in bare paddocks each day for 12 wk. Cows in each group were also individually offered dietary supplements for 12 wk in a feed trough at milking times of 5.4 kg of DM/cow per day of cracked wheat grain and 0.5 kg of DM/cow per day of cottonseed meal (control) or 2.8 kg of DM/cow per day of cracked wheat grain and 2.61 kg of DM/cow per day of WCS. The 2 diets were formulated to be similar in their concentrations of CP and ME, but the WCS diet was designed to have a higher fat concentration. Samples of rumen fluid were collected per fistula from the rumen approximately 4 h after grain feeding in the morning. Samples were taken from 8 cows (4 cows/diet) on 2 consecutive days in wk 2 of the covariate and wk 3, 6, 10, and 12 of treatment and analyzed for volatile fatty acids, ammonia-N, methanogens, and protozoa. The reduction in CH(4) emissions (g/d) because of WCS supplementation increased from 13% in wk 3 to 23% in wk 12 of treatment. Similarly, the reduction in CH(4) emissions (g/kg of DMI) increased from 5.1% in wk 3 to 14.5% in wk 12 of treatment. It was calculated that the average reduction in CH(4) emissions over the 12-wk period was 2.9% less CH(4) per 1% added fat, increasing from 1.5% in wk 3 to 4.4% less CH(4) in wk 12. There was no effect of WCS supplementation on rumen ammonia-N, rumen volatile fatty acids, rumen methanogens, and rumen protozoa. On average over the 12-wk period, supplementation with WCS decreased the yield of milk (10%), fat (11%), protein (14%), lactose (11%), and fat plus protein (12%) and BW gain (31%). The WCS supplementation had no effect on milk fat concentration but resulted in a decrease in concentration of protein (5%) and lactose (11%). The major finding from this study is that addition of WCS to the diet of lactating dairy cows resulted in a persistent reduction in CH(4) emissions (g of CH(4)/kg of DMI) over a 12-wk period and that these reductions in CH(4) are consistent with previous work that has studied the addition of oilseeds to ruminant diets.

Extending the lactation length of dairy cows beyond the traditional 10 mo toward lactations of up to 22 mo has attracted interest in the pasture-based seasonal dairying systems of Australia and New Zealand as a way of alleviating the need for cows to conceive during peak lactation, such as is required to maintain seasonally concentrated calving systems. Lactation lengths longer than 10 mo instead provide cows with more time to cycle and conceive after parturition and may therefore be more suitable systems for high-producing Holstein-Friesian cows. Before recommending such systems there is a need to evaluate the effects of long lactations on the suitability of milk for manufacture of high-quality dairy products. In the current experiment, the composition of milk from cows entering the second half of a 22-mo lactation was examined in detail and compared with that from cows undergoing a traditional 10-mo lactation. On 2 occasions, coagulation properties were measured using low amplitude strain oscillation rheometry, and Cheddar cheese was made in 250-L pilot-scale vats. Results showed that milk from extended lactations had higher concentrations of fat and protein than cows undergoing 10-mo lactations under similar management conditions and at the same time of year. The ratio of casein to true protein was not affected by lactation length and neither were the proportions of individual caseins. The increase in milk solids during extended lactations translated into a more rapid rate of coagulation and ultimately a firmer curd on one of the two occasions. Milk from extended lactations yielded more cheese per 100 kg of milk, and there were few differences in the composition or organoleptic properties of the cheese. These data are the first to show that pasture-based dairy industries could embrace the use of extended lactations without compromising the core business of producing high-quality dairy products.

The aim of this study was to measure the effect of type of diet and level of energy intake on the performance of cows undergoing extended lactations. Ninety-six Holstein-Friesian cows that calved in July and August 2004 were assigned randomly to 1 of 8 groups each of 12 cows (including 4 primiparous cows). Two of the 8 groups were assigned to each of 4 treatments that varied in lactation length (300 or 670 d) and diet (3 diets: control, high, or full total mixed ration (TMR). The 4 treatments were 1) control 300: cows were managed for a 300-d lactation and grazed pasture supplemented with grain and forage to provide a minimum daily dietary intake of 160 MJ of ME/cow; 2) control 670: as for control 300 except that cows were managed for a 670-d lactation; 3) high 670: cows were managed for a 670-d lactation and pasture was supplemented with grain and forage to provide a minimum daily dietary intake of 180 MJ of ME/cow; 4) full TMR 670: cows were managed for a TMR system that included a high body condition score at calving with cows offered a TMR during a 670-d lactation. The TMR was initially offered ad libitum indoors until about 440 DIM when the amount of TMR offered was reduced by about 2 kg of DM/d to prevent excessive weight gain. The proportions of cows still milking at the end of a 670-d lactation were similar for the control and high dietary groups. The full TMR group had fewer cows milking at 600 DIM: 17 cows milking compared with 24 cows in the control 670 group and 22 cows in the high 670 group. For the period 1 to 670 DIM, increasing the energy level in the diet (control 670 vs. high 670) resulted in a similar yield of milk and a similar fat concentration in the milk, but greater yields of milk fat and protein and greater milk protein percentage of the milk. The full TMR 670 group produced greater yields of milk and milk components (fat, protein, and lactose) and also protein percentage in the milk than the other groups. The milk solids (fat + protein) ratio for the 3 extended-lactation groups, defined as production achieved during the 24-mo calving interval divided by 2 yr (annualized production) expressed as a ratio of that produced in the normal 12-mo calving interval, was not affected by increasing the level of grain in the pasture-based diets (0.93 vs. 0.90 for control and high diets, respectively), but decreased with the TMR diet (0.79). The control 670 group produced 7.1% less milk, but only 2.4% less milk solids than the control 300 group over the 2-yr period of the study. Combining our data with that from 2 earlier studies of extended lactation demonstrated that Holstein cows with a high proportion of Northern Hemisphere genes offered pasture-based diets could achieve a high milk solids ratio, a greater proportion of cows milking at drying-off, and lower body weight gain over the lactation.

Knowledge of the inheritance of C-glycosyl flavone synthesis in maize (Zea mays L.) silk tissues has been acquired through detailed genetic studies involving primarily germplasm from the Corn Belt Dent race. To test the robustness of this genetic knowledge, we examined C-glycosyl flavone synthesis in a genetically distinct germplasm pool, popcorn. C-glycosyl flavone profiles and levels and the involvement of three specific genes/quantitative trait loci (p, pr1, and sm1) in C-glycosyl flavone synthesis were examined in popcorn germplasm representing at least two races and various diverse geographic regions. Twenty-four inbred lines and 23 hybrids involving these inbred lines and inbred line R17 were characterized for their flavone profiles and levels in silk tissues. Two F2 mapping populations were constructed to examine the involvement of p, pr1, and sm1 on C-glycosyl flavone synthesis. C-glycosyl flavone levels threefold higher than previously reported in Corn Dent Belt materials and a novel class of compounds were discovered. The gene action of sm1 was different, the functional p allele was not always dominant, and pr1 did not affect mays in synthesis. Based on this rather simplistic "model" quantitative trait, it appears that caution should be exercised when attempting to apply quantitative trait locus knowledge accumulated in one germplasm base to a germplasm base that is known to be distinctly unique.

The transepithelial transport rates of compounds after deposition as aerosolised particles onto respiratory cell layers and allowing dissolution in the cell surface secretions has not been reported in a comprehensive manner to date. Here, the twin-stage impinger (TSI) was used to deposit potentially respirable particles (aerodynamically <6.4 microm) of varying molecular weight dextrans labelled with fluorescein isothiocyanate (FITC-dex) onto Calu-3 cells, a model of the bronchial epithelium. The TSI functioned as a particle size segregator, with >96% of the deposited particles being geometrically <6.4 microm (as measured by microscopy) and the particles being deposited discretely with a uniform distribution. Cell layers tolerated particle deposition at an air flow of 60 L/min. A small reduction in transepithelial electrical resistance (TER) of <10% occurred initially, but the original TER was recovered within 10 min and there was no significant effect on apparent permeability (P(app)) of FITC-dex 4 over 4 h. Interleukin 8 (IL-8) secretion in the apical and basolateral directions over 24 h was not increased by exposure to the TSI and particle deposition. The rate of FITC-dex 4 (4 kDa) transport across the cell layer after deposition and dissolution of the particles in the cell surface secretions was approximately 20-fold higher (P<0.05) than if applied as a solution. The volume of cell surface secretions was estimated by tracer dilution (3.44+/-1.90 microl, mean+/-SEM) and this value was used to calculate the P(app) of compound once deposited as a particle. The Papp value was found to be similar to that obtained when the compound was applied in solution (P<0.05). Thus, the increased transport rate was attributable to the differences in donor chamber solute concentration rather than any change in the permeability of the cell layer itself. Following particle deposition, transport of FITC-dex with molecular weights between 4 and 70 kDa correlated well (r(2)=0.918) with reported in vivo canine pulmonary clearance after intratracheal instillation of dextrans of similar molecular weight. The use of the TSI and the Calu-3 cell line for the assessment of compound dissolution and transport rates after particle deposition may allow more realistic analyses to be made with respect to the in vivo situation.

We examined the effects of monensin, provided by controlled-release capsules, on the enteric methane emissions and milk production of dairy cows receiving ryegrass pasture and grain. In a grazing experiment, 60 Holstein-Friesian cows were assigned randomly to 1 of 2 groups (control or monensin). Cows in the monensin group received 2 controlled-release capsules, with the second capsule administered 130 d after the first. Milk production was measured for 100 d following insertion of each capsule. The sulfur hexafluoride tracer gas technique was used to measure enteric methane emissions for 4 d starting on d 25 and 81 after insertion of the first capsule, and on d 83 after insertion of the second capsule. All cows grazed together as a single herd on a predominantly ryegrass sward and received 5 kg/d of grain (as-fed basis). In a second experiment, 7 pairs of lactating dairy cows (control and monensin) were used to determine the effects of monensin controlled-release capsules on methane emissions and dry matter intake. Methane emissions were measured on d 75 after capsule insertion by placing cows in respiration chambers for 3 d. Cows received fresh ryegrass pasture harvested daily and 5 kg/d of grain. The release rate of monensin from the capsules used in both experiments was 240 +/- 0.072 mg/d, determined over a 100-d period in ruminally cannulated cows. The monensin dose was calculated to be 12 to 14.5 mg/kg of dry matter intake. There was no effect of monensin on methane production in either the grazing experiment (g/d, g/kg of milk solids) or the chamber experiment (g/d, g/kg of dry matter intake). In the grazing study, there was no effect of monensin on milk yield, but monensin increased milk fat yield by 51.5 g/d and tended to increase milk protein yield by 18.5 g/d. Monensin controlled-release capsules improved the efficiency of milk production of grazing dairy cows by increasing the yield of milk solids. However, a higher dose rate of monensin may be needed to reduce methane emissions from cows grazing pasture.

The aim of this study was to compare the reproductive performance, milk production, live weight, and body condition loss during early lactation of purebred Holstein (H) cows to Jersey x H (J x H) crossbred cows in 4 Victorian herds. Cows of H and J x H breeding were managed together within each herd, and all herds had a seasonally concentrated calving pattern that commenced in early spring (July). All crossbred cows included in the study were 25, 50, or 75% H and were considered collectively as J x H regardless of the sire and dam breeds used to reach those percentages. Each herd owner provided records of reproductive performance and milk production. Compared with H cows, J x H cows had higher first-service conception rates (52 vs. 42%), higher percentages confirmed pregnant by 6 (68 vs. 54%) and 14 wk (86 vs. 78%) after the first day of inseminating, and lower final not-in-calf rates (11 vs. 16%); however, these differences were not observed in all herds. A random selection of H and J x H cows had body condition assessed on 3 occasions between the start of calving and the first day of the artificial insemination program. The selected cows were also weighed on the final occasion. Overall, body condition scores were slightly higher for J x H cows than for H cows, but changes in body condition score between calving and the start of inseminating were similar between breed groups. The H cows were 40 kg heavier than J x H cows and had daily milk yields in early lactation that were 2.2 kg higher. Daily yields of milk fat and protein did not differ between H and J x H cows during the study period. The improved reproductive performance of J x H cows compared with H cows may render them more suitable for use in dairy herds with seasonally concentrated calving patterns. Their improved reproductive performance was not associated with differences in condition loss in early lactation.

The aim of this experiment was to quantify the milk production capacity of cows undergoing extended lactations while fed a pasture-based diet typical of those used in the seasonal-calving dairying systems of Victoria, Australia. One hundred twenty-five Holstein cows were randomly assigned to 1 of 5 groups. Breeding was progressively delayed after calving to enable management of the groups for lactation lengths of 10, 13, 16, 19, and 22 mo (equivalent to calving intervals of 12 to 24 mo). Cows were provided with a daily energy intake of at least 180 MJ of metabolizable energy/cow. This was supplied primarily by grazed pasture with supplementary cereal grain, pasture silage, and hay. Cows were dried off when milk volume fell below 30 kg/wk or when they reached 56 d before their expected calving date. Most cows (>96%) could lactate above this threshold for 16 mo, >80% for 19 mo, and >40% for 22 mo. There were negative relationships between lactation length and annual production of milk and milk solids (milk fat + protein), but losses were small until 16 mo. Annualized yields of milk solids were 497, 498, 495, 474, and 463 kg/cow for the 10, 13, 16, 19, and 22 mo groups, respectively. This reduction in annual production of milk solids with increasing lactation length was relatively less than for milk volume because during extended lactation, cows produced milk with higher concentrations of protein. Cows undergoing extended lactations also finished their lactations having gained more body weight and body condition than cows lactating for only 10 mo. The data showed that many cows on pasture-based diets were capable of lactating longer than the 10 mo that is standard for Victorian herds with seasonally concentrated calving patterns. Further, such extended lactations could be achieved with little penalty in terms of annual milk solids production.

Our study compared methane (CH4) emissions from lactating dairy cows measured using the sulfur hexafluoride (SF6) tracer and open-circuit respiration chamber techniques. The study was conducted using 16 lactating Holstein-Friesian cows. In each chamber, the cow was fitted with the SF6 tracer apparatus to measure total CH4 emissions, including emissions from the rectum. Fresh ryegrass pasture was harvested daily and fed ad libitum to each cow with a supplement of 5 kg of grain/d. The CH4 emissions measured using the SF6 tracer technique were similar to those using the chamber technique: 331 vs. 322 g of CH4/d per cow. The accuracy of the SF6 tracer technique was indicated by considering the ratio of the CH4 emission measured using the SF6 tracer to the emission measured using the chamber for each cow on each day. The calculated ratio of 102.3% (SE = 1.51) was not different from 100%. A higher variability within cow between days was found for the SF6 tracer technique [coefficient of variation (CV) = 6.1%] than for the chamber technique (CV = 4.3%). The variability among cows was substantially higher than within cows, and was higher for the SF6 technique (CV = 19.6%) than for the chamber technique (CV = 17.8%). Our CH4 emission data were compared with whole-animal chamber studies conducted in Canada and Ireland. In the Canadian study the SF6 technique did not measure CH4 emissions from the rectum and emissions were 8% lower than those measured using the chamber, indicating that emissions from the rectum may be greater than previously measured (1%). The relationship between CH4 emission and dry matter intake was examined for our data and for that reported in the Canadian study. There was a difference in the slopes of the regressions derived from our data and that from Canada; 17.1 vs. 20.8 g of CH4/kg of dry matter intake. A difference between the 2 locations was expected based on the difference in diet composition for these 2 studies. The SF6 tracer technique is reasonably accurate for inventory purposes and for evaluating the effects of mitigation strategies on CH4 emissions.

A powder formulation of live-attenuated measles vaccine is being developed for administration to the lungs. The safety and efficacy of the powder will be assessed by insufflation into cynomolgus monkeys. A Penn Century insufflator has been evaluated for powder dosing to the monkeys using an insulin formulation having similar physicochemical characteristics to the vaccine powder. Insulin pharmacokinetics were compared following dosing by powder insufflation, solution instillation into the trachea and subcutaneous injection. The insulin dosed to the lungs and trachea was more rapidly absorbed than that administered subcutaneously. Insulin bioavailability was greater from the inhaled powder than from the instilled solution. The findings confirm that the Penn Century device is suitable for vaccine powder dosing to the deep lung.
