RESUMO

Chemical investigation of the extract of the macroscopic fungus Beenakia informis led to the isolation of a previously unreported γ-pyrone and two new isoprenylated cyclohexanoids, together with speciocin N. Their structures were elucidated spectroscopically and the absolute configuration was determined by comparison of the experimental vs. calculated ECD curves. Three of the compounds showed very good to moderate activity against phytopathogenic fungi.

RESUMEN INTRODUCCIÓN: La pandemia de COVID-19 ha creado desafíos sin precedentes para los laboratorios de todo el mundo. Los gobiernos nacionales y subnacionales se vieron en la necesidad imperiosa de adaptar instituciones para la detección de SARS-CoV-2 en respuesta a la emergencia sanitaria declarada en 2020. Con el objetivo de aumentar la capacidad de diagnóstico en la provincia de Misiones, se implementó un laboratorio de diagnóstico de excelencia dentro del Instituto Misionero de Biodiversidad. Este artículo comparte los pasos realizados y los desafíos enfrentados, con el fin de que sirva de guía a otras instituciones. MÉTODOS: Las etapas para establecer el laboratorio fueron: adquisición de equipamientos/reactivos, adaptación/renovación del laboratorio, incorporación y capacitación del recurso humano, establecimiento de buenas prácticas y bioseguridad, selección de la metodología de extracción y detección, implementación de controles de calidad y habilitación. RESULTADOS: Desde su habilitación el Laboratorio de Análisis Integral procesó 1186 muestras biológicas de casos sospechosos de COVID-19 y se convirtió así en el segundo laboratorio de referencia provincial. DISCUSIÓN: La eficiente articulación entre el Instituto Misionero de Biodiversidad y el Ministerio de Salud de Misiones logró la rápida implementación de este laboratorio de alta complejidad en una región de importancia epidemiológica.


ABSTRACT INTRODUCTION: The COVID-19 pandemic has created unprecedented challenges for laboratories around the world. National and subnational governments faced the urgent need to adapt institutions for the detection of SARS-CoV-2 in response to the health emergency declared in 2020. With the aim of increasing diagnostic capacity in the province of Misiones, a diagnostic laboratory of excellence was established within the "Instituto Misionero de Biodiversidad". This article shares the steps taken and the challenges faced, in order to serve as a guide to other institutions. METHODS: The stages to establish the laboratory were: acquisition of equipment/reagents, adaptation/ renovation of the laboratory, incorporation and training of human resources, establishment of good practices and biosafety, selection of the extraction and detection methodology, implementation of quality controls and authorization. RESULTS: Since its authorization, the "Laboratorio de Análisis Integral" has processed 1186 biological samples from suspected cases of COVID-19, becoming the second reference laboratory in Misiones. DISCUSSION: The efficient articulation between the "Instituto Misionero de Biodiversidad" and the provincial Ministry of Health achieved the rapid implementation of this high complexity laboratory in a region of epidemiological importance.

Sugarcane bagasse is a natural source of phenolic compounds. However, these compounds are bound to lignocellulose components, reducing their ability to function as good antioxidants. These linkages are hydrolyzed by enzymes like ß-glucosidases, increasing free phenolics. Auricularia is a food-grade genus capable of producing ß-glucosidases. The aim of this work was (I) to determine naturally occurring species of Auricularia and (II) to obtain phenolic compounds through the solid-state bioprocessing of sugarcane bagasse. We have successfully isolated five strains that were assigned to the taxon A. fuscosuccinea. We determined ß-glucosidase activity by fluorescence plate assay of the five isolated strains and adjusted an optimal temperature for mycelial growth at 30 °C. A. fuscosuccinea LBM 243 was chosen for solid-state bioprocessing of sugarcane bagasse. ß-glucosidase activity (12.2 ± 0.62 U l-1) and protein content (51.58 ± 6.26 mg l-1) were highest on day 20 of culture. The maximum value of total phenolic content (507.5 ± 9.05 mg l-1) was obtained at day 20 and antioxidant capacity (34.44% ± 11.20) was highest at day 10, both in ethanolic extracts. The best performance of ethanol against methanol extraction in this work is highlighted considering ethanol to be a safe, efficient, and low-cost solvent.

Five new lanostanoid triterpenes were isolated from the extract of R. microporus. Three of the metabolites (1-3) present a Δ8,9 skeleton with an uncommon keto functionality at C-1. Another compound (4) has an unprecedented rearranged skeleton in which methyl-19 was transposed to C-1, with conjugated double bonds at Δ1-10 and Δ8-9. All of the compounds have hydroxylated or furane-cyclized side-chains. The structures were elucidated by spectroscopic methods, and the absolute configuration of the hydroxyl-bearing carbon in the side chain of compound 5 was established in silico. The metabolites were evaluated for their antifungal activity and the bioactivity as agonist/antagonists of the liver X receptors (LXRs). Compound 4 presents antifungal activity and compounds 3 and 5 are the agonists of LXRs.

Along many decades, protected environments were targeted by the scientific community for ecological research and for the collection of scientific information related to environmental aspects and biodiversity. However, most of the territory in hotspot regions with weak or even non legal protection has been left aside. These non-protected areas (NPA) could host high biodiversity values. This paper addresses how scientific effort on a NPA (CIAR) of 700 ha from the Atlantic Rain Forest, generates new information and tools for large-scale environmental and biodiversity management in NPAs. Information published during the last decade was summarized and complemented with subsequent novel data about biodiversity (new species, first records, DNA and chemical analyses, etc.). The results showed: 1 new genus (arachnid), 6 new species and several putative new species (fish and arthropod), 6 vulnerable species (bird and mammal) and 36 first records for Argentina (fish, arthropod, platyhelminth and fungi). When compared with protected natural areas of the same biome, the CIAR showed highly valuable aspects for fauna and environment conservation, positioning this NPA as a worldwide hotspot for some taxa. Indeed, when compared to international hotspots in a coordinated Malaise trap program, the CIAR showed 8,651 different barcode index numbers (∼species) of arthropods, 80% of which had not been previously barcoded. Molecules like Inoscavin A, with antifungal activity against phytopathogens, was isolated for the first time in Phellinus merrillii fungi. The study of major threats derived from anthropic activities measured 20 trace elements, 18 pesticides (i.e. endosulfans, chlorpyrifos, DDTs, HCHs) and 27 pharmaceuticals and drugs (i.e. benzoylecgonine and norfluoxetine) in different biotic and abiotic matrices (water, sediment, fish and air biomonitors). This integrated data analysis shows that biodiversity research in NPA is being undervalued and how multidisciplinary and multi-taxa surveys creates a new arena for research and a pathway towards sustainable development in emerging countries with biodiversity hotspots.

Six strains belonging to five species of Polyporus (P. arcularius, P. arcularioides, P. tricholoma, P. cfr. tricholoma, and P. varius), collected from an Atlantic Forest area in Misiones (Argentina), where species usually grow exposed to high temperatures and humidity, were identified by morphological and molecular analyses. P. tricholoma (BAFC 4536) and P. arcularioides (BAFC 4534) were selected by their lignin-degrading enzyme production, their ability to produce primordial of basidiomes under submerged fermentation, and the decrease in lignin content caused in Poplar wood (up to 29% after 45 days). Among several variables evaluated with a Plackett-Burman design (glucose, copper, vanillic acid and manganese concentration, incubation period, and light incidence), the most important factor affecting laccase and Mn-peroxidase (MnP) production by both strains, was light incidence. Light induced fruit body development but diminished laccase and MnP production. Moreover, a modified isoenzymatic laccase pattern was observed, showing additional isoenzymes when fungi were cultivated under darkness and differences in optimal temperature. Although the studied strains did not produce high laccase and MnP titers (uppermost detected 4230 and 90 U L-1 , respectively), their laccases showed thermal stability and optimal temperature above 70 °C, representing an interesting source in the search of thermo-tolerant enzymes for biotechnological applications.

Polyporus dictyopus, with a large number of heterotypic synonyms, has been traditionally considered a species complex, characterized by wide morphological variation and geographic distribution. Thus, neotropical specimens previously identified as P. dictyopus from Amazonia, Cerrado and Atlantic Forest biomes were studied based on detailed macro- and micromorphological examination and phylogenetic analyses, using distinct ribosomal and protein-coding genomic regions: the nuclear ribosomal internal transcribed spacer (nrITS), nuclear ribosomal large subunit (nrLSU), and RNA polymerase II second subunit (RPB2). Two unrelated generic lineages, each one represented by different species, are reported: Atroporus is recovered and re-circumscribed to include A. diabolicus and A. rufoatratus comb. nov.; Neodictyopus gen. nov. is proposed to accommodate N. dictyopus comb. nov. and two new species, N. atlanticae and N. gugliottae. Our study showed that at least five distinct species were hidden under the name P. dictyopus. Detailed descriptions, pictures, illustrations, and a key are provided for Atroporus and Neodictyopus species.

The aim of this work was to evaluate the potential of grape stalks, an agroindustrial waste, for growth and lignocellulolytic enzyme production via solid-state fermentation, using the following three white rot fungi: Trametes trogii, Stereum hirsutum and Coriolus antarcticus. The decolorization of several dyes by the above mentioned cultures was also investigated. Similar values of dry weight loss of the substrate were measured after 60 days (33-43 %). C. antarcticus produced the highest laccase and Mn-peroxidase activities (33.0 and 1.6 U/g dry solid). The maximum endoglucanase production was measured in S. hirsutum cultures (10.4 U/g), while the endoxylanase peak corresponded to T. trogii (14.6 U/g). The C. antarcticus/grape stalk system seems potentially competitive in bioremediation of textile processing effluents, attaining percentages of decolorization of 93, 86, 82, 82, 77, and 58% for indigo carmine, malachite green, azure B, remazol brilliant blue R, crystal violet and xylidine, respectively, in 5 h.

El objetivo de este trabajo fue evaluar el potencial del escobajo, un residuo agroindustrial, como sustrato para el crecimiento y la producción de enzimas lignocelulósicas de tres hongos causantes de pudrición blanca en la madera: Trametes trogii, Stereum hirsutum y Coriolus antarcticus. Para ello se utilizaron técnicas de fermentación en estado sólido. También se ensayó la decoloración de colorantes industriales sobre estos cultivos. La pérdida de peso seco del sustrato fue similar después del día 60 (33-43 %). C. antarcticus produjo las mayores actividades de lacasa y Mn-peroxidasa (33,0 y 1,6 U/g peso seco). La mayor actividad endoglucanasa fue medida en cultivos de S. hirsutum (10,4 U/g), y la mayor actividad endoxilanasa en T. trogii (14,6 U/g).
