RESUMO

The production of in vitro embryos has sped up the dissemination of superior genetic material. However, the variation among the cattle response to oocyte and embryo production is a challenging factor. This variation is even higher in the Wagyu cattle as the breed has a small effective population size. The identification of an effective marker related to reproductive efficiency would allow the selection of more responsive females to reproductive protocols. The objective of this study was to evaluate the blood levels of anti-Müllerian hormone and associate it with oocyte recovery and blastocyst rate of embryos produced in vitro in Wagyu cows, as well as observe the hormone circulating levels in males. Serum samples from 29 females with seven follicular aspirations and four bulls were used. AMH measurements were performed using the bovine AMH ELISA kit. A positive correlation was identified between oocyte production and blastocyst rate (r = 0.84, P = 9 × 10-9), and AMH levels with oocyte (r = 0.49, P = 0.006) and embryo (r = 0.39, P = 0.03) production. The mean levels of AMH were different between animals with low (11.06 ± 3.01) and high (20.75 ± 4.46) oocyte production (P = 0.01). Males showed high serological levels of AMH (3829 ± 2328 pg/ml) compared with other breeds. It is possible to use the serological measurement of AMH as a method to select Wagyu females with greater capacity for oocyte and embryo production. Further studies correlating AMH serological levels with Sertoli cell function in bulls are needed.

Housekeeping genes (HKG) are genes necessary for the maintenance of basal cellular functions, regardless of the specific roles within a tissue. It, therefore, is expected that these genes will maintain a relatively constant expression profile when there are varying physiological conditions. The identification of tissue specific reference genes is highly important for the normalization of gene expression profiles among different tissues. In this sow study, the objective was to identify stable reference genes in the uterine tissue and corpus luteum (CL), 6 days post-artificial insemination. The stability of ubiquitin (UBB), glyceraldehyde 3-phosphate dehydrogenase (GAPDH), DNA topoisomerase 2-beta (TOP2B), histone H3 (H3F3A) and hypoxanthine-guanine phosphoribosyltransferase 1 (HPRT1) abundances of mRNA were evaluated using the Bestkeeper technique. Briefly, total RNA was extracted for each tissue from 20 gilts (n = 20), processed by RT-qPCR and submitted to analysis using the Bestkeeper technique, which allowed for the evaluation of the consistency in abundance of mRNA for the reference genes. For all evaluated genes, the abundance of mRNA was relatively consistent in the uterine tissue, with the greatest abundance being for the GAPDH and TOP2B genes. The analysis of these genes in the CL, however, indicated there was a relatively greater variation of mRNA abundance for the various reference genes. Data suggest that UBB was the reference gene with the most consistent relative abundance of mRNA and that this gene could be used as a reference for corpora lutea analyses of mRNA.

In this paper, we have used two approaches to detect genetic associations with scrotal hernias in commercial pigs. Firstly, we have investigated the effects of runs of homozygosity (ROH) with the appearance of scrotal hernias, followed by a Genome Wide Association Study (GWAS). The phenotype classification was based on visual appearance of scrotal hernias. Each affected animal was matched to a healthy control from the same pen. In the total, 68 animals were genotyped using the Porcine SNP60 Beadchip, out of those, 41 animals had the presence of hernias and 27 were healthy animals. Fifteen animals were removed from the analysis due to differences in genetic background, leaving 18 healthy animals and 35 piglets with scrotal hernia. Further, the detection of extended haplotypes shared ROH were conducted for health (control) and affected (case) animals and a permutation test was used to test whether the ROH segments were more frequent in case/case pairs than non-case/case pairs. Using the ROH, we have identified an association (p = 0.019) on chromosome 2(SSC2) being segregated on animals with the presence of scrotal hernias. Using a GWAS, a region composed by 3 SNPs on the sexual chromosome X (SSCX) were associated with scrotal hernias (p < 1.6 × 10-5), this region harbors the Androgen Receptor Gene (AR).

Aims: this study sought to assess the reproductive performance of sows inseminated with sperm treated with homeopathic medicines. Materials and methods: the semen of 2 sexually mature boars age 18 months Pietrain and Duroc cross-bred with similar genetic and reproductive performance were chosen, as well as 125 sows. Sixteen samples of semen were collected and standardized through semen evaluation. Three homeopathic preparations and a placebo (control) were tested on the sperm (n=31/32 per group): Avena sativa 6cH, Pulsatilla nigricans 6cH and Avena sativa 6cH + Pulsatilla nigricans 6cH. Sows were inseminated 3 times with the same estrous diagnostic procedures. Results: there was significant difference (p<0.05) between Avena sativa 6cH and the other 3 groups regarding the return to estrus and parturition rate. There was no significant difference among the groups regarding the number of newborn piglets. Conclusion: these data suggest that homeopathic preparation Avena sativa may be used directly on sperm cells to improve the parturition rate in technified swine farms.(AU)


Objetivos: este estudo buscou avaliar o rendimento reprodutivo de suínos inseminados com esperma tratado com medicamentos homeopáticos. Materiais e métodos: o esperma de dois suínos sexualmente maduros de 18 meses de idade híbridos Pietrain e Duroc, com rendimento genético e reprodutivo similares foi selecionado, assim como 125 fêmeas. Dezesseis amostras de esperma foram coletadas e estandarizadas através de avaliação do esperma. Três preparações homeopáticas e um controle (placebo) foram testados no esperma (n=31/32 por grupo): Avena sativa 6cH, Pulsatilla nigricans 6cH and Avena sativa 6cH + Pulsatilla nigricans 6cH. As fêmeas foram inseminadas 3 vezes com os mesmos procedimentos de diagnóstico do estro. Resultados: houve diferença significativa (p< 0,05) entre Avena sativa 6cH e os outros 3 grupos no retorno ao estro e taxa de partos. Não houve diferença significativa entre os grupos no número de recém-nascidos. Conclusão: os dados sugerem que a preparação homeopática Avena sativa pode ser utilizada diretamente nas células espermáticas para melhorar a taxa de partos em fazendas tecnificadas de criação de suínos.(AU)
