RESUMO

The intensification of cytokine-producing activity by mouse bone marrow cells forming the hemopoiesis-inducing microenvironment (HIM) was demonstrated to be an obligatory component of adaptation processes developing in the body under the influence of different extreme factors (immobilization, inflammation, cytostatics, irradiation). As a rule, the first stress reaction of adhesive and nonadhesive cells consists in interleukin-1 and interleukin-3 secretion with following erythropoietic and colony-stimulating activity induction. For all this, the levels and periods of production of hemopoiesis humoral regulators are comparable under hemopoiesis-stimulating or -suppressing influences. Finally Activation of HIM functional properties expresses the interaction of central and local regulating systems, which favours the formation of an optimal response of the blood system to stress.

RESUMO

The pool of hematopoietic cells-precursors from the peripheral blood of patients with lung cancer stage III-IV was characterized by reduced number of circulating cells-precursors of granulomonocyto-(CFU-GM) and erythropoiesis (CFU-E). In the course of antitumor chemotherapy the counts of CFU-GM and CFU-E diminished 2 and 4-fold, respectively. The prednisolone functional test determined that depletion of CFU-GM and CFU-E pools in the peripheral blood was preceded with lowered pool reserve for hematopoietic precursors.

RESUMO

It was shown on the model of acute infections peritonitis in mice that the inflammation induced in the absence of mast cells was characterized by increased T-lymphocyte accumulation and macrophage-activating factor production and, in case of Thy-1,2(+)-lymphocyte elimination from bone marrow cell suspension, by earlier suppression of colony-stimulating and erythropoietic activity production by adhering and non-adhering myelokaryocytes. The results indicate that the modulatory effect of mast cells on haematopoiesis in inflammation is due in many respects to T-lymphocyte inhibition.

RESUMO

On the model of acute infectious peritonitis in mice, the inflammation was shown to accompany pronounced activation of bone marrow haemopoiesis. The increased number of committed precursor cells of erythro- and granulomonocytopoiesis, morphologically differentiated elements and enhancing colony-stimulating and erythropoietic activities of GIM cells origin were shown as well. Thy 1.2+ cells, migrating to bone marrow, were shown to play an important role in hemopoiesis stimulation inflammation. These stimulate the processes of myeloid precursor proliferation directly (by means of lymphokines) and in cooperation with monocytes-macrophages of GIM.

RESUMO

Acute infectious peritonitis simulated in mice was used to show that previous osmotic deterioration of the abdominal mast cell population significantly influenced the function of hemopoiesis-inducing microenvironment (HIM). The earlier production of the colony-stimulating and erythropoietic activities by the non-adhesive myelokaryocytes and the decreased production of these activities by the adhesive cells have been established. The results obtained evidence that mast cells modulate HIM mediating their influence on hemopoiesis under natural conditions of inflammation.

Marginal and bone marrow pools of neutrophil leucocytes were studied in 194 patients with lung cancer, stage III-IV. Reactions of neutrophil marginal pools (adrenaline test) were characterized by the development of neutrophil leucocytosis (NL) in the peripheral blood of 64.5% of cases (typical reaction), or by NL absence in 35.5% (perversed reaction). The prednisolone test conducted for evaluation of bone-marrow pool of neutrophils has revealed perversed leucocytic reaction in 39.6% of patients. Unstable marginal pool and decreased bone marrow neutrophil reserve were recorded in the course of polychemotherapy. Changes in the prednisolone test indices precede the development of neutropenia in the peripheral blood.

RESUMO

On the model of the acute infectious peritonitis in mice it is shown that the previously osmotic disruption of the peritoneal mast cell population remarkably affects the inflammatory focus, blood and bone-marrow leukocytic reactions. The initial neutrophil accumulation increase and the earlier and more intensive leukocytosis and activation of granulo-monocytopoiesis are established. At the same time the kinetics of monocytes in inflammatory focus is expressed weaker and testifies to the prolongation of the inflammatory response. Thus, in the natural inflammatory conditions mast cells of the inflammatory focus directly or indirectly modulate leukocytes and hematopoiesis.

RESUMO

A model of acute infectious peritonitis in mice demonstrated that the inflammation is attended by marked biphasic activation of bone-marrow granulomonocytopoiesis and that the activation is due in many respects to increased functional activity of elements forming the hemopoiesis-inducing microenvironment. This was suggested by increased colony-stimulating activity of the marrow mononuclear cells and the content of hemopoietic islets in the marrow. The colony-stimulating activity of peripheral blood also increased. It was established that inflammation is also characterized by activation of bone-marrow erythropoiesis, which is linked with increased erythropoietic activity of the hemopoiesis-inducing microenvironment and blood. There was a relation between the hemopoietic changes and the kinetics of leukocytes in the focus of inflammation.

A number of cytochemical characteristics and the NBT test were studied in neutrophilic granulocytes of 194 patients with diffuse pulmonary carcinoma (Stages III-IV), 31 patients with chronic nonspecific pulmonary diseases, and 20 normal subjects. Changes in the neutrophilic morphology and function were revealed in lung cancer patients, presenting as elevated alkaline phosphatase activity, reduced myeloperoxidase activity and lipid and glycogen levels, increased endogenous activation of the neutrophils in the NBT test, and decreased reaction activity in zymosan stimulation. Antitumor chemotherapy involved a lowering of the cationic protein level, as well of the acid phosphatase activity, and elevation of glycogen content. Stimulated NBT test was highly sensitive to cytostatic therapy. Tumor dissemination and morphologic variant contributed to changes in the neutrophilic morphology and function.
