Monolithic nonplanar ring oscillators (NPROs) under an applied magnetic field can operate unidirectional single-frequency lasing due to the loss differences among its four eigenpolarizations, where the minimum was empirically estimated to be 0.01%. However, this value has never been verified because the applied magnetic field is not uniformly distributed, making it hard to resolve both theoretically and experimentally. Here, we propose a method to resolve the applied magnetic field through an NPRO by combining finite-element analysis and experimental verification. By introducing the non-uniform magnetic field information to the eigenpolarization theory, the loss differences can be calculated by path integration along the optical path in the NPRO. The critical point, where the bidirectional lasing is emerging, is identified by the relative amplitude noise (RAN) of the laser and by the beating signal between the clockwise (CW) and counterclockwise (CCW) lasing. With this method, we determine that unidirectional operation is possible with loss differences as low as 0.0001% and 0.0003%, corresponding to two different NPRO designs with out-of-plane angles of 90° and 45°, respectively, which increases the precision of the loss differences for unidirectional single-frequency lasing by more than one order of magnitude. Our findings will greatly facilitate NPRO laser design with lowered magnetic field intensity requirements.

Monolithic nonplanar ring oscillators (NPROs) have achieved great success in industry, scientific applications and space missions due to their excellent narrow-linewidth, low-noise, high beam-quality, lightweight and compact performances. Here, we show that stable dual-frequency or multi-frequency fundamental-mode (DFFM or MFFM) laser can be stimulated directly by tunning pump divergence-angle and beam-waist injected to NPRO. The DFFM laser has a frequency deviation of one free spectral range of the resonator and thus can be utilized for pure microwave generation by common-mode-rejection. To demonstrate the purity of the microwave signal, a theoretical phase noise model is established, and the phase noise and the frequency tunability of the microwave signal are experimentally studied. Single sideband phase noise for a 5.7 GHz carrier is measured as low as -112 dBc/Hz at 10 kHz offset, and -150 dBc/Hz at 10 MHz offset in the free running condition of the laser, which outperforms its counterparts from dual-frequency Laguerre-Gaussian (LG) modes. The frequency of the microwave signal can be efficiently tunned through two channels, with frequency tunning coefficients of 15 Hz/V by piezo, and -60.5 kHz/K by temperature, respectively. We expect that such compact, tunable, low-cost and low-noise microwave sources can facilitate multiple applications including miniaturized atomic clocks, communication and radar, etc.

BACKGROUND: Pulmonary vein (PV) data are commonly measured on multiplanar image reformation (MPR) images and volume rendering (VR) images. PURPOSE: To compared and analyze the advantages and disadvantages of PV data based on VR images and MPR images. MATERIAL AND METHODS: A total of 94 patients with atrial fibrillation (AF) with imaging data were included in the study. The respective image postprocessing time and the three surgical interventionists' preferences for the two images were recorded. A paired t-test or chi-square test was used to compare their difference, and P < 0.05 was considered statistically significant. RESULTS: There was no statistically significant difference between the data values including the maximal and minimal ostial diameters of the left superior PV (LSPV), the left inferior PV (LIPV), the right superior PV (RSPV), and the right inferior PV (RIPV) obtained by VR and MPR images (P > 0.05). Yet, the mean postprocessing time of VR images (15.10 ± 3.05 min) was shorter compared to MPR images (16.54 ± 2.60 min) (t = 22.84, P < 0.05). All three surgical interventionists preferred VR images (accounted for 85.1%, 86.2%, and 84.0%, respectively), and there was no statistical difference in the degree of image preference among the three (chi-square = 0.596, P = 0.963). CONCLUSION: PV data measurement could be performed on both VR and MRP images; however, the data on VR images were more intuitive and more accessible for interventional surgeons.

The environmental pollution from microplastics has caused concern from the whole society due to its harm to organisms. However, the effect of microplastics on liver damage and fibrosis remains unclear in the case of long-term accumulation. The present study demonstrated that the 0.1 µm microplastic could enter hepatocytes from circulation and result liver damage even at a low concentration. Microplastic exposure could induce DNA damage in both nucleus and mitochondria, by which the dsDNA fragment was translocated into cytoplasm and triggered the DNA sensing adaptor STING. The activation of cGAS/STING pathway initiated the downstream cascade reaction, the NFκB translocated into nucleus and upregulated pro-inflammatory cytokines expression, and thus facilitating liver fibrosis eventually. Furthermore, inhibition of STING could alleviate the liver fibrosis via blocking the NFκB translocation and fibronectin expression. This study provided a valuable insight to elucidate the potential risk and mechanism of hepatic toxicity and fibrosis induced by microplastics.

Mid-infrared (mid-IR) lasers have great applications in bio-molecular sensing due to strong vibrational fingerprints in this wavelength range. However, it is a huge challenge to realize mid-IR lasers in conventional silica materials. Here, we demonstrate the generation of mid-IR Raman lasers and Kerr-frequency combs from an all-silica microresonator/fiber laser system. A single wavelength narrow-linewidth laser at ∼2 µm is first realized by using an ultrahigh Q-factor silica whispering-gallery-mode (WGM) microresonator as mode-selection mirror, and thulium-doped silica fiber as gain medium. Due to the strong intensity enhancement in the microresonator itself, multiple third-order nonlinear optical effects are observed, which include stimulated Stokes and anti-Stokes Raman scattering, and (cascaded) four-wave-mixing (FWM). The stimulated Stokes and anti-Stokes Raman scattering shift the initial 2 µm narrow-linewidth laser to as far as ∼2.75 µm and ∼1.56 µm, respectively. While the cascaded FWM helps to form a Kerr-frequency comb with a broad bandwidth of ∼900 nm and a mode spacing of twice of the microresonator free-spectral-range. This work offers a simple and effective route to realize all-silica mid-IR lasers based on enhanced optical nonlinearity in WGM microresonators.

PURPOSE: Ginkgolide B (GB) is a terpene lactone component found in Ginkgo biloba, which has a protective role on ischemia reperfusion (I/R) injury. This study was aimed at exploring the protective mechanism of GB on the myocardial I/R. PATIENTS AND METHODS: Myocardial I/R model was established on Sprague Dawley rats. The levels of cardiac troponin I, cardiac troponin T, lactic dehydrogenase, and myoglobin were determined by a 200FR NEO automatic biochemical analyzer. Histological examination was performed through HE and terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling staining. The expression levels of p-PERK, p-IRE1α, ATF6, p-AKT, and mTOR were detected by Western blot. RESULTS: The results exhibited that GB treatment suppressed the high levels of cardiac troponin I, cardiac troponin T, lactic dehydrogenase, and myoglobin and ameliorated the damaged and irregularly arranged myocardial cells induced by I/R injury significantly, indicating that GB could ameliorate myocardial I/R injury. Moreover, the high expression levels of endoplasmic reticulum (ER) stress key proteins caused by I/R injury were suppressed significantly by GB treatment, including p-PERK, p-IRE1α, and ATF6. GB treatment also decreased the number of apoptotic cells compared with I/R group. In addition, activation of ER stress by Tunicamycin treatment could counteract the protective effects of GB on I/R injury, suggesting that GB ameliorated myocardial I/R injury through inhibition of ER stress-induced apoptosis. Finally, the decreased p-AKT and p-mTOR expressions caused by I/R injury were upregulated by GB and inhibition of PI3K/AKT/mTOR pathway by LY294002 abolished the protective effects of GB on I/R injury, indicating that GB activated PI3K/AKT/mTOR pathway during I/R injury. CONCLUSION: GB protected against myocardial I/R injury through inhibiting ER stress-induced apoptosis via PI3K/AKT/mTOR signaling pathway.

OBJECTIVE: To explore the effect and mechanism of hyperoside on high glucose-induced oxidative stress injury of myocardial cells. METHODS: Oxidative stress injury of myocardial cells was simulated by treating with high glucose. Cells were divided into five groups: control group (5.5 mmol/L glucose); high glucose model group (35 mmol/L glucose); hyperoside protection groups (35 mmol/L glucose +4, 8, 20 nmol/L hyperoside). Cells were incubated for 48 h. The cell viability was detected by CCK-8. Apoptosis was measured through flow cytometry. The level of ROS was tested by Reactive Oxygen Species Assay Kit DCFH-DA with flow cytometer. The level of SOD and MDA was detected by SOD Assay Kit and MDA Assay Kit respectively. The protein levels of phosphatidylinositide 3-kinases (PI3K), protein kinase B (AKT), p-AKT, nuclear factor erythroid 2-related factor 2 (Nrf2) and p-Nrf2 were detected by Western blot. The activation of AKT was analyzed by immunofluorescence staining. RESULTS: Compared with control group, the cell viability, the levels of SOD, the expression of PI3K, the ratio of p-AKT/AKT and p-Nrf2/Nrf2 and the percentage of AKT positive cells in high glucose group were decreased with enhancive apoptosis and levels of ROS and MDA (P<0.05). Compared with high glucose group, the cell viability, the levels of SOD, the expression of PI3K, the ratio of p-AKT/AKT and p-Nrf2/Nrf2 and the percentage of AKT positive cells in hyperoside group (4, 8, 20 nmol/L) were increased with reduced apoptosis and levels of ROS and MDA (P<0.05). CONCLUSION: Hyperoside protects myocardial cells against oxidative stress injury via activation of PI3K/AKT/Nrf2 signal pathway.

Aim of this study was to investigate the effects of trimetazidine attenuating the myocardial ischemia-reperfusion injury to myocardium in rats and the underlying mechanisms. A model of myocardial ischemia reperfusion was established via ligating the left anterior descending coronary artery in 30 rats, and then they were randomly assigned to model group (n=10), low dose group (n=10) and high dose group (n=10). Moreover, additional 10 rats were collected and allocated to sham operation group, which was served as control group. Then, rats in the low dose group and high dose group were given trimetazidine with the dose of 10mg/kg and 30mg/kg respectively by intragastric administration, while rats in the control group and model group were given the equivalent volume saline. The dose was given once a day for consecutive 4 weeks in all rats. Echocardiography was applied to evaluate cardiac function, including left ventricular end-systolic dimension (LVESD), left ventricular end diastolic dimension (LVEDD) and left ventricular ejection fraction (LVEF). Next, myocardial tissue was collected, and Bax and Bcl-2 mRNA and the protein levels in the four groups were detected by RT-PCR and Western blot respectively. The level of malonaldehyde (MDA) and super oxide dismutase (SOD) activity in rat myocaridum in each group were detected by colorimetric methods, while the variables of apoptosis were measured by TUNEL methods. In comparison with the control group, LVEDD, LVEDS of rats increased significantly, LVEF decreased obviously, as well as Bax level, MDA level and the apoptotic variables in myocardial tissue increased (P<0.05), but Bcl-2 level and SOD activity decreased significantly in low dose, high dose and model group (P<0.05). Compared with model group, LVEDD, LVEDS of rats decreased obviously, LVEF increased significantly, as well as Bax level, MDA level and the apoptotic variables in myocardial tissue decreased (P<0.05), but Bcl-2 level and SOD activity increased significantly in low dose group, high dose group (P<0.05). The regulatory role of trimetazidine on above indicators of rats was in a dose-dependent manner. Trimetazidine can ameliorate rat myocardium following ischemia-reperfusion injury by effectively attenuating the injury from myocardial cell apoptosis; meanwhile, it can resist cell apoptosis through regulating Bax and bcl-2 expression, which exhibits guiding significance for the treatment of myocardial ischemia and reperfusion.

The protective effect and the mechanism of estrogen receptor ß (ERß) on myocardial infarction (MI) in mice were explored. A total of 12 female Tg-ERß transgenic mice and 12 non-transgenic littermate control (NLC) wild-type C57 mice were used for the present study. Both transgenic and wild-type mice had similar baseline data such as age, sex, and weight. The mouse model of MI was established by coronary artery ligation method, and the cardiac structure and function changes of the mouse were observed by ultrasonic echocardiography on days 1, 3 and 7 after the operation. RT-PCR method was used to detect the expression of collagen I, α-SMA, TGF-ß mRNA in the mouse heart, and Masson staining was used to detect cardiac fibrosis. At 3 days after operation, echocardiographic posterior wall thickness at end diastole (PWTD) and end systolic PWTS of Tg-ERß mice were significantly reduced, and left ventricular systolic diameter and left ventricular diastolic diameter significantly increased (P<0.05) compared with NLC mice. The levels of expression of Tg-ERß cardiac tissue collagen I, α-SMA, TGF-ß mRNA were significantly lower than those in the NLC mice (P<0.05). In conclusion, Tg-ERß exerts a protective effect on MI.

Hypoxia-induced cardiomyocyte apoptosis plays an important role in the development of ischemic heart disease. MicroRNAs (miRNAs or miRs) are emerging as critical regulators of hypoxia-induced cardiomyocyte apoptosis. miR-200c is an miRNA that has been reported to be related to apoptosis in various pathological processes; however, its role in hypoxia­induced cardiomyocyte apoptosis remains unclear. In the present study, we aimed to investigate the potential role and underlying mechanism of miR-200c in regulating hypoxia­induced cardiomyocyte apoptosis. We found that miR-200c was significantly upregulated by hypoxia in cardiomyocytes, as detected by reverse transcription-quantitative polymerase chain reaction (RT-qPCR). The lactate dehydrogenase, MTT, Annexin V/propidium iodide apoptosis and caspase-3 activity assays showed that downregulation of miR-200c markedly improved cell survival and suppressed the apoptosis of cardiomyocytes in response to hypoxia. Bioinformatics analysis and the dual-luciferase reporter assay demonstrated that miR-200c directly targeted the 3'-untranslated region of GATA-4, an important transcription factor for cardiomyocyte survival. RT-qPCR and western blot analysis showed that suppression of miR-200c significantly increased GATA-4 expression. Furthermore, downregulation of miR-200c upregulated the expression of the anti-apoptotic gene Bcl-2. However, the protective effects against hypoxia induced by the downregulation of miR­200c were significantly abolished by GATA-4 knockdown. Taken together, our results suggest that downregulation of miR-200c protects cardiomyocytes from hypoxia-induced apoptosis by targeting GATA-4, providing a potential therapeutic molecular target for the treatment of ischemic heart disease.

Endothelial cell injury plays a crucial role in the development and pathogenesis of cardiovascular disease. Vitexin is a natural flavonoid characterized by anti-oxidative and anti-inflammatory properties. The purpose of this study was to investigate the effects of vitexin on ox-LDL-induced endothelial dysfunction and to explore the underlying molecular mechanisms. In the present study, vitexin was found to play a protective role against ox-LDL-induced endothelial injury. Vitexin significantly promoted cell viability and inhibited apoptosis in ox-LDL-treated HUVECs. The up-regulation of cleaved Caspase-3, cleaved Caspase-9 and Bax induced by ox-LDL were inhibited by treatment with vitexin; meanwhile, the down-regulation of Bcl-2 was suppressed by vitexin. Pretreatment with vitexin was found to inhibit the ox-LDL-induced overexpression of IL-1ß, IL-6, TNF-α, E-selectin, ICAM1 and VCAM1. Moreover, vitexin reduced ox-LDL-induced oxidative stress by inhibiting the production of ROS and MDA, and by promoting the expression of SOD. Furthermore, we had shown that vitexin protected against the ox-LDL induced cell injury by activating autophagy. The protective effects of vitexin in ox-LDL-treated HUVECs were all reversed following treatment with the autophagy inhibitor 3-MA. In addition, we found that vitexin increased the expression of p-AMPK and decreased the expression of p-mTOR. The combination of the AMPK inhibitor Compound C plus vitexin significantly reversed the effects of vitexin in ox-LDL-treated HUVECs, such as the inhibition of autophagy, reduction in cell viability, increase in apoptosis and ROS production. In conclusion, these data suggest that vitexin ameliorates ox-LDL-mediated endothelial injury by inducing autophagy via AMPK signaling.

Stimulated Brillouin scattering (SBS) in a microcavity is usually realized by employing a wavelength tunable external cavity diode laser (TECDL) as the pump source. In this Letter, we report the observation of SBS in a high Q microcavity based on a TECDL-free scheme. The microcavity is employed as a mode-reflecting mirror for constructing a fiber-ring laser and, simultaneously, pumped by the fiber-ring lasing with intrinsic resonance latching. Several regimes are observed in a microcavity with a diameter of ∼215 µm, such as single lasing pumped SBS and multiple regular lasing pumped SBSs (single or cascaded). The microwave signals from the beat notes of the composite output lasing are measured with full-width at half-maximum on the scale of kilohertz at ∼11 and ∼22 GHz, indicating the high coherence between the pump and the Brillouin lasing.

We experimentally demonstrated a compact single-wall carbon-nanotube (SWNT)-based deep-red passively Q-switched Pr3+-doped ZBLAN all-fiber laser operating at 716 nm. A free-standing SWNT/polyvinyl alcohol composite film embedded between a pair of fiber connectors was employed as a saturable absorber (SA). The deep-red Q-switched operation is attributed to the combination of implementing a pair of fiber end-facet mirrors to achieve the linear laser resonator and incorporating a SWNT-SA into the cavity as a Q-switcher. Stable short-pulse generation with a duration of 2.3 µs was realized. When gradually increasing the incident pump power, the pulse repetition rate can be linearly tuned from 32.6 to 86.5 kHz, corresponding to a maximum average output power of 1.5 mW and the highest single-pulse energy of 18.3 nJ. To the best of our knowledge, this is the first demonstration of SWNT-based SA for a Q-switched laser at a deep-red wavelength ∼716 nm.

We propose and experimentally demonstrate a simple scheme for generating optical frequency combs (OFCs) in a fiber-ring/microresonator laser system. The ultrahigh Q whispering gallery mode microresonator is employed both as a mode reflection mirror to generate erbium lasing and as a Kerr-nonlinearity initiator that introduces optical parametric oscillation signals to form OFCs. By controlling the coupling position between the fiber taper and microresonator, optimizing the fiber polarization, as well as the pump power from a 974 nm laser diode (LD), versatile OFCs can be tuned out from single-wavelength states. The OFCs have single, multiple, or combined free spectral ranges. In addition, a Raman-gain-assisted OFC is also observed with a bandwidth of ∼230 nm. This LD-pumped and multifunctional laser system could find applications in precision spectroscopy, biochemical sensing, and optical fiber communication systems.

We demonstrate the first observation of stimulated Brillouin scattering (SBS) in a high-Q whispering gallery mode tellurite microsphere. Tellurite glass with composition of 70TeO2-20ZnO-5Na2O-5La2O3 (molar ratio) was prepared in-house using a melt-quenching technique. Moreover, tellurite microspheres with Q in excess of 13 millions at 1550 nm were fabricated by melting tellurite microwires using a CO2 laser. By pumping the tellurite microspheres with a tunable single frequency laser, SBS is further realized with a threshold as low as 0.58 mW. At last, the beat notes between the pump and the Stokes signals were measured, which indicated the Brillouin frequency shift is at the 8.2 GHz band for our tellurite glass. Our results could propel significant applications utilizing SBS by employing tellurite microspheres.

RESUMO

We experimentally demonstrate an ultralow-threshold cascaded Brillouin microlaser for tunable microwave generation in a high-Q silica microsphere resonator. The threshold of the Brillouin microlaser is as low as 8 µW, which is close to the theoretical prediction. Moreover, the fifth-order Stokes line with a frequency shift up to 55 GHz is achieved with a coupled pump power of less than 0.6 mW. Benefiting from resonant wavelength shifts driven by thermal dynamics in the microsphere, we further realized tunable microwave signals with tuning ranges of 40 MHz at an 11 GHz band and 20 MHz at a 22 GHz band. To the best of our knowledge, it was the first attempt for tunable microwave source based on the whispering-gallery-mode Brillouin microlaser. Such a tunable microwave source from a cascaded Brillouin microlaser could find significant applications in aerospace, communication engineering, and metrology.

The double transfer matrix method (DTMM) is proposed for calculating the eigenvalues of the resonant mode of a metallically coated dielectric rectangle resonator. Two-dimensional electromagnetic analyses are performed to investigate the optical influences induced by planar structure parameters. The results show that there theoretically exists a highest Q-factor resonance for both TE and TM modes at a certain length-width ratio with fixed resonant wavelength and resonator area. Due to the influence of surface plasma polaritons (SPPs) trapped at the corners of the resonator which is not considered in DTMM, the TM mode resonances are deformed and deviate severely from that of the analytical model. The geometric deformation on the resonator is introduced by replacing the four right angles with circular boundaries, and the SPP accompanied mode behaviors are corrected to the standing waves.

Plasmonic whispering gallery (WG) modes confined in metal-coated resonators are theoretically investigated by electromagnetic analyses. The resonance can be tuned from internal surface plasmonic WG modes to the hybrid state of the plasmonic mode by an introduced isolation layer. As the coated metal is reduced in size, the optical resonance is shifted out by the mode coupling of the internal and external surface plasmonic WG modes. Based on the optical leak of the plasmonic WG mode, the optical influences led by the surroundings with a variable refractive index are considered. Device performance criteria such as optical power leak, resonant wavelength shift, and threshold gain are studied. Full wave simulations are also employed and the results present good consistency with analytic solutions. The metal-coated resonator assisted by an active material is expected to provide promising performance as a refractometric sensor.

Calcitonin gene-related peptide (CGRP) is an important cardioprotective neuropeptide. Few studies have shown that calcium supplementation may increase CGRP levels transiently. However, the relationship between CGRP and calcium is poorly known. This study was to explore the correlation between serum calcium and CGRP in coronary artery disease (CAD), and observe whether short-term calcium/vitamin D supplementation would increase fasting serum CGRP. A randomized, placebo-controlled and double-blind clinical trial, and a supplementary study for further analysis of the correlations were conducted. The results showed that the correlation between serum calcium and CGRP was positive in CAD without myocardial infarction (MI) (r = 0.487, P = 0.029), but negative in acute and healing MI (r = -0.382, P = 0.003). Moreover, we found a positive correlation between lg (amino-terminal pro-B-type natriuretic peptide, NT-proBNP) and CGRP (r = 0.312, P = 0.027), but a negative correlation between lg (NT-proBNP) and serum calcium (r = -0.316, P = 0.025) in acute and healing MI. As to the clinical trial, participants subjected to CAD but without evolving or acute MI, together with blood calcium ≤ 2.4 mmol/L, were randomized into three groups. Among the groups of placebo, caltrate (600 mg elemental calcium; 125 IU vitamin D3, per tablet) 1 tablet/d and caltrate 2 tablets/d, there were no significant differences in baseline characteristics. After short-term (5 days) treatments, the results indicated that the effect of grouping was not statistically significant (P = 0.915). In conclusion, the correlations between serum calcium and CGRP in different types of CAD are inconsistent, and the main reason may be associated with elevated natriuretic peptides after acute MI. Further, our study shows that short-term calcium/vitamin D supplementation cannot significantly increase fasting serum CGRP levels.
