RESUMO

Cadmium is the most prevalent heavy metal pollutant in the environment and can be readily combined with micro/nanoplastics (M/NPs) to change their bioavailability. In the present study, we comprehensively investigated the effect of polystyrene (PS) NPs on dandelion plants grown under Cd stress. Cd exposure significantly inhibited the growth of dandelion seedlings, resulting in a decrease in seedling elongation from 26.47% to 28.83%, a reduction in biomass from 29.76% to 54.14%, and an exacerbation of lipid peroxidation and oxidative stress. The interaction between PS NPs and Cd resulted in the formation of larger aggregates, with the Cd bioavailability reduced by 12.56%. PS NPs affect ion absorption by regulating reactive oxygen production and increasing superoxide dismutase activity, thereby mitigating the adverse effects of Cd. PSCd aggregates induced significant changes in the metabolic profiles of dandelions, affecting various carbohydrates related to alcohols, organic acids, sugar metabolism, and bioactive components related to flavonoids and phenolic acids. Furthermore, based on a structural equation model, exposure to PSCd activated oxidative stress and nutrient absorption, thereby affecting plant growth and Cd accumulation. Overall, our study provides valuable insights into the effects of PS NPs on Cd bioavailability, accumulation, and plant growth, which are crucial for understanding the food safety of medicinal plants in a coexistence environment.

RESUMO

Biodegradable plastics, such as poly(butylene adipate terephthalate) (PBAT) and polylactic acid (PLA), are potential alternatives to conventional polyethylene (PE), both of which are associated with the production of microplastics (MPs). However, the toxicity of these compounds on medicinal plants and their differential effects on plant morphophysiology remain unclear. This study supplemented soils with MPs sized at 200 µm at a rate of 1% w/w and incubated them for 50 days to investigate the impact of MPs on the growth and metabolites of dandelion (Taraxacum mongolicum Hand.-Mazz.). The results demonstrated that the investigated MPs decreased the growth of dandelion seedlings, induced oxidative stress, and altered the activity of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase). Based on the comprehensive toxicity assessment results, the ecological toxicity was in the following order: PE MPs > PBAT MPs > PLA MPs. Metabolomics analyses revealed metabolic reprogramming in dandelion plants, leading to the enrichment of numerous differentially accumulated metabolites (DAMs) in the leaves. These pathways include carbohydrate metabolism, energy metabolism, and biosynthesis of secondary metabolites, suggesting that dandelions respond to MP stress by enhancing the activity of sugar, organic acid, and amino acid metabolic pathways. In addition, phenolic acids and flavonoids are critical for maintaining the balance in the antioxidant defense system. Our results provide substantial insights into the toxicity of biodegradable MPs to plants and shed light on plant defense and adaptation strategies. Further assessment of the safety of biodegradable MPs in terrestrial ecosystems is essential to provide guidance for environmentally friendly management.

RESUMO

Background: The value of semiquantitative resting myocardial perfusion imaging (MPI) in coronary artery disease (CAD) is limited. At present, quantitative MPI can be performed by a new cadmium zinc tellurium single-photon emission computed tomography (CZT-SPECT) scan. The quantitative index of resting myocardial blood flow (MBF) has received little attention, and its manifestations and clinical value in the presence of unstable coronary blood flow have not been clarified. Purpose: In patients with ST-segment elevation myocardial infarction (STEMI), whether resting MBF can provide additional value of blood flow than semi-quantitative resting MPI is not sure. We also explored the influencing factors of resting MBF. Methods: This was a retrospective clinical study. We included 75 patients with STEMI in the subacute phase who underwent resting MPI and dynamic scans after reperfusion therapy. General patient information, STEMI-related data, MPI, gated MPI (G-MPI), and resting MBF data were collected and recorded. According to the clinically provided culprit vessels, the resting MBF was divided into ischemic MBF and non-ischemic MBF. The paired Wilcoxon signed-rank test was used for resting MBF. The receiver operating characteristic (ROC) curves were used to determine the optimal threshold for ischemia, and multiple linear regression analysis was used to analyze the influencing factors of resting MBF. Results: There was a statistically significant difference between the ischemic MBF and non-ischemic MBF [0.59 (0.47-0.72) vs. 0.76 (0.64-0.93), p < 0.0001]. The ROC curve analysis revealed that resting MBF could identify ischemia to a certain extent, with a cutoff value of 0.5975, area under the curve (AUC) = 0.666, sensitivity = 55.8%, and specificity = 68.7%. Male sex and summed rest score (SRS) were influencing factors for resting MBF. Conclusion: To a certain extent, resting MBF can suggest residual ischemia after reperfusion therapy in patients with STEMI. There was a negative correlation between male sex, SRS, and ischemic MBF. A lower resting MBF may be associated with more severe myocardial ischemia.

RESUMO

Lasso peptides are a class of ribosomally synthesized and post-translationally modified peptides (RiPPs) defined by a macrolactam linkage between the N-terminus and the side chain of an internal aspartic acid or glutamic acid residue. Instead of adopting a branched-cyclic conformation, lasso peptides are "threaded", with the C-terminal tail passing through the macrocycle to present a kinetically trapped rotaxane conformation. The availability of enhanced bioinformatics methods has led to a significant increase in the number of secondary modifications found on lasso peptides. To uncover new ancillary modifications in a targeted manner, a bioinformatic strategy was developed to discover lasso peptides with modifications to tryptophan. This effort identified numerous putative lasso peptide biosynthetic gene clusters with core regions of the precursor peptides enriched in tryptophan. Parsing of these tryptophan (Trp)-rich biosynthetic gene clusters uncovered several putative ancillary modifying enzymes, including halogenases and dimethylallyltransferases expected to act upon Trp. Characterization of two gene products yielded a lasso peptide with two 5-Cl-Trp modifications (chlorolassin) and another bearing 5-dimethylallyl-Trp and 2,3-didehydro-Tyr modifications (wygwalassin). Bioinformatic analysis of the requisite halogenase and dimethylallyltransferase revealed numerous other putative Trp-modified lasso peptides that remain uncharacterized. We anticipate that the Trp-centric strategy reported herein may be useful in discovering ancillary modifications for other RiPP classes and, more generally, guide the functional prediction of enzymes that act on specific amino acids.

RESUMO

This study aimed to examine the effect of dietary cysteamine on yolk taurine content in hens during different egg production periods. In Exp. 1, China Agricultural University-3 (CAU-3) hens at the peak stage of egg production (aged 31 wks) were used to explore the effect of diets supplemented with 0.1% cysteamine on yolk taurine content, egg quality and production performance. In Exp.2, two breeds of hens (half Hy-Line Brown and half CAU-3 hens) at the late stage of egg production (68 wks) were used to investigate the influence of diets supplemented with 0, 0.02%, 0.04%, 0.08% or 0.10% cysteamine on yolk taurine content, egg quality, production performance and ovary development. In Exp.1, diets supplemented with 0.1% cysteamine significantly increased yolk taurine content (p < 0.05) without negative influence on production performance or egg quality. In Exp.2, the highest yolk taurine content was observed when cysteamine was supplemented at 0.08% (p < 0.001). However, supplemental cysteamine linearly or quadratically decreased production performance over the first few weeks of feeding, and the effects disappeared with continued feeding (p < 0.05). In conclusion, this study indicated that cysteamine supplementation benefits yolk taurine deposition in hens at both peak and late stage of egg production, but hens at the late stage of egg production show depressed production performance and egg quality.

RESUMO

Micro/nanoplastics (M/NPs) and phthalates (PAEs) are emerging pollutants. Polystyrene (PS) MPs and dibutyl phthalate (DBP) are typical MPs and PAEs in the environment. However, how dandelion plants respond to the combined contamination of MPs and PAEs remains unclear. In this study, we evaluated the individual and combined effects of PS NPs (10 mg L-1) and DBP (50 mg L-1) on dandelion (Taraxacum officinale) seedlings. The results showed that compared to control and individual-treated plants, coexposure to PS NPs and DBP significantly affected plant growth, induced oxidative stress, and altered enzymatic and nonenzymatic antioxidant levels of dandelion. Similarly, photosynthetic attributes and chlorophyll fluorescence kinetic parameters were significantly affected by coexposure. Scanning electron microscopy (SEM) results showed that PS particles had accumulated in the root cortex of the dandelion. Metabolic analysis of dandelion showed that single and combined exposures caused the plant's metabolic pathways to be profoundly reprogrammed. As a consequence, the synthesis and energy metabolism of carbohydrates, amino acids, and organic acids were affected because galactose metabolism, the citric acid cycle, and alanine, aspartic acid and glutamic acid metabolism pathways were significantly altered. These results provide a new perspective on the phytotoxicity and environmental risk assessment of MPs and PAEs in individual or coexposures.

RESUMO

Microplastic (MP) pollution is a critical environmental issue. Dandelions could be used as a biomonitor of environmental pollution. However, the ecotoxicology of MPs in dandelions remains unclear. Therefore, the toxic effects of polyethylene (PE), polystyrene (PS), and polypropylene (PP) at concentrations of 0, 10, 100, and 1000 mg L-1 on the germination and early seedling growth of dandelion were investigated. PS and PP inhibited seed germination and decreased root length and biomass while promoting membrane lipid peroxidation, increasing O2•-, H2O2, SP, and proline contents, and enhancing the activities of SOD, POD, and CAT. Principal component analysis (PCA) and membership function value (MFV) analysis indicated that PS and PP could be more harmful than PE in dandelion, especially at 1000 mg L-1. In addition, according to the integrated biological response (IBRv2) index analysis, O2•-, CAT, and proline were sensitive biomarkers of dandelion contamination by MPs. Here we provide evidence that dandelion has the potential to be a biomonitor to assess the phytotoxicity of MPs pollution, especially PS with high toxicity. Meanwhile, we believe that if dandelion is to be used as a biomonitor for MPs, attention should also be paid to the practical safety of dandelion.

RESUMO

Sitophilus zeamais (maize weevil) is one of the most destructive pests that seriously affects the quantity and quality of wheat (Triticum aestivum L.). However, little is known about the constitutive defense mechanism of wheat kernels against maize weevils. In this study, we obtained a highly resistant variety RIL-116 and a highly susceptible variety after two years of screening. The morphological observations and germination rates of wheat kernels after feeding ad libitum showed that the degree of infection in RIL-116 was far less than that in RIL-72. The combined analysis of metabolome and transcriptome of RIL-116 and RIL-72 wheat kernels revealed differentially accumulated metabolites were mainly enriched in flavonoids biosynthesis-related pathway, followed by glyoxylate and dicarboxylate metabolism, and benzoxazinoid biosynthesis. Several flavonoids metabolites were significantly up-accumulated in resistant variety RIL-116. In addition, the expression of structural genes and transcription factors (TFs) related to flavonoids biosynthesis were up-regulated to varying degrees in RIL-116 than RIL-72. Taken together, these results indicated that the biosynthesis and accumulation of flavonoids contributes the most to wheat kernels defense against maize weevils. This study not only provides insights into the constitutive defense mechanism of wheat kernels against maize weevils, but may also play an important role in the breeding of resistant varieties.

RESUMO

Elevated expression of c-Myc is associated with a variety of human cancers including clear cell renal cell carcinoma (ccRCC). Increasing evidence suggests that long noncoding RNAs (lncRNAs) are an important class of molecules that regulate both tumor initiation and progression. Here, we report the lncRNA c-Myc-induced regulator of ELF2 (MIRE) as a transcriptional target of c-Myc. MIRE functions as an oncogenic molecule in ccRCC by increasing ELF2 expression. Mechanistically, MIRE promotes phase separation of the RNA binding protein hnRNPK and facilitates the binding of hnRNPK to ELF2 mRNA, thereby resulting in the stabilization of ELF2 mRNA. Interestingly, MIRE is also under transcriptional control by ELF2, establishing an ELF2-MIRE positive feedback loop. Together, these findings provide new insights into the mechanisms by which c-Myc promotes tumorigenesis. They also implicate MIRE as an important regulator of ccRCC carcinogenesis.

RESUMO

Antibiotics have been widely used in animal husbandry, which leads to high risk of food-borne transfer of antibiotic resistance genes (ARGs). The present study investigated the distribution of ß-lactamase resistance genes (ß-RGs) on dairy farm in the Songnen Plain of western Heilongjiang Province, China, to provide mechanistic insights into food-borne transmission of ß-RGs through "meal-to-milk" chain under practically relevant circumstances. The results demonstrated that the abundance of ß-RGs (91%) was much higher than that of other ARGs in the livestock farms. The blaTEM exhibited the content as high as 94.55% among all ARGs, and higher than 98% blaTEM was detected in meal, water and milk sample. The metagenomic taxonomy analysis indicated that the blaTEM should be carried by tnpA-04 (7.04%) and tnpA-03 (1.48%) hosted in Pseudomonas genus (15.36%) and Pantoea (29.02%) genus. Both tnpA-04 and tnpA-03 in the milk sample were identified to be the key mobile genetic elements (MGEs) responsible for transferring blaTEM along the "meal-manure-soil-surface water-milk" chain. The ARGs transfer across ecological boundaries underscored the need to evaluate potential dissemination of high-risk Proteobacteria and Bacteroidetes carried by humans and animals. They were capable of producing expanded-spectrum ß-lactamases (ESBLs) and destroying commonly used antibiotics, leading to possible risk of food-borne horizontal transmission of ARGs. This study not only has important environmental implications for identifying the pathway for ARGs transfer, but also highlights the demand for appropriate policy toward safe regulation of dairy farm and husbandry products.

RESUMO

Purpose: Plant secondary metabolites are used to treat various human diseases. However, it is difficult to produce a large number of specific metabolites, which largely limits their medicinal applications. Many methods, such as drought and nutrient application, have been used to induce the biosynthetic production of secondary metabolites. Among these secondary metabolite-inducing methods, mechanical wounding maintains the composition of secondary metabolites with little potential risk. However, the effects of mechanical stress have not been fully investigated, and thus this method remains widely unused. Methods: In this study, we used metabolomics to investigate the metabolites produced in the upper and lower leaves of Catharanthus roseus in response to mechanical wounding. Results: In the upper leaves, 13 different secondary metabolites (three terpenoid indole alkaloids and 10 phenolic compounds) were screened using an orthogonal partial least squares discriminant analysis (OPLS-DA) score plot. The mechanical wounding of different plant parts affected the production of secondary metabolites. Specifically, when lower leaves were mechanically wounded, the upper leaves became a strong source of resources. Conversely, when upper leaves were injured, the upper leaves themselves became a resource sink. Changes in the source-sink relationship reflected a new balance between resource tradeoff and the upregulation or downregulation of certain metabolic pathways. Conclusion: Our findings suggest that mechanical wounding to specific plant parts is a novel approach to increase the biosynthetic production of specific secondary metabolites. These results indicate the need for a reevaluation of production practices for secondary metabolites from select commercial plants.

RESUMO

BACKGROUND: With improved resolution and sensitivity, the cadmium zinc telluride (CZT) detector measures myocardial blood flow (MBF) and myocardial flow reserve (MFR) via single photon emission computed tomography (SPECT). Recently, many studies have used vasodilator stress to obtain quantitative indexes. However, dobutamine used as a pharmaceutical stress has been rarely used to quantify myocardial perfusion using CZT-SPECT. Our study retrospectively analyzed the blood flow performance of 99mTc-Sestamibi (99mTc -MIBI) CZT-SPECT comparing dobutamine to adenosine. PURPOSE: The study aims to explore whether dobutamine stress can be used for the myocardial perfusion quantitative analysis via CZT-SPECT as well as compare dobutamine MBF and MFR to adenosine. METHODS: It was a retrospective study. A total of 68 patients with suspected or known coronary artery disease (CAD) were consecutively enrolled in this study. Thirty-four patients underwent dobutamine stress 99mTc-MIBI CZT-SPECT. Another thirty-four patients underwent adenosine stress 99mTc-MIBI CZT-SPECT. Patient characteristics, myocardial perfusion imaging (MPI) results, gated-myocardial perfusion imaging (G-MPI) results and quantitative analysis of MBF and MFR were collected. RESULTS: In dobutamine stress group, stress MBF was significantly higher than rest MBF (median [interquartile range], 1.63 [1.46-1.94] vs. 0.89 [0.73-1.06], P < 0.001). In adenosine stress group, similar results were observed (median [interquartile range], 2.01 [1.34-2.20] vs. 0.88 [0.75-1.01], P < 0.001). When comparing the dobutamine and adenosine stress group, global MFR showed significant differences (median [interquartile range], the dobutamine group: 1.88 [1.67-2.38] vs. the adenosine group: 2.19 [1.87-2.64], P = 0.037). CONCLUSION: MBF and MFR can be measured using dobutamine 99mTc -MIBI CZT-SPECT. In small sample single-center study, there was a difference in MFR produced by adenosine and dobutamine within the suspected or the known CAD population.

RESUMO

Ferns are one of the prevalent species of wild edible plants but one of the least explored terrestrial plants. Therefore, the aim of this study was to assess the nutrient composition, polyphenol profile and antioxidative properties of four wild edible ferns commonly utilized in northeastern China. We studied the content of ash, polysaccharide, protein, fat and mineral elements of the samples. Furthermore, the samples were found to have good total phenolic and total flavonoid contents and some level of antioxidant capacity as determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline) 6-sufonic acid (ABTS) and ferric reducing antioxidant power (FRAP). They also exhibited different specific accumulation of polyphenol profiles, estimated by liquid chromatography/mass spectrometry (LC/MS). Significance analysis revealed a significant correlation between individual phenolic compounds and the antioxidant activity of the ferns. The results of the study suggest that wild edible ferns are rich in nutritional value and have potential as a natural source of antioxidants.

RESUMO

BACKGROUND: Platelet lysate (PL) is considered as an alternative to fetal bovine serum (FBS) and facilitates the proliferation and differentiation of mesenchymal cells. OBJECTIVE: The aim of this study is to explore whether super activated platelet lysate (sPL), a novel autologous platelet lysate, has the ability to inhibit inflammation and promote cell proliferation, repair and osteogenesis as a culture medium. METHODS: Different concentrations of sPL on human fetal osteoblastic 1.19 cell line (hFOB1.19) proliferation and apoptotic repair were investigated; And detected proliferative capacity, inflammatory factor expressions and osteogenic differentiation of human dental pulp cells (hDPCs) stimulated by LPS under 10% FBS and 5% sPL mediums. RESULTS: sPL promoted hFOB1.19 proliferation and had repairing effects on apoptotic cells. No significant difference in proliferation and IL-1α, IL-6 and TNF-α expressions of hDPCs in FBS and sPL medium stimulated by LPS. hDPCs in sPL osteogenic medium had higher osteogenic-related factor expressions and ALP activity. LPS promoted osteogenic-related factor expressions and ALP activity of hDPCs in FBS osteogenic medium, but opposite effect showed in sPL medium. CONCLUSION: sPL promoted osteoblast proliferation and had restorative effects. Under LPS stimulation, sPL did not promote hDPCs proliferation or inhibit inflammation. sPL promotes osteogenic differentiation of hDPCs.

RESUMO

Coexisting salt and alkaline stresses seriously threaten plant survival. Most studies have focused on halophytes; however, knowledge on how plants defend against saline-alkali stress is limited. This study investigated the role of Taraxacum mongolicum in a Puccinellia tenuiflora community under environmental saline-alkali stress to analyse the response of elements and metabolites in T. mongolicum, using P. tenuiflora as a control. The results show that the macroelements Ca and Mg are significantly accumulated in the aboveground parts (particularly in the stem) of T. mongolicum. Microelements B and Mo are also accumulated in T. mongolicum. Microelement B can adjust the transformation of sugars, and Mo contributes to the improvement in nitrogen metabolism. Furthermore, the metabolomic results demonstrate that T. mongolicum leads to decreased sugar accumulation and increased amounts of amino acids and organic acids to help plants resist saline-alkali stress. The resource allocation of carbon (sugar) and nitrogen (amino acids) results in the accumulation of only a few phenolic metabolites (i.e., petunidin, chlorogenic acid, and quercetin-3-O-rhamnoside) in T. mongolicum. These phenolic metabolites help to scavenge excess reactive oxygen species. Our study primarily helps in understanding the contribution of T. mongolicum in P. tenuiflora communities on coping with saline-alkali stress.

RESUMO

The present study was conducted to investigate the dietary replacement of fish meal with poultry by-product meal (PBM) on the growth performance, immunity, antioxidant properties, and intestinal health of red swamp crayfish (Procambarus clarkia). A diet containing 20% fish meal (FM) and complex plant ingredients as the main protein resources was set as the FM group (crude protein 32%, crude lipid 6%). Four diets replacing 25%, 50%, 75%, and 100% fish meal of the FM diet with PBM were set as the PBM25, PBM50, PBM75, and PBM100 groups, respectively. Compared to the FM group, the PBM100 diet significantly decreased growth performance and feed utilization of crayfish, while markedly increasing the activity of serum aspartate aminotransferase. The immune response was depressed in crayfish fed the PBM100 diet as the activities of serum lysozyme and phenoloxidase, gene expression of anti-lipopolysaccharide factors (alf), cyclophilin A (cypa), crustin, and hemocyanin-1 (hep-1) in hepatopancreas were remarkably decreased. The activities of antioxidases and expression of antioxidant-relevant genes in the hepatopancreas were not influenced by PBM inclusion. Crayfish fed different diets exhibited no obvious symptoms of enteritis, but the PBM100 diet destructed intestinal morphology by significantly decreasing the average length of longitudinal ridges. The α-diversity and overall community structure were not significantly influenced but variations were found in the relative abundance of some genera by PBM inclusion. In summary, CAP could successfully replace 75% dietary FM in a basal diet containing 20% fish meal, while higher CAP level compromised growth performance, immunity, and intestinal histology of crayfish.

RESUMO

With the proposal of China's high-quality development strategy, how to promote regional stability and coordinated development based on a deep understanding of the main contradictions and changes in China's society has become the focus of research. High-quality development is a brand-new coordinated development concept, which aims to optimize the economic structure, transform the development model, enhance the development momentum, and take innovation as the primary driving force. How to promote the coordinated development of this region has become a hot issue considered by scholars. The Beijing-Tianjin-Hebei region is the capital economic circle of China, and the purpose of this study is to promote the coordinated and stable development of the region. On this premise, this paper firstly adopts the composite Grey Lotka-Volterra (GLV) model and Fractional GM(1,1) (FGM(1,1)) model to research Water Resources system-Economic System-Industrial System-Technology Innovation System in the Beijing-Tianjin-Hebei region. Secondly, by analyzing the research data, it is found that the relationship between the system is very complex, and the stability calculation results are all below 0. Then, the analysis of the research results shows that there is no obvious coordination among the three regions, and they have not yet reached a state of mutual promotion and stable and coordinated development. Finally, four suggestions are put forward for the coordinated development of the Beijing-Tianjin-Hebei region. This can not only provide direction for the future development of the region but also have reference significance for the development of other regions. Further, accelerate the coordination and unity of all factors of production in China and promote China's development at a deeper and higher level.

RESUMO

In this work, a novel method of solid sample pretreatment technique of bismuth fire assay (Bi-FA) combined with solid sample determination by laser ablation ICP-MS (LA-ICP-MS) was reported for the determination of ultra-trace Pt and Pd in geochemical samples. Bismuth oxide (Bi2O3) was used as fire assay collector to directly enrich Pt and Pd from solid samples, and Ag protection cupellation was employed to generate Ag granules. After cleaning, weighing and annealing, the Ag granules were compressed into thin slices and determined by LA-ICP-MS for 195Pt, 105Pd and 109Ag (109Ag was selected as the internal standard isotope). Bi2O3 provided exceptionally low blanks compared to nickel oxide and lead oxide commonly employed in fire assay procedures, and could be applied directly without purification. Different from traditional empirical coefficient method, the Chinese Certified Reference Materials (CRMs) for Pt and Pd were treated by the same procedure to obtain completely matrix matched Ag slices. And then modified empirical coefficient method and internal standard calibration strategy was used to reduce the instability of LA-ICP-MS, and random multipoint laser ablation was employed to further reduce analytical variation resulting from heterogeneity of Pt and Pd in the Ag slice. Under optimal conditions, excellent calibration curves for Pt and Pd were obtained (0.407-2958 µg g-1 and 0.407-2636 µg g-1, respectively), with correlation coefficients exceeding 0.9996. The method detection limits for Pt and Pd were 0.074 and 0.037 ng g-1, respectively. The established method was applied successfully to analysis of real geochemical samples, with determined values in good agreement with the results of traditional Pb-FA graphite furnace atomic absorption spectrometry (GF-AAS), and spiked recoveries between 87.8 and 125.0%.

RESUMO

Purpose: To evaluate the effect of super-activated platelet lysate (sPL) on wound healing of tooth extraction sockets in rats. Methods: Rat models of the tooth extraction socket were established. Thirty-six rats were divided into control and sPL groups and sacrificed on days 7, 14, and 28 after tooth extraction. Bone formation in tooth extraction sockets were observed by microscopic computed tomography (micro-CT) and hematoxylin and eosin (HE) staining; osteoprotegerin (OPG), receptor activator of nuclear factor kappa-Β ligand (RANKL), interleukin 6(IL-6), and tumor necrosis factor-alpha (TNF-α) proteins were detected by immunohistochemistry; and chemokine and osteogenic gene expressions were detected by polymerase chain reaction (PCR). Results: sPL accelerated soft tissue wound healing in the extraction socket of rats. Micro-CT showed that the amount of bone formation and bone volume fraction were higher in the sPL group than the control 14 days after extraction. HE staining showed promotion of the formation of bony trabeculae by sPL in the apical third of the extraction socket 7 days after extraction and more mature and organized bony trabeculae in the sPL group than the control 14 days after extraction; mature bony trabeculae filling most of the fossa with lesser bone porosity in the socket in the sPL group than the control 28 days after extraction. Immunohistochemistry showed that sPL induced OPG expressions 7 and 14 days after tooth extraction but did not affect the RANKL expression while transiently promoting the IL-6 expression 7 days after extraction. PCR showed that sPL promoted chemokine expressions 7 and 14 days after extraction. The expressions of osteogenesis-related factors were higher in the sPL group than the control 7 and 28 days after extraction, while the opposite trend was observed 14 days after extraction. Conclusion: sPL has a transient pro-inflammatory effect and promotes soft tissue healing and bone formation during early wound healing of extraction sockets in rats.

RESUMO

BACKGROUND: Biological and abiotic stresses such as salt, extreme temperatures, and pests and diseases place major constraints on plant growth and crop yields. Fatty acids (FAs) and FA- derivatives are unique biologically active substance that show a wide range of functions in biological systems. They are not only participated in the regulation of energy storage substances and cell membrane plasm composition, but also extensively participate in the regulation of plant basic immunity, effector induced resistance and systemic resistance and other defense pathways, thereby improving plant resistance to adversity stress. Fatty acid desaturases (FADs) is involved in the desaturation of fatty acids, where desaturated fatty acids can be used as substrates for FA-derivatives. OBJECTIVE: In this paper, the role of omega-FADs (ω-3 FADs and ω-6 FADs) in the prokaryotic and eukaryotic pathways of fatty acid biosynthesis in plant defense against stress (biological and abiotic stress) and the latest research progress were summarized. Moreover' the existing problems in related research and future research directions were also discussed. RESULTS: Fatty acid desaturases are involved in various responses of plants during biotic and abiotic stress. For example, it is involved in regulating the stability and fluidity of cell membranes, reactive oxygen species signaling pathways, etc. In this review, we have collected several experimental studies to represent the differential effects of fatty acid desaturases on biotic and abiotic species. CONCLUSION: Fatty acid desaturases play an important role in regulating biotic and abiotic stresses.
