Currently, a trend toward the commercialization of dromedary milk associated with recent intensive rearing systems has starting worldwide which impose constraints on animals affecting their behavioral repertoires and welfare status. The aim of this study was to investigate the effects of dam parity and calf sex on parturition, neonatal, and maternal behaviors in stabled Maghrebi dairy camels (Camelus dromedarius). Thirty-six (primiparas N = 10; multiparas N = 26) periparturient females were kept under video surveillance using a digital IR camera and 24-h sessions were chosen to assess calving, maternal, and neonatal behaviors in calving pens. Duration of restlessness, process of giving birth, and expulsion of fetal membranes were assessed. After first suckling, dams and their calves were moved into an individual enclosure to assess maternal behavior at 12 h, 24 h, 48 h, 72 h, and 7d postpartum. Behavior was assessed using a camcorder for 30 min after 1 h of young separation in an adjacent enclosure. Results showed an effect of parity on the duration of the birth process which was longer for primiparous than multiparous camels (P = 0.034). During this stage, primiparous females tended to raise their tails more often (P = 0.054) and spent more time standing (P = 0.001) than multiparous camels. Neonatal behavior was affected by calf sex. Female newborns took less time to raise their heads (P = 0.041) and to stand up (P = 0.048) for the first time and were the earliest to suckle their dams (P = 0.032). Multiparous dams stood up sooner (P = 0.019) after calving and suckled their calves earlier (P = 0.043) than primiparous dams. They emitted more bleats and exhibited more sniffing during the first week postpartum than primiparas. During the first postpartum week, both primiparas and multiparas showed a decrease in the total number of bleats (P < 0.001), low-pitch bleats (P < 0.001), and high-pitch bleats (P < 0.001), in sniffing frequency (P < 0.001) and sniffing duration (P < 0.001). This is the first study to describe in detail the calving, maternal, and neonatal behaviors of dromedary camels and to show the influence of parity and calf sex. Maternal care toward the newborn calf exhibited by a high level of bleating and sniffing activities in the first 2 days suggest that they play an important role in the establishment of mother-young relationship.

Although sheep are known to be seasonal breeders and give birth in winter, not all of them follow this trend. A few breeds can be mated and give birth all year round, meaning that mothers and neonates will have to face contrasting climatic conditions. The aim of this study was to test whether lambing season affects maternal and neonatal behaviors in D'man sheep. During four different lambing seasons (winter, autumn, summer and spring), periparturient ewes (n = 111) and their lambs (n = 213) were kept under 24-h-video surveillance in order to record postpartum behaviors. Mother-young preference was tested around 48 h after parturition. Lamb vigor was studied by the determination of birth weight, early postnatal behavior and rectal temperature at birth and 48 h later. Litter expulsion time was not affected by lambing season, but birth weight was biased against summer and winter born lambs. Ewes provided a higher intensity of care to their offspring in winter: latency for grooming was shorter and time spent grooming was longer compared to lambing in spring and summer (P = 0.01 in all cases). On the other hand, lambs were the most active in spring as they were faster to extend their hind legs (P = 0.01), stand up (P = 0.04) and reach the udder (P = 0.04). Rectal temperature at 48 h was affected by season of birth (P < 0.001) with higher values observed in summer. Glycemia variation between birth and 48 h was the lowest in spring born lambs and plasma levels increased less in spring born lambs than in winter (P < 0.0001), autumn (P < 0.0001) and summer born lambs (P < 0.0001). In the choice test, mothers clearly preferred their own young and no season effect was detected except that in the first minute of the test they spent less time near their own young in winter than in the other seasons (P = 0.04). Lambs also chose their mother successfully without any major effect of the season however, but winter born lambs were the least vocal (P = 0.01). Overall, this study show that maternal care, lamb behavior and vigor vary lightly according to seasons, albeit not in a consistent manner. In conclusion, a season is no more detrimental than another for the onset of mother-young relationships.

In prostate carcinogenesis, expression and/or activation of the Transient Receptor Potential Melastatin 8 channel (TRPM8) was shown to block in vitro Prostate Cancer (PCa) cell migration. Because of their localization at the plasma membrane, ion channels, such as TRPM8 and other membrane receptors, are promising pharmacological targets. The aim of this study was thus to use nanocarriers encapsulating a TRPM8 agonist to efficiently activate the channel and therefore arrest PCa cell migration. To achieve this goal, the most efficient TRPM8 agonist, WS12, was encapsulated into Lipid NanoCapsules (LNC). The effect of the nanocarriers on channel activity and cellular physiological processes, such as cell viability and migration, were evaluated in vitro and in vivo. These results provide a proof-of-concept support for using TRPM8 channel-targeting nanotechnologies based on LNC to develop more effective methods inhibiting PCa cell migration in zebrafish xenograft.

The camel industry uses traditional (i.e., iron brands and ear tags) and modern (i.e., microchips) identification (ID) systems without having performance results of reference. Previously iron-branded ( = 45; 1 yr) and microchipped ( = 59; 7 yr) camels showed problems of healing (8.6% of brands) and reading (only 42.9% of brands and 69.5% of microchips were readable), which made their use inadvisable. With the aim of proposing suitable ID systems for different farming conditions, an on-field study was performed using a total of 528 dromedaries at 4 different locations (Egypt, = 83; Spain, = 304; Saudi Arabia, = 90; and Tunisia, = 51). The ID devices tested were visual (button ear tags, 28.5 mm diameter, = 178; double flag ear tags, 50 by 15 mm, = 83; both made of polyurethane) and electronic (ear tags, = 90, and rumen boluses, = 555). Electronic ear tags were polyurethane-loop type (75 by 9 mm) with a container in which a 22-mm transponder of full-duplex technology was lodged. Electronic boluses of 7 types, varying in dimensions (50 to 76 mm length, 11 to 21 mm width, and 12.7 to 82.1 g weight) and specific gravity (SG; 1.49 to 3.86) and each of them containing a 31-mm transponder of half-duplex technology, were all administered to the dromedaries at the beginning of the study. When a low-SG bolus was lost, a high-SG bolus was readministered. Readability rates of each ID system were evaluated during 1 to 3 yr, according to device and location, and yearly values were estimated for comparison. On a yearly basis, visual ear tag readability was not fully satisfactory; it was lower for rectangular ear tags (66.3%) than for button ear tags (80.9%). Yearly readability of electronic ear tags was 93.7%. Bolus readability dramatically varied according to their SG; the SG < 2.0 boluses were fully lost after 8 mo. In contrast, the SG > 3.0 boluses were efficiently retained (99.6 to 100%) at all locations. In conclusion, according to the expected long lifespan of camels, low ID performances were observed for iron brands, injectable microchips, and ear tags (visual and electronic), making their use inadvisable as unique ID systems in camels. The high readability of dense electronic boluses recommended their use as a permanent ID device of reference in camels.

Metalloprotease-processed CD95L (cl-CD95L) is a soluble cytokine that implements a PI3K/Ca(2+) signaling pathway in triple-negative breast cancer (TNBC) cells. Accordingly, high levels of cl-CD95L in TNBC women correlate with poor prognosis, and administration of this ligand in an orthotopic xenograft mouse model accelerates the metastatic dissemination of TNBC cells. The molecular mechanism underlying CD95-mediated cell migration remains unknown. Here, we present genetic and pharmacologic evidence that the anti-apoptotic molecules BclxL and Bcl-2 and the pro-apoptotic factors BAD and BID cooperate to promote migration of TNBC cells stimulated with cl-CD95L. BclxL was distributed in both endoplasmic reticulum (ER) and mitochondrion membranes. The mitochondrion-localized isoform promoted cell migration by interacting with voltage-dependent anion channel 1 to orchestrate Ca(2+) transfer from the ER to mitochondria in a BH3-dependent manner. Mitochondrial Ca(2+) uniporter contributed to this flux, which favored ATP production and cell migration. In conclusion, this study reveals a novel molecular mechanism controlled by BclxL to promote cancer cell migration and supports the use of BH3 mimetics as therapeutic options not only to kill tumor cells but also to prevent metastatic dissemination in TNBCs.

RESUMO

The hydrochemical and isotopic investigation of the Nefzaoua aquifer system demonstrates that groundwater mineralization in is controlled by natural and anthropogenic processes including water-rock interaction and irrigation return flow. It identifies all of the water bodies that flow within the aquifer system and their circulation patterns. The isotopically depleted paleowaters, identified within the deep and intermediate aquifers, undergo significant enrichment by evaporation during irrigation and recharged the shallow aquifer by return flow. Subsequently, they infiltrate to the intermediate aquifer which receives also rainfall modern recharge.

Lasiodiplodia is a common pathogen causing dieback, gummosis, or root necrosis on the three most important fruit crops in Oman and the United Arab Emirates (UAE): date palm (Phoenix dactylifera), Citrus spp., and mango (Mangifera indica). A study was conducted to examine diversity in 64 Lasiodiplodia isolates infecting date palm (24), Citrus (11), and mango (29) in Oman and the UAE. Identification based on sequences of the internal transcribed spacer (ITS) rDNA and EF1α gene showed that date palm isolates belonged to L. hormozganensis (75% of isolates) and L. theobromae (25%); Citrus isolates belonged to L. hormozganensis (45%), L. theobromae (45%), and L. iraniensis (10%); and mango isolates belonged to L. theobromae (59%), L. iraniensis (34%), and L. hormozganensis (7%). Amplified fragment length polymorphism (AFLP) fingerprinting of the 64 isolates using four primer pair combinations produced 64 genotypes and 972 polymorphic alleles. Cluster analysis separated the isolates into four clusters representing the three species. A higher level of genetic diversity was observed in L. iraniensis (0.3105) compared to L. hormozganensis (0.2503) and L. theobromae (0.2331) in Oman. Analysis of molecular variance (AMOVA) indicated the existence of low levels of genetic differentiation among date palm populations of L. hormozganensis obtained from Oman and the UAE (FST = 0.025) and among populations of L. hormozganensis (0.0485) and L. theobromae (0.0703) from date palm, Citrus, and mango. These findings imply a high rate of movement of L. hormozganensis and L. theobromae isolates among date palm, Citrus, and mango and between the two countries. Findings from the pathogenicity test supported the AMOVA analysis and suggested a lack of host specialization in L. hormozganensis, L. iraniensis, and L. theobromae on date palm, acid lime, and mango. Although this is the first record of L. hormozganensis and L. iraniensis in Oman, the relatively moderate level of genetic diversity in the two species compared to L. theobromae suggests that the two species have been in Oman for a long time but misidentified by morphology and ITS rDNA sequences as L. theobromae. This study is also the first record of date palm and acid lime as natural hosts for L. hormozganensis and the first record of L. hormozganensis in the UAE. The diversity in Lasiodiplodia species affecting date palm, Citrus, and mango in Oman and the UAE should be taken into consideration when planning future management programs for diseases caused by these pathogens.

Effects of 4 different milking intervals (8, 12, 16, and 24 h) on milk yield and milk composition were studied in Tunisian Maghrebi dairy dromedaries (n = 6) at late lactation [240 +/- 14 days in milk (DIM), 5.84 +/- 1.62 L/d]. Camel-cows suckled their calves for 2 mo, were hand milked while suckling until mo 4 of lactation (calf weaning) and machine milked thereafter. Intravenous injection of oxytocin was administered before machine milking at each experimental milking to induce complete milk ejection and to avoid carryover effects of milking intervals. Cisternal and alveolar milk were measured at 380 +/- 16 DIM for a 24-h milking interval. Milk accumulated logarithmically (R(2) = 0.95) in the udder from 8- to 24-h milking interval without reaching a plateau. Consequently, milk secretion rate decreased exponentially (R(2) = 0.93) according to milking interval. Compared with 12-h milking interval (6.1 L/d), estimated daily milk yield was 113, 87, and 70% for 8-, 16-, and 24-h intervals, respectively. Total milk solids, milk fat content, and milk pH decreased with increasing milking interval, showing the greatest value at 8-h intervals (14.1 +/- 0.4%, 4.6 +/- 0.5%, and 6.66 +/- 0.05, respectively) and the lowest at 24-h intervals (12.3 +/- 0.9%, 2.9 +/- 0.6%, and 6.54 +/- 0.02, respectively). Milk protein (3.9 +/- 0.1%), lactose (4.5 +/- 0.2%), ash (0.84 +/- 0.01%) and density (1.028 +/- 0.01) remained constant for all milking intervals. Milk K, Ca, and Mg contents increased as milking interval increased, but Na content did not change (0.06 +/- 0.01%, on average). Milk Na:K ratio tended to decrease from 0.35 (1:2.9) to 0.22 (1:4.5) for the extreme milking intervals. Plasma lactose concentration steadied from 8- to 16-h (67 +/- 32 micromol) but increased dramatically at 24-h intervals (338 +/- 118 micromol), indicating that mammary tight junctions became permeable after 24 h of milk accumulation. Camel udders showed small cisterns (19.3% of total milk in the udder at 24 h) when compared with other dairy animals; we recommend the use of prestimulation for machine milking and selection for larger udder cisterns. Alveolar milk contained more fat (5.16 vs. 1.75%; SEM, 0.39%) and protein (3.23 vs. 2.73%; SEM, 0.15%) than cisternal milk. Despite the increase of plasma lactose during tight junction leakiness, the tendency for the Na:K ratio to decrease may be indicative of a camel's specific regulatory mechanism for controlling Na and K concentrations in milk and delaying the inhibitory effect of milk stasis on milk secretion rate. In conclusion, this short-term study proved the low storage capacity of the Tunisian Maghrebi camel udder but also showed their moderate ability to adapt to extended milking intervals at late lactation.

Browsing camels have access to different plant species in relation to the kind of pasture they can reach. A study was conducted in an arid region of Southern Tunisia to determine the dietary preference and forage quality of free ranging camels. Foliage consisting of leaves from Limonium pruinosum, Retama raetam and Stipa tenacissima, was collected during the dry season to evaluate the chemical characteristics and nutritional value of these browse fodder species. The dietary preference was studied using 15 adult camels which were selected from a herd of 50 animals appropriately marked for identification. There was a significant difference in the chemical composition and nutritional value of plant species collected. Based on crude protein (CP) content and nutritional value, the three fodder species browsed can be recommended as good-quality food source for camels under pastoral management.

BACKGROUND: The role of two recently identified polyomaviruses, KI and WU, in the causation of respiratory disease has not been established. OBJECTIVES: To determine the prevalence of KI and WU viruses (KIV and WUV) in 371 respiratory samples and evaluate their contribution to respiratory disease. STUDY DESIGN: Specimens were screened for KIV and WUV using single, multiplex or real time PCR; co-infection with other respiratory viruses was evaluated. RESULTS: Of the 371 samples analysed, 10 (2.70%) were positive for KIV and 4 (1.08%) were positive for WUV yielding an overall case prevalence of KIV and WUV infection of 3.77%. KIV and WUV were identified in patients aged<15 years (11 patients) with upper or lower respiratory tract infection and >45 years (3 patients) with upper respiratory tract infection. Co-infections were found in 5 (50%) and 3 (75%) of the KIV and WUV positive samples, respectively. CONCLUSIONS: This study supports previous conclusions that KIV and WUV detection in the respiratory tract may be coincidental and reflect reactivation of latent or persistent infection with these viruses. The age distribution of KIV and WUV infection in this study mirrors that found for the other human polyomaviruses, BK and JC.

OBJECTIVES: The present study was undertaken to assess the differential impact of insulin resistance, leptin and body composition on myocardial mass and serum markers of cardiac fibrosis in obese subjects, within a small range of elevated BMI (30-40 kg/m(2)), without pulmonary disease, cardiovascular disease, hypertension, cardiac hypertrophy or other cardiovascular disease. BACKGROUND: Obesity is an independent predictor of left ventricular mass (LVM) and is associated with disturbances in cardiac structure. The extent of the interstitial fibrosis in obese patients is not known, especially in the absence of cardiac hypertrophy. METHODS AND RESULTS: We included 160 obese subjects. The LVM was obtained using the Devereux formula. Body composition was estimated from a total body scan. Insulin sensitivity was assessed by homeostasis model assessment (HOMA), and cardiac collagen turnover by measurement of procollagen type III aminopeptide (PIIINP). PIIINP was correlated to the E/A ratio (r=0.24; P=0.012), a marker of ventricular function. PIIINP was independently correlated with glucose concentration (r=0.27; P=0.004), indexes of insulin resistance (HOMA (r=0.27; P=0.003), insulin (r=0.24; P=0.008)), and parameters associated with the insulin-resistance syndrome (HDL-cholesterol r=-0.27; P=0.004) and fat trunk/fat leg ratio (r=0.24; P=0.053)). The variable most correlated with PIIINP was HDL-cholesterol, followed by HOMA (r (2)=0.13). When HOMA was substituted for blood glucose concentration and insulinemia (Model 2), HDL-cholesterol was strongly related to lower PIIINP levels, followed by higher glucose concentration (r (2)=0.21). Regression analyses showed that LVM had the strongest independent positive correlation with fat-free mass (FFM) (r=0.39; P=0.0002), followed by systolic blood pressure (r=0.19; P=0.034). Neither adipose mass nor height independently added information to multivariate models. The ratio leptin/fat mass was correlated with LVM (r=-0.27; P=0.004), but not independently of the FFM. Markers for fibrosis were not significantly correlated with LVM. As a result, FFM was the most predictive factor of LVM in obese subjects. CONCLUSION: We found that serum levels of markers of cardiac collagen synthesis were significantly associated with insulin resistance in normotensive, nondiabetic obese subjects, and not related to the LVM. As a result, PIIINP could be a very early marker of ventricular dysfunction in these patients. Furthermore, we suggest that, for better detection of left ventricle hypertrophy in obese subjects, LVM should be indexed to FFM rather than to body surface area, or height.

RESUMO

The metabolism in mammalian is regulated by multiple levels of hormone action, with complex feedback and control mechanisms. The somatotropic axis, essentially consisting of growth hormone (GH), insulin-like growth factors (IGF-I and -II), their associated carrier proteins, and receptors, plays a key role in the control of the regulation of metabolism and physiological process. Among this axis, other hormones like insulin, leptine, glucocorticoids or thyroid hormones are involved in this mechanism by modulating GH and/or IGF-I synthesis and availability. This review summarizes the complexity of the regulation of the metabolism by the somatotropic axis using different examples such as special nutritional situations or growth promoters administration.

This paper presents data on the occurrence and pattern of inherited bleeding syndromes (IBS) in Jordan, a hitherto unexplored problem. In 1978, during the first 12 months of a prospective study at a major medical center, 91 patients from 51 families were diagnosed as having IBS. All patients were referred because of moderate-to-severe bleeding diatheses; they included 52 hemophiliacs, 27 patients with von Willebrand's disease, 4 with hemophilia B (IX-deficit), 2 with afibrinogenemia, 1 with prothrombin deficiency, and 4 were thought to have platelet dysfunction. The clinical and laboratory features of the patients observed in Jordan do not seem to be significantly different from those of patients in Western Europe or North America.
