RESUMO

On the basis of real-world clinical data, the study aimed to explore the effect and mechanisms of the treatment plan of "traditional Chinese medicine (TCM) regulating liver regeneration." A total of 457 patients with HBV-related liver failure were retrospectively collected. The patients were divided into three groups: the modern medicine control group (MMC group), patients treated with routine medical treatment; the control group combining traditional Chinese and Western medicine (CTW), patients treated with routine medical treatment plus the common TCM formula; and the treatment group of "TCM regulating liver regeneration" (RLR), patients treated with both routine medical treatment and the special TCM formula of RLR. After 8 weeks of treatment, the mortality of patients in the RLR group (12.31%) was significantly lower than those in the MMC (50%) and CTW (29.11%) groups. Total bilirubin level significantly decreased and albumin increased in the RLR group when compared with the MMC and CTW groups (P < 0.05). In addition, there were significant differences in the expression of several cytokines related to liver regeneration in the RLR group compared with the MMC group. RLR treatment can decrease jaundice, improve liver function, and significantly reduce the mortality in patients with HBV-related liver failure. The mechanism may be related to the role of RLR treatment in influencing cytokines related to liver regeneration.

RESUMO

The establishment of an animal model of liver regeneration in a state of neuro-endocrine-immune network dysfunction is an urgent need to promote the research on the regulatory mechanism of liver regeneration. The monosodium glutamate (MSG)-rat-liver regeneration model is a composite model of liver regeneration which combines the MSG-rat model with the model of partial hepatectomy (PHx) or hepatotoxicity (for example, CCl4), and it is mainly used to investigate liver regeneration and its regulatory mechanism in the state of neuro-endocrine-immune network dysfunction, mechanisms and drug screening of liver-brain syndrome, and mechanisms of disease-syndrome combination and drug efficacy. Dysregulated liver regeneration process is observed in the MSG-rat-liver regeneration model established by the MSG-rat model and PHx, which is significantly different from the liver regeneration process in normal rats after major hepatectomy. The MSG-rat-liver regeneration model established by the MSG-rat model and hepatotoxicity has a significantly higher degree of liver fibrosis than normal rats with CCl4-induced liver fibrosis. The hypothalamic-pituitary-hepatic axis is an important mechanism of the influence of the external environment on liver regeneration, and the imbalance of epithelial-mesenchymal transition/mesenchymal-epithelial transition is an important mechanism of the influence of intrahepatic microenvironment on abnormal liver regeneration (liver fibrosis). The research on MSG-rat-liver regeneration model preliminarily reveals the efficacy and mechanism of traditional Chinese medicine in the prevention and treatment of liver and related diseases (including liver-brain syndrome) by influencing the neuro-endocrine-immune-liver regeneration regulatory network and promotes the research on the combination of diseases and syndromes. The establishment and application of the MSG-rat-liver regeneration model has important scientific significance and application value in further exploration of the complex mechanism of liver regeneration and the mechanism of traditional Chinese medicine in regulating liver regeneration.

RESUMO

Objective Study on the absorption, distribution, transformation, excretion and toxicity of four forms of arsenic in rats. Methods Four thousand and fifty data were obtained from 28 days animal metabolism experiments of 81 rats exposed to the four As species.Distribution, metabolism, excretion, and subacute toxicity of 4 As species were compared by analyzing the arsenate [As(V)],arsenite [As(Ⅲ)],methyl arsenate (MMAV) and dimethyl arsenate (DMAV) content of rat feces (excrement and urine),blood, seven organs, and liver and kidney pathology. Results After oral administration of each As species,82.9% of As(Ⅲ),85.1% of As(V),95.0% of MMAV and 96.2% of DMAV were accumulatively secreted via feces and urine, while 16.2% of As(Ⅲ),14.1% of As(V),4.65% of MMAV and 0.120% of DMAV were detected in blood. The DMAV accumulated in blood and urine after dosing As(Ⅲ),As(V) and MMAV,and the content of DMAV in the blood was 8 times greater than urine.Four kinds of arsenic had effects on rat liver and kidney function, the influence on liver was acute phase effect while that on renal was cumulative effects. The effect of As(Ⅲ) was the most significant, followed by MMAV.Four kinds of arsenic could cause tissue deformation and inflammatory cells infiltration, but showed no difference between groups. Conclusion The distribution, metabolism, excretion, and toxicity of the four kinds of arsenic in rats were different in varying degrees. The research of health risk for arsenic should consider its different forms.

RESUMO

Liver fibrosis is regarded as a form or status of liver regeneration disorder. Therefore, from a new perspective of regulating liver regeneration, we put forward a new strategy for the prevention and treatment of liver fibrosis. This strategy not only focuses on confrontational reverse regulation, but also emphasizes positive induction. Moreover, this strategy applies an integrated method of promotion and inhibition, which improves liver injury and regeneration and corrects the abnormal liver regeneration and the normal liver regeneration disorder. Furthermore, this strategy also advocates the combination of prevention and treatment, which prevents the occurrence of liver fibrosis or a disorder of liver regeneration and treats the rapid progression of liver fibrosis. The imbalance between epithelial-mesenchymal transition (EMT) and mesenchymal-epithelial transition (MET) is involved in the CCl4-induced liver fibrosis in rats. Diwu Yanggan capsules can delay the progression of liver fibrosis and attenuate the degree of fibrosis, which is associated with the regulation of liver regeneration by modulating the imbalance between EMT and MET (inhibiting EMT and/or promoting MET).

RESUMO

Objective: To research the effects of Zuogui Pill on the gene expression profile of regenerate hepatic tissue in MSG-liver regeneration-rat.Methods: MSG-liver regeneration-rat models were copied;therapy group were given Zuogui Pill(5g/kg) by intragastric administration.Rats were executed at the 5th day after operation, and the regenerate liver tissues were frozen by liquid nitrogen.1176 gene chips of gene expression profile correlated to cell differentiation and multiplication were adopted to observe the changes of gene expression profile in the regenerate liver tissues. Results: There were 292 differential expression genes in therapy group compared with model group(20 genes were up-regulation expressed and 272 genes were down regulation expressed). Conclusion: Zuogui Pill has significant effects on the gene expression profile of regenerate hepatic tissue in MSG-liver regeneration-rat,and most of the genes are down regulation expressed. The result hints that Zuogui Pill could influence the liver regeneration by regulating the gene expressions of MSG-live regeneration-rat.Down regulation of the gene expressions of regenerate hepatic tissue in MSG-liver regeneration-rat possibly is one of the important molecular mechanisms of Zuogui Pill in nourishing liver and kidney.
