RESUMO

Parasitic dinoflagellates of the genus Hematodinium are known to infect various marine crustaceans worldwide, especially crabs and several species of shrimp and lobster. Some of these species are new host species and components of commercial fishery products. These parasitic species are predominantly found in the hemolymph of the host and cause pathological changes and functional damage to organs and tissues, leading to death. In recent years, these parasites have infected important commercially valuable species, particularly in European waters, US waters, Australian waters, and recently in Shandong Peninsula in China. These Hematodinium pathogens were also reported to affect wild shrimp in Chinese waters and in the English North Sea. These rapid spreads affect crustacean aquaculture industries, where they are indeed a significant threat to the sustainability of the aquaculture of important crustaceans. The fishery products industries are also under pressure from the invasion of this pathogen, as the crab meat produced has a bitter taste, which may reduce its marketability. In response to these threats, this review was aimed at providing a broader understanding of the development of parasite distribution and ecological aspects of Hematodinium. In addition, the interaction of these pathogens with their hosts, the environmental drivers of Hematodinium disease, and future research perspectives were discussed.

This study evaluated the histopathological changes in the gill, liver and kidney of African catfish (Clarias gariepinus) intoxicated with a sub-lethal dose of Melaleuca cajuputi leaves extract (MCLE) for 96 h. The acute toxicity test has been determined previously with a value of 96-h LC50 = 127 mg/L, hence the selection of sub-lethal ranges from 60 mg/L to 160 mg/L of MCLE. Degenerative alterations were prominent in all tested organs, particularly after exposure to a high concentration of MCLE. Gill exhibited haemorrhage, epithelial lifting, lamellar disorganisation, and necrosis after exposure to a high MCLE concentration. Alterations in the liver include congestion, hydropic degeneration, and vacuolation, whereas lesions in the kidney were pyknosis, vacuolation, hydropic degeneration, and tubular necrosis. The obtained data showed that the organs experienced severe changes proportional to the increase in MCLE concentration. In addition, fish exposed to higher concentrations than the LC50 value experienced irreversible lesions. The present study suggests that the use of MCLE below the LC50 is recommended to avoid severe alterations to organs, particularly in African catfish. This study demonstrated that the use of MCLE above the LC50 promotes severe damage to the gills, liver and kidney of African catfish. However, further investigations are needed to define the causing-mechanisms underlying these effects.

Gold nanoparticles (AuNPs) have emerged as a promising application in aquaculture. Their nano-sized dimensions, comparable to pathogens offer potential solutions for combating antibiotic resistance. In this study, AuNPs were synthesized by using polychaetes, Marphysa moribidii as the bio-reducing agent. Modifications were made to reduce agglomeration in green-synthesized AuNPs through ultrasonication. The antibacterial activities of AuNPs against V. parahaemolyticus were evaluated. The physicochemical characteristics of the green synthesized AuNPs were comprehensively investigated. The successful formation of AuNPs was confirmed by the appearance of a red ruby colour and the presence of surface Plasmon resonance (SPR) absorption peaks at 530 nm as observed from UV-vis spectroscopy. Scanning electron microscopy (SEM) revealed spherical-shaped AuNPs with some agglomerations. Transmission electron microscopy (TEM) showed particle size of AuNPs ranging from 10 nm to 60 nm, meanwhile dynamic light scattering (DLS) analysis indicated an average particle size of 24.36 nm. X-ray diffraction (XRD) analysis confirmed the high crystallinity of AuNPs, and no AuNPs were detected in the polychaetes extracts prior to synthesis. A brief ultrasonication significantly reduced the tendencies for AuNPs to coalesce. The green-synthesized AuNPs demonstrated a remarkable antibacterial efficacy against V. parahaemolyticus. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) tests revealed that a concentration of 0.3 g/ml of AuNPs effectively inhibited V. parahaemolyticus. These findings highlighted the potential of green-synthesized AuNPs as antibacterial agents for the prevention and management of AHPND in aquaculture.

<b>Background and Objective:</b> Studies on plant herbs as alternatives to chemical anaesthetics in fish species are numerous, but little is known on crustaceans. A study was conducted to investigate the efficacy of <i>C. citratus</i> Essential Oil (EO) on the induction and recovery of <i>M. rosenbergii</i>. <b>Materials and Methods:</b> The <i>C. citratus</i> EO was obtained by hydrodistillation and analyzed using GC-MS. The prawns were exposed to <i>C. citratus</i> EO and clove oil in 100-1000 and 200-1000 µL L<sup>1</sup>, respectively. Different stages of induction and recovery times were recorded. <b>Results:</b> In GC-MS, citral (78.47%) was detected as a major compound in <i>C. citratus</i> EO. Prawns reached loss equilibrium at 500-1000 µL L<sup>1</sup> <i>C. citratus</i> EO within 15.55-6.52 min. Exposure of prawn to <u><</u>500 µL L<sup>1</sup> <i>C. citratus</i> EO resulted in a high survival rate (100-94%). In clove oil, all tested concentrations caused significant induction in <i>M. rosenbergii</i> within 20.61-6.47 min. Recovery time and survival rate were significantly decreased with the increase of EO concentrations. The regression model showed the induction time in both anaesthetic agents was dependent on the concentration (R<sup>2 </sup>= 0.86-0.96). The recovery time of <i>C. citratus</i> EO-exposed prawn was dependent on the concentrations (R<sup>2 </sup>= 0.59). <b>Conclusion:</b> The study shows the potentiality of <i>C. citratus</i> EO as a natural anaesthetic in <i>M. rosenbergii</i>, although not as efficient as clove oil.

BACKGROUND AND OBJECTIVE: Effects of Cymbopogon citratus essential oil (EO) was tested on minimizing handling stress in Macrobrachium rosenbergii through the evaluation of their metabolite responses [glucose, lactate, glycogen, protein, Lactate Dehydrogenase (LDH), Malate Dehydrogenase (MDH), Acetylcholinesterase (AChE) and Alanine Aminotransferase (ALT)]. This study aimed to investigate the efficacy of C. citratus extract in the anaesthetization of M. rosenbergii. MATERIALS AND METHODS: Three treatments including control, prawn exposed to stress alone (T1) and prawn exposed to stress in the presence of C. citratus EO (T2) were tested. A C. citratus EO at 500 µL L-1 had been determined in a previous study and was selected as the critical dose to be applied as an anesthetic agent. Handling stress was induced into prawns by netting, at 2 min interval for 30 min and their hemolymph were collected to determine the metabolite responses. RESULTS: The increase of glucose, lactate and LDH of M. rosenbergii when exposed to handling stress alone (T1) in comparison to T2 (stress with anesthetic C. citratus EO) were identified. Further, a low glycogen level in parallel with low AChE activity was observed which indicates the involvement of secondary metabolites to cope with the energy demand in T1 over T2. CONCLUSION: This study indicates the efficiency of C. citratus EO to reduce stress during handling in M. rosenbergii.

Infection by the rhizocephalan parasite Sacculina beauforti can have detrimental effects on mud crab Scylla olivacea. However, the molecular changes that occur during rhizocephalan infection are poorly understood. Due to the disruption in the reproductive system after infection, the gonadal transcriptomic profiles of non-infected and infected Scylla olivacea were compared. A total of 686 and 843 unigenes were differentially expressed between non-infected and infected males, and females, respectively. The number of DEGs increased after infection. By comparing shared DEGs of non-infected and infected individuals, potential immune- and reproduction-related of host, and immune- and metabolism-related genes of parasite are highlighted. The only shared KEGG pathway between non-infected and infected individuals was the ribosome pathway. In summary, findings in this study provide new insights into the host-parasite relationship of rhizocephalan parasites and their crustacean hosts.

This data article presents on the ectoparasites infestation on small mammals in Peninsular Malaysia. The dataset on ectoparasites infestation is important because it raises a major medical concern regarding the spread of potentially zoonotic disease from wildlife to human. Tick and chigger are the primary ectoparasites as reservoirs of vector-borne diseases found on small mammals in Malaysia. These small mammals that are infested with ectoparasites occupy various types of habitats, including human settlements, could be of community health risks as the carriers of potentially zoonotic diseases. Field samplings were conducted from February 2015 to February 2016 in three different ecological habitats of mixed dipterocarp forest, coastal forest and insular forest, in Terengganu, Malaysia. A total of 35 and 22 species of bats and rodents respectively were captured and examined for ectoparasites. Twenty-three species of bats and 16 species of small mammal were recorded as hosts for at least one species of ectoparasites. These findings show that the highest ectoparasite infestation occurred on bat community.

AIM: The main objective of this study was to emphasize on histopathological examinations and molecular identification of Streptococcus agalactiae isolated from natural infections in hybrid tilapia (Oreochromis niloticus) in Temerloh Pahang, Malaysia, as well as to determine the susceptibility of the pathogen strains to various currently available antimicrobial agents. MATERIALS AND METHODS: The diseased fishes were observed for variable clinical signs including fin hemorrhages, alterations in behavior associated with erratic swimming, exophthalmia, and mortality. Tissue samples from the eyes, brain, kidney, liver, and spleen were taken for bacterial isolation. Identification of S. agalactiae was screened by biochemical methods and confirmed by VITEK 2 and 16S rRNA gene sequencing. The antibiogram profiling of the isolate was tested against 18 standard antibiotics included nitrofurantoin, flumequine, florfenicol, amoxylin, doxycycline, oleandomycin, tetracycline, ampicillin, lincomycin, colistin sulfate, oxolinic acid, novobiocin, spiramycin, erythromycin, fosfomycin, neomycin, gentamycin, and polymyxin B. The histopathological analysis of eyes, brain, liver, kidney, and spleen was observed for abnormalities related to S. agalactiae infection. RESULTS: The suspected colonies of S. agalactiae identified by biochemical methods was observed as Gram-positive chained cocci, ß-hemolytic, and non-motile. The isolate was confirmed as S. agalactiae by VITEK 2 (99% similarity), reconfirmed by 16S rRNA gene sequencing (99% similarity) and deposited in GenBank with accession no. KT869025. The isolate was observed to be resistance to neomycin and gentamicin. The most consistent gross findings were marked hemorrhages, erosions of caudal fin, and exophthalmos. Microscopic examination confirmed the presence of marked congestion and infiltration of inflammatory cell in the eye, brain, kidney, liver, and spleen. Eye samples showed damage of the lens capsule, hyperemic and hemorrhagic choroid tissue, and retina hyperplasia accompanied with edema. Brain samples showed perivascular and pericellular edema and hemorrhages of the meninges. Kidney samples showed hemorrhage and thrombosis in the glomeruli and tubules along with atrophy in hematopoietic tissue. Liver samples showed congestion of the sinusoids and blood vessel, thrombosis of portal blood vessel, and vacuolar (fatty) degeneration of hepatocytes. Spleen samples showed large thrombus in the splenic blood vessel, multifocal hemosiderin deposition, congestion of blood vessels, and multifocal infiltration of macrophages. CONCLUSION: Therefore, it can be concluded that pathological changes in tissues and organs of fish occur proportionally to the pathogen invasion, and because of their high resistance, neomycin and gentamicin utilization in the prophylaxis or treatment of S. agalactiae infection should be avoided.

A parasitological survey of 16 pangolins, confiscated from the Department of Wildlife and Nature Park Peninsular Malaysia (DWNP) at Kelantan and Pulau Pinang, Malaysia was conducted in 2011. Amblyomma javanense (family: Ixodidae) was the only ectoparasite found on the pangolins. The prevalence, intensity and life cycle of A. javanense were observed together with the respective pangolins' age and sex. It was found that 68.8% of the pangolins were infected, and significant difference, χ(2)(1, N=16)=4.02, p=0.05 were observed with males higher in infestation (88.9%) as compared to the females (42.9%). However, the mean intensity was higher on females (72) as compared to males (31.6). In addition, significant difference, χ(2) (2, N=16)=6.73, p=0.05 was recorded between adults and juveniles with juveniles found to be 100% infected as compared to adult (63.6%). Nevertheless, the mean intensity was slightly higher on adults (47) than juveniles (35). Adult ticks were found in higher numbers as compared to the nymph and larvae with number of male ticks higher (236) as compared to the females (53). Similarly, a high significant difference χ(2)(2, N=469)=203.47, p=0.05 was recorded in the composition of the tick's life stages with a higher number of adult ticks (61.6%) followed by nymph (30.3%) and larvae (8.1%). As such, the results of this study revealed a picture of the A. javanense life cycle which is related to the age and gender of the Malayan Pangolin.

RESUMO

A new species of Dermoergasilus Ho & Do, 1982 is described from freshwater fish hosts in the south west of Western Australia. D. occidentalis n. sp. differs from previously described species in the genus principally by the armature of the legs. The new species was found on the gills of the freshwater cobbler Tandanus bostocki Whitely and western minnow Galaxias occidentalis Ogilby in two different river systems.
