RESUMO

Background: Selenium (Se) and zinc (Zn) are essential antioxidant enzyme cofactors. Foliar Se/Zn application is a highly effective method of plant biofortification. However, little is known about the effect of such applications on the concentration of trace elements and phytochemicals with pro-oxidant or antioxidant activity in pea (Pisum sativum L.). Methods: A 2-year pot experiment (2014/2015) was conducted to examine the response of two pea varieties (Ambassador and Premium) to foliar-administered sodium selenate (0/50/100 g Se/ha) and zinc oxide (0/375/750 g Zn/ha) at the flowering stage. Concentrations of selected trace elements (Fe, Cu, and Mn), total phenolic content (TPC), total flavonoid content (TFC), and total antioxidant activity (ABTS, FRAP) of seeds were determined. Results and conclusions: Se/Zn treatments did not improve the concentration of trace elements, while they generally enhanced TPC. Among examined treatments, the highest TPC was found in Ambassador (from 2014) treated with 100 g Se/ha and 750 g Zn/ha (2,926 and 3,221 mg/100 g DW, respectively) vs. the control (1,737 mg/100 g DW). In addition, 50 g of Se/ha increased TFC vs. the control (261 vs. 151 mg/100 g DW) in Premium (from 2014), 750 g of Zn/ha increased ABTS vs. the control (25.2 vs. 59.5 mg/100 g DW) in Ambassador (from 2015), and 50 g of Se/ha increased FRAP vs. the control (26.6 vs. 18.0 mmol/100 g DW) in Ambassador (from 2015). In linear multivariable regression models, Zn, Mn, Cu, and TPC best explained ABTS (R = 0.577), while Se, Cu, and TPC best explained the FRAP findings (R = 0.696). This study highlights the potential of foliar biofortification with trace elements for producing pea/pea products rich in bioactive plant metabolites beneficial for human health.

RESUMO

Though selenium (Se) and zinc (Zn) constitute essential nutrients for human health, their deficiencies affect up to 15% and 17% of the global population, respectively. Agronomic biofortification of staple crops with Se/Zn may alleviate these challenges. Pea (Pisum sativum L.) is a nutritious legume crop that has great potential for Se/Zn biofortification. Herein, two varieties of pea (Ambassador, Premium) were biofortified via foliar application of sodium selenate (0/50/100 g of Se/ha) or zinc oxide (0/375/750 g of Zn/ha) during the flowering stage under field conditions. While no significant differences were found in Se accumulation between seed varieties upon Se treatments, selenate enhanced the accumulation of Se in the two seed varieties in a dose dependent manner. Selenium concentration was most elevated in seeds of Ambassador exposed to 100 g of Se/ha (3.93 mg/kg DW compared to the control (0.08 mg/kg DW), p < 0.001). 375 g of Zn/ha (35.7 mg/kg DW) and 750 g of Zn/ha (35.5 mg/kg DW) significantly and similarly enhanced Zn concentrations compared to the control (31.3 mg/kg DW) in Premium seeds, p < 0.001. Zinc oxide also improved accumulations of Fe, Cu, Mn, and Mg in Premium seeds. Se/Zn treatments did not significantly affect growth parameters and accumulations of soluble solids and protein in seeds. Positive and significant (p < 0.01) correlations were observed between Zn and Fe, Cu, Mn and Mg levels in Premium seeds, among others. Consuming 33 g/day of pea biofortified with Se at 50 g/ha and 266 g/day of pea biofortified with 375 g of Zn/ha could provide 100% of the RDA (55 µg) for Se and RDA (9.5 mg) for Zn in adults, respectively. These results are relevant for enhancing Se/Zn status in peas by foliar biofortification.

RESUMO

Caffeine content is a crucial attribute of coffee. Its concentration and thus maximum cups of Coffea arabica from Africa, Asia, Central America, and South America from different altitudes of growing areas, altitude, and process using different post-harvest processing (dry, wet, and pulped natural). Our results suggest that geographical origin might affect the alkaloid concentration in C. arabica. The caffeine concentration pattern in green samples was as follows: Central America > South America > Asia > Africa. Altitude affected the concentrations, lowlands > midlands > highlands, however, not significantly. Given caffeine is thermostable, the medium roasting process did not affect the concentration of caffeine directly, but a small increase was observed. Scientific opinion on the safety of habitual caffeine consumption of up to 400 mg per day does not raise safety concerns for non-pregnant adults. A cup (7 g coffee in 120 mL of water) was used for recalculation. Results suggest that mostly highlands and midlands coffee from Africa reached levels of caffeine that might be consumed in more than 5.5 cups a day.

RESUMO

The species Pleurotus ostreatus is a commercially, gastronomically, and biotechnologically important fungus. Its strain variability has been little researched. The study provides an evaluation of 59 oyster mushroom production strains in terms of the ability to accumulate selected metals in the cap and stipe. The fruiting bodies were grown under identical model conditions on straw substrate. Metal concentrations (ET-AAS) in dry fruiting bodies ranged in values 1.7-22.4 mg kg-1 for Al, 2.6-9.7 mg kg-1 Ba, 199-4560 mg kg-1 Ca, 1.7-12.0 mg kg-1 Cu, 12-120 mg kg-1 Fe, 16,000-49,500 mg kg-1 K, 876-2400 mg kg-1 Mg, 0.39-11.0 mg kg-1 Mn, 46-920 mg kg-1 Na and 11-920 mg kg-1 for Zn. More Cu, Fe, K, Mg, Mn, Zn accumulated in the cap, while in the stipe Ba was amassed. No significant difference was found between Al, Ca and Na between the accumulation in the cap and the stipe. Furthermore, the dependence of metal uptake from the substrate depending on the fortification of the substrate was confirmed. Statistically significant (p < 0.05) synergistic relationships were shown in pairs Al and Ba, Al and Fe, Ba and Na, Ba and Ca, Ca and Na, Cu and Fe, Fe and Mn, Fe and Zn, K and Mg, K and Mn, K and Zn, Mg and Mn, Mg and Na, Mg and Zn and Mn and Zn in the substrate without the addition of sodium selenate to the substrate. Altered relationships were observed after the application of sodium selenate to the substrate, synergism of Se and Ni, Se and Co and Se and Hg, Cu and Mn, Cu and Fe, Zn and Co, Zn and Ni, Zn and Hg, Mn and Fe, Mn and Cr, Co and Ni, Co and Hg, Ni and Hg, Pb and Cd. The findings of the study may help in the selection of production strains with hypercumulative properties for a particular metal and subsequent use in the addition of fortified fruiting bodies (e.g., with Zn). Based on the study the strains less sensitive to the accumulation of hazardous metals is possible to select for large-scale production, which is important from the perspective of food safety.

RESUMO

The adsorption of contaminants plays an important role in the process of their elimination from a polluted environment. This work describes the issue of loading environment with lead Pb(II) and the resulting negative impact it has on plants and living organisms. It also focuses on bentonite as a natural adsorbent and on the adsorption process of Pb(II) ions on the mixture of bentonite and bottom sediment from the water reservoir in Kolínany (SR). The equilibrium and kinetic experimental data were evaluated using Langmuir isotherm kinetic pseudo-first and pseudo-second-order rate equations the intraparticle and surface diffusion models. Langmuir isotherm model was successfully used to characterize the lead ions adsorption equilibrium on the mixture of bentonite and bottom sediment. The pseudo second-order model, the intraparticle and surface (film) diffusion models could be simultaneously fitted the experimental kinetic data.

RESUMO

In this study the kinetics and equilibrium of phenol biosorption were studied from aqueous solution using batch technique at an initial pH of 5.5. The biosorption was studied on Ca-alginate beads, on non-living mycelial pellets of Phanerochaete chrysosporium immobilized on Ca-alginate, and on free fungal biomass. Ph. chrysosporium was grown in a liquid medium containing mineral and vitamin materials with complex composition. The biosorption process followed pseudo second-order kinetics on all bioadsorbents. The bioadsorption-equilibrium on blank Ca-alginate, free and immobilized fungal biomass can be described by Langmuir, anti-Langmuir and Freundlich isotherm models using nonlinear least-squares estimation.

RESUMO

The current status of research on polyphenolic compounds in the berries of edible honeysuckle and their biological effects, including recommended utilization, are reviewed. The major classes of phenolic compounds in the blue berried honeysuckle are flavonols (quercetin, rutin, quercitrin) and flavanes (proanthocyanidins, catechins) and anthocyanins. Cyanidin-3-glucoside and cyanidin-3-rutinoside are considered as major anthocyanidins in edible honeysuckle berries. Such a high level of antioxidant activity in the berries of different species of the genus Lonicera is especially due to the high level of polyphenolic compounds, especially anthocyanins. These berries seem to be prospective sources of health-supporting phytochemicals that exhibit beneficial anti-adherence and chemo-protective activities, thus they may provide protection against a number of chronic conditions, e.g., cancer, diabetes mellitus, tumour growth or cardiovascular and neurodegenerative diseases.

RESUMO

ET-AAS and HG-AAS methods of selenium determination were compared and evaluated. The ET-AAS method has been followed with deuterium background correction and Zeeman background correction respectively. The following validation parameters were determined: accuracy (under repeatability conditions), trueness, calibration curve and linearity, limit of detection, limit of determination and combined standard uncertainty of the method. The HG-AAS method and the ET-AAS method with Zeeman correction were more suitable for the determination of low selenium concentrations in vegetables. The results were calculated by the standard addition method because of the strong matrix effect.
