Viewing body-positive content on social media can benefit women's body image. Previous research has conceptualised body positivity broadly, and therefore it remains unclear whether some components are more useful than others. This study examined the impact of body appreciation or body functionality messages (in addition to a mixed condition) overlaid across different image types to influence women's body image. Young women (17-30 years, Mage= 21.53, N = 308) completed an online survey in which they were randomised to view either fitspiration or diverse images overlaid with messaging focusing on body appreciation, body functionality, or a combination. Participants completed pre- and post-test measures of appearance and functionality satisfaction, body appreciation, and body objectification / conceptualisation. Post-test measures of social comparison and perceptions of models were also taken. Results showed that message type did not interact with image type. Rather, exposure to diverse images increased appearance satisfaction and body appreciation, with no changes to functionality satisfaction (vs a decrease for the fitspiration condition). All conditions improved in self-objectification. Further, diverse images resulted in more favourable social comparisons. Our findings consider body positive content in a controlled way and show no differences across body appreciation or body functionality themes.

CRISPR/Cas9, base editors and prime editors comprise the contemporary genome editing toolbox. Many studies have optimized the use of CRISPR/Cas9, as the original CRISPR genome editing system, in substituting single nucleotides by homology directed repair (HDR), although this remains challenging. Studies describing modifications that improve editing efficiency fall short of isolating clonal cell lines or have not been validated for challenging loci or cell models. We present data from 95 transfections using a colony forming and an immortalized cell line comparing the effect on editing efficiency of donor template modifications, concentration of components, HDR enhancing agents and cold shock. We found that in silico predictions of guide RNA efficiency correlated poorly withactivity in cells. Using NGS and ddPCR we detected editing efficiencies of 5-12% in the transfected populations which fell to 1% on clonal cell line isolation. Our data demonstrate the variability of CRISPR efficiency by cell model, target locus and other factors. Successful genome editing requires a comparison of systems and modifications to develop the optimal protocol for the cell model and locus. We describe the steps in this process in a flowchart for those embarking on genome editing using any system and incorporate validated HDR-boosting modifications for those using CRISPR/Cas9.

Historical, or 'legacy' landfills are commonly unlined and can therefore pose risks to human health and the environment via the discharge of leachate to sensitive groundwater and surface waters. Characterising the impacts on groundwater from legacy landfills located within urban re-development precincts is therefore of growing importance worldwide and is difficult using conventional indicators. At Australia's largest urban re-development precinct, Fishermans Bend, seven known legacy landfills exist, as well as numerous other contamination sources (e.g. historical industrial spillages). Conventional landfill leachate indicators (e.g. ammonia-N and bicarbonate) and perfluoroalkyl acids (PFAA) were measured in 36 bores to distinguish leachate-impacted groundwater from non-impacted areas. Whilst eleven bores showed clear leachate impacts based on conventional indicators, others did not show clearly identifiable leachate signals, particularly those installed near landfills thought to have accepted a larger component of non-putrescible waste (e.g. industrial, construction and/or demolition waste). A new index for detection of legacy landfill leachate impact on groundwater was therefore developed, incorporating perfluorooctanoate (PFOA) as a proportion of PFAA (PFOA/∑PFAA) into an existing method based on leachate to native cation ratios, ('L/N ratios'). Significant differences between the means of the leachate-impacted versus non-impacted bores were found using the 'modified L/N ratio' (p = .006), whereas no significant differences were found between the means of the two groups using the standard L/N ratio (p = .063). The modified L/N ratios also showed a statistically significant difference between the means of the bores impacted by municipal waste versus those impacted by non-putrescible waste (p = .003), indicating they are a much more sensitive indicator of both the existence and type of landfill leachate impact on groundwater than previously reported. This new index may prove particularly useful in complex urban areas where multiple potential contamination sources exist, and land use histories are either unknown or complicated. CAPSULE: Conventional methods for leachate detection in groundwater surrounding legacy landfills have been analysed and further developed via the inclusion of perfluoroalkyl acids, to better understand contaminant sources, fate and transport.

The extent of per- and polyfluoroalkyl substances (PFAS) in groundwater surrounding legacy landfills is currently poorly constrained. Seventeen PFAS were analysed in groundwater surrounding legacy landfills in a major Australian urban re-development precinct. Sampling locations (n = 13) included sites installed directly in waste material and down-gradient from landfills, some of which exhibited evidence of leachate contamination including elevated concentrations of ammonia-N (≤106 mg/L), bicarbonate (≤1,740 mg/L) and dissolved methane (≤10.4 mg/L). Between one and fourteen PFAS were detected at all sites and PFOS, PFHxS, PFOA and PFBS were detected in all samples. The sum of detected PFAS (∑14PFAS) varied from 26 ng/L at an ambient background site to 5,200 ng/L near a potential industrial point-source. Determining sources of heavy metals in soils, sediments and groundwater is important for understanding their fate and transport and mitigating human and environmental exposures. Artificially imported fill, natural sediments and groundwater from 240 ha of reclaimed land at Fishermans Bend in Australia, were analysed for heavy metals and other parameters to determine the relative contributions from different possible sources. Fishermans Bend is Australia's largest urban re-development project, however, complicated land-use history, geology, and multiple contamination sources pose challenges to successful re-development. We developed a method for heavy metal source separation in groundwater using statistical categorisation of the data, analysis of soil leaching values and fill/sediment XRF profiling. The method identified two major sources of heavy metals in groundwater: 1. Point sources from local or up-gradient groundwater contaminated by industrial activities and/or legacy landfills; and 2. contaminated fill, where leaching of Cu, Mn, Pb and Zn was observed. Across the precinct, metals were most commonly sourced from a combination of these sources; however, eight locations indicated at least one metal sourced solely from fill leaching, and 23 locations indicated at least one metal sourced solely from impacted groundwater. Concentrations of heavy metals in groundwater ranged from 0.0001 to 0.003 mg/L (Cd), 0.001-0.1 mg/L (Cr), 0.001-0.2 mg/L (Cu), 0.001-0.5 mg/L (Ni), 0.001-0.01 mg/L (Pb), and 0.005-1.2 mg/L (Zn). Our method can determine the likely contribution of different metal sources to groundwater, helping inform more detailed contamination assessments and precinct-wide management and remediation strategies.

This paper investigates the organic pollution status of shallow aquifer sediments and groundwater around Zhoukou landfill. Chlorinated aliphatic hydrocarbons, monocylic aromatic hydrocarbons, halogenated aromatic hydrocarbons, organochlorine pesticides and other pesticides, and polycyclic aromatic hydrocarbons (PAHs) have been detected in some water samples. Among the detected eleven PAHs, phenanthrene, fluorine, and fluoranthene are the three dominant in most of the groundwater samples. Analysis of groundwater samples around the landfill revealed concentrations of PAHs ranging from not detected to 2.19 µg/L. The results show that sediments below the waste dump were low in pollution, and the shallow aquifer, at a depth of 18-30 m, was heavily contaminated, particularly during the wet season. An oval-shaped pollution halo has formed, spanning 3 km from west to east and 2 km from south to north, and mainly occurs in groundwater depths of 2-4 m. For PAH source identification, both diagnostic ratios of selected PAHs and principal component analysis were studied, suggesting mixed sources of pyro- and petrogenic derived PAHs in the Zhoukou landfill. Groundwater table fluctuations play an important role in the distribution of organic pollutants within the shallow aquifer. A conceptual model of leachate migration in the Quaternary aquifers surrounding the Zhoukou landfill has been developed to describe the contamination processes based on the major contaminant (PAHs). The groundwater zone contaminated by leachate has been identified surrounding the landfill.
