RESUMO

Manure is widely used as a fertilizer and applied to agricultural land. It may contain highly active chemicals like veterinary medicinal products or biocides, which enter into the environment by this pathway. This is recognized by several regulatory frameworks, however, a detailed method for examining the transformation of chemicals in manure was lacking. This article describes the validation of a method for studying the anaerobic transformation of chemicals in pig and cattle liquid manure. Different steps are covered with an emphasis on the validation ring test and the OECD (Organisation for Economic Cooperation and Development) process that led to the recent adoption of the method as OECD Test Guideline (TG) 320.

The project nanoGRAVUR (BMBF, 2015-2018) developed a framework for grouping of nanomaterials. Different groups may result for each of the three distinct perspectives of occupational, consumer and environmental safety. The properties, methods and descriptors are harmonised between the three perspectives and are based on: Tier 1 intrinsic physico-chemical properties (what they are) or GHS classification of the non-nano-form (human tox, ecotox, physical hazards); Tier 2 extrinsic physico-chemical properties, release from nano-enabled products, in vitro assays with cells (where they go; what they do); Tier 3 case-specific tests, potentially in vivo studies to substantiate the similarity within groups or application-specific exposure testing. Amongst all properties, dissolution and transformation are least modulated by different nanoforms within one substance, whereas dustiness, dispersion stability, abiotic and especially in vitro surface reactivity vary more often between different nanoforms. The methods developed or selected by nanoGRAVUR fill several gaps highlighted in the ProSafe reviews, and are useful to implement (i) the concept of nanoforms of the European Chemicals Agency (ECHA) and (ii) the concept of discrete forms of the United States Environmental Protection Agency (EPA). One cannot assess the significance of a dissimilarity, if the dynamic range of that property is unknown. Benchmark materials span dynamic ranges that enable us to establish bands, often with order-of-magnitude ranges. In 34 case studies we observed high biological similarity within each substance when we compared different (nano)forms of SiO2, BaSO4, kaolin, CeO2, ZnO, organic pigments, especially when we compared forms that are all untreated on the surface. In contrast, different Fe2O3 or TiO2 (nano)forms differ more significantly. The same nanoforms were also integrated in nano-enabled products (NEPs) for automotive coatings, clinker-reduced cements, cosmetic sunscreen, and lightweight polymers.

BACKGROUND: Cationic polyacrylamide copolymers (PAMs) are used for sludge dewatering in municipal waste water treatment and might enter the environment by spreading of the sludge on agricultural land. Concern has been expressed since little is known about the degradation of PAMs in soils. To obtain detailed information on the polymer's fate in the soil compartment, the degradation of 14C-radiolabelled PAM in an outdoor lysimeter was studied. RESULTS: No plant uptake and leaching of radioactivity was observed. There was practically no vertical movement of polymer and no transformation products found at the end of the study. For the top 10 cm soil layer, a mass balance was established throughout the study. About 10% of applied radioactivity was not extractable from soil even with a matrix destructive method, and this was concluded to be bound residue. Characterization of extractable radioactivity by means of GPC-analysis showed a significant decrease of the molecular weight of the PAM with time. The decrease in molecular weight indicates a breakdown of the polymer backbone (the C-C-chain), and is assumed to be primary degradation. The total radioactivity content in the 10 cm top soil layer was quantified every 6 months over a period of 3 years. The results show a significant decrease of the total radioactivity over time and this is defined as ultimate degradation following the definition of OECD and EPA. Based on the data, a half-life time of 2.0 × 103 days and a rate constant of 0.00035/day were calculated. With a χ2 of 12.0 the results of the calculation are thus valid and reliable. The rate constant indicates a mineralization of 22.5% within a period of 2 years based on the total recovered radioactivity. This half-life time is solely based on mineralization and does not take into account the degradation of the polymer backbone, hydrolysis of the side chains, incorporation into the soil matrix, and thus is a conservative approach. CONCLUSIONS: 14C-PAM degrades very slowly in soil after land-spreading as a component of sewage sludge. Even in a very conservative evaluation which only considered the loss of radioactivity, a half-life time of 5.4 years was determined.

BACKGROUND: Residues of veterinary medicinal products (VMPs) enter the environment via application of manure onto agricultural areas where in particular antibiotics can cause phytotoxicity. Terrestrial plant tests according to OECD guideline 208 are part of the environmental risk assessment of VMPs. However, this standard approach might not be appropriate for VMPs which form non-extractable residues or transformation products in manure and manure-amended soil. Therefore, a new test design with a more realistic exposure scenario via manure application is needed. This paper presents an extended plant test and its experimental verification with the veterinary antibiotics florfenicol and tylosin tartrate. With each substance, plant tests with four different types of application were conducted: standard tests according to OECD 208 and three tests with application of test substance via spiked manure either without storage, aerobically incubated, or anaerobically incubated for different time periods. RESULTS: In standard tests, the lowest NOEC was <0.06 mg/kg dry soil for florfenicol and 16.0 mg/kg dry soil for tylosin tartrate. Pre-tests showed that plant growth was not impaired at 22-g fresh manure/kg dry soil, which therefore was used for the final tests. The application of the test substances via freshly spiked as well as via aerobically incubated manure had no significant influence on the test results. Application of florfenicol via anaerobically incubated manure increased the EC10 by a factor up to 282 and 540 for half-maximum and for maximum incubation period, respectively. For tylosin tartrate, this factor amounted to 64 at half-maximum and 61 at maximum incubation period. The reduction of phytotoxicity was generally stronger when using cattle manure than pig manure and particularly in tests with cattle manure phytotoxicity decreased over the incubation period. CONCLUSIONS: The verification of the extended plant test showed that seedling emergence and growth are comparable to a standard OECD 208 test and reliable effect concentrations could be established. As demonstrated in the present study, phytotoxicity of veterinary antibiotics can be significantly reduced by application via incubated manure compared to the standard plant test. Overall, the presented test design proved suitable for inclusion into the plant test strategy for VMPs.

BACKGROUND: Engineered nanomaterials (ENMs) are marketed as a substance or mixtures and are additionally used due to their active agent properties in products such as pesticides or biocides, for which specific regulations apply. Currently, there are no specific testing strategies for environmental fate and effects of ENMs within the different regulations. An environmental test and risk assessment strategy for ENMs have been developed considering the general principles of chemical assessment. RESULTS: The test strategy has been developed based on the knowledge of national and international discussions. It also takes into account the conclusions made by the OECD WPMN which held an expert meeting in January 2013. For the test strategy development, both conventional and alternative endpoints were discussed and environmental fate and effects were addressed separately. CONCLUSION: A tiered scheme as commonly used in the context of precautionary environmental risk assessment was suggested including the use of mathematical models and trigger values to either stop the procedure or proceed to the next tier. There are still several gaps which have to be filled, especially with respect to fate, to develop the test strategy further. The test strategy features a general approach. It is not specified to fulfil the information requirements of certain legislation (e.g. plant protection act, biocide regulation, REACH). However, the adaption of single elements of the strategy to the specific needs of certain legislation will provide a valuable contribution in relation to the testing of nanomaterials.

In the framework of the German environmental specimen bank one-year old spruce shoots (Picea abies) and pine shoots (Pinus sylvestris) serve as bioindicators for the atmospheric pollution. Sampling is performed in two urbanized areas in western and eastern Germany (Warndt and Duebener Heide, respectively), and in seven different rural locations. Prior to archiving conifer shoots are continuously analyzed for a set of 17 individual polycyclic aromatic hydrocarbons (PAHs). The results from the two urbanized areas show that the atmospheric contamination with PAH has declined by about 75% between 1985 and 2004 at Warndt and by about 85% between 1991 and 2004 at Duebener Heide. However, summation operatorPAH concentrations stayed virtually constant at both locations since the end of the 1990s at levels of about 100 ng g(-1) wet weight (ww). In spruce shoots from rural areas current concentrations of PAHs are significantly lower and vary between 8 and 61 ng g(-1) ww. In all shoot samples the four low molecular aromatics phenanthrene, fluoranthene, pyrene, and chrysene dominate the pattern by contributing 60 to 90% to summation operatorPAH. The group of high molecular weight aromatics is dominated by benzo[b,j,k]fluoranthene, especially in spruce shoots originating from greater altitudes remarkable amounts of six and seven ringed PAHs could be detected. Despite the strong decrease of PAH concentrations in urban areas patterns of aromatics remained nearly unchanged in the observation period 1985 to 2004.
