RESUMO

Helicoverpa armigera is a significant agricultural pest and particularly notorious for its resistance to many types of common insecticides. RNA interference (RNAi) is a mechanism of post-transcriptional gene silencing and trigged by double-strand RNA (dsRNA), has become a widely used reverse genetics and potent tool for insect pest control. In this study, the effect of ingestion and injection delivery methods of dsRNA related two important enzyme genes, α-amylase (HaAMY48, Ha-AMY49) and juvenile hormone esterase (Ha-JHE), were examined on growth and development of H. armigera. After 24, 48, 72 and 96 h of feeding bioassay, significant down regulation was observed about; 56, 68, 78, 80.75% for HaAMY48, 60, 70, 86.5 and 96.75%, for Ha-AMY49 and 14, 27.5, 23 and 31.7% for Ha-JHE, respectively. The results for injection assay was 61.5, 71.5, 74 and 95.8% for Ha-AMY48; 70, 88, 91.5 and 97.7% for Ha-AMY49 and 22, 61, 75 and 74% for Ha-JHE after 24, 48 and 72 h of last injecting, respectively. Larvae that treated with dsRNA, fed or injected, lost more than half of their weight. 50% mortality in treated larvae was observed in the case injection bioassay with dsHa-JHE and 59% of larvae that fed of dsRNA-treated cubes survived. DsHa-AMY48 and 49 have significant mortality, but mixing of them is more effective in both bioassays. Injection bioassay has a potent inhibitory effect on α-amylase-specific activity about more than 87% in treated larvae with mix of dsHa-AMY48 and 49. Adult malformation percent was evaluated for feeding (28, 35.5 and 43% for Ha-AMY48, 49 and Ha-JHE, respectively) and injection bioassay (23, 42 and 29% for Ha-AMY48, 49 and Ha-JHE, respectively). All these finding suggest that Ha-AMY48, Ha-AMY49 and Ha-JHE can be new candidates to scheming effective dsRNAs pesticide for H. armigera control.

Nine Bemisia tabaci (Gennadius) populations were collected from different regions of Iran. In all nine populations, only one biotype (B biotype) was detected. Susceptibilities of these populations to imidacloprid and acetamiprid were assayed. The lethal concentration 50 values (LC50) for different populations showed a significant discrepancy in the susceptibility of B. tabaci to imidacloprid (3.76 to 772.06 mg l-1) and acetamiprid (4.96 to 865 mg l-1). The resistance ratio of the populations ranged from 9.72 to 205.20 for imidacloprid and 6.38 to 174.57 for acetamiprid. The synergistic effects of piperonylbutoxide (PBO) and S,S,S-tributylphosphorotrithioate (DEF) were evaluated for the susceptible (RF) and resistant (JR) populations for the determination of the involvement of cytochrome P450-dependent monooxygenase and carboxylesterase, respectively, in their resistance mechanisms. The results showed that PBO overcame the resistance of the JR population to both imidacloprid and acetamiprid, with synergistic ratios of 72.7 and 106.9, respectively. Carboxylesterase, glutathione S-transferase and cytochrome P450-dependent monooxygenase were studied biochemically, for the purpose of measuring the activity of the metabolizing enzymes in order to determine which enzymes are directly involved in neonicotinoid resistance. There was an increase in the activity of cytochrome P450-dependent monooxygenase up to 17-fold in the resistant JR population (RR = 205.20). The most plausible activity of cytochrome P450-dependent monooxygenase correlated with the resistances of imidacloprid and acetamiprid, and this suggests that cytochrome P450-dependent monooxygenase is the only enzyme system responsible for neonicotinoid resistance in the nine populations of B. tabaci.

The effects of 13 soybean varieties (356, M4, M7, M9, Clark, Sahar, JK, BP, Williams, L17, Zane, Gorgan3, and DPX) on nutritional indices of the cotton bollworm, Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), were determined at 25 ± 1° C, 65 ± 5% RH and a photoperiod of 16:8 L:D. Fourth instar larvae reared on Zane showed the highest efficiency of conversion of digested food (ECD) and approximate digestibility (AD) values (0.299 and 0.867, respectively) compared with other varieties. The lowest value of ECD and food consumed (FC) was on 356 (0.133 and 53.82 mg, respectively). The highest and lowest efficiency of conversion of ingested food (ECI) of fifth instar larvae (0.235 and 0.156, respectively) were on Zane and M4, respectively. The ECI and ECD values of whole larval instars were the highest on M7 (0.524 and 0.820, respectively) and lowest on Sahar (0.279 and 0.353, respectively). However, the highest and lowest value of consumption index (CI) was on M7 (7.351) and BP (3.462). Among the different varieties of soybean, the highest AD value was on M9 (0.858), and the lowest was on Zane (0.597). The results indicated that M4, Sahar, and JK were partially resistant to H. armigera.
