RESUMO

BACKGROUND Nasopharyngeal carcinoma (NPC) is a type of head and neck cancer with very high prevalence in southern China. Phosphatase and tensin homolog (PTEN), a tumor suppressor, was reported to be downregulated in NPC patients and correlated with pathological grade and clinical stage of NPC. MATERIAL AND METHODS Luciferase reporter assay, qPCR, and Western blot analysis were used to determine if PTEN is a target of miR-152. The function of miR-152 in cell apoptosis and cell proliferation was examined as well. Tissue samples from NPC patients were also analyzed for PTEN and miR-152 expressions. RESULTS Reporter assay indicated miR-152 targets the 3'UTR of PTEN mRNA to inhibit PTEN expression. Transfection of the NPC-derived cell line with miR-152 mimic confirmed these findings. Overexpression of miRNA-152 inhibits apoptosis induced by Cisplatin in NPC cancer cells in vitro. Moreover, overexpression miR-152 also promotes NPC cancer cell invasion and proliferation. Samples from EBV-negative NPC patients demonstrated the down-regulated level of PTEN may be related with overexpression of miR-152. CONCLUSIONS The miR-152 targets PETN to inhibit cell apoptosis and promote cancer cell proliferation and migration in NPC development.

RESUMO

OBJECTIVES: To investigate the effet of nasal septum operation combined with nasal sinuses on postoperative nasal septum. METHOD: Sixty-five patients with deflection of nasal septum complicating chronic sinusitis were underwent nasal septum and nasal sinus operations under nasal endoscope. Postoperative nasal septum recover were observed and compared with patients underwent only simple nasal septum operation. RESULT: Patients underwent nasal septum operations combined with nasal sinus, postoperative symptoms were more serious at early than simple nasal septum operations. At 3 months after operation, satisfactory degree of the former group were higher than the latter, and the former group didn't have complications such as nasal septum infection. CONCLUSION: Nasal septum operations combined with nasal sinus have no harmful effect on the prognosis of nasal septum, so it was safe and reliable.

RESUMO

OBJECTIVE: To investigate the method to repair the deletion of external canal and opened mastoid space after mastoidectomy. METHOD: One stage tympanoplasty with reconstruction of the external canal with temporal cortex was completed in 58 cases (58 ears) with cholesteatoma. RESULT: During the follow-up ranging from one to three years, there was only one relapse. The configuration of external canal was almost normal and pneumatic mastoid space was maintained. Hearing was improved over 30dB in five ears, 20 to 29 dB in 11 ears, 10 to 19 dB in 30 ears, less 10 dB in eight ears and hearing wasn't improved in four ears. CONCLUSION: One stage tympanoplasty with reconstruction of the external canal after mastoidectomy for cholesteatoma is a practical way to improve hearing and decrease recurrence.
