Type 2 diabetes (T2D) and hypertension are common comorbidities and, along with hyperlipidemia, serve as risk factors for cardiovascular diseases. This study aimed to evaluate the predictive value of polygenic risk scores (PRSs) on cardiometabolic traits related to T2D, hypertension, and hyperlipidemia and the incidence of these three diseases in Taiwan Biobank samples. Using publicly available, large-scale genome-wide association studies summary statistics, we constructed cross-ethnic PRSs for T2D, hypertension, body mass index, and nine quantitative traits typically used to define the three diseases. A composite PRS (cPRS) for each of the nine traits was constructed by aggregating the significant PRSs of its genetically correlated traits. The associations of each of the nine traits at baseline as well as the change of trait values during a 3- to 6-year follow-up period with its cPRS were evaluated. The predictive performances of cPRSs in predicting future incidences of T2D, hypertension, and hyperlipidemia were assessed. The cPRSs had significant associations with baseline and changes of trait values in 3-6 years and explained a higher proportion of variance for all traits than individual PRSs. Furthermore, models incorporating disease-related cPRSs, along with clinical features and relevant trait measurements achieved area under the curve values of 87.8%, 83.7%, and 75.9% for predicting future T2D, hypertension, and hyperlipidemia in 3-6 years, respectively.

Anaerobic sludge was capable of producing anaerobic ammonium oxidation (anammox) cultures. However, the low activity of anammox bacteria in the seed sludge often led to a long time for stable anammox to initiate. The objective of this study was to investigate the influence of an extended reaction-phase time in the sequencing batch reactor (SBR) on the rapid startup of anaerobic ammonium oxidation (anammox) using anaerobic heterotrophic bacteria as the seed sludge. After the startup, suspended and attached bacteria in anammox were separately analyzed for comparison. The variations of nitrogen concentrations and shifts of the microbial community structures were studied. The results showed that anammox occurred after a long reaction-phase time in the SBR with the efficient removals of NH4+ (96.4%) and NO2- (99.8%). The effective NO2- treatment before anammox startup was attributable to inevitable denitrification or dissimilatory nitrate reduction (e.g., Denitratisoma). The occurrence of anammox was supported by the anammox stoichiometry, bacteria diversity variation, and principal component analysis. The overall nitrogen removal rate (NRR) and nitrogen removal efficiency (NRE) was 0.07 kg/m3-d and 92.8%, respectively. The relative molar quantities of NH4+ and NO2- removed as well as N2 and NO3- formed were 1(1):1.29(1.32):1.45(1.02):0.15(0.26), as the numbers in the parentheses represent the theoretical values. Denitratisoma and Desulfatiglans dominated in the seed sludge, whereas Candidatus_Jettenia abundances were significantly higher in anammox attached- (26.0%) and suspended-growth cultures (14.5%). Fifty-three genera were simultaneously identified in all samples, suggesting their importance in the startup of anammox from anaerobic sludge. Candidatus_Jettenia was observed to be more associated with the growth of anammox biofilm (the abundances were 26.0% and 14.5% in attached- and suspended-growth cultures, respectively) and supported the fine nitrogen removal performance in the attached-growth cultures.

Nitrosamines are toxic and emerging disinfection byproducts. In this study, three drinking water treatment plants (DWTPs) in southern Taiwan treating the same source water in Gaoping River with comparable technologies were selected. The objective was to evaluate the formation and fates of six nitrosamines and their formation potentials (FPs) from a surface water source to drinking water. Albeit decreased further downstream in the river, four nitrosamine-FPs were observed in the source water due to anthropogenic pollution in the upstream areas. In the DWTPs, nitrosamines were formed and NDMA was the main species. While high organic carbon concentrations indicated elevated nitrosamine-FPs in the source water, NDMA formation in the DWTPs was more positively associated with reductions of water parameters that quantify organic matters with double bonded ring structures. Although precursor removal via pre-oxidation is a viable approach to limit nitrosamine formation during post-disinfection, this study clearly indicates that a great portion of NDMA in treated water has been formed in the 1st oxidation step of drinking water treatment. The pre-oxidation simulations in the lab demonstrated the impact of pre-chlorination on nitrosamine formation. Given the limited removal in conventional treatment processes, avoiding nitrosamine-FPs in sources and/or nitrosamine formation during pre-oxidation become important issues to control the threats of nitrosamines in drinking water. Under current circumstance in which pre-oxidation is widely used to optimize the treatment effectiveness in many DWTPs, its adverse effect by forming nitrosamines needs to be carefully minimized and using technologies other than pre-chlorination (e.g., pre-ozonation) may be considered.

The reproducibility of interpretation in diagnosing bacterial vaginosis may be enhanced by adding pus cells and clue cells into two different criteria, developed by Spiegel et al. and Nugent et al. The purpose of study was designed to find out which parameter was more reproducible. 100 patients were collected with the diagnosis of bacterial vaginosis as an experimental group, while the other 100 patients who were with routine Papanicolaou smears in gynecologic clinic the collected as a control group. Two slides, including the original and reproducible ones, were obtained from vaginal smears for each patient. Three technicians read the slides randomly by using two different criteria, plus pus cells and clue cells. This showed the agreement for clue cells is the best method regardless of experimental group or control group (Kappa values between 0.708 and 1.000). The intra-observer agreement for the diagnosis of bacterial vaginosis by the method of Nugent et al. is superior to the method of Spiegel et al. Our data show the comparison of Amsel criteria versus Nugent criteria for the diagnosis of bacterial vaginosis with sensitivity of 88.9%, specificity of 55.4%, negative positive value of 62.1%, and positive predictive value of 85.8%. Moreover, our data also demonstrate the comparison of Amsel criteria versus the diagnosis either based on Nugent criteria or the presence of clue cells with sensitivity of 95.7%, specificity of 56.7%, negative positive value of 81.2%, and positive predictive value of 87.1%. The results demonstrate further adding score of the clue cells can enhance the reproducible diagnosis of bacterial vaginosis, which is superior to the methods of Nugent et al. and Spiegel et al.
