The purpose of this experiment was to study the effect of the physical form of rapeseed fat on methane (CH4) mitigation properties, feed digestion, and rumen fermentation. Four lactating ruminal-, duodenal-, and ileal-cannulated Danish Holstein dairy cows (143 d in milk, milk yield of 34.3 kg) were submitted to a 4×4 Latin square design with 4 rations: 1 control with rapeseed meal (low-fat, CON) and 3 fat-supplemented rations with either rapeseed cake (RSC), whole cracked rapeseed (WCR), or rapeseed oil (RSO). Dietary fat concentrations were 3.5 in CON, 5.5 in RSC, 6.2 in WCR, and 6.5% in RSO. The amount of fat-free rapeseed was kept constant for all rations. The forage consisted of corn silage and grass silage and the forage to concentrate ratio was 50:50 on a dry matter basis. Diurnal samples of duodenal and ileal digesta and feces were compiled. The methane production was measured for 4 d in open-circuit respiration chambers. Additional fat reduced the CH4 production per kilogram of dry matter intake and as a proportion of the gross energy intake by 11 and 14%, respectively. Neither the total tract nor the rumen digestibility of organic matter (OM) or neutral detergent fiber were significantly affected by the treatment. Relating the CH4 production to the total-tract digested OM showed a tendency to decrease CH4 per kilogram of digested OM for fat-supplemented rations versus CON. The acetate to propionate ratio was not affected for RSC and WCR but was increased for RSO compared with CON. The rumen ammonia concentration was not affected by the ration. The milk and energy-corrected milk yields were unaffected by the fat supplementation. In conclusion, rapeseed is an appropriate fat source to reduce the enteric CH4 production without affecting neutral detergent fiber digestion or milk production. The physical form of fat did not influence the CH4-reducing effect of rapeseed fat. However, differences in the volatile fatty acid pattern indicate that different mechanisms may be involved.

The objective was to assess the ability of the in situ mobile nylon bag method for predicting small intestinal and total tract starch digestibility. Starch disappearance was measured for 18 samples of different cereals and legumes subjected to different physical and chemical processing methods and compared with coherent in vivo digestibility. Starch disappearance was measured both with and without initial ruminal pre-incubation during 4 h. Bags were retrieved from either the ileal cannula or faeces. Two dry Danish Holstein cows fitted with rumen cannulas were used for rumen pre-incubations and two lactating Danish Holstein cows fitted with duodenal and ileal cannulas were used for intestinal incubations. Rumen pre-incubation had no significant effect on disappearance from bags recovered in faeces. The disappearance of legume starch was lower, both in the rumen and small intestine, compared with starch from barley, wheat, oats, ear maize and maize. Transit times of the mobile bags from duodenum to ileum were not significantly different between feeds. A weak positive correlation was found between in vivo small intestinal and total tract digestibility of starch and disappearance obtained using the mobile bag technique across a broad range of starch sources. Omitting two less conventional starch sources (NaOH wheat and xylose-treated barley) resulted in a high (0.87) correlation between total tract in vivo digestibility and mobile bag disappearance. The use of the mobile bag method for estimation of in vivo starch digestibility will therefore depend on the starch type.

Methane is a greenhouse gas with a significant anthropogenic contribution from cattle production. A demand exists for techniques that facilitate evaluation of mitigation strategies for dairy cows. Therefore, a low-cost system facilitating the highest possible animal welfare was constructed and validated. The system uses the same principles as systems for open-circuit indirect calorimetry, but to lower the costs, the chamber construction and air-conditioning system were simpler than described for other open-circuit systems. To secure the highest possible animal welfare, the system is located in the cow's daily environment. The system consists of 4 transparent polycarbonate chambers placed in a square so that the cows are facing each other. The chamber dimensions are 183 (width), 382 (length), and 245 cm (height) with a volume of 17 m(3). Flow and concentrations of O(2), CO(2), CH(4), and H(2) are measured continuously in the outlet. Flow is measured with a mass flow meter, O(2) with a paramagnetic sensor, CO(2) and CH(4) with infrared sensors, and H(2) with an electrochemical sensor. Chamber inlet is placed in the barn and background concentrations may differ between chambers, but delta values between background and outlet concentrations for all chambers were within instrument tolerance. Average recovery rates of CO(2) and CH(4) were (mean ± SD) 101 ± 4 and 99 ± 7%, respectively. This is within the expected tolerance of the whole system (gas sensors and flow meters). Feed dry matter intakes were not affected by confining the animals in chambers, as dry matter intake before and during chamber stay were similar. It was concluded that the system delivers reliable values, and the transparent construction in combination with the location in the barn environment prevent negative impact on animal welfare and, thereby, data quality.

The objective of the present study was to examine if near infrared reflectance spectroscopy (NIRS) could be used to predict degradation parameters and effective degradation from scans of original forage samples. Degradability of dry matter (DM), crude protein (CP) and neutral detergent fibre (NDF) of 61 samples of perennial ryegrass (Lolium perenne L.) and orchardgrass (Dactylis glomerata L.) was tested by using the in situ technique. The grass samples were harvested at three different stages, early vegetative growth, early reproductive growth and late reproductive growth. Degradability was described in terms of immediately rumen soluble fraction (a fraction, for DM and CP only as NDF does not contain a soluble fraction), the degradable but not soluble faction (b fraction) and the rate of degradation of the b fraction (c value). Overall effective degradability of DM, CP and NDF was also calculated. Near infrared reflectance spectroscopy was examined for its ability to predict degradation parameters and to make a direct prediction of effective degradation from scans of the original samples of perennial ryegrass and orchardgrass. Prediction of effective degradation of the different feed fractions showed different accuracy. The coefficients of determination (R(2)) from regressions of predicted vs. measured effective degradation, using a cross-validation method, were 0.92 for DM, 0.78 for CP and 0.61 for NDF. The attempt to predict the degradation parameters (a, b and c) by NIRS was less successful as the coefficients of determination for the degradation parameters were low. Concentrations of CP and NDF in the original samples were predicted by using NIRS and the validated R(2) value was 0.98 for CP and 0.92 for NDF. It is concluded that using NIRS predictions from scans of original samples is a promising method to obtain values for the effective degradation of DM, CP and NDF in ruminant feeds, but that larger calibration sets are necessary for obtaining improved accuracy.

The objective of this digestibility assessment was to determine whether there are significant differences in the digestibility of Roundup Ready (glyphosate-tolerant) and conventional sugar beet, fodder beet, and beet pulp produced from sugar beet varieties when fed to sheep (seven wethers per treatment group). Three experiments were conducted in this assessment. Experiment 1 (35 wethers) compared one glyphosate-tolerant fodder beet variety with four conventional varieties, Exp. 2 (42 wethers) compared one glyphosate-tolerant sugar beet variety with five conventional varieties, and Exp. 3 (42 wethers) compared beet pulp derived from glyphosate-tolerant sugar beet with beet pulp from five European locations. The experimental phase consisted of a 2-wk preliminary period followed by a 1-wk collection period for Exp. 1 and 2, and a 1-wk preliminary period followed by a 1-wk digestibility collection period for Exp. 3. Diets were comprised of grass hay at 30, 30, and 20% of DM for Exp. 1, 2, and 3, respectively, with the balance being beet components. Urea and sodium sulfate were supplemented (8 and 2.9 g, respectively, for Exp. 1 and 2; and 6 g and 2.16 g, respectively, for Exp. 3) to supply sufficient dietary N and S. Each diet was fed to sheep (96 +/- 0.9 kg) in the three experiments to at or near maintenance energy levels. Treatment differences were considered significant at P < 0.05. Apparent digestibilities of DM, OM, CP, NDF, ADF, and DE for glyphosate-tolerant fodder and sugar beets did not differ from those for commercial fodder and sugar beets in Exp. 1 and 2. There were differences (P < 0.05) in DM, OM, CP, NDF, ADF, and DE digestibilities influenced by the different varieties of beet pulp in Exp. 3, but these were not unique to just the Roundup Ready sugar beet variety. Digestibilities and feeding values of Roundup Ready fodder beet, sugar beet, and beet pulp produced from Roundup Ready sugar beet varieties were not influenced by the introduction of the Roundup Ready trait compared with conventional varieties.

The effect of increased postruminal supply of lysine and methionine was investigated in a production trial involving 64 dairy cows in early lactation. Within each of two basal rations, based on either corn silage or grass silage, rations were either naturally deficient in lysine or fortified with 24 g of lysine in a rumen-protected form and naturally deficient in methionine or fortified with 12 g of methionine in a rumen-protected form. The data were analyzed separately for the four lysine and the four methionine treatment groups. Milk production, body weight gain, and plasma concentrations of insulin-like growth factor-I, bovine somatotropin, insulin, glucose, nonesterified fatty acids, and urea were monitored over a 12-wk period. Supplementation with protected methionine led to increases in milk fat and protein contents of 2.4 and 1.8 g/kg of milk, respectively. Supplementation with protected lysine or methionine numerically increased protein yield comparable to values reported in the literature, but the treatment effects were not statistically significant. Efficiency of utilization of absorbed amino acids for milk protein synthesis and efficiency of utilization of metabolizable energy for milk production were not significantly altered in response to increased postruminal lysine and methionine flow, but a numerically increased efficiency of utilization of total amino acids was observed. No significant effect of lysine or methionine supplementation was observed on endocrine parameters nor on plasma metabolite concentrations. However, across treatment groups, high milk yield was correlated with low plasma insulin-like growth factor-I concentrations (r = -0.44) and partially with low plasma nonesterified fatty acids concentration and insulin levels (r = -0.26), while body weight gain was negatively correlated (r = -0.33) with elevated plasma bovine somatotropin concentrations.

To investigate the metabolism of 1,2-propanediol (PPD) in lactating cows independently of normal rumen microbial metabolism, three ruminally cannulated lactating Holstein cows were subjected to three experimental infusion protocols under washed reticulo-ruminal conditions in a Latin square design. Reticulo-ruminal absorption rates were maintained for 420 min by continuous intraruminal infusion of VFA and PPD. With the control treatment, 1,246 +/- 39 mmol/ h of acetate and 213 +/- 5 mmol/h of butyrate were absorbed from the reticulorumen. With the propionate treatment, 1,148 +/- 39 mmo/h of acetate, 730 +/- 23 mmol/h of propionate and 196 +/- 5 mmol/h of butyrate were absorbed from the reticulorumen. With PPD treatment, 1,264 +/- 39 mmol/h of acetate, 220 +/- 5 mmol/h of butyrate and 721 +/- 17 mmol/h of PPD were absorbed from the reticulorumen. Glucose irreversible loss rate (ILR), as well as the relative enrichment of plasma lactate and alanine, were determined by primed continuous infusion of [U-13C]glucose in a jugular vein. Treatments did not affect (P > 0.10) the plasma concentrations of glucose (4.2 +/- 0.1 mmoVL), alanine (0.14 +/- 0.01 mmol/L), or insulin (80 +/- 25 pmol/L). The plasma concentration of lactate was higher (P < 0.05) with both propionate (0.84 +/- 5 mmol/L) and PPD treatment (0.81 +/- 5 mmol/ L) compared with the control treatment (0.29 +/- 0.5 mmol/L). The plasma concentration of pyruvate was higher (P < 0.05) with the propionate treatment (0.09 +/- 0.01 mmol/L) compared with the control treatment (0.03 +/- 0.01 mmol/L). The plasma concentration of 3-hydroxybutyrate was lower (P < 0.05) with the propionate treatment (0.15 +/- 0.03 mmol/L) compared with the control treatment (0.40 +/- 0.03). With the PPD treatment, the plasma concentrations of pyruvate and 3-hydroxybutyrate were in between the other treatments and tended (P < 0.10) to be different from both. The plasma concentration of PPD increased throughout the infusion period with the PPD treatment and reached a concentration of 4.9 +/- 0.6 mmol/L at 420 min. The ILR of glucose was not affected (P > 0.10) by treatments (441 +/- 35 mmol/h). The relative 13C enrichment of plasma lactate compared with that of glucose decreased (P < 0.05) with the PPD treatment compared with the control treatment (44 to 21 +/- 3%). It was concluded that PPD has a low rate of metabolism in cows without a normal functioning rumen, although about 10% of the absorbed PPD was metabolized into lactate.

The small intestinal digestibility of microbial amino acids and the apparent re-absorption of endogenous amino acids were estimated in three ruminal, duodenal and ileal fistulated lactating dairy cows. The cows were assigned to a 3 x 3 Latin square and fed diets low in amino acids, composed of straw, starch, molasses and urea. The treatments were three straw to concentrate ratios. The digestibilities and re-absorptions were estimated from duodenal and ileal flows of amino acids from feed, microbial and endogenous sources. The duodenal distribution of amino acids between different origins was determined by the difference method and by the amino acid profile method. The ileal distribution was estimated by a simple method where the feed fraction was estimated by the mobile bag method, the endogenous fraction as the water soluble fraction and the microbial fraction as the rest. In ileal samples, 8.0 +/- 1.1, 52.2 +/- 2.9 and 39.8 +/- 2.6% of total amino acid nitrogen (AAN) were of feed, microbial and endogenous origin, respectively, compared with 5.1 +/- 0.9, 61.6 +/- 6.6 and 32.7 +/- 6.9% in duodenal samples. The small intestinal digestibility of microbial AAN was estimated to be 75.1 +/- 1.8% and the apparent re-absorption of endogenous AAN to be 62.3 +/- 11.1%.

Three cannulated, lactating cows were used in a 3 x 3 Latin square design to determine the effect of roasting or NaOH treatment of barley on ruminal fermentation and site and extent of digestion of nutrients. Experimental treatments were rolled barley, roasted (exit temperature, 135 degrees C) and rolled barley, and treated with 4% NaOH and 220 L of H2O/tonne of barley. Diets also consisted of grass silage and soybean meal. Treatment with NaOH reduced concentrations of several AA, starch, and NDF in the barley. Starch digestibility in the rumen was lower for barley that was treated with NaOH but was unaffected for roasted barley. Digestibilities of N and starch in the small intestine were reduced for barley treated with NaOH, but values for rolled and roasted barley were similar. Apparent total tract digestibility of starch was reduced for the NaOH treated barley. Treatment of barley with NaOH tended to have a detrimental effect on feed intake, digestibility, and milk production. Roasting of barley did not appear to affect the site or extent of carbohydrate digestion, but roasting protected N from ruminal degradation. The protective effect on the carbohydrate fraction would be expected to be greater if the grain were cooled prior to rolling so that the protein matrix of the starch granule remained intact.

To study relationships of high undegradable intake protein and dietary fat on intestinal AA supply, the ruminal, intestinal, and total digestibilities of diets with or without added fat (5% of DM) and animal protein (blood meal: hydrolyzed feather meal, 1:1; 8% of DM) were examined with four cows in a 2 x 2 factorial design in a 4 x 4 Latin square experiment. Ruminal degradabilities were 14.9 and 18.6%, and intestinal digestibilities were 98.9 and 68.3%, respectively, for CP in blood meal and feather meal. Treatment effects on ruminal digestibilities were small. Protein supplementation increased total N intake by 29%, duodenal AA N flow by 39%, and AA N absorbed by 37%; absorption of Leu and Lys increased 60 and 33%, and absorption of Ile and Met increased 11 and 7%, respectively. Measured duodenal AA N flow (Cr2O3 marker) was 33% higher in cows cannulated adjacent to the pylorus compared with cows cannulated 100-cm distal to the pylorus, but only when cows were fed protein-supplemented diets; the estimates from those diets caused calculated microbial protein efficiency to exceed theoretical values. We postulated that blood meal and feather meal segregated near the pylorus, yielding high estimates of duodenal AA N flow. Removal of data for protein-supplemented diets obtained from cows cannulated at the pylorus yielded estimates of microbial protein synthetic efficiency consistent with literature values. Microbial synthesis of AA N was related linearly to ruminal digestion of carbohydrate. Location of intestinal cannulas may influence accuracy of nutrient flow estimates.
