RESUMO

MOTIVATION: In eukaryotes, the family of non-coding RNA genes includes a number of genes encoding small nucleolar RNAs (mainly C/D and H/ACA snoRNAs), which act as guides in the maturation or post-transcriptional modifications of target RNA molecules. Since in Drosophila melanogaster (Dm) only few examples of snoRNAs have been identified so far by cDNA libraries screening, integration of the molecular data with in silico identification of these types of genes could throw light on their organization in the Dm genome. RESULTS: We have performed a computational screening of the Dm genome for C/D snoRNA genes, followed by experimental validation of the putative candidates. Few of the 26 confirmed snoRNAs had been recognized by cDNA library analysis. Organization of the Dm genome was also found to be more variegated than previously suspected, with snoRNA genes nested in both the introns and exons of protein-coding genes. This finding suggests that the presence of additional mechanisms of snoRNA biogenesis based on the alternative production of overlapping mRNA/snoRNA molecules. Mechanical properties, diametral tensile strength (DTS) and flexural strength (FS) of six fluoride releasing materials were measured and compared. The samples were prepared and tested according to ISO specifications. The materials included a glass ionomer (Fuji IX), a resin-modified glass ionomer (Photac-Fil), two compomers (F 2000; Dyract AP) and two composites (Solitaire; Tetric Ceram). The tests were performed after the materials were stored in distilled water (DTS) and phosphate buffered saline solution (FS) at 37 degrees C for 24 hours and one week. Fluoride-releasing composite resin had the highest flexural and diametral tensile strengths and were statistically stronger than compomers, followed by resin-modified glass ionomer and conventional glass ionomer. However, a notable exception to this general trend was Solitaire, a fluoride-releasing composite resin.

Six fluoride-releasing materials of shade A3 were tested: one glass ionomer (Fuji IX), one resin-modified glass ionomer (Photac-Fil), two compomers (F 2000 and Dyract AP) and two composites (Tetric Ceram and Solitaire). Disk-shaped specimens of each material were prepared according to manufacturer's instructions, polished and L*a*b* baseline measurements taken. Specimens were randomly divided into two groups and given four different treatments of UV light exposure and immersion in a staining solution. Chromo Meter color measurements were taken following each treatment. Two-way ANOVA and Duncan Multiple Range post-hoc tests were used to compare color changes as a function of the four treatment conditions and one-way ANOVA was used to compare materials for each treatment separately. The results showed significant difference in shade A3 between products. In general, the hydrophobic materials showed greater color stability and stain resistance than the hydrophilic materials. Tetric ceram had the best color stability and stain resistance, while Fuji IX had the least.

RESUMO

The aim of this research was to evaluate the effectiveness of the Silicoater MD System for bonding resin to four different Brazilian alloys. The alloys included the following proprietary materials: Duracast, Durabond II, Palad and Premium. Biobond II, a popular nickel-chromium alloy has been used as control. Dentacolor served as the veneering agent. All specimens were thermo-cycled for 24 hours between 41 degrees and 131 degrees C at a dwell time of one minute per temperature. Shear-bond strengths ranged from 1667 to 2842 psi. Scanning electron microscopic evaluation demonstrated no gap formations at the resin/metal interface for all of the aforementioned systems, the Silicoater MD System demonstrated acceptable retention efficiency in five types of metallic alloys.

RESUMO

SUMMARY: VIRTLAB is a self-training program based on PBL (Problem-Based-Learning Pathway) built to simulate a molecular biology laboratory. It has been designed to stimulate students in the biological sciences to analyse and solve molecular biology problems using standard laboratory techniques (e.g. restriction enzyme digestions, analytical and preparative agarose gels, DNA cloning and sequencing, etc.) and can thus be viewed as a teaching aid.
