Maitake polysaccharides, after hydrothermal processing, were fermented with Lactobacillus acidophilus CCFM202 (L.A.) and Lactobacillus plantarum CCFM6392 (L.P.). The degradation of molecular weight of polysaccharides by hydrothermal processing under acidic conditions was obviously enhanced, which turned part of the water-insoluble-polysaccharides (WIP) into water-soluble-polysaccharides (WSPs). The pH value of water-soluble-polysaccharides (WSPs) and water-insoluble-polysaccharides (WIPs) were intensely dropped (4- 5) after 24 h fermentation. The optical density (O.D.) was increased (1.4- 2.3) due to bacterial growth, and short-chain fatty acids also followed this trend. LA-WSP predominantly produced acetic acid, 3- 4 folds to lactic acid, while LP-WIP groups produced dominant butyric acid (15- 17 folds). Hydrothermal processing induced the growth of L.A. and L.P., where the highest abundance was 2.5 × 104. From the Venn diagram, WSP-1 produced the most elevated metabolites (874). Therefore, experimental results show a significant impact on making WSPs fragments, whereas temperature and pH influence the WSPs degradation, withstand to higher fermentation efficacy.

RESEARCH BACKGROUND: Cardiovascular diseases and diabetes are the biggest causes of death globally. Bioactive peptides derived from many food proteins using enzymatic proteolysis and food processing have a positive impact on the prevention of these diseases. The bioactivity of Chinese pond turtle muscle proteins and their enzymatic hydrolysates has not received much attention, thus this study aims to investigate their antioxidant, antidiabetic and cytotoxic activities. EXPERIMENTAL APPROACH: Chinese pond turtle muscles were hydrolysed using four proteolytic enzymes (Alcalase, Flavourzyme, trypsin and bromelain) and the degrees of hydrolysis were measured. High-performance liquid chromatography (HPLC) was conducted to explore the amino acid profiles and molecular mass distribution of the hydrolysates. The antioxidant activities were evaluated using various in vitro tests, including 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), hydroxyl radical scavenging activity, reducing capacity, chelating Fe2+ and lipid peroxide inhibition activity. Antidiabetic activity was evaluated using α-amylase inhibition and α-glucosidase inhibition assays. Besides, cytotoxic effect of hydrolysates on human colon cancer (HT-29) cells was assessed. RESULTS AND CONCLUSIONS: The amino acid composition of the hydrolysates revealed higher mass fractions of glutamic, aspartic, lysine, hydroxyproline and hydrophobic amino acids. Significantly highest inhibition of lipid peroxidation was achieved when hydrolysate obtained with Alcalase was used. Protein hydrolysate produced with Flavourzyme had the highest radical scavenging activity measured by DPPH (68.32%), ABTS (74.12%) and FRAP (A 700 nm=0.300) assays, α-glucosidase (61.80%) inhibition and cytotoxic effect (82.26%) on HT-29 cell line at 550 µg/mL. Hydrolysates obtained with trypsin and bromelain had significantly highest (p<0.05) hydroxyl radical scavenging (92.70%) and Fe2+ metal chelating (63.29%) activities, respectively. The highest α-amylase (76.89%) inhibition was recorded when using hydrolysates obtained with bromelain and Flavourzyme. NOVELTY AND SCIENTIFIC CONTRIBUTION: Enzymatic hydrolysates of Chinese pond turtle muscle protein had high antioxidant, cytotoxic and antidiabetic activities. The findings of this study indicated that the bioactive hydrolysates or peptides from Chinese pond turtle muscle protein can be potential ingredients in pharmaceuticals and functional food formulations.

Grass turtle muscle was hydrolyzed with papain enzyme to produce protein hydrolysate (PH) and the degree of hydrolysis (DH) was determined. Under optimal conditions, the highest DH was 19.52% and the yield was recorded as 17.26%. Protein content of the hydrolysates was ranged from 73.35% to 76.63%. Total amino acids were more than 96.77% for each PH. The PH obtained at DH 19.52% achieved excellent solubility and emulsifying activity which were 95.56% and 108.76 m2/g, respectively at pH 6. Foam capacity amounted 100% in PH of DH 19.52% at pH 2, and water-holding capacity was 4.38 g/g. The antioxidant activity showed the strongest hydroxyl radical scavenging activity (95.25%), ABTS (84.88%), DPPH (75.89%), iron chelating (63.25%), and cupper chelating (66.90%) at DH 11.96%, whereas reducing power (0.88) at DH 19.52%. Thus, the findings indicated that utilization of grass turtle muscle protein hydrolysate is a potential alternative protein resource to improve the nutritional and functional properties in food ingredients and product formulations.

The crude polysaccharides (GFP) were isolated from the Maitake fruiting body (Grifola frondosa) and purified by DEAE Cellulose-52 ionic-exchange chromatography and Sephadex G-25 gel filtration chromatography in that order. Five main fractions, GFP-1 to GFP-5 were obtained through the isolation and purification steps. Free sugars were isolated by G-25 gel filtration chromatography and identified glucose and (α,α)-trehalose by nuclear magnetic resonance (NMR). GC-MS and methylation analysis that linkages were mainly ß-1,3 and ß-1,6, ß-1,4 and ß-1,2 bonds in WIP. Seven main oligomer products were detected and their structures characterized by mass spectrum. Experimental results shown the similarity in structure between water soluble polysaccharides (WSP) and water insoluble polysaccharides (WIP), thus WSP can be the product of cell wall by breakdown.

Haematological malignancies can have devastating effects on the patients' physical, emotional, psycho-sexual, educational and economic health. With the improvement of therapies patients with these malignancies are living longer, however significant proportion these patient show poor quality of life (QoL) due to various physical and psychological consequences of the disease and the treatments. Health-related QoL (HRQoL) is multi-dimensional and temporal, relating to a state of functional, physical, psychological and social/family well-being. Compared with the general population, HRQoL of these patients is worse in most dimensions. However without routine holistic need assessment (HNA), clinicians are unlikely to identify patients with clinically significant distress. Surviving cancer is a chronic life-altering condition with several factors negatively affecting their QoL, such as psychological problems, including depression and excessive fear of recurrence, as well as social aspects, such as unemployment and social isolation. These need to be adequately understood and addressed in the healthcare of long-term survivors of haematological cancer. Applying a holistic approach to patient care has many benefits and yet, only around 25% of cancer survivors in the UK receive a holistic needs assessment. The efforts of the last decade have established the importance of ensuring access to psychosocial services for haematological cancer survivors. We need to determine the most effective practices and how best to deliver them across diverse settings. Distress, like haematological cancer, is not a single entity, and one treatment does not fit all. Psychosocialoncology needs to increase its research in comparative effectiveness.

This cross-sectional study aimed to determine the age at menarche and its socioeconomic determinants among urban female students (n=680) in Bangladesh. The mean age of the respondents was 14±1.43years. Majority of the respondents were unmarried (98.4%). The mean age at menarche was 11.6±3.6years, median 12years. Almost one-third (35.7%) of the participants had menarche at the age of 12years. There was no statistically significant difference between age at menarche before and after 12years with the socio-economic characteristics, except education (p=<0.001). In the multivariate model, only higher education was statistically significant predictor of age at menarche.

Bangladesh shares a common border with India in the west, north and east and with Myanmar in the southeast. These borders cut across 57 rivers that discharge through Bangladesh into the Bay of Bengal in the south. The upstream courses of these rivers traverse India, China, Nepal and Bhutan. Transboundary flows are the important sources of water resources in Bangladesh. Among the 57 transboundary rivers, the Teesta is the fourth major river in Bangladesh after the Ganges, the Brahmaputra and the Meghna and Bangladesh occupies about 2071 km2. The Teesta River floodplain in Bangladesh accounts for 14% of the total cropped area and 9.15 million people of the country. The objective of this study was to investigate trends in both maximum and minimum water flow at Kaunia and Dalia stations for the Teesta River and the coping strategies developed by the communities to adjust with uncertain flood situations. The flow characteristics of the Teesta were analysed by calculating monthly maximum and minimum water levels and discharges from 1985 to 2006. Discharge of the Teesta over the last 22 years has been decreasing. Extreme low-flow conditions were likely to occur more frequently after the implementation of the Gozoldoba Barrage by India. However, a very sharp decrease in peak flows was also observed albeit unexpected high discharge in 1988, 1989, 1991, 1997, 1999 and 2004 with some in between April and October. Onrush of water causes frequent flash floods, whereas decreasing flow leaves the areas dependent on the Teesta vulnerable to droughts. Both these extreme situations had a negative impact on the lives and livelihoods of people dependent on the Teesta. Over the years, people have developed several risk mitigation strategies to adjust with both natural and anthropogenic flood situations. This article proposed the concept of 'MAXIN (maximum and minimum) flows' for river water justice for riparian land.

Introduction and Aim. Injecting drug users (IDUs) are amongst the most vulnerable people to acquisition of HIV/AIDS. This study aims to collect information on IDUs and their health seeking behavior in Bangladesh. Design and Methods. A cross-sectional study was conducted among 120 IDUs attending a drug rehabilitation center in Dhaka, Bangladesh. Data were collected on sociodemographics, drug use, health seeking behavior, knowledge of injecting drugs, and sexual behavior. Results. The mean ± SD and median (IQR) age of the participants were 32.5 ± 21.3 and 33 (27-38) years, respectively, with only 9.2% females. Injection buprenorphine was the drug of choice for 40% of participants, and 58% of the participants first started drug use with smoking cannabis. 73.3% of participants shared needles sometimes and 57.5% were willing to use the needle exchange programs. 60% of the participants had no knowledge about the diseases spread by injection. Condom use during the last intercourse with regular partners was 11.7% and with any partners 15.8%. Conclusion. IDUs in Bangladesh are a high-risk group for HIV/AIDS due to lack of knowledge and risky behaviors. Education and interventions specifically aimed at IDUs are needed, because traditional education may not reach IDUs or influence their behavior.

BACKGROUND: Hypertension is a major risk factor for several cardiovascular diseases (CVD). The prevalence of hypertension is increasing in Bangladesh, especially in urban areas. The objective of this study was to estimate the prevalence of hypertension and its risk factors in an urban area of Bangladesh. METHODS: We conducted a cross-sectional survey involving participants aged ≥ 25 years in an urban area in Dhaka between June-December 2012, using multi-stage random sampling. Data on socioeconomic status, tobacco use, physical activity, diet, extra-salt use, family history of hypertension, CVD, anthropometric measurements and blood pressure were collected using modified WHO-STEPS protocol. Hypertension and pre-hypertension were defined according to JNC-7. Multiple logistic regressions models were used to identify risk factors associated with hypertension. RESULTS: The overall age-adjusted prevalence hypertension and pre-hypertension among 730 participants was 23.7% and 19%, respectively, which was higher among males compared to females (23.6% vs 21.71% and 21.7% vs 17.0%, respectively). Bivariate analysis showed significant relationship of hypertension with age, BMI, no physical activity, tobacco use, extra salt intake and family history of stroke/cardiovascular disease. In the multivariate model, factors significantly associated with hypertension were older age (OR 19.18, 95% CI 13.58-28.11), smoking (OR 3.47, 95% CI 2.85-5.19), extra salt intake (OR 1.13, 95% CI 1.04-2.21), and high waist circumference (OR 3.41, 95% CI 2.81-5.29). CONCLUSIONS: The prevalence of hypertension and pre-hypertension was high among our study participants. Population-based intervention programs and policies for increased awareness about the risk factors, and life-style modification are essential for prevention of hypertension.

Assessment of growth regulator and NPK fertilization effects are important tools for flower stimulation and yield improvement in cucurbits. This investigation demonstrates the comparative male-female flower induction and fruit yield of small sized bitter gourd treated with NPK fertilizers and plant growth regulators. Namely, two experiments having three replicates were conducted in a Randomized Complete Block Design (RCBD) with NPK fertilization and plant growth regulators-GA3, NAA and Ethophon application on small sized bitter gourd-genotype BG5 at the research field of the Bangabandhu Sheikh Mujibur Rahman Agricultural University (BSMRAU). In experiment 1, different doses of NPK fertilizers comprised of 10 treatments and in that of experiment 2, different levels of plant growth regulators indicated 10 treatments. The results indicated that application of different doses of NPK fertilizer and plant growth regulators significantly (< or = 0.05) influenced over the flower initiation and fruit setting. The application of N90-P45-K60 fertilizer along with Ethophon spraying resulted in the better yield of small sized bitter gourd.

We have studied physiological parameters in a living cell using fluorescence lifetime imaging of endogenous chromophores. In this study, pH dependence of the fluorescence lifetime of flavin adenine dinucleotide (FAD), that is a significant cofactor exhibiting autofluorescence, has been investigated in buffer solution and in cells. The fluorescence lifetime of FAD remained unchanged with pH 5 to 9 in solution. However, the fluorescence lifetime in HeLa cells was found to decrease with increasing intracellular pH, suggesting that pH in a single cell can be estimated from the fluorescence lifetime imaging of FAD without adding exogenous fluorescent probes.

Stem cell transplantation forms an integral part of the treatment for multiple myeloma. This paper reviews the current role of transplantation and the progress that has been made in order to optimize the success of this therapy. Effective induction chemotherapy is important and a combination regimen incorporating the novel agent bortezomib is now favorable. Adequate induction is a crucial adjunct to stem cell transplantation and in some cases may potentially postpone the need for transplant. Different conditioning agents prior to transplantation have been explored: high-dose melphalan is most commonly used and bortezomib is a promising additional agent. There is no well-defined superior transplantation protocol but single or tandem autologous stem cell transplantations are those most commonly used, with allogeneic transplantation only used in clinical trials. The appropriate timing of transplantation in the treatment plan is a matter of debate. Consolidation and maintenance chemotherapies, particularly thalidomide and bortezomib, aim to improve and prolong disease response to transplantation and delay recurrence. Prognostic factors for the outcome of stem cell transplant in myeloma have been highlighted. Despite good responses to chemotherapy and transplantation, the problem of disease recurrence persists. Thus, there is still much room for improvement. Treatments which harness the graft-versus-myeloma effect may offer a potential cure for this disease. Trials of novel agents are underway, including targeted therapies for specific antigens such as vaccines and monoclonal antibodies.

Fluorescence lifetime images of reduced nicotinamide adenine dinucleotide (NADH) that is a key cofactor in cellular metabolism were obtained in a cell at various values of intracellular pH. The average fluorescence lifetime of NADH is found to become shorter monotonically with increasing pH, indicating that pH in a single cell can be determined by fluorescence lifetime imaging of NADH without adding exogenous fluorescent probes. The magnitude of the pH-induced lifetime change is higher in cells than that in buffer solution. The fluorescence lifetime of NADH is not uniform inside a cell; the fluorescence lifetime of nuclear NADH is shorter than that of mitochondrial NADH at each pH, and the magnitude of the pH-induced change is larger in nuclei than in other areas. The local electric field effect on the fluorescence lifetime is discussed since this effect may be one of the strong possibilities for the nonuniformity of the autofluorescence lifetime of NADH in cells.

Fluorescence decays of flavin adenine dinucleotide (FAD) that is a typical autofluorescent species in cells and tissues have been measured in a mixture of alcohol and water in the femtosecond and nanosecond time range. The fluorescence lifetimes of both the stacked conformation between the isoalloxazine and adenine moieties in close proximity and the extended open conformation in water are affected by the addition of alcohol. The nanosecond fluorescence lifetime of the open conformation increases with decreasing dielectric constant of the medium, contributing to the enhancement of the fluorescence intensity of FAD in less dielectric media. The fluorescence lifetime of the open conformation is also affected by medium viscosity, suggesting that the photoexcited open conformation is quenched by the dynamic interaction between the two aromatic rings. The fluorescence component decaying in tens of picoseconds is attributed to the stacked conformation that shows the efficient fluorescence quenching due to the intramolecular electron transfer. The picosecond fluorescence lifetime of the stacked conformation increases with decreasing dielectric constant, suggesting the shift of the distribution of the stacked conformation to a longer intramolecular distance between the two aromatic rings in less dielectric media. The pre-exponential factor of the picosecond decaying component relative to that of the nanosecond one decreases with the increase of the alcohol concentration in the femtosecond time-resolved fluorescence, which demonstrates the increase in the population of the open conformation with the reduction of the dielectric constant. The possibility to evaluate the polar environment in a cell by the fluorescence lifetime of FAD is discussed based on the results obtained.

Hematological cancers and non-malignant hematological disorders are biologically diverse conditions and are treated differently. We compared the pattern of EBV infections following allogeneic stem cell transplantation between the above two groups of hematological disorders. Eighty-three transplants were evaluated over a consecutive 7-year period at a single center. No difference was found in the incidence of post-transplant EBV infections between the two groups, though a higher median peak viral load was noted in the non-malignant group (P=0·04). Pre-transplant immunosuppressive therapy with antithymocyte globulin (ATG) significantly increased the risk of post-transplant EBV infections (P=0·04) in the non-malignant group patients. No significance was found for prior cytotoxic chemotherapy among the malignant group of patients. Alemtuzumab based conditioning was not associated with an increased risk for EBV infections in either of the groups. Treatment with two or more courses of ATG was found to be significantly associated with post-transplant EBV-related PTLD (P=0·01). Post-transplant EBV infections did not influence overall survival (non-malignant, P=0·66; malignant, P=0·41) in either of the subgroups. There were no deaths directly attributable to EBV infections.

The haematological indications for allogeneic stem cell transplantation can be broadly divided into non-malignant and malignant disorders. We compared the incidence and risk factors for post-transplant cytomegalovirus (CMV) infections between these two biologically diverse subgroups of haematological conditions. Out of 105 allogeneic transplants, 64 and 41 were for underlying non-malignant and malignant indications respectively. CMV infections were significantly more frequent (P=0.016) in the malignant subgroup. Pre-transplant recipient CMV seropositivity in both subgroups (negative versus positive; non-malignant,P=0.023; malignant, p<0.001), donor seropositivity (P=0.002) and acute graft-versus-host disease (GVHD) (P=0.02) in the non-malignant subgroup and > or =3 courses of previous cytotoxic therapy (P=0.023) in the malignant subgroup were found to be associated with an increased risk of CMV infections. On multivariate analysis, donor seropositivity in the non-malignant patients (negative versus positive,P=0.022; odds ratio: 0.18) and recipient seropositivity in patients with malignancies (negative versus positive; P=0.001, odds ratio: 0.01) were identified to be significant factors for risk of CMV infection.
