RESUMO

A precise and reproducible method of quantitative determination of the photosensitizer Photolon in hepatic tissue of the liver of rats by means of spectrophotometric assay with preliminary chemical extraction was developed for validation of the method of in vivo measurement of a fluorescent signal of the tissue after injection of the photosensitizer. Using the ex vivo spectrophotometric technique a quantitative assessment of the results of spectrofluorescent measurements performed in vivo was made. A high correlation (R2=0.99) between results estimated by two different techniques was shown. Also validity of the simple and easy-to-use method of in vivo fluorescence intensity measurements for the investigation of the pharmacokinetics and biodistribution of different photosensitizers was demonstrated.

RESUMO

A method for measuring the magnetic susceptibility of separate cells has been theoretically developed and implemented in experiments. It is based on fitting the videocomputer and predicted data for the integral motion curve of cells that settle in a fluid along a thin ferromagnetic bar by the action of a uniform magnetic field. The magnetic susceptibility of HeLa tumor cells and culture medium 199 has been measured.

RESUMO

Radionuclide angiography was performed in rats with transplantable tumors (sarcoma 45 and Walker carcinosarcoma 256). Induced hyperglycemia was shown to result in blood flow inhibition in tumor and normal tissues of tumor-bearing rats. Some differences were revealed in a degree of reversibility of blood flow disorders in tissues of the above strains. The results obtained confirmed the advisability of radiation therapy at the height of a decrease in tumor blood flow.

RESUMO

Artificial hyperglycemia (AHG) used in the cancer treatment considerably increases the insulin and S-peptide content in blood, decreases the glucagon concentration and tissue glucose tolerance. The changes are reversible, have a functional character, that indicates the absence of a disbalance in hormonal mechanisms of carbohydrate metabolism control as well as marked disorders of the pancreas function. In order to achieve a wider range of indications for using AHG in complex schemes of the cancer treatment, it is necessary to take measures on optimization of the carbohydrate metabolism state which should be controlled by the suggested complex of laboratory tests.

RESUMO

In the experiments on the C57Bl mice the authors studied the effect of artificial hyperglycemia (AH), amputation of the extremities with tumors as well as combinations of these effects on the intensity of metastatic spreading of carcinoma RL-67 to the lungs. AH did not prove to intensify the process of metastasizing if it was conducted on the 1, 3, 5, 7, 9 and 11th days. The average number of metastases did not differ from that in the control group. AH which was conducted one day before amputation of the extremity with the tumor caused a more significant inhibition of metastasizing than a surgical intervention.

RESUMO

The injection of sarcolysine, methotrexate, thiophosphamide, cyclophosphamide, adryamycin and platidinum during glucose infusion is shown to increase their antitumour activity. The antitumour effectiveness of 5-FU did not increase under conditions of artificial hyperglycemia, while that of vincristine decreased. Intensification of the activity of some chemotherapeutic drugs in combination with hyperglycemia was observed against the background of their increased general toxic effects.

RESUMO

The effect of multiple artificial hyperglycemia treatments on metastatic spreading and tumour growth depending on the stage of the tumour development is studied in experiments on mice with transplanted subcutaneous lung cancer LC-67. Intensification of the metastatic spreading and of the primary tumour growth under hyperglycemia treatment of early and advanced diseases is detected. Artificial hyperglycemia does not influence the tumour progress at the terminal stage of the disease.
