RESUMO

The objective of this study was to investigate the pattern of age-associated oxidative post-translational modifications in the skeletal muscles of a mammalian species and to address whether the modifications result in the loss of function of the oxidatively modified protein(s). Accordingly, proteins in the mitochondrial matrix of the hind limb of C57BL/6Nnia mice were examined for modifications by carbonylation--an established marker of oxidative post-translational modifications--by Western blotting using anti-2,4-dinitrophenyl antibodies and tritiated sodium borohydride methods. An age-associated increase in carbonylation of mitochondrial matrix proteins was observed, but not all proteins were equally susceptible. A 55 kDa protein, identified as the α-subunit of the F1 complex of ATP synthase (ATP phosphohydrolase [H(+)-transporting]), had approximately 17% and 27% higher levels of protein carbonyls in adult and old animals, respectively, in comparison to the young controls as estimated using tritiated sodium borohydride. In addition, an age-associated decline in its activity was observed, with approximately 9% and 28% decrease in the activity in the adult and old animals, respectively, in comparison to young controls. It may be concluded that such oxidative post-translational modifications and the resultant attenuation of the protein activity may contribute to the age-related energy loss and muscular degeneracy.

Portal vein thrombosis (PVT) is a rare disorder that is associated with a variety of underlying condition of which liver cirrhosis, malignancy and myeloproliferative disorders are the most common. It is of two types, acute and chronic portal vein thrombosis. Anticoagulation therapy is recommended for all patients with acute portal vein thrombosis. Chronic portal vein thrombosis is characterised by the development o f portal hypertension. Bleeding from ruptured varices is the main complication. In the absence of bleeding, continuous anticoagulation therapy should be considered for chronic portal vein thrombosis in whom an underlying prothrombotic factor is to be identified. Here in this report a 13-year-old girl presented with haematemesis. The spleen was hugely enlarged. Her Hb was 8.38 g/dl. Grade III oesophageal varices were found in oesophagogastroduodenostomy. CT abdomen showed portal cavernoma formation with increased splenic collateral. Protein C activity was 45% and protein S activity was 40%. She was treated with beta-blocker, endoscopic variceal ligation followed by low molecular weight heparin and warfarin.

A study was done on 30 patients admitted in the department of medicine, RG Kar MCH to know the incidence and prevalence of systemic lupus erythematosus (SLE) patients and among them whether there were any cases of renal involvement, time of onset of clinicopathological features of renal involvement and renal changes if already been started in them who had no renal problem clinically. The cases were selected on the basic of revised ARA criteria. Total clinical examination and biochemical tests were done followed by renal biopsy in 28 cases. In rest 2 case, renal biopsy could not be performed because one case presented with severe azotaemia and another case presented with end stage renal disease with severe pallor. There were 28 females and 2 males in the present study presenting chiefly with joint pain and fever. Although renal symptoms were not very common in their presentation, in the present study, some form of renal abnormality (either on urine analysis or histology) was found in 26 patient. Among renal involvement, type IV diffuse proliferative lupus nephritis was more common. Incidence and prevalence of SLE was more common in this part of the world. When worldwise incidence of SLE is considered, renal involvement was quite high (96%) in the present study, type IV diffuse proliferative lupus nephritis was commonest type and in severe form.

OBJECTIVE: To study the changing incidence of blood transfusion-related viral infections consequent to compulsory screening of blood and greater awareness of the problem, over the last five years. METHODS: This is a cross-sectional study carried out at Medical College, Calcutta. Three groups each consisting of 100 subjects were selected for this study. Group A comprised multiple transfused patients who have also received transfusion before 1995. Group B comprised patients who had received transfusions only since 1995. Group C comprised of control patients who have never been transfused. The incidence of HBsAg +ve, anti-HCV +ve and HIV +ve cases were calculated and expressed as percentages and compared using the chi square test. RESULTS: The incidences of HBsAg +ve and anti-HCV +ve cases in the three groups were 20% and 16% in Group A, 7% and 6% in Group B and 4% and 2% in Group C. The difference between Group A and Group B were statistically significant. CONCLUSION: The incidence of HBsAg and anti-HCV positive cases among the multi-transfused has decreased over the last five years.

Geometrical isomers of testosterone 3-(O-carboxymethyl)oxime and their histamine derivatives were purified on reverse-phase high pressure liquid chromatography, and their antibody binding characteristics were studied. Using a competitive testosterone enzyme immunoassay, the unfractionated mixture of the oximes showed 75% cross-reactivity with respect to testosterone, whereas the isolated 3Z- and 3E-isomers showed 124% and 26% cross-reactivity, respectively. The cross-reactivity was increased in the histamine derivatives, but the difference in cross-reactivity of the two isomers was reduced. Suppression of the ionization of the carboxyl group by lowering the pH of the incubation mixture in the antigen-antibody binding step raised the cross-reactivity of the mixture of free oximes to 128%, at pH 4.0. Thus, the geometry and ionization state of the carboxymethyl oxime group has a profound effect on the affinity of the isomers for the antibody.

Chromatographic resolution of polyclonal antibodies is a challenging analytical problem and a successful method may have many applications. We have resolved polyclonal antibodies against testosterone 3-(O-carboxymethyl)oxime on a homologous testosterone-Sepharose matrix by using a ternary gradient system of propionate-1 M propionic acid-2 M propionic acid. Nineteen peaks were detected, sixteen of which were characterized by steroid binding activity and IgG assay. The relative affinities of the pooled fractions, determined by ELISA and by thiocyanate elution, correlated well with their retention times. The slopes of the displacement curves in antigen-immobilized testosterone ELISA increased with retention times of the fractions; testosterone required for 50% displacement being, 90, 5.8 and 24 ng/well for fraction I, fraction XVI and total antiserum, respectively. Cross-reactivity of the fractions towards 5alpha-dihydrotestosterone varied from 31 to 74% compared to 47% in the total antiserum.
