Genetic parameters for ultimate pH of pectoralis major muscle (PM-pHu) and sartorius muscle (SART-pHu); color parameters L*, a*, b*; logarithm of drip loss (LogDL) of pectoralis major (PM) muscle; breast meat yield (BMY); thigh and drumstick yield (TY); abdominal fat percentage (AFP); and BW at 6 wk (BW6) were estimated in 2 lines of broiler chickens divergently selected for PM-pHu. Effects of selection on all the previous traits and on glycolytic potential, pectoralis major muscle pH at 15 min postmortem, curing-cooking yield (CCY), cooking loss (CL), and Warner-Bratzler shear force (WBSF) of the PM muscle were also analyzed after 5 generations. Strong genetic determinism of PM-pHu was observed, with estimated h(2) of 0.57 ± 0.02. There was a significant positive genetic correlation (rg) between PM-pHu and SART-pHu (0.54 ± 0.04), indicating that selection had a general rather than a specific effect on energy storage in skeletal muscles. The h(2) estimates of L*, a*, and b* parameters were 0.58 ± 0.02, 0.39 ± 0.02, and 0.48 ± 0.02, respectively. Heritability estimates for TY, BMY, and AFP were 0.39 ± 0.04, 0.52 ± 0.01, and 0.71 ± 0.02, respectively. Our results indicated different genetic control of LogDL and L* of the meat between the 2 lines; these traits had a strong rg with PM-pHu in the line selected for low ultimate pH (pHu) value (pHu-; -0.80 and -0.71, respectively), which was not observed in the line selected for high pHu value (pHu+; -0.04 and -0.29, respectively). A significant positive rg (0.21 ± 0.04) was observed between PM-pHu and BMY but not between PM-pHu and BW6, AFP, or TY. Significant phenotypic differences were observed after 5 generations of selection between the 2 lines. The mean differences (P < 0.001) in pHu between the 2 lines were 0.42 and 0.21 pH units in the breast and thigh muscle, respectively. Breast meat in the pHu+ line exhibited lower L* (-5 units; P < 0.001), a* (-0.22 units; P < 0.001), b* (-1.53 units; P < 0.001), and drip loss (-1.6 units; P < 0.001) than in the pHu- line. Breast meat of the pHu+ line was also characterized by greater CCY (+6.1 units; P < 0.001), lower CL (-1.66 units; P < 0.01), and lower WBSF after cooking (-5.1 units; P < 0.001) compared to the pHu- line. This study highlighted that selection based on pHu can be effective in improving the processing ability of breast meat and reducing the incidence of meat quality defects without affecting chicken growth performance.

The structural and metabolic characteristics of the pectoralis major (P. major) muscle (i.e., breast muscle) and the quality of the resulting meat were studied in relation to breast muscle fiber development in broiler chickens. Six hundred birds originating from a commercial, grand parental, male heavy line (Hubbard-Europe, Châteaubourg, France) were kept under conventional breeding methods until their usual marketing age of 6 wk. For all birds, the plasma creatine kinase activity and the P. major muscle fiber cross-sectional area (CSA), glycolytic potential, lactate content, pH at 15 min postmortem, as well as the ultimate pH, CIELAB color parameters [lightness (L*), redness (a*), and yellowness (b*)], and drip loss of breast meat, were measured. Increased breast weight and yield were associated with increased fiber CSA, reduced muscle glycolytic potential, and reduced lactate content at 15 min postmortem. Therefore, P. major muscle exhibiting larger fiber CSA exhibited greater pH at 15 min postmortem and ultimate pH, produced breast meat with lower L* and reduced drip loss, and was potentially better adapted to further processing than muscle exhibiting small fiber CSA.

RESUMO

1. The aim of this study was to compare the behavioural and physiological responses to hanging and acute heat stress in three different chicken breeds. Chicks were obtained from a slow-growing French 'Label Rouge' line (SGL), a fast-growing standard line (FGL) and a heavy line (HL). The SGL, FGL and HL birds were slaughtered at their respective market ages of 12, 6 and 6 weeks, in an attempt to achieve similar body weights. Before stunning, birds were either shackled by their legs on the moving line for 2 min (shackling stress: SH) or placed in a room at 35 degrees C and 60% of humidity for 3.5 h and then shackled for 2 min (acute heat stress plus shackling: H + SH) or subjected to minimal stress by shackling for 10 s before stunning (control group: C). 2. Bird physiological responses to the three pre-slaughter treatments were estimated by measuring blood corticosterone, glycaemia, creatine kinase activity, acid-base status and electrolyte concentration as well as lactate content and glycolytic potential in the breast (Pectoralis major) and thigh (Ilio tibialis) muscles. Behavioural responses to shackling stress were evaluated by measuring wing flapping duration, straightening up attempts and vocalisations. 3. Blood corticosterone was higher in SH and H+SH groups than in the C group, regardless of genotype. The struggling activity on the shackle line differed among chicken breeds. It was more intense and occurred more rapidly after hanging in the SGL birds than in both other breeds. Furthermore, SGL struggling activity was not affected by hanging duration while it increased with hanging duration in FGL and HL birds. 4. Wing flapping duration was negatively correlated with blood pH, bicarbonate concentration and positively correlated with breast muscle lactate content, indicating that struggling stimulated antemortem glycolysis activity in breast muscle. Acute heat stress affected blood Ca2+ and Na+ concentration and increased glycaemia and glycolytic potential of thigh muscle. 5. Both acute heat stress and shackling before slaughter were experienced as stressful events by all types of birds.

1. Pectoralis major (P. major) muscle pH and meat quality traits were studied in relation to bird response to ante-mortem stress in three chicken lines: a fast-growing standard line (FGL), a slow-growing French 'Label Rouge' line (SGL) and a heavy line (HL). Ninety-nine birds of the three genetic types were slaughtered at their usual marketing age (6, 12 and 6 weeks for FGL, SGL and HL birds, respectively) on the same day. The birds of each line were divided into three different ante-mortem treatment groups: minimum stress (shackling for 10 s) (C), shackling for 2 min (SH) and acute heat plus shackling stress (exposure to 35 degrees C for 3.5 h and shackling for 2 min before stunning) (H + SH). 2. Regardless of chicken line, wing flapping duration (WFD) between hanging and stunning was strongly negatively related to P. major muscle pH at 15 min post-mortem. It was also moderately negatively related to P. major muscle glycolytic potential (GP), which represents glycogen level at death. Increasing WFD induced an increased ultimate pH (pHu) only in HL. The consequences of increased WFD for breast meat traits were dependent on the chicken line: it induced lower L* and b* and higher a* and drip loss in SGL while it only increased breast a* in HL birds. By contrast, WFD variations did not alter breast meat quality traits of FGL birds. Regardless of the chicken line, increased GP was associated with lower pHu and higher L* and drip loss. In SGL, it also increased b* and decreased curing-cooking yield of breast meat. 3. Struggling activity on the shackle line and muscle glycogen content at death could partly explain line and pre-slaughter variations in breast meat pH and quality traits. The water holding capacity of the raw and cooked meat was impaired by long shackling in the case of SGL birds while it was barely affected by ante-mortem conditions in the two standard lines. In conditions which minimised bird struggling (C), SGL and FGL birds had meat with a better water holding ability than that of broilers from the heavy line. However, when broilers were subjected to SH or H + SH conditions, the breast meat water holding capacity of SGL birds was lowered to the same level as that of the heavy line birds.

Ad libitum feeding reduces livability and reproductive fitness in broiler breeder hens. Two genotypes, a standard (S) and an experimental dwarf broiler breeder (E), were fed ad libitum (SA and EA, respectively), restricted at 55% of ad libitum feed intake (intermediate restriction) from 6 to 15 wk of age (SI and EI, respectively), or restricted (SR and ER, respectively) to match a standard growth curve with a diluted mash feed (2,400 kcal/kg). The experiment was repeated at 2 locations (experiment 1 = 672 hens in pens from 0 to 40 wk; experiment 2 = 420 hens in pens and cages from 0 to 53 wk). Feed restriction reduced adult BW by 20% compared with ad libitum feeding, delayed sexual maturity by 2 to 4 wk, and improved livability. Hens fed the intermediate diet immediately compensated after 15 wk of age to reach BW, sexual maturity, and livability close to those of ad libitum-fed hens. The E genotype exhibited better tolerance to ad libitum feeding than the S genotype in all measured aspects. Average laying rate during the first 24 wk of lay was 66.4, 77.4, 69.9, 47.2, 57.9, and 72.4% for EA, EI, ER, SA, SI, and SR respectively in experiment 2. Egg abnormalities (double yolk, shell problems) decreased after the peak of lay but remained consistently higher for S compared with E, and for ad libitum and intermediate diets compared with the restricted diet. Yolk deposition rate was measured by a double dye technique. Duration of yolk rapid growth was 8.8 d in E and 9.3 d in S hens (P < 0.001), but this difference did not explain the observed variations in laying rate. The potential to increase feed allowances even with a diluted diet in broiler breeder hens requires adapted genotypes.

(1) Tolerance to ad libitum feeding was compared in three genotypes of broiler breeder hens: a standard broiler breeder fed ad libitum (SA) or restricted (SR), a slow growing 'label' broiler breeder (L) and an experimental dwarf heavy broiler breeder (E). Two similar experiments were conducted in two distinct research centres. (2) Feed intake and body weight were measured every 3 weeks from hatch to 40 to 49 weeks of age. Egg production and egg abnormalities were recorded. The number of yellow follicles in ovaries was counted at the age of 32 weeks. (3) Body weight was stabilised at 2.2, 3.7 and 5.4 kg after 24 weeks of age in L, E and SA hens, respectively. Growth of the SR hens was similar to that of L up to 20 weeks and stabilised at a similar level to that of E hens after 30 weeks of age. (4) Sexual maturity was delayed by 6 weeks in restricted breeders compared to ad libitum fed hens that started to lay at 20 weeks. SA hens had low egg production and a high proportion of defective eggs, which was largely compensated for by feed restriction. However, productivity of SR hens remained lower than that of L breeders. (5) Compared to the low viability and reproductive fitness observed with SA hens, the E dwarf broiler breeder tolerated ad libitum feeding and had better egg production, fewer egg abnormalities and yellow follicles per ovary and a higher egg production. However, laying rate was still lower than that of the SR and L groups. Energy conversion (kJ/g egg) from 32 to 40 weeks of age was much higher in the SA group than in the other three groups. 6. The feasibility of feeding a dwarf broiler breeder ad libitum calls for further research on implications of specific IGF and GH-receptor expression at the level of the ovary in dw chickens.

In broiler breeder management, stringent feed restriction is practiced to reduce body size in order to improve egg production and meet broiler production demand, but this practice has raised welfare issues. The potential for the dwarfing (dw) gene to reduce feed intake and body size of breeders under ad libitum feeding or less stringent restriction while maintaining improved egg production has been reported. In this study, we compared embryo physiology, quality of chicks, and performance of broilers from eggs of dwarf breeders with those from a standard broiler breeder. Hatching eggs from 3 commercial lines of broiler breeders were compared for incubation parameters, 1-d-old chick weight, chick quality, and broiler growth to 41 d of age. The lines included a standard heavy (S) line, an experimental (E) line, and a label-type (L) line. The E and L line breeders carry the sex-linked dw gene and are being used to assess the potential for dw to reduce feed intake or lower feed restriction and improve reproductive performance in heavy female broiler parent stock. Two separate experiments were conducted. All female parent stocks were mated to Cornish males, and fertile eggs were collected. In the first experiment, eggs were incubated for 21 d under standard conditions to determine, during final stages of incubation, corticosterone and thyroid hormone levels (triiodothyronine, T3; thyroxine, T4) in embryos and hatchlings, CO2 partial pressure (pCO2), and O2 partial pressure (pO2) in air cells, heat production by eggs and 1-d-old weights. In the second experiment, eggs were incubated for 21 d to compare chick quality, chick weights at 1 d of age, and broiler growth to 7 and 41 d. Average egg weights were higher for the S and L lines than the E line, but weight loss during incubation was lowest for the E line. Plasma T3 and T3/T4 ratio was similar between lines at IP, but corticosterone was higher in the S line. At hatch, T3/T4 ratio was higher in the S line compared with the E and L lines, but corticosterone was higher in the S and E lines than in the L line. Heat production by embryos was different among lines (S > E > L). The pCO2 was also higher in the S line than the E and L lines. These incubation parameters suggest different metabolic rates among lines (S > E > L). Incubation duration was shortest for the S line. Chick weights at 1 d old were not different between lines. Chick quality scores were also not different when expressed as a percentage of high-quality chicks or as an overall average score of each line. However, broiler BW at 7 and 41 d were different among lines (S > E > L). Chicks of higher quality (score of 100) in all lines had higher BW than those of lower quality (score of < 100). For corresponding quality groups between lines, the S line had higher BW, and those of the L had the lowest. These data suggest a link between the levels of embryo metabolism and growth potentials of the lines. We concluded that the dw gene has potential for reducing feed intake in heavy broiler parent stock (as in the E line), improving reproductive performance (as in the L and E lines), and maintaining progeny broiler chick weights at 1 d of age and quality but with lower weight at slaughter.

The present study was aimed at estimating the genetic variability between lines of breast and thigh meat quality (pH decline, color, drip loss, and curing-cooking yield) by comparing a slow-growing French label-type line (SGL) and a fast-growing standard line (FGL) of chickens exposed to different preslaughter stress conditions. The birds were slaughtered under optimal conditions or after exposure to 2 h of transport or acute-heat stress (2 h at 35 degrees C). Relationships between meat quality and stress sensitivity were investigated by measuring struggle during shackling and tonic immobility (TI) duration, 1 wk before slaughter, as an indicator of the basal level of fear of the birds. Although most of the meat quality indicators varied between the 2 lines, differences were muscle dependent. In concordance with a lower ultimate pH, curing-cooking yield of thigh meat was decreased for the FGL birds. In contrast, these birds had a higher curing-cooking yield and a lower drip loss of breast meat resulting from a less rapid pH decline in this muscle compared with SGL birds. Thigh meat characteristics were influenced by both preslaughter stresses, but no significant effects were detected for breast meat. The main effect of heat stress in thigh meat was a decrease of the ultimate pH and led to paler color and lower curing-cooking yield; opposite effects were obtained for transport. Breast meat was much more sensitive to physical activity of birds on the shackle line. Longer durations of wing flapping on the shackle line gave more rapid initial pH decline. Whatever the line, no relationship between TI duration and meat quality characteristics or activity was observed. The present study suggested that SGL birds could be at disadvantage due to more struggle during shackling and accelerated postmortem glycolysis, which is detrimental to the quality of breast meat.
