RESUMO

Objective To investigate the role and mechanism of the small-molecule inhibitor CIL56 in the death of esophageal squamous cell carcinoma cells. Methods SRB method and plate-cloning method were used to detect the effect of CIL56 on the proliferation of esophageal squamous cell carcinoma. The effect of CIL56 on the migration of esophageal squamous cell carcinoma cells was investigated by scratch-healing test. The effect of CIL56 on the concentration of iron ions in esophageal squamous cell carcinoma was detected with an iron-detection kit. A total glutathione test kit was used to examine the effect of CIL56 on glutathione concentration in esophageal squamous cell carcinoma. Western blot was used to investigate the effect of CIL56 on the expression of xCT and GPX4 proteins related to iron death, as well as YAP1 protein, in esophageal squamous cell carcinoma. Results CIL56 could significantly inhibit the proliferation (P < 0.05) and migration (P < 0.001) of esophageal squamous cell carcinoma. With the increased CIL56 concentration, the iron concentration in esophageal squamous cell carcinoma increased (P < 0.05). CIL56 could reduce the glutathione content in esophageal squamous cell carcinoma (P < 0.01). CIL56 could reduce the expression of xCT and GPX4 proteins related to iron death and decrease the level of YAP1 protein in esophageal squamous cell carcinoma (both P < 0.001). Conclusion The small-molecule inhibitor CIL56 can significantly inhibit the proliferation and migration of esophageal squamous cell carcinoma cells and reduce the expression of the iron-death-related proteins xCT and GPX4, as well as YAP1 protein.

RESUMO

SARS-CoV-2 is a single-stranded RNA betacoronavirus with a high mutation rate. The rapidly emerged SARS-CoV-2 variants could increase the transmissibility, aggravate the severity, and even fade the vaccine protection. Although the coinfections of SARS-CoV-2 with other respiratory pathogens have been reported, whether multiple SARS-CoV-2 variants coinfection exists remains controversial. This study collected 12,986 and 4,113 SARS-CoV-2 genomes from the GISAID database on May 11, 2020 (GISAID20May11) and April 1, 2021 (GISAID21Apr1), respectively. With the single-nucleotide variants (SNV) and network clique analysis, we constructed the single-nucleotide polymorphism (SNP) coexistence networks and noted the SNP number of the maximal clique as the coinfection index. The coinfection indices of GISAID20May11 and GISAID21Apr1 datasets were 16 and 34, respectively. Simulating the transmission routes and the mutation accumulations, we discovered the linear relationship between the coinfection index and the coinfected variant number. Based on the linear relationship, we deduced that the COVID-19 cases in the GISAID20May11 and GISAID21Apr1 datasets were coinfected with 2.20 and 3.42 SARS-CoV-2 variants on average. Additionally, we performed Nanopore sequencing on 42 COVID-19 patients to explore the virus mutational characteristics. We found the heterozygous SNPs in 41 COVID-19 cases, which support the coinfection of SARS-CoV-2 variants and challenge the accuracy of phylogenetic analysis. In conclusion, our findings reported the coinfection of SARS-CoV-2 variants in COVID-19 patients, demonstrated the increased coinfected variants number in the epidemic, and provided clues for the prolonged viral shedding and severe symptoms in some cases.

A prospective randomized controlled study was conducted in 120 patients with hydronephrosis-free lower calyceal calculi in Jiangshan People′s Hospital from June 2020 to August 2021. Patients were divided into drug group, vibration group and combination group by random number table method with 40 cases in each group. The drug group was treated with potassium hydrogen citrate sodium, the vibration group was treated with external physical vibration lithotripsy (EPVL), and the combination group was treated with EPVL combined with potassium hydrogen citrate sodium. The renal stone discharge rate and complications were observed and compared among three groups 1, 2 and 4 weeks after treatment. The overall discharge rate in 120 patients was 39.2%(47/120). The discharge rate of vibration group and combination group were higher than that of drug group ( P<0.05). There were no significant differences in stone discharge rate at 1 and 2 weeks after treatment between vibration and combination group ( P=0.63 and 0.14), while the calculi discharge rate at 4 weeks after treatment in combination group was higher than that in vibration group ( P=0.03).The overall incidence of complications in 120 patients was 12.5% (15/120), that of drug group, vibration group and combination group was 7.5%(3/40), 15.0%(6/40), and 15.0% (6/40), respectively ( P<0.05). No serious complication occurred, and all patients recovered well after treatment. The results suggest that EPVL can promote the discharge of small calyceal calculi in hydronephrosis-free patients, and potassium hydrogen citrate sodium can further facilities the discharge of renal stone.

Objective To investigate the expression and clinical significance of P-selectin in colorectal cancer cells. Methods The protein expression of P-selectin was detected by immunohistochemistry in colorectal cancer tissues ( n=116) and corresponding non-tumor normal mucosa(n=57). The differences of P-selectin expression in colorectal cancer tissues, corre?sponding non-tumor normal mucosa and the clinical pathological significance were compared and analysed in two groups. Results The expression of P-selectin protein was found not only in vascular endothelial cells but also in colorectal cancer cells. The positive rate of P-selectin expression was apparently higher in colorectal cancer cells (77.6%, 90/116) than that in normal mucosa (17.5%, 10/57,χ2=56.49, P0.05). Conclusion The high expression of P-seletin in colorectal cancer cells is an important characteristic of colorectal cancer.

Objective To study the effects of Xinshuai mixture on mitochondrial function of hypertrophy cardiomyocyte cell in vitro.Methods The primary neonatal rat myocardial cells were cultured to establish hypertrophic myocardial model by AngII.The levels of superoxide dismutase(SOD), methane dicarboxylic aldehyde (MDA), monoamine oxidase(MAO) and cyclooxygenase (COX) were detected by ELISA, as the mitochondrial function and oxidative stress damage.The expression level of caspase-3 mRNA was detected by Real-time PCR.Results Compared with control group, the SOD level in model group reduced markedly, MDA level increased significantly, the activities of MAO increased, the activities of COX decreased, the expression of caspase-3 level increased significantly(P<0.01).Compared with the model group, the SOD level increased and MDA level decreased in Xinshuai mixture group and blank serum group.But the SOD level and activities of MAO, COX in Xinshuai mixture group and blank serum group had significant difference(P<0.05,P<0.01).The expression of caspase-3 mRNA level in Xinshuai mixture was significantly higher than the blank serum group(P<0.01).Conclusion Xinshuai mixture could significantly improve the fuction of mitochondrial in hypertrophy cardiomyocyte cell, reduce the energy metabolism and oxidative stress injury, and protect cardiac myocyte through lower the expression of caspase-3.

This study was aimed to investigate the effects of Shen-Y uan Y i-Qi Huo-Xue (SYYQHX) capsule com-bined with early percutaneous coronary intervention (PCI) on near-term quality of life (QOL) in unstable angina (UA) patients. Seventy-eight patients diagnosed with UA were selected and randomly divided into the treatment group and control group, with 39 patients in each group. Early PCI was undergone after coronary angiography. Before PCI, the control group was given routine western medication. The treatment group was given routine western medication plus SYYQHX capsule, three pills once, three times daily. The treatment lasted for 30 days. The QOL scores were evalu-ated among patients from both groups before and after treatment in order to determine the efficacy on angina, electro-cardiogram (ECG) and traditional Chinese medicine (TCM) main symptom. The results showed that compared to pre-treatment, the scores of physical limitation (PL), angina stability (AS), angina frequency (AF), and treatment satisfac-tion ( TS ) were significantly increased after treatment ( P < 0 . 05 ) . Compared with the control group , after treat-ment with SYYQHX capsule, the AF, duration time and symptoms of palpitation, fatigue and shortness of breath were obviously improved (P< 0.05). The scores of AS, AF and TS in the treatment group were significantly increased (P < 0.05). It was concluded that SYYQHX capsule combined with early PCI can improve the near-term QOL and TCM main symptoms among UA patients.
