RESUMO

OBJECTIVE: This study aimed to investigate the effects of dietary supplementation with Agaricus blazei polysaccharide (ABP) at varying concentrations on the performance, egg quality, blood biochemistry, intestinal morphology, and microflora of quail. METHODS: The study involved a total of 2,700 Korean quails, which were randomly divided into three groups. The measured variables encompassed productive performance, egg parameters, carcass parameters, serum metabolites, immune response parameters, antioxidative properties, and gut microbiome. RESULTS: The addition of ABP did not have a significant effect on average daily feed intake. However, it was found to increase the average daily egg weight and egg production rate, reduce the feed-egg ratio. There were no significant impacts on egg quality measures such as egg shape index, egg yolk index and color, egg yolk and protein content. However, ABP supplementation significantly increased the Hough unit (p<0.01) and decreased the rate of unqualified eggs (p<0.01). Regarding serum parameters, the inclusion led to an increase in total protein concentration (p<0.05) and a reduction in low-density lipoprotein cholesterol (p<0.05). There were no significant effects observed on immune indicators such as immunoglobulin A (IgA) and IgM. ABP supplementation increased the levels of serum antioxidant indicators, including glutathione peroxidase, total superoxide dismutase (p<0.05), and total antioxidant capacity colorimeter (p<0.05). Furthermore, ABP supplementation significantly elevated the intramuscular fatty acid content in quail meat. Additionally, ABP supplementation demonstrated a significant improvement in the diversity of gut microbiota and induced alterations in the composition of the gut microbiota. CONCLUSION: The findings of this study indicate that dietary supplementation of ABP enhanced production performance and antioxidant capacity while increasing the levels of polyunsaturated fatty acids in quail muscle.

RESUMO

BACKGROUND: The widespread use of antibiotics has led to the emergence of many drug-resistant strains; thus, the development of new antibacterial drugs is essential with antimicrobial peptides becoming the focus of research. This study assessed the antibacterial effect of a novel antimicrobial peptide, named LL-1 on Escherichia coli (E.coli) by determining the minimum inhibitory concentration (MIC) and the antibacterial curve. The interaction between LL-1 and E. coli DNA was then detected by nucleic acid gel electrophoresis. The effect of LL-1 on the E. coli cell membrane was assessed by detecting the leakage of ß-galactosidase, nucleic acid and protein. The influence of LL-1 on the intracellular ATP of E. coli was analysed by determining the concentration of intracellular ATP. Finally, the bacteria and colonies of E. coli treated with LL-1 were observed using scanning and transmission electron microscopy. RESULTS: The results suggested that the MIC value was 3.125 µg/ml, and the antibacterial effect was dose-dependent. LL-1 dose-dependently combined with E. coli DNA. LL-1 resulted in the leakage of intracellular ß-galactosidase, nucleic acid and protein, and decreased intracellular ATP concentrations of E. coli. Two MIC of LL-1 caused E. coli to shrink, resulting in a rough surface, plasmolysis, and bacterial adhesion. CONCLUSION: This study indicated that LL-1 had a good bactericidal effect on E. coli by mainly increasing the permeability of the cell membrane, leading to leakage of the intracellular content. This will lay the foundation for an in-depth study on the antibacterial mechanism of LL-1 against E. coli and its clinical application.

RESUMO

Excess application with the poor efficiency of fertilizers is one of the major problems confronting the sustainable agricultural development. Whereas, loss of N and P nutrients from farmlands to surface waters is considered as one of the major sources inducing eutrophication. In order to work out a minimum fertilizer application rate for both keeping conventional yield and reducing nutrient loading of non point source pollution, an experiment of formulated fertilization scheme on rice at different rates and nutrient ratios by incorporating the soil supply potential was conducted in a paddy soil Wunitu in Kunshan City, Suzhou. The soil nutrient can supply a yield at 6.75 t/hm2 at least for one cropping season. Among the fertilization treatments, a maximum yield of 9.79 t/hm2 was obtained at fertilization (N + P) rate of 300 kg/hm2kg N and P at a ratio of N to P being 3:2 or 1:1. However, with much lower fertilization (N + P) rate at 240 kg/hm2 with the same ratio a high yield of 9.69 t/hm2 was reached. The fertilizer efficiency of the later was 3 times as that of the former rate. Thus, a economical and environmentally sound mode of fertilization in such paddy soils as total N and P 240 kg/hm2, with N:P:Kratio being 3:2:0 or 1:1:0 was developed. Further study would be directed to a innovation of combination of reducing fertilization rate with rotated fertilization for alternative crops.
