RESUMO

Partitioning defective protein 3 (Par3) can activate the Tiam1/Rac pathway to inhibit invasion and metastasis in many cancers; however, the role of Par3 in lung adenocarcinoma remains unknown. Here we show that Par3 is downregulated in lung adenocarcinoma tissues and is associated with higher rates of lymph node metastasis and recurrence. Our functional study demonstrated that knock-down of Par3 promoted lung adenocarcinoma cell growth, cell migration, tumor formation, and metastasis, all of which were effectively inhibited when 14-3-3ζ was silenced. We found that Par3 binded with 14-3-3ζ protein and also showed that Par3 abrogated the binding of 14-3-3ζ to Tiam1, which was responsible for Rac1 activation. Knock-down of 14-3-3ζ inhibited Tiam1/Rac-GTP activation and blocked the invasive behavior of cells lacking Par3. These data suggest that loss of Par3 promotes metastatic behavior in lung adenocarcinoma cells through 14-3-3ζ protein.

RESUMO

Objective: To study the changing rules of Glycyrrhizae Radix et Rhizoma Praeparata cum Melle (GRRPM) with the active components alignment (ACA, liquiritin-liquiritigenin-glycyrrhizin acid-isoliquiritinin) of tonifying spleen and replenishing qi. Methods: Taking single herbs, their medicine pairs, or their medicine compounds as test sample, the sample solution was prepared by traditional water decoction and determined by HPLC. Results: The contents of liquiritin-liquiritigenin-glycyrrhizin acid-isoliquiritinin in GRRPM were (12.14±0.31), (3.25±0.19), (38.75±1.77), and (1.40±0.25) mg/mL; The ACA contents in GRRPM-Ginseng Radix et Rhizoma (GRR) were (2.87±0.04), (0.74±0.02), (6.42±0.18), and (0.37±0.03) mg/mL; The ACA contents in GRRPM-Atractylodis Macrocephalae Rhizoma (AMR) were (5.99±0.01), (2.16±0.09), (22.40±1.80), and (0.77±0.02) mg/mL; The ACA contents in Sijunzi Decotion (SD) were (2.99±0.03), (0.81±0.04), (10.05±0.31), and (0.32±0.03) mg/mL. Conclusion: Four main active components in the efficacy of the test components have almost the same change rule: GRRPM≤GRRPM-AMR≤SD≤GRRPM-GRR; The percentage change intervals of the ACA (liquiritin-liquiritigenin-glycyrrhizin acid-isoliquiritinin) are (7.76-9.34):(2.00-2.81):(17.35-31.41):1. The total contents in single herbs are three times as much as in compatibility components, so the Chinese materia medica (CMM) are often used in the type of formula, which indicates that the utilized space of ACA resources is larger, ACA has the obvious effect of correspondence, and the quality standard of CMM should be marked ACA and medicinal components of biological potency as indicators.

RESUMO

OBJECTIVETo obtain sufficient recombinant VP2 protein of human Bocavirus and establish it's seroepidemiology assying metbord. METHORD: Tbe capsid protein VP2 DNA genes of HBoV1 and 2 were optimized in accordance with tbe usage of the favorite codons in K coil so as to enhance its protein expression in prokaryotic expressing system. The protein was purified by Ni-NTA column, and its antigenicity was determined by Western Blot. Then establish ELISA to detect the specific anti-VP2 IgG antibodies against HBoV1 and 2 in healthy children aged 3-6 years in Nanjing, China.

RESULTSThe recombinant protein 6 x His-VP2 was produced in a larger quantity at 25 degrees C induced by IPTG (1 mmol/L) over night and purified by Ni-NTA column. Seropositive rates of HBoV1 and 2 were 62.2% and 55.5% and their mixed seropositivity was 37%.

CONCLUSIONThe optimizing expression of the capsid protein VP2 from human Bocavirus constructed successfully and get a high yield under certain conditions. The established ELISA could be used to further analyze seroepidemiology of HBoV in China.

RESUMO

OBJECTIVETo evaluate plastic surgical procedures for the extensive soft tissue hemangiomas in the oral and maxillofacial regions in children and to explore prevention methods from surgical complications.

METHODS134 patients with hemangiomas were surgically treated. The defects of soft tissue were repaired with pedicle flaps or skin grafts. Of the 134 patients, 16 were newborns, 91 infant, 16 at preschool age and 11 at school age. The location of hemangiomas included parotid gland (39.6%), lip (20.9%), buccal (20.1%), suborbital (9%), orbital (7.5%) and nasal region (3%).

RESULTSFollow-up from 6 months to 5 years showed that the cure rate was 80.6%, apparently effective rate was 15.7%, effective rate was 2.2% and ineffective rate was 1.5%. No major surgical complications were found.

CONCLUSIONExtensive hemangiomas in the oral and maxillofacial regions in children were treated safely using plastic surgical procedures with satisfactory results esthetically and functionally.
