OBJECTIVES: This study describes the outbreaks of H5N8 highly pathogenic avian influenza (HPAI) in Korea during the first wave, from January 16, 2014 through July 25, 2014. Its purpose is to provide a better understanding of the epidemiology of H5N8 HPAI. METHODS: Information on the outbreak farms and HPAI positive wild birds was provided by the Animal and Plant Quarantine Agency. The epidemiological investigation sheets for the outbreak farms were examined. RESULTS: During the 7-month outbreak period (January-July 2014), H5N8 HPAI was confirmed in 212 poultry farms, 38 specimens from wild birds (stools, birds found dead or captured). Ducks were the most frequently infected poultry species (159 outbreak farms, 75.0%), and poultry in 67 (31.6%) outbreak farms was asymptomatic. CONCLUSION: As in the previous four H5N1 epidemics of HPAI that occurred in Korea, this epidemic of H5N8 proved to be associated with migratory birds. Poultry farms in Korea can hardly be free from the risk of HPAI introduced via migratory birds. The best way to overcome this geographical factor is to reinforce biosecurity to prevent exposure of farms, related people, and poultry to the pathogen.

In January 2014, an outbreak of infection with highly pathogenic avian influenza (HPAI) A(H5N8) virus began on a duck farm in South Korea and spread to other poultry farms nearby. During this outbreak, many sick or dead wild birds were found around habitats frequented by migratory birds. To determine the causes of death, we examined 771 wild bird carcasses and identified HPAI A(H5N8) virus in 167. Gross and histologic lesions were observed in pancreas, lung, brain, and kidney of Baikal teals, bean geese, and whooper swans but not mallard ducks. Such lesions are consistent with lethal HPAI A(H5N8) virus infection. However, some HPAI-positive birds had died of gunshot wounds, peritonitis, or agrochemical poisoning rather than virus infection. These findings suggest that susceptibility to HPAI A(H5N8) virus varies among species of migratory birds and that asymptomatic migratory birds could be carriers of this virus.

Highly pathogenic H5N8 avian influenza viruses (HPAIVs) were introduced into South Korea during 2014, thereby caused outbreaks in wild birds and poultry farms. During the 2014 outbreak, H5N8 HPAIVs were isolated from 38 wild birds and 200 poultry farms (up to May 8, 2014). To better understand the introduction of these viruses and their relationships with wild birds and poultry farm, we analyzed the genetic sequences and available epidemiological data related to the viruses. Genetic analysis of 37 viruses isolated from wild birds and poultry farms showed that all of the isolates belonged to clade 2.3.4.6 of the hemagglutinin (HA) gene, but comprised two distinct groups. During the initial stage of the outbreak, identical isolates from each group were found in wild birds and poultry farms near Donglim Reservoir, which is a resting site for migratory birds, thereby indicating that two types of H5N8 HPAIVs were introduced into the lake at the same time. Interestingly, the one group of H5N8 HPAIV predominated around Donglim Reservoir, and the predominant virus was dispersed by wild birds among the migratory bird habitats in the western region of South Korea as time passed, and it was also detected in nearby poultry farms. Furthermore, compared with the results of the annual AIV surveillance of captured wild birds, which has been performed since 2008, more HPAIVs were isolated and H5 sero-prevalence was also detected during the 2014 outbreak. Overall, our results strongly suggest that migratory birds played a key role in the introduction and spread of viruses during the initial stage of the 2014 outbreak.

The immune defense against FMDV has been correlated to the antibody mediated component. However, there are occasions when some animals with high virus neutralising (VN) antibody are not protected following challenge and some with low neutralising antibody which do not succumb to disease. The importance of cell mediated immunity in clinical protection is less clear and so we investigated the source and production of interferon-gamma (IFN-γ) in re-stimulated whole blood of FMDV immunized cattle and its correlation to vaccine induced protection and FMDV persistence. We were able to show a positive correlation between IFN-γ response and vaccine induced protection as well as reduction of long term persistence of FMD virus. When combining this IFN-γ response in re-stimulated blood with virus neutralizing antibody titer in serum on the day of challenge, a better correlation of vaccine-induced protection with IFN-γ and VN antibody was predicted. Our investigations also showed that CD4+ T-cells are the major proliferating phenotype and IFN-γ producing cells.

This retrospective study of the patterns and efforts of the extensive vaccination programs for canine rabies control (EVPCRC) compared the prevalence rate (PR) of rabies under two different vaccination programs for dogs in Korea between 1962 and 2007. A total of 753 cases of rabid dogs were reported between 1962 and 2007, and there were 350 human cases reported in the same period. There were 518 cases (PR, 4.11) of rabid dogs reported from 1962 to 1979, when the low-egg-passage Flury strain vaccine was used, and a total of 342 human cases were observed over the same period. However, there were 235 rabid dogs (PR, 0.83) identified from 1980 to 2007 when the Evelyn-Rokitnicki-Abelseth strain vaccine was used, and only 8 human cases of rabies were observed during the same period. The prevalence of rabies in dogs during the two periods was significantly different (P < 0.01). Moreover, the year with the largest outbreak, 1975, contained 91 cases, and the PR value was 6.3. In 1963, for humans, there were 103 cases, and the PR value was 0.4. However, in recent years, raccoon dogs have increased as a reservoir of rabies in the mountain areas linking North Korea with South Korea near the demilitarized zone, reflecting areas with poor EVPCRC.

Necrotizing sialometaplasia (NS) is a self-limiting, benign, ischemic, inflammatory disease that is most often described in the submandibular glands of dogs, with clinical and histologic features that resemble malignancy. Unilateral swelling of the parotid salivary gland in a 7-year-old Cocker Spaniel dog was diagnosed as NS. The dog also had otitis externa on the same side as the parotid gland lesions. The main histologic features were included lobular necrosis of salivary tissue; fibrinoid necrosis of some arteries; marked squamous metaplasia of duct and/or acinar epithelium, with intercellular bridge formation; preservation of salivary lobular morphology; and variable inflammation and fibrosis. Etiologic factors for NS in both humans and animals remain obscure.

Over a 6-day period beginning on 15 October 2008, 93 dead or sick wild waterfowl, including Mallards, Spotbills, and teal species, were found along the shore of a branch stream of the Hangang River, which flows through Seoul, Korea, and were submitted to the National Veterinary Research and Quarantine Service (NVRQS) for diagnosis. Clinically, the affected birds showed flaccid paralysis of the legs and wings and paralysis of the neck. Grossly, no bird showed any lesions, but all had almost empty stomachs. Histopathologic findings included mild lymphocytic hepatitis and mild lymphocytic interstitial nephritis. Clostridium botulinum type C toxin was identified in sera collected from the birds using a mouse bioassay for botulinum toxins; however, no bacteria were isolated from any of the affected birds. In addition, a low-pathogenic avian influenza virus was isolated from two Spotbills, and pesticides such as diazinon and phorate, were detected in seven Mallards. The cause of this outbreak is not clear, but an increase in organic materials from sewage due to drought, increased temperatures, and an increased number of aquatic carcasses resulting from pesticide contamination may have increased the replication of C. botulinum, contributing to the release of botulinum toxins into the waterfowl food chain.

A total of 628 Escherichia coli isolates recovered from 877 intestinal samples of stray pet dogs (n = 565) and hospitalized pet dogs (n = 312) in Korea were analyzed for resistance to 15 antimicrobial agents. Most common resistance observed in E. coli isolated from both groups of dogs was to tetracycline (52.4-53.6%), streptomycin (35.8-41.7%), ampicillin (32.9-47.1%), nalidixic acid (21.6-37.4%), and trimethoprim/sulfamethoxazole (19.7-36.4%). Resistance to chloramphenicol, gentamicin, and ciprofloxacin was observed in 19.4% (17.1-24.3%), 18% (16.1-21.8%), and 16.1% (13.5-21.4%) of the isolates, respectively. No E. coli isolated from hospitalized dogs showed resistance to imipenem and cefepime, whereas three (0.7%) isolates from stray dogs were resistant to cefepime. Some of the isolates from both groups showed resistance to cefotaxime (2.4-3.9%) and amikacin (0.5-1.5%). In general, the frequency of resistance tended to be higher in isolates from hospitalized dogs than isolates from stray dogs against most antimicrobials tested. Around 39% (162/422) and 27% (55/206) of E. coli isolates from stray dogs and hospitalized dogs were susceptible to all antimicrobials tested, respectively. Multiresistance (> or = 3 subclasses of antimicrobials) was observed in 32% and 48% of E. coli isolates from stray dogs and hospitalized dogs, respectively. More attention should be paid to the use of antimicrobials and the occurrence of antimicrobial resistance in companion animals.

Clove (Eugenia caryophyllata) is a well-known medicinal plant used for diarrhea, digestive disorders, or in antiseptics in Korea. Eugenol is the main active ingredient of clove and has been chosen as a marker compound for the chemical evaluation or QC of clove. This paper reports the development and validation of an HPLC-diode array detection (DAD) method for the determination of eugenol in clove. HPLC separation was accomplished on an XTerra RP18 column (250 x 4.6 mm id, 5 microm) with an isocratic mobile phase of 60% methanol and DAD at 280 nm. Calibration graphs were linear with very good correlation coefficients (r2 > 0.9999) from 12.5 to 1000 ng/mL. The LOD was 0.81 and the LOQ was 2.47 ng/mL. The method showed good intraday precision (%RSD 0.08-0.27%) and interday precision (%RSD 0.32-1.19%). The method was applied to the analysis of eugenol from clove cultivated in various countries (Indonesia, Singapore, and China). Quantitative analysis of the 15 clove samples showed that the content of eugenol varied significantly, ranging from 163 to 1049 ppb. The method of determination of eugenol by HPLC is accurate to evaluate the quality and safety assurance of clove, based on the results of this study.

In spite of intensive surveillance programs for the control of HPAI, an outbreak of highly pathogenic avian influenza (HPAI) H5N1 in Korea in April 2008 caused serious damage to poultry farms, as did previous outbreaks in 2003/2004 and 2006/2007. Six viruses were selected from the Korean 2008 isolates for genetic analysis, and all eight gene segments from each of the influenza viruses were sequenced. A phylogenetic analysis showed that all of the viruses were of the same virus type and that the hemagglutinin (HA) gene was clustered with that of clade 2.3.2 viruses. However, the internal and neuraminidase (NA) genes were closely related to those of the clade 2.3.4 viruses (recent human and bird isolates from Southeast Asia).

Gram-negative bacteria (GNB) with multidrug resistance pose a serious threat to public health. They are environmental pathogens frequently isolated from raw milk and mastitis in dairy cattle. This study was to examine the in vitro antimicrobial activities against 225 isolates belonging to six important species of GNB from mastitic raw milk samples of dairy herds in the Republic of Korea: Acinetobacter baumannii (n = 17), Citrobacter freundii (n = 19), Enterobacter cloacae (n = 54), Klebsiella pneumoniae (n = 55), Pseudomonas aeruginosa (n = 45), and Serratia marcescens (n = 35). In general, amikacin, gentamicin, and piperacillin exhibited strong antimicrobial activities against all bacterial species tested, whereas rifampin, cephalothin, cefazolin, and ampicillin were ineffective against most of the bacterial species tested. Wide differences were observed in the patterns of resistance among the bacterial species; in particular, resistance to kanamycin, streptomycin, tetracycline, and chloramphenicol was highly variable among the strains belonging to different bacterial species. Almost half of the GNB isolates (45.3%, 102/225) were resistant to 5 or more of 12 antimicrobial agents tested: P. aeruginosa (86.6%, 39/45) showed the highest resistance rate, followed by S. marcescens (65.7%, 23/35). This study indicates that multiple antimicrobial resistances are prevalent among GNB isolates from mastitic milk samples of dairy cattle in the Republic of Korea.

Nine isolates of rabbit hemorrhagic disease virus (RHDV) were used for the genetic characterization of RHDV strains collected from rabbits in Korea between 2006 and 2008. A phylogenetic analysis of the complete VP60 region was performed and the sequences were divided mainly into two groups. The one group consisted of original RHDV and the other contained antigenic variant strain known as RHDVa strains. Most of the Korean isolates clustered with Chinese RHDV strains and belonged to the RHDVa subtype. A comparison of the amino acid sequences among RHDVa strains and original RHDV strains revealed significant substitutions of two amino acids in the A region, two in the B region, two in the F region, and nine amino acids in the E region. Taken together, the recent RHDVa strains have gradually replaced the original RHDV and are the predominant strains in Korea.

A simple lateral-flow assay (LFA) based on a monoclonal antibody (MAb 70-17) was developed for the detection of foot-and-mouth disease virus (FMDV) under nonlaboratory conditions. The LFA was evaluated with epithelial suspensions (n = 704) prepared from current and historical field samples which had been submitted to the Pirbright Laboratory (United Kingdom) and from negative samples (n = 100) collected from naïve animals in Korea. Four FMDV serotypes (type O, A, Asia 1, and C) were detected in the LFA, but not the remaining three FMDV serotypes (SAT 1, SAT 2, and SAT 3). The diagnostic sensitivity of the LFA for FMDV types O, A, C, and Asia 1 was similar, at approximately 87.3%, to that of 87.7% obtained with antigen enzyme-linked immunosorbent assay (Ag-ELISA). The diagnostic specificity of the LFA was 98.8%, compared to 100% for the Ag-ELISA. These results demonstrate that the LFA using the FMDV MAb 70-17 to detect FMDV is a supportive method for taking rapid measurements at the site of a suspected foot-and-mouth disease outbreak in Asia before diagnosing the disease in the laboratory, thereby offering the possibility of implementing control procedures more rapidly.

The aim of this study was to determine the antimicrobial resistance of streptococci isolated from mastitic bovine milk samples. A total of 178 isolates belonging to 6 different Streptococcus species were examined: S. uberis (n = 99), S. bovis (n = 30), S. oralis (n = 24), S. salivarius (n = 13), S. intermedius (n = 7), and S. agalactiae (n = 5). Only 8.9% (16/178) of the isolates were susceptible to all antimicrobial agents tested in this study, and S. agalactiae and S. intermedius isolates were all resistant to at least 1 antimicrobial agent tested. Overall, the most frequently observed resistance was to tetracycline (61.2%), followed by lincomycin (43.2%), gentamycin (35.3%), oxacillin (34.3%), and erythromycin (28.6%). Cephalothin and penicillin were the only antimicrobial agents to which most of the streptococci (>or=92%) were susceptible. Wide differences in the prevalence of resistance are apparent among the individual species: S. salivarius displayed exceptionally high resistance to cephalothin (23.0%) and oxacillin (76.9%) and S. agalactiae (20%) and S. intermedius (14.2%) to penicillin. Streptococcus salivarius and S. agalactiae were all susceptible to erythromycin, but others showed various rates of resistance ranging from 12.5% to 42.8%. Resistance to 3 or more of 7 antimicrobial agents was observed in all species (37.6%, 67/178).

An outbreak of fatal hemorrhagic pneumonia with 70-90% morbidity and 50% mortality occurred in an animal shelter in Yangju, Gyeonggi Province, Korea. Clinically, the affected dogs showed severe respiratory distress within 48 h after arriving in the shelter. The dead were found mainly with nasal bleeding and hematemesis. At necropsy, hemothorax and hemorrhagic pneumonia along with severe pulmonary consolidation was observed, though histopathological analysis showed mainly hemorrhagic bronchopneumonia. Lymphoid depletion was inconsistently seen in the spleen, tonsil and bronchial lymph node. Gram-positive colonies were shown in blood vessels or parenchyma of cerebrum, lung, liver, spleen, and kidney. Also, Streptococcus (S.) equi subsp. zooepidemicus was isolated from the various organs in which the bacterium was microscopically and histologically detected. In addition, approximately 0.9 Kb specific amplicon, antiphagocytic factor H binding protein, was amplified in the bacterial isolates. In this study, we reported an outbreak of canine hemorrhagic bronchopneumonia caused by S. equi subsp. zooepidemicus in an animal shelter in Yangju, Korea.

Thirty-six bovine viral disease viruses (BVDVs) were identified in bovine feces (n=16), brains (n=2), and aborted fetuses (n=18) in Korea. To reveal the genetic diversity and characteristics of these Korean strains, the sequences of their 5'-untranslated regions (5'-UTRs) were determined and then compared with published reference sequences. Neighbor-joining phylogenetic analysis revealed that most of the Korean viruses were of the BVDV subtypes 1a (n=17) or 2a (n=17). The remaining strains were of subtypes 1b (n=1) and 1n (n=1). This analysis indicates that the 1a and 2a BVDV subtypes are predominant and widespread in Korea. In addition, the prevalence of BVDV-2 was markedly higher in aborted fetuses than in other samples and was more often associated with reproductive problems and significant mortality in cattle.

Ectopic expression of the structural protein Pr55(Gag) of HIV-1 has been limited by the presence of inhibitory sequences in the gag coding region that must normally be counteracted by HIV-1 Rev and RRE. Here, we describe a cytoplasmic RNA replicon based on the RNA genome of Japanese encephalitis virus (JEV) that is capable of expressing HIV-1 gag without requiring Rev/RRE. This replicon system was constructed by deleting all three JEV structural protein-coding regions (C, prM, and E) from the 5'-proximal region of the genome and simultaneously inserting an HIV-1 gag expression cassette driven by the internal ribosome entry site of encephalomyocarditis virus into the 3'-proximal noncoding region of the genome. Transfection of this JEV replicon RNA led to expression of Pr55(Gag) in the absence of Rev/RRE in the cytoplasm of hamster BHK-21, human HeLa, and mouse NIH/3T3 cells. Production of the Pr55(Gag) derived from this JEV replicon RNA appeared to be increased by approximately 3-fold when compared to that based on an alphavirus replicon RNA. Biochemical and morphological analyses demonstrated that the Pr55(Gag) proteins were released into the culture medium in the form of virus-like particles. We also observed that the JEV replicon RNAs expressing the Pr55(Gag) could be encapsidated into single-round infectious JEV replicon particles when transfected into a stable packaging cell line that provided the three JEV structural proteins in trans. This ectopic expression of the HIV-1 Pr55(Gag) by JEV-based replicon RNAs/particles in diverse cell types may represent a useful molecular platform for various biological applications in medicine and industry.

Porcine reproductive and respiratory syndrome virus (PRRSV) isolates belonging to the European genotype 1 have recently emerged in South Korea, suggesting potential problems for disease control. In the present study, we attempted to determine the complete nucleotide sequence of the first Korean type 1 PRRSV isolate, designated KNU-07. The full-length genome of KNU-07 was found to be 15,038 nucleotides in length, which was 60 nucleotides shorter than the type 1 prototype strain Lelystad due to a notable 60-bp deletion within the nonstructural protein 2 (NSP2). The KNU-07 genome was shown to consist of a 221-nucleotide (nt) 5' untranslated region (UTR), a 14,703-nt protein-coding region, and a 114-nt 3' UTR, followed by a 42-73-bp poly(A) tail. A nucleotide sequence comparison of the KNU-07 genome with 20 complete PRRSV genomes revealed a 10.5-13.3% and 39.5-40.3% divergence from type 1 and type 2 strains, respectively, at the genome level, indicating a high similarity to the virus strains commonly identified as the European genotype. In order to investigate genetic variation and to understand the molecular evolution of the type 1 isolate in Korea, extensive phylogenetic analyses were performed using the ORF5 and ORF7 nucleotide sequences of published type 1 PRRSV isolates. The data further indicated that the newly emerging type 1 isolate KNU-07 belongs to the recently proposed pan-European subtype 1. Taken together, the results of this study describe the genomic characterization of the type 1 PRRSV isolated in South Korea, suggesting a recent introduction of the virus typical for this genotype that has commonly appeared worldwide.
