RESUMO

The CRISPR-Cas system stands out as a promising genome editing tool due to its cost-effectiveness and time efficiency compared to other methods. This system has tremendous potential for treating various diseases, including genetic disorders and cancer, and promotes therapeutic research for a wide range of genetic diseases. Additionally, the CRISPR-Cas system simplifies the generation of animal models, offering a more accessible alternative to traditional methods. The CRISPR-Cas9 system can be used to cleave target DNA strands that need to be corrected, causing double-strand breaks (DSBs). DNA with DSBs can then be recovered by the DNA repair pathway that the CRISPR-Cas9 system uses to edit target gene sequences. High cleavage efficiency of the CRISPR-Cas9 system is thus imperative for effective gene editing. Herein, we explore several factors affecting the cleavage efficiency of the CRISPR-Cas9 system. These factors include the GC content of the protospacer-adjacent motif (PAM) proximal and distal regions, single-guide RNA (sgRNA) properties, and chromatin state. These considerations contribute to the efficiency of genome editing.

Two-dimensional transition-metal dichalcogenide (TMD) materials have attracted increasing attention in efforts to overcome fundamental issues faced by the complementary metal-oxide-semiconductor industry. Multilayer TMD materials such as MoS2 can be used for high-performance transistor-based applications; the drive currents are high and the materials handle low-frequency (LF) noise well. We fabricated double-gated multilayer MoS2 transistors using the h-BN dielectric for the top gate and silicon dioxide for the bottom gate. We systemically investigated the bottom gate voltage (Vb)-controlled electrical characteristics and the top/bottom interface-coupling effects. The effective thickness of the MoS2 channel (tMoS2_eff) was well modulated by Vb, and tMoS2_eff reduction by negative Vb dramatically improved the Ion/Ioff ratio. Numerical simulation and analytical modeling with a variation of the depletion depth under different bias conditions verified the experimental results. We were also the first to observe Vb-tuned LF noise characteristics. Here, we discuss the Vb-affected series resistance and carrier mobility in detail. Our findings greatly enhance the understanding of how double-gated multilayer MoS2 transistors operate and will facilitate performance optimization in the real world.

Salmonellosis is a disease of critical concern for public health, and the use of bacteriophages is among the most promising approaches to combating Salmonella. As Salmonella has various serotypes and strains, and bacteriophages are virulent to specific hosts, it is important to isolate phages and evaluate interactions with their hosts. In the present study, a novel Salmonella-infecting bacteriophage, pSal-SNUABM-01, was isolated and characterized. Transmission electron microscopy revealed that the bacteriophage is a member of the family Podoviridae and possesses an elongated head and a short tail. The phage genome is circular and 89,500 bp in size. A total of 162 open reading frames were predicted, eight of which were tRNAs. Morphological and genomic analysis revealed that pSal-SNUABM-01 is closely related to phage 7-11. In phylogenetic analysis, pSal-SNUABM-01 and 7-11 did not cluster together with the members of any established genus, suggesting that these two phages comprise a novel genus. The results of this study enhance our understanding of the phylogeny of the family Podoviridae and might be applicable to the development of bacteriophage treatments against Salmonella infections.

Generally, a high-temperature postannealing process is required to enhance the photoelectrochemical (PEC) performance of hematite nanorod (NR) photoanodes. However, the thermal annealing time is limited to a short duration as thermal annealing at high temperatures can result in some critical problems, such as conductivity degradation of the fluorine-doped tin oxide film and deformation of the glass substrate. In this study, selective laser processing is introduced for hematite-based PEC cells as an alternative annealing process. The developed laser-induced phase transformation (LIPT) process yields hematite NRs with enhanced optical, chemical, and electrical properties for application in hematite NR-based PEC cells. Owing to its improved properties, the LIPT-processed hematite NR PEC cell exhibits an enhanced water oxidation performance compared to that processed by the conventional annealing process. As the LIPT process is conducted under ambient conditions, it would be an excellent alternative annealing technique for heat-sensitive flexible substrates in the future.

RESUMO

Intravenous fat emulsion (IFE) therapy is an adjunct therapy administered to hemodynamically compromised patients with glyphosate-surfactant intoxication when they respond poorly to conventional therapies such as fluid resuscitation or vasopressors [1,2]. However, the use of IFE as an adjunct therapy in collapsed patients with glyphosate intoxication has not been reported previously. Here, we describe the case of a patient with glyphosate-surfactant­induced cardiovascular collapse who responded to IFE.

OBJECTIVES: We investigated whether the superimposed-thumb technique could reduce the chest compression area in infant cardiopulmonary resuscitation (CPR). METHODS: Charts and multidirectional computed tomography images of infants presented to four hospitals from January 2007 to September 2010 were reviewed retrospectively. We measured at the point of maximal anterior-posterior heart diameter the width of the sternum meter (S(ap)), vertical heart length from S(ap), length and width of the superimposed-thumb technique and length and width of the alongside-thumb technique. We studied the structures located underneath thumbs superimposed and thumbs alongside at S(ap) and S(nipple) (the sternum of the inter-nipple line). RESULTS: In the 84 infants enrolled, the width of the sternum at S(ap), and the vertical heart length from S(ap) were 0.85 ± 0.31 and 1.71 ± 0.47 cm, respectively. The length and width of the superimposed-thumb technique were 1.65 ± 0.13 and 2.73 ± 0.22 cm, respectively. The length and width of the alongside-thumb technique were 3.00 ± 0.48 and 3.77 ± 0.24 cm, respectively. The liver was situated underneath thumbs superimposed at S(ap) in 59.5% infants. The livers and lungs of 73.8% and 64.3% infants, respectively, were underneath thumbs alongside at S(nipple). CONCLUSION: In this study, we confirmed that the superimposed-thumb technique may reduce chest compression area in infant CPR. The lungs or livers were located more often underneath thumbs alongside at S(nipple) than underneath thumbs superimposed at S(ap). However, further studies are needed to validate the efficiency and safety of this technique.

The purpose of this study was to find out morbidity rate and utilization pattern of medical services of sailors on a voyage. The subjects of this study were 470 sailors who are sailors' trainee and safety-trainee for embarkation in a ship. Data were collected by means of personal survey conducted from September 5 to October 30, 1988. The subjects were sailed 12.6 months on the average. During the voyage, 49.4% of the sailors had sickness more than once, and 24.7%' was unable to work more than a day because of illness. The average monthly morbidity rate was 85.7 per 1,000 and the monthly prevalence rate of disease that was unable to work more than a day was 21.2 per 1,000. The sailors of 40-49 years old had the highest morbidity rate of 103.6 per 1,000/month. The number of days activity restricted due to sickness was 93.4 days per 1,000 persons, and the average duration of illness was 11.2 days per sickness. The proportion of treatment days to sickness days was 48.8%. Considering morbidity unable to work more than a day, the number of those who had traumatic injury were 17.2%, which revealed the highest rate, influenza or cold were 15.5%, and the digestive disease 11.2%. Less than half(42.1%) of patients were treated with medicine kept in the cabin. However, 2.1% was transferred by helicopter and 29L% was treated at hospitals or clinics in nearby port. In the cabin, 67.4% of patients was cured completely.
