RESUMO

The role of steric hindrance in controlling the binding mode of propylene oxide to a novel vanadyl salen-type complex N,N'-bis(5-tert-butylsalicylidene)-1,2-cyclohexanediamino-vanadium(IV) oxide, [VO(3)], has been investigated using CW/pulse EPR, ENDOR and HYSCORE spectroscopy and compared to that of the parent complex N,N'-bis(3,5-di-tert-butylsalicylidene)-1,2-cyclohexanediamino-vanadium(IV) oxide, [VO(1)]. The single-crystal X-ray structure of [VO(3)]·HCCl(3) has been determined by X-ray analysis and is complemented by DFT calculations and circular dichroism measurements. The structure of the complex in frozen solution, as revealed by the EPR methods, is in good agreement with the X-ray and DFT analyses. Removal of the 'inner'tert-butyl groups from the salicylidene rings reduces the steric hindrance between the ligand and epoxide substrate. As a result the selectivity for binding single enantiomers of propylene oxide in these complexes is reversed in [VO(3)] relative to [VO(1)].

BACKGROUND: Several epidemiologic studies in the United States and Europe have linked Alternaria sensitivity to both persistence and severity of asthma. In this study, we examined T cell responses and HLA class II alleles in children with moderate-severe asthma. METHODS: Ninety-six children with moderate-severe asthma were compared to 90 children with mild asthma. HLA class II genotyping was performed to determine HLA allelic frequencies. Th1/Th2 Alternaria-specific T cell cytokine responses were determined by the use of Alternaria-stimulated cultures. HLA class II restriction was examined by inhibition of Alternaria-stimulated lymphoproliferative responses with blocking anti-HLA class II monoclonal antibodies. RESULTS: Children with moderate-severe asthma had significantly increased sensitivities to Aspergillus fumigatus; sensitivities to Alternaria were similar in both moderate-severe and mild asthmatics. The frequency of HLA-DRB1*13 alleles were increased in mold-sensitive moderate-severe asthmatic children. HLA-DRB1*03 tended to be increased in mold-sensitive moderate-severe asthmatics. The frequency of HLA-DQB1*03 alleles was significantly decreased in mold and Alternaria-sensitive moderate-severe asthma. HLA class II blocking monoclonal antibodies demonstrated HLA-DR restriction. Alternaria-stimulated IL-5 and IL-13 synthesis was significantly increased in moderate-severe asthmatics. IL-5 and IL-13 synthesis was significantly increased in Alternaria-stimulated lymphocyte cultures of HLA-DQB1*03- asthmatics compared to HLA-DQB1*03+ asthmatics. CONCLUSIONS: In children with Alternaria-sensitive moderate-severe asthma, there was increased Th2 sensitivity to Alternaria stimulation. This was associated with HLA-DR restriction and with increased frequency of HLA-DRB1*13 and HLA-DRB1*03. There was decreased frequency of HLA-DQB1*03 in Alternaria-sensitive moderate-severe asthma, suggesting HLA-DQB1*03 may be protective of the development of Alternaria-sensitive severe asthma.

New tris(heterocyclic beta-diketonato)europium(III) complexes of the general formula Eu(PBI)3.L [where HPBI = 3-phenyl-4-benzoyl-5-isoxazolone and L = H2O, 2,2'-bipyridine (bpy), 4,4'-dimethoxy-2,2'-bipyridine (dmbpy), 1,10-phenanthroline (phen), or 4,7-diphenyl-1,10-phenanthroline (bath)] were synthesized and characterized by elemental analysis, Fourier transform infrared spectroscopy (FT-IR), 1H NMR, high-resolution mass spectrometry, thermogravimetric analysis, and photoluminescence (PL) spectroscopy. Single-crystal X-ray structures have been determined for the complexes Eu(PBI)3.H2O.EtOH and Eu(PBI)3.phen. The complex Eu(PBI)3.H2O.EtOH is mononuclear, and the central Eu3+ ion is coordinated by eight oxygen atoms to form a bicapped trigonal prism coordination polyhedron. Six oxygens are from the three bidentate HPBI ligands, one is from a water molecule, and another is from an ethanol molecule. On the other hand, the crystal structure of Eu(PBI)3.phen reveals a distorted square antiprismatic geometry around the europium atom. The room-temperature PL spectra of the europium(III) complexes are composed of the typical Eu3+ red emission, assigned to transitions between the first excited state (5D0) and the multiplet (7F0-4). The results demonstrate that the substitution of solvent molecules by bidentate nitrogen ligands in Eu(PBI)3.H2O.EtOH richly enhances the quantum yield and lifetime values. To elucidate the energy transfer process of the europium complexes, the energy levels of the relevant electronic states have been estimated. Judd-Ofelt intensity parameters (Omega2 and Omega4) were determined from the emission spectra for Eu3+ ion based on the 5D0 --> 7F2 and 5D0 --> 7F4 electronic transitions, respectively, and the 5D0 --> 7F1 magnetic dipole allowed transition was taken as the reference. The high values obtained for the 4f-4f intensity parameter Omega2 for europium complexes suggest that the dynamic coupling mechanism is quite operative in these compounds.

Zn(O3PCH2OH) (1) has been formed by reaction of zinc acetate with diethyl hydroxymethylphosphonate. The acidity of the zinc solution effects hydrolysis of the phosphonate to produce phosphonic acid in situ. 1 crystallizes in the trigonal spacegroup R3, with a = 15.9701(2) A, c = 7.783(2) A, and Z = 18. The compound has channels in the [001] direction, formed by phosphonate groups bridging the octahedral coordinated zinc atoms. The zinc atoms are coordinated by the three oxygens of the phosphonate group and the oxygen of the hydroxy group.

Experiments are described to show some of the potential of the synchrotron radiation Laue method for the study of structural change within single crystals. In the metastable tetragonal crystals of P(4)N(4)Cl(8) the eight-membered P(4)N(4) ring is in a boat conformation, with symmetry {\bar 4}. On heating to ca 340 K the crystals transform, slowly to a second tetragonal form in which the ring conformation is a chair, its symmetry {\bar 1}. Both structures are known [Hazekamp, Migchelsen & Vos (1962). Acta Cryst. 15, 539-543; Wagner & Vos (1968). Acta Cryst. B24, 707-713]. In the transformation the molecular packing, unit-cell dimensions and crystal quality remain almost unchanged. To study this transformation, series of Laue diffraction patterns were recorded at 2-3 min intervals over a period of 30-40 min, while the temperature was raised to 373 K. For two series, reflection intensities were measured and they allowed determination and refinement of the fraction of boat and chair molecules present in a mixed boat/chair model of the structure. No significant change in the crystal occurs below ca 340 K; at or above 340 K, 40-50% of the molecules are converted from boat to chair conformations within 5 min, but the remainder of the conversion is much slower, even when the temperature is raised towards 370 K.
