RESUMO

Cancer remains one of the major causes of morbidity and mortality worldwide. Scientists from different fields are working to devise an efficient treatment strategy in order to reduce the global burden of cancer. Commonly used treatment approaches for cancer treatment include chemotherapy, immunotherapy, photodynamic therapy, radiation, surgery, etc. These treatment procedures have several pitfalls, such as toxicity, limited bioavailability, rapid elimination, poor specificity, and high cost. On the other side, plant-derived anticancer compounds exhibit several advantages and can overcome these shortcomings. Plant-based anticancer compounds are safer, potent, easily available, and comparatively cost-effective. The current review discusses pure plant- based compounds that are used as a therapeutic remedy for anticancer application. The proposed mechanisms of action, through which these compounds inhibit cancer cell growth, tumor growth, angiogenesis, instigate apoptosis, cytotoxicity, mitochondrial membrane degradation, and reduce cell viability as well as cell cycle progression, are also reviewed. These naturally occurring compounds exhibit great therapeutic potential and could be used as candidate drugs in clinical applications.

RESUMO

The genus Berberis includes about 500 different species and commonly grown in Europe, the United States, South Asia, and some northern areas of Iran and Pakistan. Leaves and fruits can be prepared as food flavorings, juices, and teas. Phytochemical analysis of these species has reported alkaloids, tannins, phenolic compounds and oleanolic acid, among others. Moreover, p-cymene, limonene and ocimene as major compounds in essential oils were found by gas chromatography. Berberis is an important group of the plants having enormous potential in the food and pharmaceutical industry, since they possess several properties, including antioxidant, antimicrobial, anticancer activities. Here we would like to review the biological properties of the phytoconstituents of this genus. We emphasize the cultivation control in order to obtain the main bioactive compounds, the antioxidant and antimicrobial properties in order to apply them for food preservation and for treating several diseases, such as cancer, diabetes or Alzheimer. However, further study is needed to confirm the biological efficacy as well as, the toxicity.

ETHNO-PHARMACOLOGICAL RELEVANCE: To best of our knowledge it is first quantitative ethno-botanical study from Alpine and Sub-alpine, Western Himalaya of Pakistan. The study aims to report, compare the uses and highlight the ethno-botanical significance of medicinal plants for treatment of various diseases. METHODS: A total of 290 (278 males and 12 females) informants including 14 Local Traditional Healers (LTHs) were interviewed. Information was collected using semi-structured interviews, analyzed and compared by quantitative ethno-botanical indices such as Informant Consensus Factor (ICF), Relative frequency of citation (RFC), use value (UV), Fidelity Level (FL) and Jaccard index (JI). RESULTS: A total of 125 plant species (Gymnosperms 7 species, Monocotyledons 2 and 116 Di-cotyledons) belonging to 41 families are collected, identified and ethno-botanically assessed. The most dominant family is Ranunculaceae (20 species) followed by Rosaceae (14 species). In diseases treated, gastrointestinal tract (GIT) diseases have highest proportion (27.5%) followed by respiratory diseases (20%) in the mountain communities. The most dominant life form of plants used is herbs (78%) followed by shrubs (19%) while the most commonly used plant parts are leaves (44 reports) followed by underground part, the roots (37 reports). The highest ICF (0.68) is found for ear, nose and eye disease category followed by respiratory disorders (0.46). There are 15 medicinal plants having 100% FL. Use value (UV) and Relative frequency of citation (RFC) range from 0.03 to 0.53 and 0.04 to 0.23 respectively. In comparison, maximum similarity index is found in the studies with JI 19.52 followed by 17.39. Similarity percentage of plant uses range from 1.69% to 19.52% while dissimilarity percentage varies from 0% to 20%. CONCLUSIONS: The Alpine and Sub-alpine regions of Pakistan are rich in medicinal plants and still need more research exploration. On the other hand, ethno-botanical knowledge in study areas is decreasing day by day due to high emigration rates resulting from after effects of wars during the last few decades. Historically developed ethno-botanical heritage should be preserved and promoted on global level for analyzing phytochemical, pharmaceutical and other biological activities for future drug discovery.

ETHNOPHARMACOLOGICAL RELEVANCE: Bergenia himalaica Boriss is mainly distributed in the temperate Himalayas between altitudes of 900 and 3000m ranging from the southeastern regions in central Asia and northern regions in South Asia. The plant has a long history of its use in traditional medicine for the treatment of various diseases such as diabetes, urinary complaints, kidney stones, hemorrhagic diseases and epilepsy. The aim of this study is to isolate pure compounds from Bergenia himalaica Boriss, elucidate their structures and determine their blood glucose lowering activity to obtain additional scientific evidence for its usage in traditional medicine for the management of diabetes. MATERIALS AND METHODS: The crude methanolic extract from the aerial parts of Bergenia himalaica Boriss was separated into EtOAc and water sub-extracts and the EtOAc sub-extract was further divided into petroleum ether soluble and insoluble fractions. The pet-ether insoluble fraction was subjected to fractionation through column chromatography followed by prep. TLC. The blood glucose lowering activity of the 2 new compounds was evaluated in streptozotocin-nicotinamide induced diabetic rats. Additionally, glucose-stimulated insulin secretion was measured on isolated mice islets. RESULTS: Two new compounds bergenicin and bergelin were isolated and their structures determined on the basis of spectral analysis. Significant decrease of blood glucose was observed at 1-h (1.0mg/kg) and 2-h (0.5mg/kg), after bergenicin administration to the diabetic rats and at 2-h (1.0mg/kg) and 3-h (0.5mg/kg), after bergelin administration. Bergenicin, but not bergelin, enhanced glucose-stimulated insulin secretion in isolated pancreatic islets. CONCLUSIONS: In the present studies two new compounds, bergenicin and bergelin were isolated from Bergenia himalaica Boriss and their structures were elucidated. Both the compounds showed anti-hyperglycemic effects in streptozotocin-nicotinamide induced diabetic rats. Bergenicin showed insulinotropic effect; suggesting that the anti-hyperglycemic effect is mostly due to enhancement of insulin secretion from pancreatic ß-cells.
