RESUMO

Adapted immune cells are known to develop memory functions that increase resistance to subsequent infections after initial pathogen exposure, however, it is unclear whether non-immune cells, like tissue-resident stem cells, have similar memory functions. Here, it is found that tissue-resident stem cells crucial for tissue regeneration show diminished adverse effects on diverse stem cell functions against successive exposure to foreign antigen (ß-glucan) to maintain tissue homeostasis and stability both in vitro and in vivo. These data suggest that endometrial stem cells may possess a robust memory function, in contrast, fully differentiated cells like fibroblasts and vesicular cells do not show these memory mechanisms upon consecutive antigen exposure. Moreover, the pivotal role of Angiopoietin-like 4 (ANGPTL4) in regulating the memory functions of endometrial stem cells is identified through specific shRNA knockdown in vitro and knockout mice in vivo experiments. ANGPTL4 is associated with the alteration of diverse stem cell functions and epigenetic modifications, notably through histone H3 methylation changes and two pathways (i.e., PI3K/Akt and FAK/ERK1/2 signaling) upon consecutive antigen exposure. These findings imply the existence of inherent self-defense mechanisms through which local stem cells can adapt and protect themselves from recurrent antigenic challenges, ultimately mitigating adverse consequences.

CONTEXT: In applying good energy conservation strategies to relieve cancer-related fatigue, it is critical to first identify cancer patients who are at a high risk for poor energy conservation. However, instruments have not been developed to evaluate energy conservation strategies in an oncology setting. OBJECTIVES: The aim of this study was to validate an instrument that cancer patients may use to evaluate energy conservation strategies to overcome cancer-related fatigue. METHODS: The questionnaire development followed a four-phase process: 1) item generation and reduction, 2) construction, 3) pilot testing, and 4) field testing. Using relevant and priority criteria, as well as pilot testing, we developed a 25-item questionnaire. After field testing, five items were discarded. Finally, 20 items were included in the Energy Conservation Strategies Inventory (ECSI). Factor analysis, multitrait scaling analysis, and Cronbach's α were used to determine the construct validity and reliability. RESULTS: Factor analyses of data from 140 cancer patients resulted in the ECSI, which covers activities related to planning, overcoming distractions, labor saving, burden reducing, and comfort. All subscales (Cronbach's α range, 0.69-0.78) and total scores (Cronbach's α=0.87) were found to possess acceptable internal consistency. CONCLUSIONS: The good psychometric properties of the ECSI instrument show that it may be useful for measuring the frequency of energy conservation strategies used by cancer patients.

This study was performed to investigate the effect of ethanol extract from Saururus chinensis (Lour.) Baill on liver function, plasma lipid composition and antioxidant system with high-fat diet for 4 weeks. Rats were divided into the following five groups; untreated control group (normal), treated with 0.5% SE (normal + 0.5% SE), high-fat group (high-fat), high-fat group treated with 0.1% SE (high-fat + 0.1% SE), or 0.5% SE (high-fat + 0.5% SE). Weight gains showed a tendency to decrease in rat with high-fat + SE. Plasma total cholesterol showed a tendency to decrease with ethanol extract from S. chinensis (Lour.) Baill. LDL-cholesterol contents were lower in ethanol extract group than that of control group. Aspartate amino transferase and alanine amino transferase activities were increased by high-fat diet, and were decreased by 0.5% SE. Lipid peroxide level showed a tendency to increase in high-fat diet group than that of normal group. In ethanol extract from S. chinensis (Lour.) Baill groups, lipid peroxide level decreased significantly and SOD activity was also decreased progressively. These results demonstrated that the ethanol extract of S. chinensis (Lour.) Baill lowered serum cholesterol levels, tissue lipid contents and accumulation of cholesterol in the rat.

In the course of screening for reactive oxygen species scavengers from natural products, an antioxidant was isolated from the mycelial culture broth of Phellinus linteus and identified as hispidin. The hispidin content was reached its maximum level at 12 days after onset of inoculation. About 2.5 mg/mL of hispidin was produced by P. linteus in a yeast-malt medium (pH 5.8, 25 degrees C). Hispidin inhibited 22.6 and 56.8% of the super oxide anion radical, 79.4 and 95.3% of the hydroxyl radical, and 28.1 and 85.5% of the DPPH radical at 0.1 and 1.0 mM, respectively. The positive control alpha-tocopherol scavenged 25.6 and 60.3%, 74.6 and 96.3%, and 32.7 and 77.5% of each radical, respectively, at the same concentrations. However, hispidin showed no significant activity on the hydrogen peroxide radical.
