Introduction: While increasing evidence points toward the benefits of musical activities in promoting cognitive and emotional well-being in older adults, more longitudinal studies are needed to establish their long-term effects and uncover the mechanisms through which musical activities affect well-being. Most previous research has focused on instrumental musical activities, but little is currently known about the long-term effects of singing, even though neuroimaging evidence suggests that it is a versatile activity for the brain, involving a multitude of neural processes that are potentially beneficial for well-being. Methods: We conducted a 2-year follow-up study to assess aging-related changes in cognitive functioning and emotional and social well-being with self-report questionnaires and standardized tests in 107 older adult choir singers and 62 demographically matched non-singers. Data were collected at baseline (T1), and at 1-year (T2) and 2-year (T3) follow-ups using questionnaires on subjective cognitive functioning, depression, social engagement, and quality of life (QOL) in all participants and neuropsychological tests in a subgroup of participants (45 choir singers and 41 non-singers). Results: The results of linear mixed model analysis showed that in verbal flexibility (phonemic fluency task), the choir singers had higher scores already at T1 and showed no change over time, whereas the non-singers showed enhancement from T1 to T3. Furthermore, active retrieval of word knowledge (WAIS-IV Vocabulary task) showed significantly different changes from T1 to T2 between the groups (enhancement in choir singers and decline in non-singers), however lacking significant change within groups. Similar opposite trajectories of QOL related to social inclusion and safety of the environment (WHOQOL-Bref Environmental subscale) were significant from T1 to T3, but these changes were not significant within groups or at each timepoint. Within the choir singers, shorter experience in choir singing was associated with greater improvement in the vocabulary task over the follow-up period, suggesting that initiation of choir singing at older age induces some verbal benefits. There were no group differences in any other questionnaire or neuropsychological measure over time. Discussion: In conclusion, our results suggest that choir singing at older age is associated with a sustained enhancement of phonemic fluency, while the effects on other verbal skills and quality of life are less clear.

Aging is accompanied by difficulties in auditory information processing, especially in more complex sound environments. Choir singing requires efficient processing of multiple sound features and could, therefore, mitigate the detrimental effects of aging on complex auditory encoding. We recorded auditory event-related potentials during passive listening of sounds in healthy older adult (≥ 60 years) choir singers and nonsinger controls. We conducted a complex oddball condition involving encoding of abstract regularities in combinations of pitch and location features, as well as in two simple oddball conditions, in which only either the pitch or spatial location of the sounds was varied. We analyzed change-related mismatch negativity (MMN) and obligatory P1 and N1 responses in each condition. In the complex condition, the choir singers showed a larger MMN than the controls, which also correlated with better performance in a verbal fluency test. In the simple pitch and location conditions, the choir singers had smaller N1 responses compared to the control subjects, whereas the MMN responses did not differ between groups. These results suggest that regular choir singing is associated both with more enhanced encoding of complex auditory regularities and more effective adaptation to simple sound features.

Parkinson's disease (PD) is a neurodegenerative disorder, well-known for its motor symptoms; however, it also adversely affects cognitive functions, including language, a highly important human ability. PD pathology is associated, even in the early stage of the disease, with alterations in the functional connectivity within cortico-subcortical circuitry of the basal ganglia as well as within cortical networks. Here, we investigated functional cortical connectivity related to spoken language processing in early-stage PD patients. We employed a patient-friendly passive attention-free paradigm to probe neurophysiological correlates of language processing in PD patients without confounds related to active attention and overt motor responses. MEG data were recorded from a group of newly diagnosed PD patients and age-matched healthy controls who were passively presented with spoken word stimuli (action and abstract verbs, as well as grammatically correct and incorrect inflectional forms) while focussing on watching a silent movie. For each of the examined linguistic aspects, a logistic regression classifier was used to classify participants as either PD patients or healthy controls based on functional connectivity within the temporo-fronto-parietal cortical language networks. Classification was successful for action verbs (accuracy = 0.781, p-value = 0.003) and, with lower accuracy, for abstract verbs (accuracy = 0.688, p-value = 0.041) and incorrectly inflected forms (accuracy = 0.648, p-value = 0.021), but not for correctly inflected forms (accuracy = 0.523, p-value = 0.384). Our findings point to quantifiable differences in functional connectivity within the cortical systems underpinning language processing in newly diagnosed PD patients compared to healthy controls, which arise early, in the absence of clinical evidence of deficits in cognitive or general language functions. The techniques presented here may aid future work on establishing neurolinguistic markers to objectively and noninvasively identify functional changes in the brain's language networks even before clinical symptoms emerge.

Learning a new language requires the acquisition of morphological units that enable the fluent use of words in different grammatical contexts. While accumulating research has elucidated the neural processing of native morphology, much less is known about how second-language (L2) learners acquire and process morphology in their L2. To address this question, we presented native speakers as well as beginning and advanced learners of Finnish with spoken (1) derived words, (2) inflected words, (3) novel derivations (novel combinations of existing stem + suffix), and (4) pseudo-suffixed words (existing stem + pseudo-suffix) in a passive listening EEG experiment. An early (60 msec after suffix deviation point) positive ERP response showed no difference between inflections and derivations, suggesting similar early parsing of these complex words. At 130 msec, derivations elicited a lexical ERP pattern of full-form memory-trace activation, present in the L2 beginners and advanced speakers to different degrees, implying a shift from lexical processing to more dual parsing and lexical activation of the complex forms with increasing proficiency. Pseudo-suffixed words produced a syntactic pattern in a later, 170-240 msec time-window, exhibiting enhanced ERPs compared to well-formed inflections, indicating second-pass syntactic parsing. Overall, the L2 learners demonstrated a gradual effect of proficiency towards L1-like responses. Advanced L2 learners seem to have developed memory traces for derivations and their neurolinguistic system is capable of early automatic parsing. This suggests that advanced learners have already developed sensitivity to morphological information, while such knowledge is weak in beginners. Discrepancies in ERP dynamics and topographies indicate partially differing recruitment of the language network in L1 and L2. In beginners, response differences between existing and novel morphology were scarce, implying that representations for complex forms are not yet well-established. The results suggest successful development of brain mechanisms for automatic processing of L2 morphology, capable of gradually attaining L1-like functionality with increasing proficiency.

Developmental dyslexia is characterised as an inability to read fluently. Apart from literacy problems, dyslexics have other language difficulties including inefficient speech encoding and deficient novel word learning. Yet, the neural mechanisms underlying these impairments are largely unknown. We tracked online formation of neural memory traces for a novel spoken word-form in dyslexic and normal-reading children by recording the brain's electrophysiological response dynamics in a passive perceptual exposure session. Crucially, no meaning was assigned to the new word-form nor was there any task related to the stimulus, enabling us to explore the memory-trace formation of a purely phonological form in the absence of any short-term or working memory demands. Similar to previously established neural index of rapid word learning in adults, the control children demonstrated an early brain response enhancement within minutes of exposure to the novel word-form that originated in frontal cortices. Dyslexic children, however, lacked this neural enhancement over the entire course of exposure. Furthermore, the magnitude of the rapid neural enhancement for the novel word-form was positively associated with reading and writing fluency. This suggests that the rapid neural learning mechanism for online acquisition of novel speech material is associated with reading skills. Furthermore, the deficient online learning of novel words in dyslexia, consistent with poor rapid adaptation to familiar stimuli, may underlie the difficulty of learning to read.

Research into neurobiological mechanisms of morphosyntactic processing of language has suggested specialised systems for decomposition and storage, which are used flexibly during the processing of complex polymorphemic words (such as those formed through affixation, e.g., boy + s = noun + plural marker or boy + ish = noun plus attenuator). However, neural underpinnings of acquisition of novel morphology are still unknown. We implicitly trained our participants with new derivational affixes through a word-picture association task and investigated the neural processes underlying formation of neural memory traces for new affixes. The participants' brain activity was recorded using magnetoencephalography (MEG), as they passively listened to the newly trained and untrained suffixes combined with real word and pseudoword stems. The MEG recording was repeated after a night's sleep using the same stimuli, to test the effects of overnight consolidation. The newly trained suffixes combined with real stems elicited stronger source activity in the left inferior frontal gyrus (LIFG) at ∼50 msec after the suffix onset than untrained suffixes, suggesting memory trace formation for the newly learned suffixes already on the same day. The following day, the suffix learning effect spread to the left superior temporal gyrus (STG) where it was again manifest as a response enhancement, particularly at ∼200-300 msec after the suffix onset, which might reflect an additional effect of overnight consolidation. Overall, the results demonstrate the rapid and dynamic processes of both immediate build-up and longer-term consolidation of neocortical memory traces for novel morphology, taking place after a short period of exposure to novel morphology and involving fronto-temporal perisylvian language circuitry.

Mastering multiple languages is an increasingly important ability in the modern world; furthermore, multilingualism may affect human learning abilities. Here, we test how the brain's capacity to rapidly form new representations for spoken words is affected by prior individual experience in non-native language acquisition. Formation of new word memory traces is reflected in a neurophysiological response increase during a short exposure to novel lexicon. Therefore, we recorded changes in electrophysiological responses to phonologically native and non-native novel word-forms during a perceptual learning session, in which novel stimuli were repetitively presented to healthy adults in either ignore or attend conditions. We found that larger number of previously acquired languages and earlier average age of acquisition (AoA) predicted greater response increase to novel non-native word-forms. This suggests that early and extensive language experience is associated with greater neural flexibility for acquiring novel words with unfamiliar phonology. Conversely, later AoA was associated with a stronger response increase for phonologically native novel word-forms, indicating better tuning of neural linguistic circuits to native phonology. The results suggest that individual language experience has a strong effect on the neural mechanisms of word learning, and that it interacts with the phonological familiarity of the novel lexicon.

A unique feature of human communication system is our ability to rapidly acquire new words and build large vocabularies. However, its neurobiological foundations remain largely unknown. In an electrophysiological study optimally designed to probe this rapid formation of new word memory circuits, we employed acoustically controlled novel word-forms incorporating native and non-native speech sounds, while manipulating the subjects' attention on the input. We found a robust index of neurolexical memory-trace formation: a rapid enhancement of the brain's activation elicited by novel words during a short (~30min) perceptual exposure, underpinned by fronto-temporal cortical networks, and, importantly, correlated with behavioural learning outcomes. Crucially, this neural memory trace build-up took place regardless of focused attention on the input or any pre-existing or learnt semantics. Furthermore, it was found only for stimuli with native-language phonology, but not for acoustically closely matching non-native words. These findings demonstrate a specialised cortical mechanism for rapid, automatic and phonology-dependent formation of neural word memory circuits.

The spatiotemporal dynamics of the neural processing of spoken morphologically complex words are still an open issue. In the current study, we investigated the time course and neural sources of spoken inflected and derived words using simultaneously recorded electroencephalography (EEG) and magnetoencephalography (MEG) responses. Ten participants (native speakers) listened to inflected, derived, and monomorphemic Finnish words and judged their acceptability. EEG and MEG responses were time-locked to both the stimulus onset and the critical point (suffix onset for complex words, uniqueness point for monomorphemic words). The ERP results showed that inflected words elicited a larger left-lateralized negativity than derived and monomorphemic words approximately 200 ms after the critical point. Source modeling of MEG responses showed one bilateral source in the superior temporal area ∼100 ms after the critical point, with derived words eliciting stronger source amplitudes than inflected and monomorphemic words in the right hemisphere. Source modeling also showed two sources in the temporal cortex approximately 200 ms after the critical point. There, inflected words showed a more systematic pattern in source locations and elicited temporally distinct source activity in comparison to the derived word condition. The current results provide electrophysiological evidence for at least partially distinct cortical processing of spoken inflected and derived words. In general, the results support models of morphological processing stating that during the recognition of inflected words, the constituent morphemes are accessed separately. With regard to derived words, stem and suffix morphemes might be at least initially activated along with the whole word representation.

How are words represented in the human brain and can these representations be qualitatively assessed with respect to their structure and properties? Recent research demonstrates that neurophysiological signatures of individual words can be measured when subjects do not focus their attention on speech input. These automatic activations, which take the form of negative deflections of event-related potentials, can appear surprisingly early (within ~200 ms) and are based on robust connections within neuronal memory circuits encoding individual words that ignite even when attentional resources are scarce. A new and critical prediction of this framework is that words with high frequency of occurrence have especially strong connections of their underlying memory circuits and should thus yield more negative ERPs compared with rarer words. We tested this prediction by presenting our subjects, in passive non-attend conditions, with acoustically matched high- and low-frequency words along with pseudo-words. Using factorial and correlation analyses, we found that already at ~120 ms after the spoken stimulus information was available, amplitude of brain responses was modulated by the words' lexical frequency. Topographic mapping and source analysis suggested that this early automatic frequency effect originates from the left inferior-frontal cortices. While, at this early latency, lexical differences between words and pseudo-words (more negative-going potential for meaningful words) could be seen only for the most frequent word stimuli, later-on (~270 ms), a more global lexicality effect with bilateral perisylvian sources was found for all stimuli, suggesting faster access to more frequent lexical entries. Our results support the account of word memory traces as interconnected neuronal circuits, and suggest that speed and magnitude of their activation are determined by their internal connection strength, which, in turn, is determined by the everyday language use.
