RESUMO

The effect of hot water treatment of beef trimmings on the processing characteristics, shelf-life and consumer acceptability of ground beef was evaluated. Hot water treatment (85°C for 40s) substantially enhanced the microbial quality of trimmings during refrigerated storage and this was independent of the fat level of the trimmings. Treatment had no effect on the oxidative stability of trimmings stored up to 7days, ground beef displayed in a retail cabinet for up to 3days, and had minimal effect on textural properties. Instrumental results demonstrate that ground beef from hot water treated trimmings was slightly lighter and tended to have less red color compared to non-treated beef. These color differences did not impact the consumer acceptance of raw patties, and in addition, hot water treatment did not significantly affect the consumer acceptability of cooked patty attributes.

RESUMO

Convective structures characterized by E×B motion are observed in a dipole-confined plasma. Particle transport rates are calculated from density dynamics obtained from multipoint measurements and the reconstructed electrostatic potential. The calculated transport rates determined from the large-scale dynamics and local probe measurements agree in magnitude, show intermittency, and indicate that the particle transport is dominated by large-scale convective structures.

Blood, liver, kidney, lymph nodes and muscle tissue were obtained from the carcasses of five cows with advanced Johne's disease. Samples from the raw tissues, from cooked muscle tissues and from cooked hamburger patties that contained chopped mesenteric lymph nodes were collected aseptically. Each sample was divided into two portions, one of which was decontaminated. Both portions were homogenized. Homogenates were spread on selective agar for the recovery of Mycobacterium avium subsp. paratuberculosis (Map) and inoculated into a Map growth medium with the organism being detected in the cultures by PCR procedures and Ziehl-Neelsen staining. Map were recovered at numbers > 10(3) cfu/g from 7 of 15 liver and mesenteric and ileocaecal lymph node samples; and at lesser numbers from 5 of 15 kidney and superficial inguinal and prescapular lymph node samples. The numbers recovered from decontaminated and not decontaminated portions of each sample were generally similar. Map was recovered from 1 and detected in 6 of 50 not decontaminated portions of samples of raw, chilled or frozen meat; and detected in 1 of 15 not decontaminated samples of meat cooked to 61 degrees C, and in 1 of 40 samples of meat cooked to >/=70 degrees C. Map was detected in 2 of 4 samples of mesenteric lymph nodes cooked to 61 degrees C, but not in samples cooked to > or =70 degrees C. The findings indicate that Map may be present in meat from infected animals at low numbers, but that any such organisms are likely to be inactivated when meat is cooked to a well done condition.

Despite advances in the therapy of chronic hepatitis C for some hepatitis C virus (HCV) genotypes interferon and ribavirin combination therapy is effective in less than 50% of patients. Abnobaviscum Quercus (AQ) is a mistletoe preparation containing defined amounts of mistletoe lectins (ML). It has shown immunomodulatory properties in vitro and in vivo. In small clinical trials AQ resulted, within an anthroposophical treatment concept, in a biochemical or virological response in up to 40% of patients with chronic hepatitis C. In order to evaluate the effect of this preparation we conducted an individually controlled cohort study. 25 patients with chronic hepatitis C (mean duration 147 +/- 80 months) and elevated alanine aminotransferase (ALT) levels were included in the study. As control they were observed for 6 months pre-treatment. This pre-treatment period was followed by 6 months of active treatment in which the mistletoe preparation was subcutaneously injected three times a week. Main outcome parameters were normalization of ALT and viral load. Hepatitis C associated signs and symptoms like tiredness, fullness in the right upper abdomen and musculoskeletal pain were assessed monthly in a standardized questionnaire. All 25 patients completed the study and most of the patients wanted to continue treatment. Mean duration of treatment was 9.1 months. None of the patients had complete or partial normalization of ALT or HCV RNA levels during pre-treatment or treatment period. Mean ALT did not change during the study. Tiredness, fullness in the right upper abdomen and musculoskeletal pain were present in 18, 8 and 4 patients respectively. They significantly improved within two months of treatment. A significant eosinophilia (p=0.0001) occurred between month 2 and 6 during treatment. 9 month treatment with a ML containing mistletoe preparation has no effect on viral load or ALT as markers of activity in patients with chronic hepatitis C. However, frequency and intensity of clinical signs and symptoms in our patients decreased significantly, similar to reports of improved quality of life in tumour patients treated with such preparations. A significant eosinophilia suggests that ML containing mistletoe preparations induce a T-helper 2 immune response.

Infection with the human immunodeficiency virus (HIV) and the subsequent derangement of host immunity place affected patients at risk for secondary infections. Some of the secondary pathogens occur with such frequency or are so rare in the non-immunosuppressed population that they have become part of the Centers for Disease Control and Prevention (CDC) classification for HIV/acquired immune deficiency syndrome (AIDS). Other infectious agents not yet included in the CDC definition are being reported in the HIV-infected population with increased frequency. General observations of the degree of immunosuppression associated with specific secondary infections have been useful in developing classification systems for HIV disease such as that of the CDC. However, the specific alterations in host immunity that promote infection with specific secondary pathogens are generally unknown. Geographic differences in the types and frequency of secondary infections also have been reported. Variation in strains of HIV, effect of malnutrition, lack of appropriate medical treatment, prevalence of virulent infectious diseases, and epidemiologic differences are possible contributing factors. Some infections that seemed likely to be closely associated with HIV infection have not occurred more frequently in HIV-infected patients. This review summarizes the histopathology of infectious conditions in the current CDC classification and highlights some conditions seen in HIV-infected individuals that are not currently HIV/AIDS-defining infections, yet may be seen by practicing pathologists.

We constructed replication-competent human adenovirus type 5 (HAd5) recombinants (HAd5-HN and HAd5-F) containing the bovine parainfluenza virus type 3 (BPIV3) hemagglutinin-neuraminidase (HN) or fusion (F) gene under the control of the simian virus 40 (SV40) regulatory sequences. These genes were inserted in the early region 3 (E3) of the HAd5 genome in the E3 parallel orientation. Expression of HN or F in HAd5-HN- or HAd5-F-infected cell extracts, respectively, was observed by immunoprecipitation using a BPIV3-specific polyclonal antiserum. Our results suggest that HN and F expressed by HAd5 recombinants were functionally similar to the native HN and F expressed in BPIV3-infected cells.

Cessation of cerebral blood flow results in severe damage to neurons and other brain structures. This is secondary to a combination of energy loss, excessive excitation promoting intracellular Ca2+ buildup, relative lack of inhibitory responses, generation of oxygen free radicals, especially during the reperfusion period and several other destructive cascades. Therapies aimed at decreasing the ill effects of glutamate are either not effective or have serious side-effects. Ca2+ entry blockers are generally not effective in cerebral ischemia, and data with protective effects of oxygen free radical scavengers in the post-ischemic period have shown conflicting results. There is recent interest with the use of agents that increase cerebral inhibitory responses after an ischemic insult. Such agents are effective when used before, during or up to 4 h after the ischemic insult. Many such medications have few side-effects and are in clinical use for other indications. This review will summarize inhibitory mechanisms that may be important in cerebral ischemia, and provide experimental evidence for their potential efficacy.

We report a developing technique capable of making continuous time-series measurements of naturally occurring minor-species concentrations. The high repetition rate of the mode-locked laser used in this technique allows for the study of transient combustion events, such as turbulence, and their effect on minor-species concentrations. The technique is applied to make CH fluorescence time-series measurements and to calculate power spectral densities in a turbulent nonpremixed flame. To our knowledge, the reported time series represents the first such measurement for a naturally occurring minor species in a turbulent flame.

Diagnostic anatomic pathologists play a crucial role in the battle against acquired immunodeficiency syndrome (AIDS). Not only are they intimately involved in the treatment of individual patients with human immunodeficiency virus (HIV) infection, but also they make important observations that result in the expansion of the scientific understanding of its pathogenesis. Pathologists studying tissue from patients with HIV infection should be familiar with the conditions to which these patients are susceptible. Although opportunistic infections are important causes of morbidity and mortality, noninfectious conditions frequently make substantial contributions to the disease course. Patients with HIV infection may be at increased risk for neoplastic disease. They do not, however, have an increased incidence of the most common tumors affecting the general population, such as breast, colon, and prostate carcinoma. Immunodeficiency results in increased susceptibility to malignant neoplasms, both by decreased immunologic response to abnormal cells and increased susceptibility to infection by viruses. All of the malignant neoplastic diseases that are Centers for Disease Control and Prevention (CDC) AIDS indicator conditions have been shown to have an association with a virus: Kaposi sarcoma (KS) with herpes hominis virus 8 (HHV-8), malignant lymphoma with Epstein-Barr virus (EBV), and cervical carcinoma with human papilloma virus (HPV). Patients with HIV infection also can develop reactive processes that are attributable to direct effects of HIV or immune system alterations. Such conditions include salivary gland cystic lymphoepithelial lesion, lymphadenopathy, lymphocytic interstitial pneumonitis, encephalopathy, enteropathy, nephropathy, hepatic conditions, dermatologic conditions and anemia.

The genes for the F and HN glycoprotein of a pathogenic field isolate of bovine parainfluenza virus type 3 (BPIV3) were isolated, converted to cDNA, and sequenced using dideoxynucleotides. The resulting nucleotide sequences were converted to protein sequence and were compared to previously sequenced glycoprotein genes with amino acid differences in the glycoproteins of isolates expressing different phenotypes. The HN glycoprotein, involved in the attachment and release of the virus, and the F glycoprotein, involved in penetration and spread of the virus, have been shown to affect pathogenicity of the virus and are the immunodominant proteins of the virus. Both the F and HN proteins have been shown to be required for syncytium formation. Our results suggest that BPIV3 viruses that exhibit greater syncytium-inducing activity in vitro have greater pathogenicity in vivo. By determining which epitopes are involved in syncytium formation and comparing the sequences and enzymatic activities of different strains of virus, it may be possible to design subunit vaccines that protect against disease.

This chapter provides an update on HIV and AIDS and discusses the pathologic evaluation of various respiratory conditions in the HIV-infected patient.

Picosecond time-resolved laser-induced fluorescence (PITLIF) can potentially be used to obtain measurements of minor species concentrations in rapidly fluctuating flames. Previous studies demonstrated this potential for atomic sodium by monitoring the temporal fluorescence signal with both an equivalent-time and a real-time sampling method. In this developmental study, PITLIF is used to determine hydroxyl concentrations in laminar CH(4)-O(2)-N(2) flames by the measurement of both the integrated fluorescence signal and the fluorescence lifetime. The quenching environment can be monitored with real-time sampling, and thus the necessary quenching rate coefficient is obtained in 348 us, which is fast enough for use in many turbulent flows. Fluorescence lifetimes of OH are also measured at different equivalence ratios in laminar flames by the use of the equivalent-time sampling technique. These results compare favorably with predicted lifetimes based on relevant quenching cross sections and calculated species concentrations.

We experimentally investigate the influence of O(2) interferences on laser-induced fluorescence measurements of NO in lean methane-fueled flames at a range of pressures for both narrow-band and broadband fluorescence detection. We identify NO excitation schemes that minimize O(2) interferences. From detection scans we obtain interference spectra for the different NO excitation schemes. We then identify optimum excitation-detection schemes for narrow-band detection measurements of NO. To simulate broadband detection experiments, we numerically apply five different filter combinations to the experimentally obtained detection scans. We develop filter-assessment parameters to judge the effectiveness of the different filtering schemes, and we establish a methodology for evaluating broadband excitation-detection strategies. From this research we identify optimum excitation-detection schemes for broadband detection measurements of NO.

Cotton rats were used to study the replication and pathogenesis of bovine parainfluenza virus type 3 (bPIV3) and to test the efficacy of the F and HN glycoproteins in modulating infection. In vitro cultures of cotton rat lung cells supported the growth of bPIV3 as shown by virus recovery, immunofluorescence, immunoprecipitation, and syncytium induction. Intranasal (i.n.) inoculation of cotton rats with 10(7) PFU resulted in peak recovery of virus after 2 days (8 x 10(4) PFU/g of lung tissue) and significant bronchiolitis with lymphocyte infiltration 5 to 7 days postinfection. Immunohistochemical staining of lungs and trachea demonstrated that virus antigen-positive cells increased in frequency over the course of infection to a maximum on day 5. Serum antibody responses were evaluated by enzyme-linked immunosorbent assays (ELISA), hemagglutination inhibition (HAI), and serum neutralization (SN). Following a single i.n. inoculation, serum antibody levels were 1/40,960, 1/32, and 1/80, as detected by ELISA, HAI, and SN, respectively. When an intramuscular inoculation of 10(7) PFU was administered 10 days prior to the i.n. inoculation, a secondary response which resulted in an ELISA titer of 1/163,000, an HAI titer of 1/640, and an SN titer of 1/512 was induced. IN inoculation of recombinant adenoviruses type 5 containing the bPIV3 F or HN protein or a combination of the two viruses protected cotton rats from bPIV3 challenge. Protection was evaluated serologically by ELISA, HAI, and SN titers, histopathology, immunohistochemistry, and virus recovery.

An unusual case of group B streptococcal meningitis in an adult is described. The evidence presented suggests that early vascular involvement during the meningitic process resulted in cerebral infarction, thereby explaining the patient's sudden deterioration and atypical presentation.

Picosecond time-resolved laser-induced fluorescence (PITLIF) has the potential to provide rapid measurements of minor-species concentrations by correction for local quenching conditions on the time scale of turbulence. Previous studies demonstrated that this technique could provide laser-induced fluorescence data and local quenching rates in flames but used equivalent-time sampling to obtain the required fluorescence decays. This precludes the use of PITLIF in turbulent systems. Fluorescence decays of sodium seeded into a laminar H(2)-O(2)-Ar diffusion flame are obtained from real-time data with an acquisition rate on the time scale of turbulence. The results obtained with this method are shown to be similar to those obtained from equivalent-time sampling.

RESUMO

An eighty-three-year-old nulliparous woman hospitalized for orthopedic surgery developed postoperative upper gastrointestinal bleeding and hypotension resulting in her death. Postmortem examination revealed a ruptured splenic artery aneurysm. This is the second reported case of gastrointestinal bleeding due to ruptured splenic artery aneurysm in a nulliparous woman and the first such case resulting in death. The incidence, pathophysiology, clinical presentation, and diagnosis of splenic artery aneurysm are discussed.
