RESUMO

The total and unsedimentable activity of acid DNase, RNase, phosphatase and arylsulfatases A and B was examined in the rat kidneys during long-term compression of soft tissues in the presence of high excitability of the sympathoadrenal system. Injection of adrenalin to rats with trauma reduced the total activity of DNase, acid phosphatase and arylsulfatases A and B, particularly at the late periods of soft tissue compression, whereas the total activity of acid RNase slightly increased as compared with control. Compression of soft tissues after adrenalin preinjection was accompanied by a substantial rise of unsedimentable activity of the lysosomal enzymes under study in the kidneys. The activity of the enzymes in cytosol progressively ascended as the time of soft tissue injury increased.

RESUMO

The functions of rat hepatic and renal lysosomes were explored during severe mechanical trauma caused by soft tissue compression in the presence of acetylcholinesterase induction by multiple acetylcholine administration. The increase of the trauma severity entailed the accumulation of non-hydrolyzed acetylcholine, thereby producing an injurious action on lysosomal membranes. Varying intensity of the changes in the activity of lysosomal hydrolases under study is likely to be related to the structural features of hepatic and renal lysosomes.

RESUMO

The effect of hyperreactivity of the sympathoadrenal system on rat liver lysosomal function during severe mechanical injury induced by compression of soft tissues was studied. Parenteral administration of adrenaline before the animals were exposed to the injury produced no qualitative differences in the response to the lysosomal injury of the organ under study. The response manifested in quantitative alterations--activation of the test lysosomal hydrolases and impairment of membrane stability followed by release of the enzymes to the hepatocyte cytoplasm and then to the systemic circulation.

The content of 5 lysosomal hydrolases was examined in the rat liver and blood serum after compression of hind limb soft tissues in the presence of a long-term intake of excess doses of pyridoxine, riboflavin and glutamic acid. It was shown that the 14-day application of the drug complexes dramatically increased the overall content of cathepsin C, arylsulfatases A and B, beta-glucuronidase and p-acetyl-beta-D-galactosaminidase and reduced the overall content of cathepsin D in the rat liver. The non-sedimented content of the enzymes did not practically differ from the control magnitudes. In the blood serum, the content of cathepsin C and B1 approximated that seen in the control, while that of arylsulfatases A and B and p-acetyl-beta-D-galactosaminidase decreased, whereas the beta-glucuronidase content was 75% higher as compared to the basic characteristics. In the presence of administering the drug complexes, severe mechanical injury entailed the lowering of the content of the majority of rat liver lysosomal hydrolases. Besides, one could observe an essential fall of the non-sedimented content of cathepsin C and arylsulfatases A and B. The blood serum demonstrated an appreciable decrease in the content of cathepsins C and B1, p-acetyl-beta-D-galactosaminidase and arylsulfatases A and B. Thus, the fall of the non-sedimented content and diminished release of lysosomal hydrolases into the systemic circulation attest to the preservation of the structural and functional integrity of the liver cell lysosomal system during severe mechanical injury in the presence of combined excess intake of pyridoxine, riboflavin and glutamic acid.

RESUMO

A study was made of the pattern of changes in overall and non-sedimented activity of lysosomal hydrolases of the liver of rats injected with serotonin under grave mechanical trauma. Lability of the lysosomal membranes was found to be twice as increased, with remarkable hyperfermentemia being seen starting from the 3rd hour of compression in the presence of an appreciable rise of overall enzymatic activity. The role of serotonin in the mechanism of exocytosis of lysosomal enzymes is discussed.
