Diabetes mellitus (DM) belongs to the category of socially significant diseases with epidemic rates of increases in prevalence. Diabetic nephropathy (DN) is a specific kind of kidney damage that occurs in 40% of patients with DM and is considered a serious complication of DM. Most modern methods for treatments aimed at slowing down the progression of DN have side effects and do not produce unambiguous positive results in the long term. This fact has encouraged researchers to search for additional or alternative treatment methods. Hyperglycemia has a negative effect on renal structures due to a number of factors, including the activation of the polyol and hexosamine glucose metabolism pathways, the activation of the renin-angiotensin-aldosterone and sympathetic nervous systems, the accumulation of advanced glycation end products and increases in the insulin resistance and endothelial dysfunction of tissues. The above mechanisms cause the development of oxidative stress (OS) reactions and mitochondrial dysfunction, which in turn contribute to the development and progression of DN. Modern antioxidant therapies for DN involve various phytochemicals (food antioxidants, resveratrol, curcumin, alpha-lipoic acid preparations, etc.), which are widely used not only for the treatment of diabetes but also other systemic diseases. It has also been suggested that therapeutic approaches that target the source of reactive oxygen species in DN may have certain advantages in terms of nephroprotection from OS. This review describes the significance of studies on OS biomarkers in the pathogenesis of DN and analyzes various approaches to reducing the intensity of OS in the prevention and treatment of DN.

Soil pollution with oil as a result of accidents at oil pipelines and oil refineries is a frequent occurrence in the south of Russia. To restore such polluted lands, it is necessary to carry out soil remediation measures. This work aimed to evaluate the use of ameliorants of various natures (biochar, sodium humate, and microbial preparation Baikal EM-1) to restore the ecological state of oil-contaminated soils with different properties (Haplic Chernozem, Haplic Arenosols, Haplic Cambisols). To assess the ecological state of soils, the following physicochemical and biological indicators were studied: residual oil content, redox potential, and medium reaction (pH). Changes in enzymatic activity were also studied, including catalase, dehydrogenases, invertase, urease, and phosphatase. The greatest decomposition of oil in Haplic Chernozem and Haplic Cambisols was provided by Baikal EM-1 (56 and 26%), and in Haplic Arenosols, this was provided by biochar (94%) and sodium humate (93%). In oil-contaminated Haplic Cambisols, the content of easily soluble salts with the addition of biochar and Baikal EM-1 increased by 83 and 58%, respectively. The introduction of biochar caused an increase in pH from 5.3 (Haplic Cambisols) to 8.2 (Haplic Arenosols). The introduction of oil-contaminated Haplic Arenosols of biochar, humate, and Baikal stimulated the activity of catalase and dehydrogenases by 52-245%. The activity of invertase was stimulated in the Haplic Chernozem after the introduction of ameliorants by 15-50%. The activity of urease was stimulated after the introduction of ameliorants into borax and Arenosol by 15-250%. The most effective ameliorant for restoring the ecological state of Haplic Cambisols after oil pollution was biochar. For Haplic Arenosols, this was sodium humate, and for Haplic Chernozem, the effectiveness of biochar and sodium humate did not differ. The most informative indicator for the remediation of Haplic Chernozem and Haplic Cambisols was the activity of dehydrogenases, and for Haplic Arenosols, this was the activity of phosphatase. The results of the study should be used to biomonitor the ecological state of oil-contaminated soils after bioremediation.

Oxidative stress plays a leading role in the pathogenesis of diabetic nephropathy. However, many aspects of oxidative stress reactions in the initial stages of this disease are not fully understood. The men cohort is of particular interest because of the severe effects of diabetes on their urogenital system. The aim of this study is to assess the intensity of lipids, proteins, DNA oxidative damage, blood antioxidant defense enzymatic, and activity of non-enzymatic components in men with type 1 diabetes mellitus (T1DM) in the early stages of diabetic nephropathy using receiver operator characteristic (ROC) analysis. This study included eighty-nine reproductive-age men in the initial stages of diabetic nephropathy (DN) and thirty-nine age- and sex-matched individuals not suffering from glycemic disorders. The DN patients were divided into two subgroups: stage 1 patients (urinary albumin < 30 mg/day and albumin/creatinine ratio < 3 mg/mmol (n = 45)) and stage 2 patients (urinary albumin 30−300 mg/day and albumin/creatinine ratio 3−30 mg/mmol (n = 44)). Levels of oxidative damage products (conjugated dienes (CDs), thiobarbituric acid reactants (TBARs), methylglyoxal (MGO), and 8-hydroxy-2'-deoxyguanosine (8-OHdG)) and antioxidants (glutathione peroxidase (GPx), glutathione S-transferases π (GSTp), glutathione reductase (GR), copper and zinc-containing superoxide dismutase 1 (SOD-1), total antioxidant status (TAS), α-tocopherol, retinol, reduced glutathione (GSH), and oxidative glutathione (GSSG)) were estimated in plasma and erythrocytes. Oxidative damage to cellular structures (higher values of median CDs (1.68 µmol/L; p = 0.003), MGO (3.38 mg/L; p < 0.001) in the stage 1 group and CDs (2.28 µmol/L; p < 0.0001), MGO (3.52 mg/L; p < 0.001), 8-OHdG (19.44 ng/mL; p = 0.010) in the stage 2 group) and changes in the antioxidant defense system (lower values of TAS (1.14 units; p = 0.011), α-tocopherol (12.17 µmol/L; p = 0.009), GPx (1099 units; p = 0.0003) and elevated levels of retinol (1.35 µmol/L; p < 0.001) in the group with stage 1; lower values of α-tocopherol (12.65 µmol/L; p = 0.033), GPx (1029.7 units; p = 0.0001) and increased levels of GR (292.75 units; p < 0.001), GSH (2.54 mmol/L; p = 0.010), GSSG (2.31 mmol/L; p < 0.0001), and retinol (0.81 µmol/L; p = 0.005) in the stage 2 group) were identified. The ROC analysis established that the following indicators have the highest diagnostic significance for stage 1 diabetic nephropathy: CDs (AUC 0.755; p < 0.0001), TBARs (AUC 0.748; p = 0.0001), MGO (AUC 0.720; p = 0.0033), retinol (AUC 0.932; p < 0.0001), GPx (AUC 0.741; p = 0.0004), α-tocopherol (AUC 0.683; p = 0.0071), and TAS (AUC 0.686; p = 0.0052) and the following for stage 2 diabetic nephropathy: CDs (AUC 0.714; p = 0.001), TBARs (AUC 0.708; p = 0.001), 8-OHdG (AUC 0.658; p = 0.0232), GSSG (AUC 0.714; p = 0.001), and GSH (AUC 0.667; p = 0.0108). We conclude that changes in indicators of damage to lipids, proteins, DNA, and the insufficiency of antioxidant defense factors already manifest in the first stage of diabetic nephropathy in men with T1DM. The ROC established which parameters have the greatest diagnostic significance for stages 1 and 2 of diabetic nephropathy, which may be utilized as additional criteria for defining men with T1DM as being in the risk group for the development of initial manifestations of the disease and thus allow for substantiating appropriate approaches to optimize preventive measures.

The content of various chemical elements such as metals, metalloids, and nonmetals in the environment is associated with natural and anthropogenic sources. It is necessary to normalize the content of metals, metalloids, and nonmetals as potentially toxic elements (PTE) in the Haplic Chernozem. The soils of the Southern Russia are of high quality and fertility. However, this type of soil, like Haplic Chernozem, is subject to contamination with a wide range of PTE. The aim of the work was to rank metals, metalloids, and nonmetals by ecotoxicity in Haplic Chernozem. To assess the ecotoxicity of chernozem, data for 15 years (2005-2020) were used. Biological indicators used to assess the ecotoxicity of Haplic Chernozem: catalase activity, cellulolytic activity, number of bacteria, Azotobacter spp. abundance, to change of length of radish's roots. Based on these biological indicators, an integral indicator of the state of Haplic Chernozem was calculated. The ecotoxicity of 23 metals (Cd, Hg, Pb, Cr, Cu, Zn, Ni, Co, Mo, Mn, Ba, Sr, Sn, V, W, Ag, Bi, Ga, Nb, Sc, Tl, Y, Yb), 5 metalloids (B, As, Ge, Sb, Te) and 2 nonmetals (F, Se) as priority pollutants. It is proposed to distinguish three hazard classes of metals, metalloids, and nonmetals to Haplic Chernozem: I class - Te, Ag, Se, Cr, Bi, Ge, Sn, Tl, Hg, Yb, W, Cd; II class - As, Co, Sc, Sb, Cu, Ni, B, Nb, Pb, Ga; III class - Sr, Y, Mo, Zn, V, Ba, Mn, F. It is advisable to use the results of the study for predictive assessment of the impact of metals, metalloids, and nonmetals on the ecological state of the soil during pollution.

An increase in the penetration of metal-based nanoparticles (NPs) into the environment requires an assessment of their ecotoxicity as they impair the critical activity of plants, animals, bacteria, and enzymes. Therefore, the study aimed to observe the effects of metal-based NPs, including copper (Cu), nickel (Ni), and zinc (Zn), on the Cambisols, which cover a significant part of the earth's soil and play an important role in the biosphere. Metal-based NPs were introduced into the soil at concentrations of 100, 1000, and 10,000 mg/kg. The biological properties of the soil are being investigated as the most sensitive to external contamination. The highest ecotoxicity of the studied pollutants introduced into the soil at the same concentrations was shown by Cu (up to 34%) and Zn (up to 30%) NPs, while Ni NPs showed less (up to 22%). Microbiological (total number of bacteria, Azotobacter sp. abundance) and phytotoxic properties (radish seed germination and length of roots) of Cambisols were more sensitive (22-53%) to pollution by NPs of Cu, Zn, and Ni, while enzymatic activity (catalase and dehydrogenases) showed less sensitivity (14-32%). The present results could be useful for biomonitoring the state of contaminated soils, especially by NPs.

In recent years, silver nanoparticles (AgNPs) are increasingly used in various industries due to their antibacterial properties, which lead to an increase in pollution of the environment and soil ecosystems. However, the ecological effects of soil pollution by AgNPs were poorly studied than that with AgNPs of other metal-based NPs. The aim of this study is to assess the influence of AgNPs on the biological properties of Haplic Chernozem. Silver was introduced into the soil in the form of AgNPs with a concentration of 0.5; 1; 5; 10; 50, and 100 mg/kg in laboratory conditions. The influence of AgNPs on the biological properties of Haplic Chernozem was assessed 30 days after contamination. The degree of reduction in biological properties depends on the AgNPs concentration in the soil. This study showed that the sensitivity to contamination by AgNPs in the total number of bacteria and enzymatic activity was more than that in the abundance of bacteria of the genus Azotobacter. The integrated index of biological state (IIBS) of Haplic Chernozem was decreased by contamination with AgNPs. Silver nanoparticles in the concentration of 10 mg/kg caused a decrease in the indicator by 13% relative to the control. It also decreased IIBS by doses of 50 and 100 mg/kg by 22 and 27% relative to the control. All used biological indicators could be used for biomonitoring, biodiagnosis, bioindication, and regulation of ecological condition of soil contamination by AgNPs.

The presence of heavy metals in the soil could impose serious problems on soil-plant systems due to the accumulation of heavy metals in plants. Even vital elements such as Cu and Zn have a toxic effect in the case of excessive intake by living organisms. The present work aimed to investigate the content of loosely bound (exchangeable, complexed, and specifically sorbed) compounds of Cu and Zn and their availability to spring barley (Hordeum sativum distichum) in contaminated Haplic Chernozem soil under the conditions of a model experiment (five approximate permissible concentrations (APC) and 10 APC of metal). Changes in the bioavailability of the metals upon application of carbon sorbents were observed. An increase in loosely bound metal compounds has been shown under conditions of soil contamination with metals (up to 57% of the total content). The increase in the availability of Cu in the soil was mainly due to the formation of complexed metal forms with organic matter (up to 17%). The availability of Zn was found to be associated with an increase in exchangeable (up to 21%) and specifically sorbed compounds (up to 27%). Granular activated carbon (GAC) and biochar have high sorption properties. A decrease in the content of loosely bound compounds of metals was established, especially in the most mobile forms such as exchangeable and complexed forms. The introduction of sorbents into the soil opened up a new venue for binding heavy metals in situ, eventually leading to a decrease in their bioavailability. The inactivation of Cu and Zn in the soil upon the application of sorbents led to a decrease in metal absorption by spring barley. The highest efficiency of biochar application was established at a dose of 2.5% and 5% in soil contaminations of 5 APC and 10 APC of Cu or Zn. The efficiency of the use of sorbents was more influenced by the dose of application than by the type of sorbent. There was no significant difference between biochar and GAC. Stabilization and inactivation of metals may improve soil fertility and plant growth.

Triple-negative breast cancers are often characterized by aggressive behavior and short clinical course once they become chemotherapy-resistant. We describe below a patient who has shown a response to combination of chemotherapy with Elenagen, a plasmid encoding p62. Elenagen was tested in a previous phase I/II study in patients with refractory solid tumors and shown to be safe. Also, plasmid ability to halt tumor progression and restore sensitivity to chemotherapy was found. Preclinical data supports effects on tumor grade and change the tumor's microenvironment in spontaneous canine breast cancers. We describe here a 48-year old female with triple-negative and BRCA1/2-negative breast cancer who had a primary resistance to chemotherapy and negative dynamics despite the use of multiple lines of treatments. Elenagen was applied intramuscularly at a dose of 1 mg weekly in combination with standard chemotherapy scheme CMF (cyclophosphamide, methotrexate, fluorouracil). In this patient we observed partial tumor regression (by 33%) and 19 weeks of progression-free survival. This first observed objective response to a combination of Elenagen with chemotherapy demonstrates that even in heavily pretreated chemo-resistant triple-negative tumor, the addition of Elenagen to a chemotherapy regimen can cause an objective response and increase in progression-free survival. Such a regimen is worthy of further study in a larger number of patients.

Lipid profile comparative analysis was performed to reveal the interdependence of lipids with Circadian locomoter output cycles protein kaput (CLOCK) 3111T/C gene polymorphism in menopausal women with/without a body mass index (BMI) of ≥25 kg/m2. Methods: A total of 193 female volunteers aged 45 to 60 years were divided into two groups: Those with BMI < 25 kg/m2 (control) and those with BMI ≥ 25 kg/m2. Each group was then divided into two subgroups: Those with the CLOCK TT-genotype and those with the CLOCK TC-, CC-genotypes. Lipid metabolism parameters were determined by the enzymatic method. Single-nucleotide polymorphisms (SNPs) were detected via polymerase chain reaction-restriction fragment length polymorphism technology. Results: There were no differences in CLOCK 3111T/C genotypes or allele frequency between the control and main groups. In addition, there were no differences in lipid profile parameters between women of the control group and different CLOCK 3111T/C genotypes. The total cholesterol (p = 0.041) and low-density lipoprotein cholesterol (p = 0.036) levels were higher in the subgroup of women with a BMI ≥ 25 kg/m2 and CLOCK TT-genotype as compared to the subgroup with a BMI ≥ 25 kg/m2 and minor allele 3111C. Conclusions: SNP 3111T/C of the CLOCK gene is not associated with BMI however, data suggest that the minor allele of the CLOCK 3111T/C gene polymorphism may have a protective role in atherogenic lipid levels in women with a BMI greater than or equal to 25 kg/m2.

BACKGROUND: Few studies have examined the effects of low-dose alcohol consumption on the "lipid peroxidation-antioxidant defense" (LPO-AOD) system of mothers and infants, and on infant growth and development. This study examined effects of alcohol consumption on the LPO-AOD system of pregnant women and newborns and infant development. METHODS: A total of 209 pregnant women were recruited for this prospective study at the first prenatal visit and followed until delivery: 112 consumed alcohol and 97 reported no alcohol use during pregnancy. Infants were evaluated at birth, and at 6 and 12 months of age. The study controlled for the confounding effect of maternal smoking. RESULTS: Biomarkers of lipid peroxidation, for example, thiobarbituric acid reactants, were higher and the activity of the antioxidant defense system was lower in drinkers and their infants. Higher rates of pathological conditions and slower postnatal growth were observed among infants who were prenatally exposed to alcohol. Low-dose alcohol use and tobacco smoking were associated with lower postnatal infant growth trajectories, resulting in restricted growth at 6 and 12 months among infants born to mothers who drank or smoked during pregnancy. Alcohol had a broad effect on the infant and maternal LPO-AOD system, while the effect of smoking was limited in this study to maternal glutathione peroxidase. CONCLUSIONS: Small amounts of alcohol consumed during pregnancy are associated with dysfunction of the LPO-AOD system and development of oxidative stress in women and their children. Identification and preventive interventions are needed for pregnant women who use alcohol in any amount.

Previously, we reported that the administration of a p62/SQSTM1-encoding plasmid demonstrates high safety and signs of clinical benefits for human cancer patients. The treatment also suppressed tumor growth and metastasis in dogs and mouse models. Here we investigated some mechanistic aspects of these effects. In mammary tumors bearing-dogs, i.m. injections of p62 plasmid reduced tumor sizes and their aggressive potential in 5 out of 6 animals, with one carcinoma switching to adenoma. The treatment increased levels of smooth muscle actin in stroma cells and type III collagen in the extracellular matrix, which correlate with a good clinical prognosis. The p62 treatment also increased the abundance of intratumoral T-cells. Because of the role of adaptive immunity cannot be tested in dogs, we compared the protective effects of the p62 plasmid against B16 melanoma in wild type C57BL/6J mice versus their SCID counterpart lacking lymphocytes. The plasmid was only protective in the wild type strain. Also, p62 plasmid amplified the anti-tumor effect of T-cell transfer from tumor-bearing animals to animals challenged with the same tumors. We conclude that the plasmid acts via re-modeling of the tumor microenvironment, making it more favorable for increased anti-cancer immunity. Thus, the p62-encoding plasmid might be a new adjuvant for cancer treatments.

A comparative analysis of lipid peroxidation processes and antioxidant defense system in Caucasian menopausal women with/without insomnia depending on the genotype of Clock 3111T/C gene polymorphism was performed. Two hundred and fourteen Caucasian menopausal women divided into control (without insomnia) and main group (with insomnia) were examined. Lipid peroxidation (conjugated dienes, thiobarbituric acid reactants) and antioxidant defense system parameters (?-tocopherol, retinol, reduced and oxidized glutathione, glutathione S-transferase, glutathione peroxidase, glutathione reductase, superoxide dismutase) were determined by spectrofluorophotometer and immunoenzymometric methods. Patients with insomnia carriers of the TT-genotype had a significantly higher thiobarbituric acid reactants level and glutathione peroxidase activity as compared to group with insomnia carriers of the minor 3111C-allele (p < .05). A comparative analysis of the parameters in the women of the main and control groups showed higher conjugated dienes, thiobarbituric acid reactants levels and lower retinol, reduced glutathione levels, glutathione reductase activity in women with insomnia carriers of the TT-genotype (p < .05). The carriers of the minor allele with insomnia had a higher conjugated dienes levels and lower glutathione peroxidase activity as compared to control (p < .05). Thus, lipid peroxidation and antioxidant system parameters in Caucasian menopausal women with insomnia depend on the Clock 3111T/C gene polymorphism.

P62/SQSTM1, a multi-domain protein that regulates inflammation, apoptosis, and autophagy, has been linked to age-related pathologies. For example, previously we demonstrated that administration of p62/SQSTM1-encoding plasmid reduced chronic inflammation and alleviated osteoporosis and metabolic syndrome in animal models. Herein, we built upon these findings to investigate effect of the p62-encoding plasmid on an age-related macular degeneration (AMD), a progressive neurodegenerative ocular disease, using spontaneous retinopathy in senescence-accelerated OXYS rats as a model. Overall, the p62DNA decreased the incidence and severity of retinopathy. In retinal pigment epithelium (RPE), p62DNA administration slowed down development of the destructive alterations of RPE cells, including loss of regular hexagonal shape, hypertrophy, and multinucleation. In neuroretina, p62DNA prevented gliosis, retinal thinning, and significantly inhibited microglia/macrophages migration to the outer retina, prohibiting their subretinal accumulation. Taken together, our results suggest that the p62DNA has a strong retinoprotective effect in AMD.

Ilizarov is an accepted technique for lengthening short amputation stumps to improve prosthetic function and rehabilitation. The relation of stump lengthening and phantom limb pain (PLP) has not been reported in literature. We present here a case report of a transfemoral amputee who had a flare up of PLP following stump lengthening. He responded well to a combination of pharmacological therapy and soft tissue manipulative techniques and desired length was achieved. This report alerts the possibility of aggravation of PLP following stump lengthening and discusses its management.

Thermodynamic calculations of the optimal compositions of oxide catalysts with different natures are performed based on the theory of catalysis by polyhedra. The obtained compositions of the active catalysts agree with experimental data. The electrostatic potential generated by polyhedra of metal-oxide catalysts in a variety of directions is calculated. The dependence of the sign and magnitude of the potential on the distance from the central metal ion towards the vertex of the polyhedron, the middle of its edge or the centre of the face is estimated. It is assumed that the magnitude of the potential can serve as a reference point for determining active centres, which produce adsorption complexes and intermediate compounds.

A high-calorie diet (HCD) induces two mutually exacerbating effects contributing to diet-induced obesity (DIO): impaired glucose metabolism and increased food consumption. A link between the metabolic and behavioral manifestations is not well understood yet. We hypothesized that chronic inflammation induced by HCD plays a key role in linking together the two components of diet-induced pathology. Based on this hypothesis, we tested if a plasmid (DNA vaccine) encoding p62 (SQSTM1) would alleviate DIO including its metabolic and/or food consumption abnormalities. Previously we reported that injections of the p62 plasmid reduce chronic inflammation during ovariectomy-induced osteoporosis. Here we found that the p62 plasmid reduced levels of pro-inflammatory cytokines IL-1ß, IL-12, and INFγ and increased levels of anti-inflammatory cytokines IL-4, IL-10 and TGFß in HCD-fed animals. Due to this anti-inflammatory response, we further tested whether the plasmid can alleviate HCD-induced obesity and associated metabolic and feeding impairments. Indeed, p62 plasmid significantly reversed effects of HCD on the body mass index (BMI), levels of glucose, insulin and glycosylated hemoglobin (HbA1c). Furthermore, p62 plasmid partially restored levels of the satiety hormone, serotonin, and tryptophan, simultaneously reducing activity of monoamine oxidase (MAO) in the brain affected by the HCD. Finally, the plasmid partially reversed increased food consumption caused by HCD. Therefore, the administering of p62 plasmid alleviates both metabolic and behavioral components of HCD-induced obesity.

Background. There are limited published data in regard to the relationship between obstructive sleep apnea (OSA) and hypertension and neurobehavioral and mental status in adolescence. The aim of our study was to evaluate neurobehavioral patterns and cognitive functions in adolescents with hypertension according to absence or presence of OSA. Methods. This was a retrospective cohort study completed at the Scientific Center for Family Health and Human Reproduction Problems. Participants included adolescents aged 14-17 years and referred for 24-hour ambulance blood pressure monitoring (ABPM) and polysomnographic (PSG) studies between 2007 and 2009, inclusive. Results. 18 hypertensive OSA (the 1st group) and 20 hypertensive non-OSA adolescents (the 2nd group) were included in the study. Significant changes of neurobehavioral functioning in OSA patients were shown. Cognitive abilities also were impaired. Verbal and visual memory indexes and attention index were 2.1 and 2.2 times lower, accordingly, in the 1st group than in the 2nd group (P < 0.05). Speech index was significantly 2.8 times lower in OSA patients than in non-OSA patients (P < 0.05). In hypertensive OSA adolescents more significant Spearman correlations between classic sleep parameters and cognitive measures were found compared to patients without OSA. Conclusions. These results suggest that OSA is closely associated with neurobehavioral and cognitive functioning in hypertensive adolescents.
